1In vivo flow cytometry reveals a circadian rhythm of circulating tumor cells显示文摘Circulating tumor cells(CTCs)is an established biomarker of cancer metastasis.The circulation dynamics of CTCs are important for understanding the mechanisms underlying tumor cell dissemination.Although studies have revealed that the circadian rhythm may disrupt the growth of tumors,it is generally unclear whether the circadian rhythm controls the release of CTCs.In clinical examinations,the current in vitro methods for detecting CTCs in blood samples are based on a fundamental assumption that CTC counts in the peripheral blood do not change significantly over time,which is being challenged by recent studies.Since it is not practical to draw blood from patients repeatedly,a feasible strategy to investigate the circadian rhythm of CTCs is to monitor them by in vivo detection methods.Fluoresce nee in vivo flow cytometry(IVFC)is a powerful optical technique that is able to detect fluoresce nt circulating cells directly in living animals in a noninvasive manner over a long period of time.In this study,we applied fluorescence IVFC to monitor CTCs noninvasively in an orthotopic mouse model of human prostate cancer.We observed that CTCs exhibited stochastic bursts over cancer progression.The probability of the bursting activity was higher at early stages than at late stages.We longitudinally monitored CTCs over a 24-h period,and our results revealed striking daily oscillations in CTC counts that peaked at the onset of the night(active phase for rodents),suggesting that the release of CTCs might be regulated by the circadian rhythm.Xi Zhu Yuanzhen Suo Yuting Fu Fuli Zhang Nan Ding Kai Pang Chengying Xie Xiaofu Weng Meilu Tian Hao He Xunbin Wei 2021Light(Science & Applications)2021,10,6:6
2In vivo dissolution of poorly water-soluble drugs:Proof of concept based on fluorescence bioimaging显示文摘In vitro-in vivo correlation(IVIVC)of solid dosage forms should be established basically between in vitro and in vivo dissolution of active pharmaceutical ingredients.Nevertheless,in vivo dissolution profles have never been accurately portrayed.The current practice of IVIVC has to resort to in vivo absorption fractions(Fa).In this proof-of-concept study,in vivo dissolution of a model poorly watersoluble drug fenofbrate(FNB)was investigated by fuorescence bioimaging.FNB crystals were frst labeled by near-infrared fuorophores with aggregation-caused quenching properties.The dyes illuminated FNB crystals but quenched immediately and absolutely once been released into aqueous media,enabling accurate monitoring of residual drug crystals.The linearity established between fuorescence and crystal concentration justifed reliable quantifcation of FNB crystals.In vitro dissolution was frst measured following pharmacopoeia monograph protocols with well-documented IVIVC.The synchronicity between fuorescence and in vitro dissolution of FNB supported using fuorescence as a measure for determination of dissolution.In vitro dissolution correlated well with in vivo dissolution,acquired by either live or ex vivo imaging.The newly established IVIVC was further validated by correlating both in vitro and in vivo dissolution with Faobtained from pharmacokinetic data.Yinqian Yang Yongjiu Lv Chengying Shen Tingting Shi Haisheng He Jianping Qi Xiaochun Dong Weili Zhao Yi Lu Wei Wu 2021Acta Pharmaceutica Sinica B2021,11,4:3
3Reply to Comment on “In vivo flow cytometry reveals a circadian rhythm of circulating tumor cells”显示文摘Dear Editor,We thank Niedre et al.for their correspondence regarding our recent paper1.They proposed a point that the temporal distribution of circulating tumor cells(CTCs)monitored by diffuse in vivo flow cytometry in a multiple myeloma mouse model in their previous work2 might be different from our results.Niedre et al.claimed that CTC detection statistics deviated from Poisson but did not found circadian variations in CTC numbers in a multiple myeloma mouse model.They also cite another literature by Juratli.et al.3,in which the authors reported that CTC numbers did not always correlate with tumor size during cancer progression.However,by establishing an orthotopic mouse model of prostate cancer and utilizing the technology in vivo flow cytometry(IVFC),we found CTCs exhibited bursting activity and daily oscillation in an orthotopic model of prostate cancer.Xi Zhu Yuanzhen Suo Yuting Fu Fuli Zhang Nan Ding Kai Pang Chengying Xie Xiaofu Weng Meilu Tian Hao He Xunbin Wei 2021Light(Science & Applications)2021,10,10:2
4Preventing and Limiting Exposure to Geo-Hazards:Some Lessons from Two Mountain Villages Destroyed by the Wenchuan Earthquake显示文摘This paper examines the experiences of two mountain communities- Yinchanggou and Donghekou in the Wenchuan earthquake of May 12, 2008, where Yinchanggou's tourism economy and natural park system was destroyed and Donghekou was buried by a landslide. We conducted research surveys on both the communities, interviewing survivors and local officials, and observed the destruction/reconstruction, geological, and living conditions. We suggest that protracted educational processes be put into place so that mountain communities possess a knowledge base to consider long-term disaster prevention when building the economy in the fragile and geo-hazardous conditions of the Longmenshan. 9Daily administration times of Canhuang tablet based on a pharmacodynamic/pharmacokinetic model in jaundiced rats显示文摘OBJECTIVE: To study the daily administration times of Canhuang tablet(CHT) for treating jaundice in rats based on a pharmacodynamic/ pharmacokinetic model.METHODS: Rats were modeled by 4% 1-Naphthylisothiocyanate(75 mg/kg, p.o.). After 48 h, CHT was given(p.o.) at 0.75 g/kg once a day, 0.375 g/kg twice a day, and 0.25 g/kg three times a day. Blood was collected from the orbital sinus at different intervals. Levels of liver enzymes and bilirubin were detected using these blood samples. Bile was collected and determined after the first administration of CHT. High-performance liquid chromatography was used to determine the concentration of berberine in bile simultaneously. Time-effect and time-dose curves were then obtained.RESULTS: Compared with rats taking CHT twiceand three times a day, the total amount of bile within 10 h of rats taking CHT once a day were 1.32-and 1.47-fold higher, respectively. There was good consistency between the pharmacokinetics of berberine and the pharmacodynamics of the effect on liver enzymes and bilirubin in vivo. The pharmacokinetic analyses showed that rats administered CHT once daily maintained a higher concentration of berberine in bile for a longer period than rats administered CHT two- and three-times daily.CONCLUSION: In jaundiced rats, taking CHT once a day is better than taking CHT twice or three times a day.
