2The contribution of absorption of integral nanocrystals to enhancement of oral bioavailability of quercetin显示文摘In this study,self-discriminating hybrid nanocrystals was utilized to explore the biological fate of quercetin hybrid nanocrystals(QT-HNCs)with diameter around 280 nm(QT-HNCs-280)and550 nm(QT-HNCs-550)following oral and intravenous administration and the contribution of integral nanocrystals to oral bioavailability enhancement of QT was estimated by comparing the absolute exposure of integral QT-HNCs and total QT in the liver.Results showed that QT-HNCs could reside in vivo as intact nanocrystals for as long as 48 h following oral and intravenous administration.A higher accumulation of integral QT-HNCs in liver and lung was observed for both oral and intravenous administration of QT-HNCs.The particle size affects the absorption and biodistribution of integral QT-HNCs and total QT.As compared to QT-HNCs-550,QT-HNCs-280 with smaller particle size is more easily absorbed,but dissolves faster in vivo,leading to higher distribution of QT(146.90 vs.117.91 h·mg/mL)but lower accumulation of integral nanocrystals(6.82 e10 vs.15.27 e10 h·[p/s]/[μW/cm2])in liver following oral administration.Due to its slower dissolution and enhanced recognition by RES,QT-HNCs-550 with larger diameter shows higher liver distribution for both of QT(1015.80 h·mg/mL)and integral nanocrystals(259.63 e10 h·[p/s]/[μW/cm2])than those of QT-HNCs-280(673.82&77.66 e10 h·[p/s]/[μW/cm2])following intravenous administration.The absolute exposure of integral QT-HNCs in liver following oral administration of QT-HNCs are 8.78%for QT-HNCs-280 and 5.88%for QT-HNCs-550,while the absolute exposure of total QT for QT-HNCs-280 and QT-HNCs-550 are 21.80%and 11.61%,respectively.Owing to imprecise quantifcation method,a surprisingly high contribution of integral QT-HNCs to oral bioavailability enhancement of QT(40.27%for QT-HNCs-280 and 50.65%for QT-HNCs-550)was obtained.These results revealed signifcant difference in absorption and biodistrbution between integral nanocrystals and overall drugs following oral and intravenous administration of QT-HNCs,and provided a meaningful reference for the contribution of integral nanocrystals to overall bioavailability enhancement.Baode Shen Chengying Shen Weifeng Zhu Hailong Yuan 2021Acta Pharmaceutica Sinica B2021,11,4:10

4In vivo flow cytometry reveals a circadian rhythm of circulating tumor cells显示文摘Circulating tumor cells(CTCs)is an established biomarker of cancer metastasis.The circulation dynamics of CTCs are important for understanding the mechanisms underlying tumor cell dissemination.Although studies have revealed that the circadian rhythm may disrupt the growth of tumors,it is generally unclear whether the circadian rhythm controls the release of CTCs.In clinical examinations,the current in vitro methods for detecting CTCs in blood samples are based on a fundamental assumption that CTC counts in the peripheral blood do not change significantly over time,which is being challenged by recent studies.Since it is not practical to draw blood from patients repeatedly,a feasible strategy to investigate the circadian rhythm of CTCs is to monitor them by in vivo detection methods.Fluoresce nee in vivo flow cytometry(IVFC)is a powerful optical technique that is able to detect fluoresce nt circulating cells directly in living animals in a noninvasive manner over a long period of time.In this study,we applied fluorescence IVFC to monitor CTCs noninvasively in an orthotopic mouse model of human prostate cancer.We observed that CTCs exhibited stochastic bursts over cancer progression.The probability of the bursting activity was higher at early stages than at late stages.We longitudinally monitored CTCs over a 24-h period,and our results revealed striking daily oscillations in CTC counts that peaked at the onset of the night(active phase for rodents),suggesting that the release of CTCs might be regulated by the circadian rhythm.Xi Zhu Yuanzhen Suo Yuting Fu Fuli Zhang Nan Ding Kai Pang Chengying Xie Xiaofu Weng Meilu Tian Hao He Xunbin Wei 2021Light(Science & Applications)2021,10,6:6
