5Acute effects of human protein S administration after traumatic brain injury in mice显示文摘Despite years of effort,no effective acute phase treatment has been discovered for traumatic brain injury.One impediment to successful drug development is entangled secondary injury pathways.Here we show that protein S,a natural multifunctional protein that regulates coagulation,inflammation,and apoptosis,is able to reduce the extent of multiple secondary injuries in traumatic brain injury,and therefore improve prognosis.Mice subjected to controlled cortical impact were treated acutely(10–15 minutes post-injury)with a single dose of either protein S(1 mg/kg)or vehicle phosphate buffered saline via intravenous injection.At 24 hours post-injury,compared to the non-treated group,the protein S treated group showed substantial improvement of edema and fine motor coordination,as well as mitigation of progressive tissue loss.Immunohistochemistry and western blot targeting caspase-3,B-cell lymphoma 2(Bcl-2)along with terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assay revealed that apoptosis was suppressed in treated animals.Immunohistochemistry targeting CD11 b showed limited leukocyte infiltration in the protein S-treated group.Moreover,protein S treatment increased the ipsilesional expression of aquaporin-4,which may be the underlying mechanism of its function in reducing edema.These results indicate that immediate intravenous protein S treatment after controlled cortical impact is beneficial to traumatic brain injury prognosis.Animal Use Protocols(AUPs)were approved by the University Committee on Animal Resources(UCAR)of University of Rochester Medical Center(approval No.UCAR-2008-102 R)on November 12,2013.Xiaowei Wang Jing Tong Xiaodi Han Xiaoming Qi Jun Zhang Erxi Wu Jason H.Huang 2020Neural Regeneration Research2020,15,11:1

8DNA aptamer S11e recognizes fibrosarcoma and acts as a tumor suppressor显示文摘Fibrosarcoma is a serious malignant mesenchymal tumor with strong invasiveness,high recurrence,and poor prognosis.Currently,surgical resection is the main treatment for fibrosarcoma.However,due to the lack of specific biomarkers,the inability to accurately diagnose fibrosarcoma can lead to sub-optimal surgical outcomes and decreased survival.Here,we seek to address this translational barrier and we show that DNA aptamer S11e was able to recognize fibrosarcoma cells(HT1080)but not human embryonic lung fibroblast cells with Kd values in the nanomolar range.In addition,we found that S11e discerned tumors in HT1080 xenograft mouse models and tumor tissues from fibrosarcoma patients.Furthermore,we demonstrated that S11e internalized into HT1080 cells independent of the lysosome pathway and located in mitochondria.Moreover,we revealed that S11e promoted the apoptosis of HT1080 cells and inhibited HT1080 cell migration.Finally,we investigated the biologically functional cellular target of S11e using a mass spectrometry approach,and identified that Diablo/SMAC protein is a cellular binding protein of S11e,by interacting to which S11e inhibited HT1080 cell migration and invasion.Taken together,these results provide the evidence that S11e may be useful for early diagnosis,targeted therapy,and prognostication of fibrosarcoma.Yunyi Liu Cheng Peng Hui Zhang Juan Li Hailong Ou Yang Sun Chaoqi Wen Dan Qi Xiaoxiao Hu Erxi Wu Weihong Tan 2022Bioactive Materials2022,7,6:0
9Development of phosphonate-terminated magnetic mesoporous silica nanoparticles for pH-controlled release of doxorubicin and improved tumor accumulation显示文摘In this study,phosphonate-terminated magnetic mesoporous nanoparticles(pMMSNs)was designed by incorporation of MNPs in the center of mesoporous silica nanoparticles(MSNs)and followed by grafting phosphonate group on to the surface of MMSNs.The carrier exhibited a typical superparamagnetic feature and the saturation magnetization was 4.89 emu/g measured by vibrating sample magnetometer(VSM).pMMSNs had a spherical morphology and a pore size of 2.2 nm.FromN2 adsorption-desorption analysis,pMMSNs had a surface area of 613.4 m^(2)/g and a pore volume of 0.78 cm^(3)/g.Phosphonate modification improved the colloidal stability