| 1 | Material transportation and fluid-melt activity in the subduction channel: Numerical modeling显示文摘The subduction channel is defined as a planar to wedge-like area of variable size,internal structure and composition,which forms between the upper and lower plates during slab subduction into the mantle.The materials in the channel may experience complex pressure,temperature,stress and strain evolution,as well as strong fluid and melt activity.A certain amount of these materials may subduct to and later exhume from>100 km depth,forming high to ultra-high pressure rocks on the surface as widely discovered in nature.Rock deformation in the channel is strongly assisted by metamorphic fluids activities,which change composition and mechanical properties of rocks and thus affect their subduction and exhumation histories.In this study,we investigate the detailed structure and dynamics of both oceanic and continental subduction channels,by conducting highresolution petrological-thermomechanical numerical simulations taking into account fluid and melt activities.The numerical results demonstrate that subduction channels are composed of a tectonic rock melange formed by crustal rocks detached from the subducting slab and the hydrated mantle rocks scratched from the overriding plate.These rocks may either extrude sub-vertically upward through the mantle wedge to the crust of the upper plate,or exhume along the subduction channel to the surface near the suture zone.Based on our numerical results,we first analyze similarities and differences between oceanic and continental subduction channels.We further compare numerical models with and without fluid and melt activity and demonstrate that this activity results in strong weakening and deformation of overriding lithosphere.Finally,we show that fast convergence of orogens subjected to fluid and melt activity leads to strong deformation of the overriding lithosphere and the topography builds up mainly on the overriding plate.In contrast,slow convergence of such orogens leads to very limited deformation of the overriding lithosphere and the mountain building mainly occurs on the subducting plate. | LI ZhongHai LIU MingQi Taras GERYA | 2015 | Science China Earth Sciences2015,58,8: | 18 |
| 2 | The Immune System and the Role of Inflammation in Perinatal Depression显示文摘Major depression during pregnancy is a common psychiatric disorder that arises from a complex and multifactorial etiology. Psychosocial stress, sex, hormones, and genetic vulnerability increase the risk for triggering mood disorders. Microglia and toll-like receptor 4 play a crucial role in triggering wide and varied stress-induced responses mediated through activation of the inflammasome; this leads to the secretion of inflammatory cytokines, increased serotonin metabolism, and reduction of neurotransmitter availability along with hypothalamic–pituitary–adrenal axis hyperactivity. Dysregulation of this intricate neuroimmune communication network during pregnancy modifies the maternal milieu, enhancing the emergence of depressive symptoms and negative obstetric and neuropsychiatric outcomes. Although several studies have clearly demonstrated the role of the innate immune system in major depression, it is still unclear how the placenta, the brain, and the monoaminergic and neuroendocrine systems interact during perinatal depression. Thus, in the present review we describe the cellular and molecular interactions between these systems in major depression during pregnancy, proposing that the same stress-related mechanisms involved in the activation of the NLRP3 inflammasome in microglia and peripheral myeloid cells in depressed patients operate in a similar fashion in the neuroimmune placenta during perinatal depression. | 3 | Stagnant lid tectonics:Perspectives from silicate planets,dwarf planets, large moons,and large asteroids显示文摘To better understand Earth's present tectonic style-plate tectonics—and how it may have evolved from single plate(stagnant lid) tectonics, it is instructive to consider how common it is among similar bodies in the Solar System. Plate tectonics is a style of convection for an active planetoid where lid fragment(plate) motions reflect sinking of dense lithosphere in subduction zones, causing upwelling of asthenosphere at divergent plate boundaries and accompanied by focused upwellings, or mantle plumes;any other tectonic style is usefully called 'stagnant lid' or 'fragmented lid'. In 2015 humanity completed a 50+ year effort to survey the 30 largest planets, asteroids, satellites, and inner Kuiper Belt objects,which we informally call 'planetoids' and use especially images of these bodies to infer their tectonic activity. The four largest planetoids are enveloped in gas and