2Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157

4Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual.Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang 2020Protein & Cell2020,11,4:44
5Isotopic evidence for the moisture origin and composition of surface runoff in the headwaters of the Heihe River basin显示文摘We investigated the moisture origin and contribution of different water sources to surface runoff entering the headwaters of the Heihe River basin on the basis of NECP/NCAR(National Centers for Environmental Prediction/National Center for Atmospheric Research) re-analysis data and variations in the stable hydrogen and oxygen isotope ratios(δ D and δ 18O) of precipitation,spring,river,and melt water. The similar seasonality in precipitation δ 18O at different sites reveals the same moisture origin for water entering the headwaters of the Heihe River basin. The similarity in the seasonality of δ 18O and d-excess for precipitation at Yeniugou and Urumchi,which showed more positive δ 18O and lower d-excess values in summer and more negative δ 18O and higher d-excess values in winter,indicates a dominant effect of westerly air masses in summer and the integrated influence of westerly and polar air masses in winter. Higher d-excess values throughout the year for Yeniugou suggest that in arid inland areas of northwestern China,water is intensively recycled. Temporal changes in δ 18O,δ D,and d-excess reveal distinct contributions of different bodies of water to surface runoff. For example,there were similar trends for δ D,δ 18O,and d-excess of precipitation and river water from June to September,similar δ 18O trends for river and spring water from December to February,and similar trends for precipitation and runoff volumes. However,there were significant differences in δ 18O between melt water and river water in September. 7Caspase-11/4 and gasdermin D-mediated pyroptosis contributes to podocyte injury in mouse diabetic nephropathy显示文摘Diabetic nephropathy(DN)is characterized by sterile inflammation with continuous injury and loss of renal inherent parenchyma cells.Podocyte is an essential early injury target in DN.The injury and loss of podocytes are closely associated with proteinuria,the early symptom of renal injury in DN.However,the exact mechanism for podocyte injury and death in DN remains ambiguous.In this study we investigated whether pyroptosis,a newly discovered cell death pathway was involved in DN.Diabetic mice were generated by high-fat diet/STZ injections.We showed that the expression levels of caspase-11 and cleavage of gasdermin D(GSDMD-N)in podocytes were significantly elevated,accompanied by reduced expression of podocyte makers nephrin and podocin,loss and fusion in podocyte foot processes,increased inflammatory cytokines NF-κB,IL-1β,and IL-18,macrophage infiltration,glomerular matrix expansion and increased urinary albumin to creatinine ratio(UACR).All these changes in diabetic mice were blunted by knockout of caspase-11 or GSDMD.Cultured human and mouse podocytes were treated with high glucose(30 mM),which significantly increased the expression levels of caspase-11 or caspase-4(the homolog of caspase-11 in human),GSDMD-N,NF-κB,IL-1β,and IL-18,and decreased the expression of nephrin and podocin.Either caspase-4 or GSDMD knockdown by siRNA significantly blunted these changes.In summary,our results demonstrate that caspase-11/4 and GSDMD-mediated pyroptosis is activated and involved in podocyte loss under hyperglycemia condition and the development of DN.Qian Cheng Jing Pan Zhuan-li Zhou Fan Yin Hong-yan Xie Pan-pan Chen Jing-yao Li Pei-qing Zheng Li Zhou Wei Zhang Jun Liu Li-min Lu 2021Acta Pharmacologica Sinica2021,42,6:33

9Oral health status in Sichuan Province: findings from the oral health survey of Sichuan, 2015–2016显示文摘To investigate oral health status in the residents of Sichuan Province, southwest China, a cross-sectional study was performed using the latest Oral Health Survey Basic Methods recommended by the World Health Organization. A multistage stratified random cluster-sampling method was used to enroll participants from the following three groups: children aged 3–5 years,adolescents aged 12 years, and people aged 65–74 years. In these three groups, the mean numbers of teeth that were affected by caries were 3.28, 0.86 and 5.13, respectively, resulting in a prevalence of 63.47%, 37.20% and 83.20%, respectively.Relative to the high rate of decayed teeth, the prevalence of fillings was very low in all age groups(0.97%, 7.24% and 5.43%,respectively). In the 12-year-old adolescent group, only 3.61% had good pit and fissure sealing. In