1The favorable impact of PIK3CA mutations on survival: an analysis of 2587 patients with breast cancer显示文摘The phosphatidylinositol-3 kinase(PI3K) pathway regulates a number of cellular processes, including cell survival, cell growth, and cell cycle progression. Consequently, this pathway is commonly deregulated in cancer. In particular, mutations in the gene PIK3CA that encodes the p110α catalytic subunit of the PI3K enzymes result in cell proliferation and resistance to apoptosis in vitro and induce breast tumors in transgenic mice. These data underscore the role of this pathway during oncogenesis. Thus, an ongoing, large-scale effort is underway to develop clinically active drugs that target elements of the PI3K pathway. However, conflicting data suggest that gain-of-function PIK3CA mutations may be associated with either a favorable or a poor clinical outcome, compared with the wild-type PIK3CA gene. In the current study, we performed a systematic review of breast cancer clinical studies. Upon evaluation of 2587 breast cancer cases from 12 independent studies, we showed that patients with tumors harboring a PIK3CA mutation have a better clinical outcome than those with a wild-type PIK3CA gene. Importantly, this improved prognosis may pertain only to patients with mutations in the kinase domain of p110α and to postmenopausal women with estrogen receptor-positive breast cancer. We propose three potential explanations for this paradoxical observation. First, PIK3CA mutations may interfere with the metastasis process or may induce senescence, which results in a better outcome for patients with mutated tumors. Secondly, we speculate that PIK3CA mutations may increase early tumor diagnosis by modification of the actin cytoskeleton in tumor cells. Lastly, we propose that PIK3CA mutations may be a favorable predictive factor for response to hormonal therapy, giving a therapeutic advantage to these patients. Ultimately, an improved understanding of the clinical impact of PIK3CA mutations is critical for the development of optimally personalized therapeutics against breast cancer and other solid tumors. This effort will be important to prevent or explain therapeutic failures and select patients who are most likely to respond to new therapies that inhibit the PI3K pathway.Amaury G. 2Bilateral versus unilateral cryptorchidism in nonobstructive azoospermia:testicular sperm extraction outcomes显示文摘Cryptorchidism is one of the most frequent causes of nonobstructive azoospermia(NOA)in adulthood.Although it is well known that spermatogenesis is more impaired in bilateral than in unilateral cryptorchidism,previous studies have only described small cohorts or inhomogeneous population.Consequently,we analyzed a cohort of 225 men with only a history of cryptorchidism as sole etiopathogenetic factor for NOA,and compared testicular sperm extraction(TESE)outcomes between men with bilateral versus unilateral cryptorchidism.Our results show no difference in follicle-stimulating hormone(FSH)levels and testicular volumes between men with a history of bilateral cryptorchidism compared to unilateral cryptorchidism(median:21.3 IU I^-1 vs 19.3 IU I^-1,P=0.306;and 7.2 ml vs 7.9 ml,P=0.543,respectively).In addition,sperm retrieval rates were similar(66.2%vs 60.0%,P=0.353).Using multivariate analysis,we have found that only a low inhibin B level(above the assay's detection limit)was positively associated with successful sperm retrieval(P<0.05).Regarding intracytoplasmic sperm injection outcomes,we found that cumulative pregnancy rate and live birth rate per cycle were not statistically different between the two groups(17.4%vs 27.8%,P=0.070;and 16.1%vs 26.4%,P=0.067,respectively).Unexpectedly,there was no significant difference in hormonal profiles(FSH,luteinizing hormone[LH],testosterone,and inhibin B levels)and TESE outcomes between unilateral versus bilateral cryptorchidism.This suggests that a history of unilateral cryptorchidism could reflect a bilateral testicular impairment.Interestingly,inhibin B level might be a predictor of successful TESE.Anne-Laure Barbotin Anais Dauvergne Agathe Dumont Nassima Ramdane Valerie Mitchell Jean-Marc Rigot Florence Boitrelle Geoffroy Robin 2019Asian Journal of Andrology2019,21,5:4

