| 1 | Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12 |

| 4 | Prognostic Factors of Ampulla of Vater Carcinoma after Radical Surgery显示文摘OBJECTIVE Ampullary carcinoma is a rare disease with betterprognosis than other periampullary neoplasms. This studyinvestigated the association between clinicopathologic factors andprognosis after radical resection of ampulla of Vater carcinoma.METHODS Clinical data from 105 patients who underwentradical pancreaticoduodenectomy from January 1990 to December2005 were retrospectively analyzed by the Kaplan-Meier method,log-rank test, and the Cox proportional hazard model.RESULTS The in-hospital mortality rate was 8.6%, the lymphnode metastasis rate was 37.1%, and the five-year survival ratewas 42.8%. Pancreatic involvement (P = 0.027), tumor diameter (P= 0.008), T stage (P = 0.003), TNM stage (P < 0.001), and number ofmetastatic lymph nodes (P < 0.001) were associated with prognosiswhen the univariate analysis was used. | 10 | A cluster positioning architecture and relative positioning algorithm based on pigeon fock bionics显示文摘Unmanned clusters can realize collaborative work,fexible confguration,and efcient operation,which has become an important development trend of unmanned platforms.Cluster positioning is important for ensuring the normal operation of unmanned clusters.The existing solutions have some problems such as requiring external system assistance,high system complexity,poor architecture scalability,and accumulation of positioning errors over time.Without the aid of the information outside the cluster,we plan to construct the relative position relationship with north alignment to adopt formation control and achieve robust cluster relative positioning.Based on the idea of bionics,this paper proposes a cluster robust hierarchical positioning architecture by analyzing the autonomous behavior of pigeon focks.We divide the clusters into follower clusters,core clusters,and leader nodes,which can realize fexible networking and cluster expansion.Aiming at the core cluster that is the most critical to relative positioning in the architecture,we propose a cluster relative positioning algorithm based on spatiotemporal correlation information.With the design idea of low cost and large-scale application,the algorithm uses intra-cluster ranging and the inertial navigation motion vector to construct the positioning equation and solves it through the Multidimensional Scaling(MDS)and Multiple Objective Particle Swarm Optimization(MOPSO)algorithms.The cluster formation is abstracted as a mixed direction-distance graph and the graph rigidity theory is used to analyze localizability conditions of the algorithm.We designed the cluster positioning simulation software and conducted localizability tests and positioning accuracy tests in diferent scenarios.Compared with the relative positioning algorithm based on Extended Kalman Filter(EKF),the algorithm proposed in this paper has more relaxed positioning conditions and can adapt to a variety of scenarios.It also has higher relative positioning accuracy,and the error does not accumulate over time. | Zhongliang Deng Hang Qi Chengfeng Wu Enwen Hu Runmin Wang | 2023 | Satellite Navigation2023,4,1: | 0 |

| 12 | Development of a rapid neutralizing antibody test for SARS-CoV-2 and its application for neutralizing antibody screening and vaccinated serum testing显示文摘Background:Since the outbreak of coronavirus disease(COVID-19),the high infection rate and mutation fre-quency of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),the causative agent,have contributed to the ongoing global pandemic.Vaccination has become the most effective means of controlling COVID-19.Tra-ditional neutralizing tests of sera are complex and labor-intensive,therefore,a rapid test for detecting neutralizing antibodies and antibody status post-immunization is needed.Methods:Based on the fact that antibodies exhibit neutralizing activity by blocking the binding of the S protein receptor-binding domain(S-RBD)to ACE2,we developed a rapid neutralizing antibody test,ACE2-Block-ELISA.To evaluate the sensitivity and specificity,we used 54 positive and 84 negative serum samples.We also tested the neutralizing activities of monoclonal antibodies(mAbs)and 214 sera samples from healthy individuals im-munized with the inactivated SARS-CoV-2 vaccine.Results:The sensitivity and specificity of the ACE2-Block ELISA were 96.3%and 100%,respectively.For neu-tralizing mAb screening,ch-2C5 was selected for its ability to block the ACE2-S-RBD interaction.A plaque assay confirmed that ch-2C5 neutralized SARS-CoV-2,with NT 50 values of 4.19,10.63,and 1.074μg/mL against the SARS-CoV-2 original strain,and the Beta and Delta variants,respectively.For the immunized sera samples,the neutralizing positive rate dropped from 82.14%to 32.16%within 4 months post-vaccination.Conclusions:This study developed and validated an ACE2-Block-ELISA to test the neutralizing activities of an-tibodies.As a rapid,inexpensive and easy-to-perform method,this ACE2-Block-ELISA has potential applications in rapid neutralizing mAb screening and SARS-CoV-2 vaccine evaluation. | Yuchang Li Mingyue Wang Hongzhen Wu Hui Zhao Lei Dong Yunfei Li Xiaofeng Li Ying Tang Sen Zhang Jing Li Chengfeng Qin Tao Jiang Yongqiang Deng Xiaoping Kang | 2022 | Infectious Medicine2022,1,2: | 0 |