addition, the rate of dental fluorosis was 24.80%, and the Community Fluorosis Index value was 0.39. In the elder group, the community periodontal index was2.92. The prevalence in the elderly of having lost at least one tooth was 75.54%. Additionally, 4.44% of these participants were edentulous. The incidence of dental prosthesis was 51.75%, the proportion with a removable partial denture, a fixed denture, full dentures, dental implants and an informal fixed bridge was 21.59%, 11.45%, 4.64%, 0 and 16.67%, respectively. In this study,8.2% of the elderly participants were affected by different types of oral mucosal lesions. Among such lesions, recurrent aphthous ulcers were most prevalent(2.69%) and oral lichen planuses were second(1.6%). The conclusion presented in this survey is that oral diseases, especially dental caries and periodontal disease, are frequent and common in Sichuan province, China. 10Climate changes in East China since the Late-glacial inferred from high-resolution mountain peat humification records显示文摘High-resolution peat humification records were obtained from Dajiuhu of the Shennongjia Mountains and Qianmutian of the Tianmu Mountains to study climate changes in East China.The analyses of pollen,organic matters,TOC,and Rb/Sr indicate a high degree of peat humification and thus strong decomposition of organic matter when climate was dry.Conversely,when climate was humid,the degree of humification is low because peat was preserved in a waterlogged condition.Peat humification from Dajiuhu occurred not only during the Younger Dryas(about 11.4-12.6 cal ka BP),the Blling-Allerd Warm Period(12.6-15.2 cal ka BP),and the Oldest Dryas(about 15.2-16.0 cal ka BP),but also during the early Holocene(about 11.4-9.4 cal ka BP),the 8.2 cal ka BP cold event,and the Holocene Optimum(about 7.0-4.2 cal ka BP).Both peat humification records since nearly 5 ka BP are consistent,showing that mountain peatland has synchronous responses to the East Asia monsoon-induced precipitation.The LOI data confirm the above observation.The monsoon precipitation since nearly 5 ka BP recorded in these two peat profiles can be divided into three phases.During 4.9-3.5 ka BP,precipitation amount was high but fluctuated greatly.During 3.5-0.9 ka BP,precipitation amount was low.During 0.9-0 ka BP,degree of humification reduced gradually,indicating the increase of monsoon precipitation.Contrast to other high-resolution records from East China monsoon region shows that the monsoon precipitation records of the two peat profiles since nearly 16 ka BP are controlled by a common forcing mechanism of summer solar radicalization in the Northern Hemisphere.MA ChunMei ZHU Cheng ZHENG ChaoGui YIN Qian ZHAO ZhiPing 2009Science China Earth Sciences2009,52,1:22
11A study of preoperative methionine-depleting parenteral nutrition plus chemotherapy in gastric cancer patients显示文摘AIM To investigate the interference ofmethionine.free parenteral nutrition plus 5-Fu(-MetTPN+5-Fu)in gastric cancer cell kineticsand the side effects of the regimen.METHODS Fifteen patients with advancedgastric cancer were randomly divided into twogroups,7 patients were given preoperatively aseven-day course of standard parenteralnutrition in combination with a five-day courseof chemotherapy(sTPN+5-Fu),while the other8 patients were given methionine-deprivedparenteral nutrition and 5-Fu(-MetTPN+5-Fu).Cell cycles of gastric cancer and normal mucosawere studied by flow cytometry(FCM).Bloodsamples were taken to measure the serumprotein,methionine(Met)and cysteine(Cys)levels,and liver and kidney functions.RESULTS As compared with the resultsobtained before the treatment,the percentage ofG0/G1 tumor cells increased and that of S phasedecreased in the-MetTPN+5-Fu group,while thecontrary was observed in the sTPN+5-Fu group.Except that the ALT,AST and AKP levels wereslightly increased in a few cases receiving-MetTPN+5-Fu,all the other biochemicalparameters were within normal limits.Serum Cys level decreased slightly after the treatmentin both groups.Serum Met level of patientsreceiving sTPN+5-Fu was somewhat higher aftertreatment than that before treatment;however,no significant change occurred in the -MetTPN+5-Fu group,nor operative complications in bothgroups.CONCLUSION -MetTPN+5-Fu exerted asuppressive effect on cancer cell proliferation,probably through a double mechanism ofcreating a state of'Met starvation'adverse tothe tumor cell cycle,and by allowing 5-Fu to killspecifically cells in S phase.Preoperative short-term administration of -MetTPN+5-Fu had littleundesirable effect on host metabolism.Cao WX Cheng QM Fei XF Li SF Yin HR Lin YZ 2000World Journal of Gastroenterology2000,6,2:22

