| 1 | Anti-inflammatory properties of the short-chain fatty acids acetate and propionate:A study with relevance to inflammatory bowel disease显示文摘AIM: To compare the anti-inflammatory properties of butyrate with two other SCFAs,namely acetate and propionate,which have less well-documented effects on inflammation. METHODS: The effect of SCFAs on cytokine release from human neutrophils was studied with ELISA. SCFA-dependent modulation of NF-κB reporter activity was assessed in the human colon adenocarcinoma cell line,Colo320DM. Finally,the effect of SCFAs on gene expression and cytokine release,measured with RT-PCR and ELISA,respectively,was studied in mouse colon organ cultures established from colitic mice. RESULTS: Acetate,propionate and butyrate at 30 mmol/L decreased LPS-stimulated TNFα release from neutrophils,without affecting IL-8 protein release. All SCFAs dose dependently inhibited NF-κB reporter activity in Colo320DM cells. Propionate dose-dependently suppressed IL-6 mRNA and protein release from colon organ cultures and comparative studies revealed that propionate and butyrate at 30 mmol/L caused a strong inhibition of immune-related gene expression,whereas acetate was less effective. A similar inhibition was achieved with the proteasome inhibitor MG-132,but not the p38 MAPK inhibitor SB203580. All SCFAs decreased IL-6 protein release from organ cultures. CONCLUSION: In the present study propionate and butyrate were equipotent,whereas acetate was less effective,at suppressing NF-κB reporter activity,immune-related gene expression and cytokine release in vitro. | 3 | Intracellular distribution of amyloid beta peptide and its relationship to the lysosomal system显示文摘Background:Amyloid beta peptide(Aβ)is the main component of extraneuronal senile plaques typical of Alzheimer’s disease(AD)brains.Although Aβis produced by normal neurons,it is shown to accumulate in large amounts within neuronal lysosomes in AD.We have recently shown that under normal conditions the majority of Aβis localized extralysosomally,while oxidative stress significantly increases intralysosomal Aβcontent through activation of macroautophagy.It is also suggested that impaired Aβsecretion and resulting intraneuronal increase of Aβcan contribute to AD pathology.However,it is not clear how Aβis distributed inside normal neurons,and how this distribution is effected when Aβsecretion is inhibited.Methods:Using retinoic acid differentiated neuroblastoma cells and neonatal rat cortical neurons,we studied intracellular distribution of Aβby double immunofluorescence microscopy for Aβ40 or Aβ42 and different organelle markers.In addition,we analysed the effect of tetanus toxin-induced exocytosis inhibition on the intracellular distribution of Aβ.Results:Under normal conditions,Aβwas found in the small cytoplasmic granules in both neurites and perikarya.Only minor portion of Aβwas colocalized with trans-Golgi network,Golgi-derived vesicles,early and late endosomes,lysosomes,and synaptic vesicles,while the majority of Aβgranules were not colocalized with any of these structures.Furthermore,treatment of cells with tetanus toxin significantly increased the amount of intracellular Aβin both perikarya and neurites.Finally,we found that tetanus toxin increased the levels of intralysosomal Aβalthough the majority of Aβstill remained extralysosomally.Conclusion:Our results indicate that most Aβis not localized to Golgi-related structures,endosomes,lysosomes secretory vesicles or other organelles,while the suppression of Aβsecretion increases intracellular intra-and extralysosomal Aβ. | Lin Zheng Angel Cedazo-Minguez Martin Hallbeck Fredrik Jerhammar Jan Marcusson Alexei Terman | 2012 | Translational Neurodegeneration2012,1,1: | 10 |
| 4 | Cholesterol metabolism in cholestatic liver disease and liver transplantation:From molecular mechanisms to clinical implications显示文摘The aim of this review is to enlighten the critical roles that the liver plays in cholesterol metabolism. Liver transplantation can serve as gene therapy or a source of gene transmission in certain conditions that affect cholesterol metabolism, such as low-density-lipoprotein(LDL) receptor gene mutations that are associated with familial hypercholesterolemia. On the other hand, cholestatic liver disease often alters cholesterol metabolism. Cholestasis can lead to formation of lipoprotein X(Lp-X), which is frequently mistaken for LDL on routine clinical tests. In contrast to LDL, Lp-X is non-atherogenic, and failure to differentiate between the two can interfere with cardiovascular risk assessment, potentially leading to prescription of futile lipid-lowering therapy. Statins do not effectively lower Lp-X levels, and cholestasis may lead to accumulation of toxic levels of statins. Moreover, severe cholestasis results in poor micellar formation, which reduces