5Isolation and characterization of ferrous-and sulfur-oxidizing bacteria from Tengchong solfataric region,China显示文摘Microbial oxidation and reduction of iron and sulfur are important parts of biogeochemical cycles in acidic environments such as geothermal solfataric regions.Species of Acidithiobacillus and Leptospirillum are the common ferrous-iron and sulfur oxidizers from such environments.This study focused on the Tengchong sofataric region,located in Yunnan Province,Southwest China.Based on cultivation,9 strains that grow on ferrous-iron and sulfuric compounds were obtained.Analysis of 16S rRNA genes of the 9 strains indicated that they were affliated to Acidithiobacillus,Alicyclobacillus,Sulfobacillus,Leptospirillum and Acidiphilium.Physiological and phylogenetic studies indicated that two strains(TC-34 and TC-71) might represent two novel members of Alicyclobacillus.Strain TC-34 and TC-71 showed 94.8%-97.1%16S rRNA gene identities to other species of Alicyclobacillus.Different from the previously described Alicyclobacillus species,strains TC-34 and TC-71 were mesophilic and their cellular fatty acids do not containω-cyclic fatty acids.Strain TC-71 was obligately dependent on ferrous-iron for growth.It was concluded that the ferrous-iron oxidizers were diversified and Alicyclobacillus species were proposed to take part in biochemical geocycling of iron in the Tengchong solfataric region.JIANG Chengying LIU Ying LIU Yanyang GUO Xu LIU Shuang-Jiang 2009Journal of Environmental Sciences2009,21,9:5
6Adaptive PI Control Strategy for Flat Permanent Magnet Linear Synchronous Motor Vibration Suppression显示文摘Due to low damping ratio,flat permanent magnet linear synchronous motor's vibration is difficult to be damped and the accuracy is limited.The vibration suppressing results are not good enough in the existing research because only the longitudinal direction vibration is considered while the normal direction vibration is neglected.The parameters of the direct-axis current controller are set to be the same as those of the quadrature-axis current controller commonly.This causes contradiction between signal noise and response.To suppress the vibration,the electromagnetic force model of the flat permanent magnet synchronous linear motor is formulated first.Through the analysis of the effect that direct-axis current noise and quadrature-axis current noise have on both direction vibration,it can be declared that the conclusion that longitudinal direction vibration is only related to the quadrature-axis current noise while the normal direction vibration is related to both the quadrature-axis current noise and direct-axis current noise.Then,the simulation test on current loop with a low-pass filter is conducted and the results show that the low-pass filter can not suppress the vibration but makes the vibration more severe.So a vibration suppressing strategy that the proportional gain of direct-axis current controller adapted according to quadrature-axis reference current is proposed.This control strategy can suppress motor vibration by suppressing direct-axis current noise.The experiments results about the effect of K p and T i on normal direction vibration,longitudinal vibration and the position step response show that this strategy suppresses vibration effectively while the motor's motion performance is not affected.The maximum reduction of vibration can be up to 40%.In addition,current test under rated load condition is also conducted and the results show that the control strategy can avoid the conflict between the direct-axis current and the quadrature-axis current under typical load.Adaptive PI control strategy can effectively suppress the flat permanent magnet linear synchronous motor's vibration without affecting the motor's performance.MENG Fanwei LIU Chengying LI Zhijun WANG Liping 2013Chinese Journal of Mechanical Engineering2013,26,1:4
7In vivo dissolution of poorly water-soluble drugs:Proof of concept based on fluorescence bioimaging显示文摘In vitro-in vivo correlation(IVIVC)of solid dosage forms should be established basically between in vitro and in vivo dissolution of active pharmaceutical ingredients.Nevertheless,in vivo dissolution profles have never been accurately portrayed.The current practice of IVIVC has to resort to in vivo absorption fractions(Fa).In this proof-of-concept study,in vivo dissolution of a model poorly watersoluble drug fenofbrate(FNB)was investigated by fuorescence bioimaging.FNB crystals were frst labeled by near-infrared fuorophores with aggregation-caused quenching properties.The dyes illuminated FNB crystals but quenched immediately and absolutely once been released into aqueous media,enabling accurate monitoring of residual drug crystals.The linearity established between fuorescence and crystal concentration justifed reliable quantifcation of FNB crystals.In vitro dissolution was frst measured following pharmacopoeia monograph protocols with well-documented IVIVC.The synchronicity between fuorescence and in vitro dissolution of FNB supported using fuorescence as a measure for determination of dissolution.In vitro dissolution correlated well with in vivo dissolution,acquired by either live or ex vivo imaging.The newly established IVIVC was further validated by correlating both in vitro and in vivo dissolution with Faobtained from pharmacokinetic data.Yinqian Yang Yongjiu Lv Chengying Shen Tingting Shi Haisheng He Jianping Qi Xiaochun Dong Weili Zhao Yi Lu Wei Wu 2021Acta Pharmaceutica Sinica B2021,11,4:3