13Protective effects of Yindanxinnaotong capsule in a rat model of myocardial ischemia/reperfusion injury显示文摘OBJECTIVE: To investigate the effects of Yindanxinnaotong capsule(YDXNTC) and main components compatibility and ratios on myocardium against ischemia/reperfusion injury and the effect's underlying mechanism.METHODS: Myocardial ischemia/reperfusion injury(MIRI) was induced by ischemia for 30 min and reperfusion for 30 min. Electrocardiogram data and coronary flow were recorded, and superoxide dismutase(SOD), malondialdehyde(MDA), lactate dehydrogenase, creatine kinase-MB, cardiac troponin T and I(cT nT, cT n I) and interleukin-1β, interleukin-8,interleukin-18(IL-1β, IL-8, IL-18) in myocardium were measured. Hypoxia/reoxygenation and hydrogen peroxide(H2O2) injury were induced by hypoxia for 3 h/reoxygenation for 2 h, and 100 μM H2O2 for 1 h, respectively, in vitro rat myocardial cells(H9c2). Cell viability, SOD, MDA, cT nT and inflamma-tory factors(IL-1β, IL-8 and IL-18) were determined,and Toll-like receptor 4(TLR-4) expression was measured by western blotting.RESULTS: In the isolated heart experiment, elevated heart function, coronary flow and SOD levels,and decreased MDA levels and inflammatory factors were noted in the YDXNTC, main components and main components compatibility groups. Ventricular tachycardia/ventricular fibrillation occurrence decreased in the ginkgo biloba extract(GBE),and GBE and salvia miltiorrhiza ethanol extract compatibility(SM-E, GSEC) groups. Lactic dehydrogenase levels decreased in the YDXNTC and aqueous extract of salvia miltiorrhiza(SM-H) groups. Creatine kinase-MB decreased with GBE, SM-E, SM-H and GSEC treatment, and cT n I and cT nT levels decreased with GSEC. In the in vitro cell study,YDXNTC and main components ratios improved cell viability and SOD levels, and suppressed MDA,cT nT and inflammatory factors. TLR-4 expression was down-regulated.CONCLUSION: YDXNTC and main components compatibility showed protective effects on MIRI in this rat model and in vitro study. 14Identification of long-range transport pathways and potential sources of PM2.5 and PM10 in Beijing from 2014 to 2015显示文摘Trajectory clustering, potential source contribution function(PSCF) and concentration-weighted trajectory(CWT) methods were applied to investigate the transport pathways and identify potential sources of PM_(2.5)and PM_(10) in different seasons from June 2014 to May 2015 in Beijing.The cluster analyses showed that Beijing was affected by trajectories from the south and southeast in summer and autumn. In winter and spring, Beijing was not only affected by the trajectories from the south and southeast, but was also affected by trajectories from the north and northwest. In addition, the analyses of the pressure profile of backward trajectories showed that backward trajectories, which have important influence on Beijing, were mainly distributed above 970 h Pa in summer and autumn and below 950 h Pa in spring and winter. This indicates that PM_(2.5)and PM_(10) were strongly affected by the near surface air masses in summer and autumn and by high altitude air masses in winter and spring. Results of PSCF and CWT analyses showed that the largest potential source areas were identified in spring, followed by winter and autumn,then summer. In addition, potential source regions of PM_(10) were similar to those of PM_(2.5). There were a clear seasonal and spatial variation of the potential source areas of Beijing and the airflow in the horizontal and vertical directions. 15Effectiveness of 5-flurouracil-based neoadjuvant chemotherapy in locally-advanced gastric/gastroesophageal cancer:A meta-analysis显示文摘AIM:To investigate the effectiveness of 5-flurouracilbased neoadjuvant chemotherapy(NAC) for gastroesophageal and gastric cancer by meta-analysis.METHODS:MEDLINE and manual searches were performed to identify all published randomized controlled trials(RCTs) investigating the efficacy of the flurouracilbased NAC for gastroesophageal and gastric cancer,and RCTs of NAC for advanced gastroesophageal and gastric cancer vs no therapy before surgery.Studies that included patients with metastases at enrollment were excluded.Primary endpoint was the odds ratio(OR) for improving overall survival rate of patients with gastroesophageal and gastric cancer.Secondary endpoints were the OR of efficiency for down-staging tumor and increasing R0 resection in patients with gas-troesophageal and gastric cancer.Safety analyses were also performed.The OR was the principal measurement of effect,which was calculated as the treatment group(NAC plus surgery) vs control