ice(Jupiter, Saturn, Uranus, and Neptune)and are not considered. The other 26 planetoids range in mass over 5 orders of magnitude and in diameter over 2 orders of magnitude, from massive Earth down to tiny Proteus; these bodies also range widely in density, from 1000 to 5500 kg/m^3. A gap separates 8 silicate planetoids with ρ = 3000 kg/m^3 or greater from 20 icy planetoids(including the gaseous and icy giant planets) with ρ = 2200 kg/m^3 or less. We define the 'Tectonic Activity Index'(TAI), scoring each body from 0 to 3 based on evidence for recent volcanism, deformation, and resurfacing(inferred from impact crater density). Nine planetoids with TAI = 2 or greater are interpreted to be tectonically and convectively active whereas 17 with TAI <2 are inferred to be tectonically dead. We further infer that active planetoids have lithospheres or icy shells overlying asthenosphere or water/weak ice. TAI of silicate(rocky) planetoids positively correlates with their inferred Rayleigh number. We conclude that some type of stagnant lid tectonics is the dominant mode of heat loss and that plate tectonics is unusual. | 6 | Neural progenitor cells but not astrocytes respond distally to thoracic spinal cord injury in rat models显示文摘Traumatic spinal cord injury(SCI) is a detrimental condition that causes loss of sensory and motor function in an individual. Many complex secondary injury cascades occur after SCI and they offer great potential for therapeutic targeting. In this study, we investigated the response of endogenous neural progenitor cells,astrocytes, and microglia to a localized thoracic SCI throughout the neuroaxis. Twenty-five adult female Sprague-Dawley rats underwent mild-contusion thoracic SCI(n = 9), sham surgery(n = 8), or no surgery(n = 8). Spinal cord and brain tissues were fixed and cut at six regions of the neuroaxis. Immunohistochemistry showed increased reactivity of neural progenitor cell marker nestin in the central canal at all levels of the spinal cord. Increased reactivity of astrocyte-specific marker glial fibrillary acidic protein was found only at the lesion epicenter. The number of activated microglia was significantly increased at the lesion site,and activated microglia extended to the lumbar enlargement. Phagocytic microglia and macrophages were significantly increased only at the lesion site. There were no changes in nestin, glial fibrillary acidic protein,microglia and macrophage response in the third ventricle of rats subjected to mild-contusion thoracic SCI compared to the sham surgery or no surgery. These findings indicate that neural progenitor cells, astrocytes and microglia respond differently to a localized SCI, presumably due to differences in inflammatory signaling. These different cellular responses may have implications in the way that neural progenitor cells can be manipulated for neuroregeneration after SCI. | 7 | Effect of medicinal mushrooms on blood cells under conditions of diabetes mellitus显示文摘Diabetes mellitus(DM) is the third most common noninfectious disease leading to early disability and high mortality. Moreover, the number of patients is growing every year. The main symptom of DM is hyperglycemia. Increased levels of blood glucose activate polyol, hexosamine, and protein kinase metabolic pathways cause the intensification of non-enzymatic glycosylation and nitration of macromolecules. This, in turn, leads to the development of oxidative and nitrative stresses and secondary complications, such as different kinds of microand macroangiopathies. Metabolic disorders caused by insulin deficiency in diabetes significantly impede the functioning of a homeostasis system, which change the physical, biochemical, morphological, and functional properties of blood cells. As a result, the oxygen-transport function of red blood cells(RBCs), rheological properties of the blood, and functions of immunocompetent cells as well as the process of apoptosis are primarily affected. Modern pharmacotherapy focuses on the search for new preparations that aim to decrease blood glucose levels. Undesirable side effects and adverse reactions caused by synthetic medicines led to the search and investigation of new preparations of natural origin. Medicinal mushrooms play an important role among such new preparations. They are a source of a large number of high-and lowmolecular compounds with pronounced biological effects. Our investigations show pronounced hypoglycemic and anti-anemic action of submerged cultivated mycelium powder of medicinal mushrooms Agaricus brasiliensis(A. brasiliensis) and Ganoderma lucidum(G. lucidum) on streptozotocin-induced DM in rats. Also, we showed that mycelium powders have membrane protective properties as evidenced by the redistribution of RBC populations