| 1 | Surveilling Russell body Helicobacter pylori-negative gastritis: A case report and review of literature显示文摘BACKGROUND Russell body gastritis(RBG)is very rare type of chronic inflammation of gastric mucosa.The pathologic hallmark of the disease is Russell bodies(RB)which represent accumulation of eosinophilic cytoplasmic inclusions in endoplasmic reticulum of mature plasma cells(Mott cells).Most published cases are associated with Helicobacter pylori(H.pylori)infection because of correlation between plasma cell activation and antigenic stimulation.There are insufficient data about H.pylori-negative RBG and very little is known about the natural course of the disease.CASE SUMMARY A 51-year-old male patient underwent endoscopic screening for mild iron deficiency anemia.Gastroscopy revealed diffuse hyperemia,edema and nodularity of the fundic and corpus mucosa.Due to non-specific endoscopic findings and iron-deficiency anemia our preliminary diagnosis was diffuse type of gastric carcinoma or gastric lymphoma.Biopsy specimens of gastric mucosa showed inflammatory infiltrate rich in Mott cells,consisting entirely of cytoplasmic RB.Absence of nuclear atypia and mitosis of the plasma cells,polyclonal pattern of the Mott cells and negative staining for cytokeratins favored diagnosis of RBG.The patient was treated with proton-pump inhibitor for 8 wk.Long-term clinical and endoscopic surveillance was scheduled.Albeit,there was no improvement in endoscopic features of the gastric mucosa in three consecutive gastroscopies,histopathological findings demonstrated that the chronic inflammatory infiltrate in the fundic mucosa is less pronounced,rich in plasma cells,with almost absent RB and Mott cells.CONCLUSION The prognosis of this entity is uncertain,that is why these patients are subjects of continuous follow up. | Milena Peruhova Monika Peshevska-Sekulovska Viktoriya Georgieva Gabriela Panayotova Dorian Dikov | 2020 | World Journal of Gastroenterology2020，26，33： | 5 |
| 2 | 比较胺碘酮加β受体阻断剂、单用索他洛尔与单用β受体阻断剂预防埋藏式复律除颤器电击的随机试验:OPTIC研究显示文摘Context: Implantable cardioverter defibrillator(ICD) therapy is effective but is associated with high-voltage shocks that are painful. Objective: To determine whether amiodarone plus β -blocker or sotalol are better than β -blocker alone for prevention of ICD shocks. Design, Setting, and Patients: A randomized controlled trial with blinded adjudication of events of 412 patients from 39 outpatient ICD clinical centers located in Canada, Germany, United States, England, Sweden, and Austria, conducted from January 13, 2001, to September 28, 2004. Patients were eligible if they had received an ICD within 21 days for inducible or spontaneously occurring ventricular tachycardia or fibrillation. Intervention: Patients were randomized to treatment for 1 year with amiodarone plus β -blocker, sotalol alone, or β -blocker alone. Main Outcome Measure: Primary outcome was ICD shock for any reason. Results: Shocks occurred in 41 patients(38.5% ) assigned to β -blocker alone, 26(24.3% ) assigned to sotalol, and 12(10.3% ) assigned to amiodarone plus β -blocker. A reduction in the risk of shock was observed with use of either amiodarone plus β -blocker or sotalol vs β -blocker alone(hazard ratio[HR], 0.44; 95% confidence interval[CI], 0.28- 0.68; P< .001). Amiodarone plus β -blocker significantly reduced the risk of shock compared with β -blocker alone(HR, 0.27; 95% CI, 0.14- 0.52; P< .001) and sotalol(HR, 0.43; 95% CI, 0.22- 0.85; P=.02). There was a trend for sotalol to reduce shocks compared with β -blocker alone(HR, 0.61; 95% CI, 0.37- 1.01; P=.055). The rates of study drug discontinuation at 1 year were 18.2% for amiodarone, 23.5% for sotalol, and 5.3% for β -blocker alone. Adverse pulmonary and thyroid events and symptomatic bradycardia were more common among patients randomized to amiodarone. Conclusions: Despite use of advanced ICD technology and treatment with a β -blocker, shocks occur commonly in the first year after ICD implant. Amiodarone plus β -blocker is effective for preventing these shocks and is more effective than sotalol but has an increased risk of drug-related adverse effects. | 3 | An overview of resistance to chemotherapy in osteosarcoma and future perspectives显示文摘Osteosarcoma(OS)is the most common type of bone sarcoma.Despite the availability of multimodal treatment with surgery and chemotherapy,the clinical results remain unsatisfactory.The main reason for the poor outcomes in patients with OS is the development of resistance to methotrexate,cisplatin,doxorubicin,and ifosfamide.Molecular and cellular mechanisms associated with resistance to chemotherapy include DNA repair and cell-cycle alterations,enhanced drug efflux,increased detoxification,resistance to apoptosis,autophagy,tumor extracellular matrix,and angiogenesis.This versatility of cells to generate chemoresistance has motivated the use of anti-angiogenic therapy based on tyrosine kinase inhibitors.This approach has shown that other therapies,along with standard chemotherapy,can improve responses to therapy in patients with OS.Moreover,microRNAs may act as predictors of drug resistance in OS.This review provides insight into the molecular and cellular mechanisms involved in the development of resistance during the treatment of OS and discusses promising novel therapies(e.g.,afatinib and palbociclib)for overcoming resistance to chemotherapy in OS. | Dorian Yarih Garcia-Ortega Sara Aileen Cabrera-Nieto Haydee Sarai Caro-Sanchez Marlid Cruz-Ramos | 2022 | Cancer Drug Resistance2022，5，3： | 2 |
| 4 | Genome-wide association studies of lactation yields of milk, fat, protein and somatic cell score in New Zealand dairy goats显示文摘Background: Identifying associations between genetic markers and traits of economic importance will provide practical benefits for the dairy goat industry, enabling genomic prediction of the breeding value of individuals, and facilitating discovery of the underlying genes and mutations. Genome-wide association studies were implemented to detect genetic regions that are significantly associated with effects on lactation yields of milk(MY), fat(FY),protein(PY) and somatic cell score(SCS) in New Zealand dairy goats.Methods: A total of 4,840 goats were genotyped with the Caprine 50 K SNP chip(Illumina Inc., San Diego, CA).After quality filtering, 3,732 animals and 41,989 SNPs were analysed assuming an additive linear model. Four GWAS models were performed, a single-SNP additive linear model and three multi-SNP Bayes C models. For the single-SNP GWAS, SNPs were fitted individually as fixed covariates, while the Bayes C models fit all SNPs simultaneously as random effects. A cluster of significant SNPs were used to define a haplotype block whose alleles were fitted as covariates in a Bayesian model. The corresponding diplotypes of the haplotype block were then fit as class variables in another Bayesian model.Results: Across all four traits, a total of 43 genome-wide significant SNPs were detected from the SNP GWAS. At a genome-wide significance level, the single-SNP analysis identified a cluster of variants on chromosome 19 associated with MY, FY, PY, and another cluster on chromosome 29 associated with SCS. Significant SNPs mapped in introns of candidate genes(45%), in intergenic regions(36%), were 0–5 kb upstream or downstream of the closest gene(14%) or were synonymous substitutions(5%). The most significant genomic window was located on chromosome 19 explaining up to 9.6% of the phenotypic variation for MY, 8.1% for FY, 9.1% for PY and 1% for SCS.Conclusions: The quantitative trait loci for yield traits on chromosome 19 confirms reported findings in other dairy goat populations.