4Monolithic crystalline silicon solar cells with SiN_x layers doped with Tb^(3+) and Yb^(3+) rare-earth ions显示文摘In this study, we propose the fabrication of monolithic crystalline silicon solar cells with Tb^(3+) and Yb^(3+)-doped silicon nitride(SiN_x) layers by low-cost screen-printing methods. The performances of c-Si solar cells can be enhanced by rare-earth ions doped SiN_x layers via the mechanism of spectrum conversion.These SiN_x doped and codoped thin films were deposited by reactive magnetron co-sputtering and integrated as the antireflection coating layers in c-Si solar cells. The characterizations of SiN_x, SiN_x:Tb^(3+) tand SiN_x:Tb^(3+)-Yb^(3+) thin films were conducted by means of photoluminescence, Rutherford backscattering spectroscopy, Ellipsometry spectroscopy and Fourier transform infrared measurements. Their composition and refractive index was optimized to obtain good anti-reflection coating layer for c-Si solar cells.Transmission electron microscopy performs the uniform coatings on the textured emitter of c-Si solar cells. 7Toluene degradation by a water/silicone oil mixture for the design of two phase partitioning bioreactors显示文摘Toluene degradation performances were studied in a 10 L Two-Phase Partitioning Bioreactor(TPPB).The liquid phase consisted of a mixture of water and PDMS 50(Poly Di Methyl Siloxane,i.e.silicone oil,viscosity of 46 m Pa·s) in the volume ratio of 75%/25%.Two series of experiments were carried out:in the first,the reactor was sequentially supplied with toluene whereas in the second,toluene was continuously supplied.Activated sludge from the wastewater treatment plant of Beaurade(Rennes,France) was used at an initial concentration of 0.5 dry mass g·(mixture L)^(-1).The elimination capacity(EC) was investigated as well as the change in biomass concentration over time.Toluene biodegradation was very ef ficient(removal ef ficiency,RE=100%) for toluene flows ranging from 0.2 to 1.2 ml·h^(-1),corresponding to elimination capacities of up to 104 g·m^(-3)·h^(-1).For a toluene flow of 1.2 ml·h^(-1),the biomass concentration measured at the end of the experiment was 4.7 dry mass g·(mixture L)^(-1).The oxygen concentration in the liquid phase was clearly not a limiting factor in these operating conditions.Based on these results,an extrapolation leading to the design of a large-scale pilot TPPB can now be considered to study toluene degradation performances in industrial conditions.Maxime Guillerm Annabelle Couvert Abdeltif Amrane Edith Norrant Audrey Breton éric Dumont 2017Chinese Journal of Chemical Engineering2017,25,10:2

16用心脏磁共振成像解释肥厚型心肌病的心电图异常显示文摘Aims: To clarify the mechanisms of electrocardiographic abnormalities in hypertrophic cardiomyopathy, 102 patients were examined with cardiac magnetic resonance. Distribution and magnitude of hypertrophy and late-enhancement were correlated with electrocardiographic abnormalities. Methods and results: Abnormal Q waves were associated with greater upper anterior septal thickness(22±7 mm vs. 18±5 mm, P=0.001) and increased ratios of upper anterior septum to mean inferolateral(P=0.01), anterolateral(P=0.002), apical(P=0.001), and right ventricular(P=0.001) wall thickness. There was no relation between abnormal Q waves and late-enhancement, except for Q waves ≥40 ms(P=0.02). Conduction disturbances and absent septal Q waves were associated with late-enhancement(89 vs. 45%, P=0.01 and 75 vs. 39%, P=0.002, respectively). The depth of negative T waves was related to an increased ratio of the mean thickness between apical and basal level(P=0.01), and to the presence of apical late-enhancement(P=0.03). Conclusion: Abnormal Q waves reflect the interrelation between upper anterior septal thickness and other regions of the left and right ventricles, and wider Q waves are associated with late-enhancement. Conduction disturbances and absent septal Q waves are associated with late-enhancement. The depth of negative T waves is related to craniocaudal asymmetry and apical late-enhancement.Dumont C.A.