towards the growth of full functional cell numbers. Normalization of parameters of leukocyte formula and suppression of apoptosis of white blood cells in diabeticrats treated with A. brasiliensis and G. lucidum mycelia indicates pronounced positive effects of these strains of mushrooms. Thus, the use of medicinal mushrooms for treatment of DM and in prevention development of its secondary complications might be a new effective approach of this disease's cure. | 9 | Squamous cell cancer of the rectum显示文摘Squamous cell carcinoma of the rectum is a rare malignancy.It appears to be associated with chronic inflammatory conditions and infections.The clear association seen between Human Papilloma Virus and various squamous cancers has not been firmly established for the squamous cell cancer of the rectum. The presentation is nonspecific and patients tend to present with advanced stage disease.Diagnosis relies on endoscopic examination with biopsy of the lesion.Distinction from squamous cell cancer of the anus can be difficult,but can be facilitated by immunohistochemical staining for cytokeratins.Staging of the cancer with endoscopic ultrasound and computed tomography provides essential information on prognosis and can guide therapy.At present,surgery remains the main therapeutic option;however recent advances have made chemoradiation a valuable therapeutic addition. Squamous cell carcinoma of the rectum is a distinct entity and it is of crucial importance for the practicing Gastroenterologist to be thoroughly familiar with this disease.Compared to adenocarcinoma of the rectum and squamous cell cancer of the anal canal,squamous cell carcinoma of the rectum has different epidemiology, etiology,pathogenesis,and prognosis but,most importantly,requires a different therapeutic approach. | 11 | Helicobacter pylori infection in bleeding peptic ulcer patients after non-steroidal antiinflammatory drug consumption显示文摘AIM:To establish the prevalence of Helicobacter pylori (H. pylori ) infection in patients with a bleeding peptic ulcer after consumption of non-steroidal antiinflamma- tory drugs (NSAIDs). METHODS:A very early upper endoscopy was performed to find the source of upper gastrointestinal bleeding and to take biopsy specimens for analysis of H. pylori infection by the rapid urease (CLO) test, histological examination, and bacterial culture. IgG anti-CagA were also sought. The gold standard for identifying H. pylori infection was positive culture of biopsy specimens or contemporary positivity of the CLO test and the presence of H. pylori on tissue sections. RESULTS:Eighty patients, 61 males (76.3%), mean age 61.2 ± 15.9 years, were consecutively enrolled. Forty-seven (58.8%) patients occasionally consumed NSAIDs, while 33 (41.3%) were on chronic treatment with low-dose aspirin (LD ASA). Forty-four (55.0%) patients were considered infected by H. pylori . The infection rate was not different between patients who occasionally or chronically consumed NSAIDs. The culture of biopsy specimens had a sensitivity of 86.4% and a specificity of 100%; corresponding figures for histological analysis were 65.9% and 77.8%, for the CLO test were 68.2% and 75%, for the combined use of histology and the CLO test were 56.8% and 100%, and for IgG anti-CagA were 90% and 98%. The highest accuracy (92.5%) was obtained with the culture of biopsy specimens.CONCLUSION:Patients with a bleeding peptic ulcer after NSAID/LD ASA consumption frequently have H. pylori infection. | 12 | Contrasted continental rifting via plume-craton interaction: Applications to Central East African Rift显示文摘The East African Rift system(EARS)provides a unique system with the juxtaposition of two contrasting yet simultaneously formed rift branches,the eastern,magma-rich,and the western,magma-poor,on either sides of the old thick Tanzanian craton embedded in a younger lithosphere.Data on the pre-rift,syn-rift and post-rift far-field volcanic and tectonic activity show that the EARS formed in the context of the interaction between a deep mantle plume and a horizontally and vertically heterogeneous lithosphere under far-field tectonic extension.We bring quantitative insights into this evolution by implementing high-resolution 3D thermo-mechanical numerical deformation models of a lithosphere of realistic rheology.The models focus on the central part of the EARS.We explore scenarios of plumelithosphere interaction with plumes of various size and initial position rising beneath a tectonically pre-stretched lithosphere.We test the impact of the inherited rheological discontinuities(suture zones)along the craton borders,of the rheological structure,of lithosphere plate thickness variations,and of physical and mechanical contrasts between the craton and the embedding