8The Salt Reduction and Yield Increase Effects of Smashing Ridging Transforming Saline-alkali Land and Its Development Prospects显示文摘Using smashing ridging tillage machine and smashing ridging technology invented by the authors,transformation test of saline-alkali land by smashing ridging was conducted respectively in Xinjiang and Shaanxi during 2015-2016. 10Proteomic and molecular investigations revealed that Acidithiobacillus caldus adopts multiple strategies for adaptation to NaCl stress显示文摘Acidithiobacillus caldus plays an important role in commercial bioleaching.To understand how NaCl stress adaptation occurs in A.caldus,we grew A.caldus strain SM-1 in media containing high NaCl concentrations.SM-1 grew at concentrations of up to 1.0-mol L^(-1)NaCl,but growth was severely inhibited at higher concentrations.Proteomic analysis showed that SM-1 used multiple strategies to respond to NaCl stress.In addition to several heatshock proteins,enzymes involved in proline biosynthesis increased under NaCl stress.In addition,two DNA-binding proteins and a third protein of unknown function(Atc_(1291)),which was subsequently identified as a putative single-stranded DNA-binding protein,were up-regulated in the presence of NaCl stress.These DNA-binding proteins might play a role in response to osmotic stress.Atc_(1291)was cloned and expressed in Escherichia coli.Surprisingly,we found that E.coli BL21/pET28a-atc_(1291)grew to higher cell densities than E.coli BL21/pET28a,regardless of NaCI stress.Homologs to Atc_(1291)were identified in several groups of Proleohacleria.The role of Atc_(1291)in enhancing cell growth needs further investigation.Xu Guo Chengying Jiang Yuanming Luo Mingjiang Zhang Ansgar Poetsch Shuangjiang Liu 2014Chinese Science Bulletin2014,59,3:2
11Effects of novel Fufang Biejia Ruangan Tablets with sheep placenta as substitute for Hominis Placenta on CCl4-induced liver fibrosis显示文摘Objective:Fufang Biejia Ruangan Tablet(FBRT) is widely used for the treatment of liver fibrosis.However,Hominis Placenta(HP),as an important adjuvant of FBRT,has been restricted for medicinal using due to the limited availability,ethical controversy and safety issues.The present study aimed to investigate the therapeutic effects of novel FBRT(N-FBRT) with sheep placenta(SP) as substitute for HP on liver fibrosis and explore its possible mechanisms.Different dosages of SP in N-FBRT were also evaluated.Methods:Rats were subcutaneously injected with CCl_(4)to induce liver fibrosis and then treated with NFBRT and FBRT.The anti-hepatic fibrosis effect was determined based on biomarkers analysis of liver function and hepatic fibrosis,and the liver pathology was visualized by H&E staining and Masson staining.The oxidative stress and inflammatory cytokines were also detected.Immunohistochemical staining of a-SMA,real time PCR and Western blotting were performed to evaluate hepatic stellate cells(HSCs)activation and TGF-β1/Smad signaling pathway.Results:N-FBRT and FBRT could ameliorate CCl_(4)-induced liver fibrosis and improve liver function,as evidenced by lowering serum biomarkers levels of liver function and hepatic fibrosis,and decreasing hepatic Hyp content and collagen deposition,and improving the hepatic morphology and architecture changes.Moreover,the anti-liver fibrosis effect was better when the dosage of SP used in N-FBRT was 1/2 of HP in FBRT.Administration of N-FBRT markedly alleviated oxidative stress and inflammatory cytokines,and inhibited a-SMA expression.Furthermore,the mRNA expression of Col Ⅰ,Col Ⅲ,a-SMA and TGF-β1,and proteins expression of a-SMA,TGF-β1,Smad2/3 and p-Smad2/3 were significantly down-regulated by N-FBRT treatment.Conclusion:SP can be used as substitute for HP to prepare N-FBRT for the treatment of liver fibrosis and the anti-liver fibrosis effect of N-FBRT is achieved by eliminating oxidative stress and inflammation,and inhibiting HSCs activation and ECM production by blocking TGF-β1/Smad signaling pathway.Baode Shen Li Deng Yuan Liu Ruisheng Li Chengying Shen Xiao Liu Yinchao Li Hailong Yuan 2022Chinese Herbal Medicines2022,14,1:2
12Preventing and Limiting Exposure to Geo-Hazards:Some Lessons from Two Mountain Villages Destroyed by the Wenchuan Earthquake显示文摘This paper examines the experiences of two mountain communities- Yinchanggou and Donghekou in the Wenchuan earthquake of May 12, 2008, where Yinchanggou's tourism economy and natural park system was destroyed and Donghekou was buried by a landslide. We conducted research surveys on both the communities, interviewing survivors and local officials, and observed the destruction/reconstruction, geological, and living conditions. We suggest that protracted educational processes be put into place so that mountain communities possess a knowledge base to consider long-term disaster prevention when building the economy in the fragile and geo-hazardous conditions of the Longmenshan.