cholesterol absorption, potentially impairing the cholesterol-lowering effect of ezetimibe. Apolipoprotein B-100 measurement can help distinguish between atherogenic and non-atherogenic hypercholesterolemia. Furthermore, routine serum cholesterol measurements alone cannot reflect cholesterol absorption and synthesis. Measurements of serum non-cholesterol sterol biomarkers- such as cholesterol precursor sterols, plant sterols, and cholestanol- may help with the comprehensive assessment of cholesterol metabolism. An adequate cholesterol supply is essential for liver-regenerative capacity. Low preoperative and perioperative serum cholesterol levels seem to predict mortality in liver cirrhosis and after liver transplantation. | 7 | Manufacture and characterization of graphene membranes with suspended silicon proof masses for MEMS and NEMS applications显示文摘Graphene’s unparalleled strength,chemical stability,ultimate surface-to-volume ratio and excellent electronic properties make it an ideal candidate as a material for membranes in micro-and nanoelectromechanical systems(MEMS and NEMS).However,the integration of graphene into MEMS or NEMS devices and suspended structures such as proof masses on graphene membranes raises several technological challenges,including collapse and rupture of the graphene.We have developed a robust route for realizing membranes made of double-layer CVD graphene and suspending large silicon proof masses on membranes with high yields.We have demonstrated the manufacture of square graphene membranes with side lengths from 7µm to 110µm,and suspended proof masses consisting of solid silicon cubes that are from 5µm×5µm×16.4µm to 100µm×100µm×16.4µm in size.Our approach is compatible with wafer-scale MEMS and semiconductor manufacturing technologies,and the manufacturing yields of the graphene membranes with suspended proof masses were>90%,with>70%of the graphene membranes having>90%graphene area without visible defects.The measured resonance frequencies of the realized structures ranged from tens to hundreds of kHz,with quality factors ranging from 63 to 148.The graphene membranes with suspended proof masses were extremely robust,and were able to withstand indentation forces from an atomic force microscope(AFM)tip of up to~7000nN.The proposed approach for the reliable and large-scale manufacture of graphene membranes with suspended proof masses will enable the development and study of innovative NEMS devices with new functionalities and improved performances. | Xuge Fan Anderson D.Smith Fredrik Forsberg Stefan Wagner Stephan Schroder Sayedeh Shirin Afyouni Akbari Andreas C.Fischer Luis Guillermo Villanueva Mikael Ostling Max C.Lemme Frank Niklaus | 2020 | Microsystems & Nanoengineering2020,6,1: | 4 |

| 12 | Efficacy and safety of granulocyte, monocyte/macrophage adsorptive in pediatric ulcerative colitis显示文摘AIM: To investigate efficacy and safety for granulocyte, monocyte apheresis in a population of pediatric patients with ulcerative colitis.METHODS: The ADAPT study was a prospective, openlabel, multicenter study in pediatric patients with moderate, active ulcerative colitis with pediatric ulcerative colitis activity index(PUCAI) of 35-64. Patients received one weekly apheresis with Adacolumn granulocyte, monocyte/macrophage adsorptive(GMA) apheresis over 5 consecutive weeks, optionally followed by up to 3 additional apheresis treatments over 3 consecutive weeks. The primary endpoint was the change in mean PUCAI between baseline and week 12; the secondary endpoint was improvement in PUCAI categorized as(Significant Improvement, PUCAI decrease of ≥ 35), Moderate Improvement(PUCAI decrease of 20 < 35), Small Improvement(PUCAI decrease of 10 < 20) or No change(PUCAI decrease of < 10). RESULTS: Twenty-five patients(mean age 13.5 years; mean weight 47.7 kg) were enrolled. In the intention-to-treat set(ITT), the mean value for PUCAI improvement was 22.3 [95%CI: 12.9-31.6; n = 21]. In the per-protocol(PP) set, the mean improvement was 36.3 [95%CI: 31.4-41.1; n = 8]. Significant Improvement was recorded for 9 out of 20 patients(45%); 5 out of 20 patients(25%) had Moderate Improvement and one patient(5%) had No Change in PUCAI score at week 12. In the PP set, six out of eight patients(75%) showed Significant Improvement; and in two out of eight patients(25%) Moderate Improvement was recorded. The endoscopic activity index(EAI) decreased by 3 points on average. Seven(7) out of 21(33%) patients in ITT and 4 out of 8(50%) patients in PP have used steroids during the clinical investigation. The mean steroid dosage for these patients in the ITT set decreased from a mean 12.4 mg to 10 mg daily on average from Baseline to week 12.CONCLUSION: Adacolumn; GMA apheresis treatment was effective in pediatric patients with moderate active Ulcerative Colitis. No new safety signals were reported.
