1Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). Our work dem on strates the utility of the con versi on betwee n hepatocyte and liver proge nitor-like cells for studying HBV biology and an tiviral therapies. 2Effects of Shuanghuanglian oral liquids on patients with COVID-19:a randomized,open-label,parallel-controlled,multicenter clinical trial显示文摘We conducted a randomized,open-label,parallel-controlled,multicenter trial on the use of Shuanghuanglian(SHL),a traditional Chinese patent medicine,in treating cases of COVID-19.A total of 176 patients received SHL by three doses(56 in low dose,61 in middle dose,and 59 in high dose)in addition to standard care.The control group was composed of 59 patients who received standard therapy alone.Treatment with SHL was not associated with a difference from standard care in the time to disease recovery.Patients with 14-day SHL treatment had significantly higher rate in negative conversion of SARS-CoV-2 in nucleic acid swab tests than the patients from the control group(93.4%vs.73.9%,P=0.006).Analysis of chest computed tomography images showed that treatment with high-dose SHL significantly promoted absorption of inflammatory focus of pneumonia,which was evaluated by density reduction of inflammatory focus from baseline,at day 7(mean difference(95%CI),−46.39(−86.83 to−5.94)HU;P=0.025)and day 14(mean difference(95%CI),−74.21(−133.35 to−15.08)HU;P=0.014).No serious adverse events occurred in the SHL groups.This study illustrated that SHL in combination with standard care was safe and partially effective for the treatment of COVID-19.Li Ni Zheng Wen Xiaowen Hu Wei Tang Haisheng Wang Ling Zhou Lujin Wu Hong Wang Chang Xu Xizhen Xu Zhichao Xiao Zongzhe Li Chene Li Yujian Liu Jialin Duan Chen Chen Dan Li Runhua Zhang Jinliang Li Yongxiang Yi Wei Huang Yanyan Chen Jianping Zhao Jianping Zuo Jianping Weng Hualiang Jiang Dao Wen Wang 2021Frontiers of Medicine2021,15,5:7

4Cis-2-dodecenoic Acid Mediates Its Synergistic Effect with Triazoles by Interfering with Efflux Pumps in Fluconazole-resistant Candida albicans显示文摘Objective To evaluate the synergy of the Burkholderia signaling molecule cis-2-dodecenoic acid(BDSF) and fluconazole(FLU) or itraconazole(ITRA) against two azole-resistant C. albicans clinical isolates in vitro and in vivo. Methods Minimum inhibitory concentrations(MICs) of antibiotics against two azole-resistant C. albicans were measured by the checkerboard technique, E-test, and time-kill assay. In vivo antifungal synergy testing was performed on mice. Analysis of the relative gene expression levels of the strains was conducted by quantitative reverse-transcription polymerase chain reaction(qR T-PCR). Results BDSF showed highly synergistic effects in combination with FLU or ITRA with a fractional inhibitory concentration index of ≤ 0.08. BDSF was not cytotoxic to normal human foreskin fibroblast cells at concentrations of up to 300 μg/mL. The qR T-PCR results showed that the combination of BDSF and FLU/ITRA significantly inhibits the expression of the efflux pump genes CDR1 and MDR1 via suppression of the transcription factors TAC1 and MRR1, respectively, when compared with FLU or ITRA alone. No dramatic difference in the mR NA expression levels of ERG1, ERG11, and UPC2 was found, which indicates that the drug combinations do not significantly interfere with UPC2-mediated ergosterol levels. In vivo experiments revealed that combination therapy can be an effective therapeutic approach to treat candidiasis. 