of MMSNs,and the hydrodynamic diameter of pMMSNs was around 175 nm.The hydrophilic phosphonate group significantly enhanced the negative surface charge of MMSNs from -19.3 mV to -28.8 mV pMMSNs with more negative surface charge had a 2.3-fold higher drug loading capacity than that of MMSNs.In addition,the rate and amount of release of doxorubicin(DOX)from DOX/pMMSNs was pH-dependent and increased with the decrease of pH.At pH 7.4,the release amount was quite low and only approximately 17wt%ofDOXwasreleasedin48h.AtpH5.0and3.0,the release rate increased significantly and the release amount achieved 31 wt%and 60 wt%in 48 h,respectively.To evaluate the magnetic targeting performance ofpMMSNs,FITC labeledpMMSNswas injected into mice bearing S180 solid tumor.FITC labeledpMMSNscontrolled by an external magnetic field showed higher tumor accumulation and lower normal tissue distribution.Erxi Che Long Wan Ying Zhang Qinfu Zhao Xiling Han Jia Li Jia Liu Siling Wang 2014Asian Journal of Pharmaceutical Sciences2014,9,6:0
10Astrocytic calcium waves:unveiling their roles in sleep and arousal modulation显示文摘Neuron-astrocyte interactions are vital for the brain’s connectome.Understanding astrocyte activities is crucial for comprehending the complex neural network,particularly the population-level functions of neurons in different cortical states and associated behaviors in mammals.Studies on animal sleep and wakefulness have revealed distinct cortical synchrony patterns between neurons.Astrocytes,outnumbering neurons by nearly fivefold,support and regulate neuronal and synaptic function.Recent research on astrocyte activation during cortical state transitions has emphasized the influence of norepinephrine as a neurotransmitter and calcium waves as key components of ion channel signaling.This summary focuses on a few recent studies investigating astrocyte-neuron interactions in mouse models during sleep,wakefulness,and arousal levels,exploring the involvement of noradrenaline signaling,ion channels,and glutamatergic signaling in different cortical states.These findings highlight the significant impact of astrocytes on large-scale neuronal networks,influencing brain activity and responsiveness.Targeting astrocytic signaling pathways shows promise for treating sleep disorders and arousal dysregulation.More research is needed to understand astrocytic calcium signaling in different brain regions and its implications for dysregulated brain states,requiring future human studies to comprehensively investigate neuron-astrocyte interactions and pave the way for therapeutic interventions in sleep-and arousal-related disorders.Erxi Wu Dan Qi Damir Nizamutdinov Jason H.Huang 2024Neural Regeneration Research2024,19,5:0
11Identification of new aptamer BC-3 targeting RPS7 from rapid screening for bladder carcinoma显示文摘Aptamers,short single DNA or RNA oligonucleotides,have shown immense application potential as molecular probes for the early diagnosis and therapy of cancer.However,conventional cell-SELEX technologies for aptamer discovery are time-consuming and laborious.Here we discovered a new aptamer BC-3 by using an improved rapid X-Aptamer selection process for human bladder carcinoma,for which there is no specific molecular probe yet.We show that BC-3 exhibited excellent affinity in bladder cancer cells but not normal cells.We demonstrate that BC-3 displayed high selectivity for tumor cells over their normal counterparts in vitro,in mice,and in patient tumor tissue specimens.Further endocytosis pathway analysis revealed that BC-3 internalized into bladder cancer cells via clathrin-mediated endocytosis.Importantly,we identified ribosomal protein S7(RPS7)as the binding target of BC-3 via an integrated methodology(mass spectrometry,colocalization assay,and immunoblotting).Together,we report that a novel aptamer BC-3 is discovered for bladder cancer and its properties in the disease are unearthed.Our findings will facilitate the discovery of novel diagnostic and therapeutic strategies for bladder cancer.Yunyi Liu Juan Li Hailong Ou Dan Qi Bei Hu Yuxi Xu Jian Hu Yi Xiong Luling Xia Jason HHuang Xiaoxiao Hu Erxi Wu 2023Genes & Diseases2023,10,5:0
