| 1 | Validation of Zebrafish (Danio rerio) Reference Genes for Quantitative Real-time RT-PCR Normalization显示文摘The normalization of quantitative real time RT-PCR (qRT-PCR) is important to obtain accurate gene expression data. The most common method for qRT-PCR normalization is to use reference, or house- keeping genes. However, there is emerging evidence that even reference genes can be regulated under different conditions, qRT-PCR has only recently been used in terms of zebrafish gene expression studies and there is no validated set of reference genes. This study characterizes the expression of nine possible reference genes during zebrafish embryonic development and in a zebrafish tissue panel. All nine reference genes exhibited variable expression. The β-actin, EF1α and Rp113α genes comprise a validated reference gene panel for zebrafish developmental time course studies, and the EF1α, Rp113α and 18S rRNA genes are more suitable as a reference gene panel for zebrafish tissue analysis. | 6 | CAD-VR geometry and meta data synchronization for design review applications显示文摘In this paper we present the MEMPHIS middleware framework for the integration of CAD geometries and assemblies with derived Virtual Reality (VR) models and its specific meta data and attributes. The goal of this work is to connect real time VR applications, especially for the Design Review, with enterprise software storing and managing CAD models (Product Data Man- agement—PDM). The preparation of VR models requires expert knowledge, is time consuming, and includes selection of required CAD data, tessellation, healing of unwanted gaps, applying materials and textures, and special surface and light effects. During the Design Review process, decisions are made concerning the choice of materials and surface forms. While materials can be switched directly on the VR model, the modification of part geometries must be made on the CAD model. Our system synchronizes modi- fications of the original CAD geometries and of attributes that are relevant for the realistic rendering using the PLM Services standard. | 13 | Surface Ozone in Jiuzhaigou National Park, Eastern Rim of the Qinghai-Tibet Plateau, China显示文摘Located in southwestern China, Jiuzhaigou National Park is one of the most popular tourism destinations in China, famous for its unique aquatic ecosystems and beautiful forests. However, plants in the park may be at high ozone risk as a result of the intensive use of diesel tour buses in the park. In addition, Jiuzhaigou is close to a region with relatively high regional anthropogenic NO x emissions. During the growing season, also the peak season of tourism, we measured ozone concentration at two sites within the Park and these were: Jiuzhaigou Bureau (JB) and Long Lake (LL). The results indicate that ozone concentrations were not high enough to cause foliar injury during the monitoring period, although the risk of ozone to plants was higher in spring than in summer and autumn. Diurnal ozone cycles at JB and LL displayed significantly higher ozone concentrations in the daytime than in the nighttime, suggesting photochemical production of ozone during the day and ozone deposition during the night as a result of the nocturnal boundary layer. In parallel with the seasonal change of background surface ozone in the Northern Hemisphere, maximum daily 8-h average ozone concentration (MDA8) and daily ozone concentration decreased from spring to autumn at the two sites. This temporal variation in Jiuzhaigou was most likely associated with the downward mixing of ozone-rich air from the free troposphere, because all the high-ozone events (MDA8 > 70.0 ppb) were observed in spring and ozone-rich air from the free troposphere was the dominant cause.