6Calcipotriol abrogates cancer-associated fibroblast-derived IL-8-mediated oxaliplatin resistance in gastric cancer cells via blocking PI3K/Akt signaling显示文摘Activation of vitamin D receptor(VDR)in cancer-associated fibroblasts(CAFs)has been implicated in hesitating tumor progression and chemoresistance of several human malignancies.Yet,the role of VDR in CAF-induced chemotherapy resistance of gastric cancer(GC)cells remains elusive.In this study we first conducted immunohistochemistry analysis on tissue microarrays including 88 pairs of GC and normal mucosa samples,and provided clinical evidence that VDR was mainly expressed in gastric mucous cells but almost invisible in CAFs,and VDR expression was negatively correlated with malignant clinical phenotype and advanced stages,low VDR expression confers to poor overall survival rate of patients with GC.In a co-culture system of primary CAFs and cancer cells,we showed that treatment of HGC-27 and AGS GC cells with VDR ligand calcipotriol(Cal,500 nM)significantly inhibited CAF-induced oxaliplatin resistance.By using RNA-sequencing and Human Cytokine Antibody Array,we demonstrated that IL-8 secretion from CAFs induced oxaliplatin resistance via activating the PI3K/AKT pathway in GC,whereas Cal treatment greatly attenuated the tumor-supportive effect of CAF-derived IL-8 on GC cells.Taken together,this study verifies the specific localization of VDR in GC tissues and demonstrates that activation of VDR abrogates CAF-derived IL-8-mediated oxaliplatin resistance in GC via blocking PI3K/Akt signaling,suggesting vitamin D supplementation as a potential strategy of enhancing the anti-tumor effect of chemotherapy in GC.Zhen-xiong Zhao Yan-qiu Zhang Hui Sun Zi-qi Chen Jin-jia Chang Xin Wang Xu Wang Cong Tan Shujuan Ni Wei-wei Weng Meng Zhang Lei Wang Dan Huang Yun Feng Wei-qi Shen Mi-die Xu 2023Acta Pharmacologica Sinica2023,44,1:2

10TLSmell: Direct Identification on Malicious HTTPs Encryption Traffic withSimple Connection-Specific Indicators显示文摘Internet traffic encryption is a very common traffic protection method.Most internet traffic is protected by the encryption protocol called transport layersecurity (TLS). Although traffic encryption can ensure the security of communication, it also enables malware to hide its information and avoid being detected.At present, most of the malicious traffic detection methods are aimed at the unencrypted ones. There are some problems in the detection of encrypted traffic, suchas high false positive rate, difficulty in feature extraction, and insufficient practicability. The accuracy and effectiveness of existing methods need to be improved.In this paper, we present TLSmell, a framework that conducts maliciousencrypted HTTPs traffic detection with simple connection-specific indicators byusing different classifiers based online training. We perform deep packet analysisof encrypted traffic through data pre-processing to extract effective features, andthen the online training algorithm is used for training and prediction. Withoutdecrypting the original traffic, high-precision malicious traffic detection and analysis are realized, which can guarantee user privacy and communication security.At the same time, since there is no need to decrypt the traffic in advance, the effi-ciency of detecting malicious HTTPs traffic will be greatly improved. 14Integrated scanning spectrometer with a tunable micro-ring resonator and an arrayed waveguide grating显示文摘Integrated spectrometers with both wide optical bandwidths and high spectral resolutions are required in applications such as spectral domain optical coherence tomography(SD-OCT).Here we propose a compact integrated scanning spectrometer by using a tunable micro-ring resonator(MRR)integrated with a single arrayed waveguide grating for operation in the 1265-1335-nm range.The spectral resolution of the spectrometer is determined by the quality factor of the MRR,and the optical bandwidth is defined by the free spectral range of the arrayed waveguide grating.The spectrometer is integrated with on-chip germanium photodetectors,which enable direct electrical readout.A 70-nm optical bandwidth and a 0.2-nm channel spacing enabled by scanning the MRR across one free spectral range are demonstrated,which offer a total of350 wavelength channels with 31-kHz wavelength scanning speed.The integrated spectrometer is applied to measure different spectra and the interference signals from an SD-OCT system,which shows its great potential for future applications in sensing and imaging systems.ZUNYUE ZHANG YI WANG JIARUI WANG DAN YI DAVID WENG U CHAN WU YUAN HON KI TSANG 2022Photonics Research2022,10,5:1