lithosphere.Our experiments indicate that the ascending plume material is deflected by the cratonic keel and preferentially channeled along one of its sides,leading to the formation of a large rift zone along the eastern side of the craton,with significant magmatic activity and substantial melt amount derived from the mantle plume material.We show that the observed asymmetry of the central EARS,with coeval amagmatic(western)and magmatic(eastern)branches,can be explained by the splitting of warm material rising from a broad plume head whose initial position is slightly shifted to the eastern side of the craton.In that case,neither a mechanical weakness of the contact between the craton and the embedding lithosphere nor the presence of second plume are required to produce simulations that match observations.This result reconciles the passive and active rift models and demonstrates the possibility of development of both magmatic and amagmatic rifts in identical geotectonic environments. | Alexander Koptev Evgueni Burov Eric Calais Sylvie Leroy Taras Gerya Laurent Guillou-Frottier Sierd Cloetingh | 2016 | Geoscience Frontiers2016,7,2: | 4 |

| 15 | Liver-specific deletion of mechanistic target of rapamycin does not protect against acetaminophen-induced liver injury in mice显示文摘Background:Acetaminophen(APAP)overdose can cause liver injury and liver failure,which is one of the most common causes of drug-induced liver injury in the United States.Pharmacological activation of autophagy by inhibiting mechanistic target of rapamycin(mTOR)protects against APAP-induced liver injury likely via autophagic removal of APAP-adducts and damaged mitochondria.In the present study,we aimed to investigate the role of genetic ablation of mTOR pathways in mouse liver in APAP-induced liver injury and liver repair/regeneration.Methods:Albumin-Cre(Alb-Cre)mice,mTOR^(f/f) and Raptor^(f/f) mice(C57BL/6J background)were crossbred to produce liver-specific mTOR knockout(L-mTOR KO,Alb Cret/-,mTOR^(f/f))and liver-specific Raptor KO(L-Raptor,Alb Cret/-,Raptor^(f/f))mice.Alb-Cre littermates were used as wild-type(WT)mice.These mice were treated with APAP for various time points for up to 48 h.Liver injury,cell proliferation,autophagy and mTOR activation were determined.Results:We found that genetic deletion of neither Raptor,an important adaptor protein in mTOR complex 1,nor mTOR,in the mouse liver significantly protected against APAP-induced liver injury despite increased hepatic autophagic flux.Genetic deletion of Raptor or mTOR in mouse livers did not affect APAP metabolism and APAP-induced c-Jun N-terminal kinase(JNK)activation,but slightly improved mouse survival likely due to increased hepatocyte proliferation.Conclusions:Our results indicate that genetic ablation of mTOR in mouse livers does not protect against APAP-induced liver injury but may slightly improve liver regeneration and mouse survival after APAP overdose. | Hua Sun Hong-Min Ni Jennifer M.McCracken Jephte Y Akakpo Sam Fulte Tara McKeen Hartmut Jaeschke Hua Wang Wen-Xing Ding | 2021 | Liver Research2021,5,2: | 3 |

| 19 | Toward three-dimensional in vitro models to study neurovascular unit functions in health and disease显示文摘The high metabolic demands of the brain require an efficient vascular system to be coupled with neural activity to supply adequate nutrients and oxygen.This supply is coordinated by the action of neurons,glial and vascular cells,known collectively as the neurovascular unit,which temporally and spatially regulate local cerebral blood flow through a process known as neurovascular coupling.In many neurodegenerative diseases,changes in functions of the neurovascular unit not only impair neurovascular coupling but also permeability of the blood-brain barrier,cerebral blood flow and clearance of waste from the brain.In order to study disease mechanisms,we need improved physiologicallyrelevant human models of the neurovascular unit.Advances towards modeling the cellular complexity of the neurovascular unit in vitro have been made using stem-cell derived organoids and more recently,vascularized organoids,enabling intricate studies of non-cell autonomous processes.Engineering and design innovations in microfluidic devices and tissue engineering are progressing our ability to interrogate the cerebrovasculature.These advanced models are being used to gain a better understanding of neurodegenerative disease processes and potential therapeutics.Continued innovation is required to build more physiologically-relevant models of the neurovascular unit encompassing both the cellular complexity and designed features to interrogate neurovascular unit functionality. | Tara M.Caffrey Emily B.Button Jerome Robert | 2021 | Neural Regeneration Research2021,16,11: | 3 |