group(surgery alone) and was presented as a point estimate with 95% confidence intervals(CI).All calculations and statistical tests were performed using RevMan 5.1 software.RESULTS:Seven RCTs were included for the analysis.A total of 1249 patients with advanced gastroesophageal and gastric cancer enrolled in the seven trials were divided into treatment group(n = 620) and control group(n = 629).The quality scores of the RCTs were assessed according to the method of Jadad.The RCT quality scores ranged from 2 to 7(5-point scale),with a mean of 3.75.The median follow-up time in these studies was over 3 years.The meta-analysis showed that NAC improved the overall survival rate(OR 1.40,95%CI 1.11-1.76;P = 0.005),which was statistically significant.The 3-year progression-free survival rate was significantly higher in treatment group than in control group(37.7% vs 27.3%)(OR 1.62,95%CI 1.21-2.15;P = 0.001).The tumor down-stage rate was higher in treatment group than in control group(55.76% vs 41.38%)(OR 1.77,95%CI 1.27-2.49;P = 0.0009) and the R0 resection rate of the gastroesophageal and gastric cancer was higher in treatment group than in control group(75.11% vs 68.56%)(OR 1.38,95%CI 1.03-1.85;P = 0.03),with significant differences.No obvious safety concerns about mortality and complications were raised in these trials.There were no statistically significant differences in perioperative mortality(5.08% vs 4.86%)(OR 1.05,95%CI 0.57-1.94;P = 0.87 fixed-effect model) and in the complication rate between the two groups(13.25% vs 9.66%)(OR 1.40,95%CI 0.91-2.14;P = 0.12 fixed-effect model).Trials showed that patients from Western countries favored NAC compared with those from Asian countries(OR 1.40,95%CI 1.07-1.83).Monotherapy was inferior tomultiple chemotherapy(OR 1.40,95%CI 1.07-1.83).Intravenous administration of NAC was more advantageous than oral route(OR 1.41,95%CI 1.09-1.81).CONCLUSION:Flurouracil-based NAC can safely improve overall survival rate of patients with gastroesophageal/gastric cancer.Additionally,NAC can down the tumor stage and improve R0 resection.Lei Ge Hai-Jiang Wang Dong Yin Cheng Lei Jin-Feng Zhu Xiao-Hui Cai Guo-Qing Zhang 2012World Journal of Gastroenterology2012,18,48:19
16Immediate Effects of Hegu Needling on Adhesive Scapulohumeral Periarthritis显示文摘Objective:To observe the immediate effects of Hegu needling at Ashi points in patients with adhesive scapulohumeral periarthritis (SP). Methods:A total of 60 SP cases were randomized into a treatment group and a control group, 30 in each group. Cases in the treatment group were treated with Hegu needling at Ashi points and normal needling at other local points. Cases in the control group were treated with normal needling at local points alone. The therapeutic efficacy was then evaluated and analyzed using short-form of McGill pain questionnaire (SF-MPQ) and Japanese Orthopaedic Association (JOA) shoulder scoring system. Results:Hegu needling at Ashi points can substantially alleviate pain in patients with adhesive SP, showing a statistically significant difference when compared with the control group (P<0.01 or P<0.05). In addition, cases in the treatment group obtained better effects in abductor muscle strength, endurance, arm raising, internal rotation, external rotation, activities of daily living (ADL) and joint stability than those in the control group (P<0.01 or P<0.05). 18On controllability and stabilizability of probabilistic Boolean control networks显示文摘The controllability of probabilistic Boolean control networks(PBCNs)is first considered.Using the input-state incidence matrices of all models,we propose a reachability matrix to characterize the joint reachability.Then we prove that the joint reachability and the controllability of PBCNs are equivalent,which leads to a necessary and sufcient condition of the controllability.Then,the result of controllability is used to investigate the stability of probabilistic Boolean networks(PBNs)and the stabilization of PBCNs.A necessary and sufcient condition for the stability of PBNs is obtained first.By introducing the control-fixed point of Boolean control networks(BCNs),the stability condition has finally been developed into a necessary and sufcient condition of the stabilization of PBCNs.Both necessary and sufcient conditions for controllability and stabilizability are based on reachability matrix,which are easily computable.Hence the two necessary and sufcient conditions are straightforward verifiable.Numerical examples are provided from case to case to demonstrate the corresponding theoretical results.ZHAO Yin CHENG DaiZhan 2014Science China(Information Sciences)2014,57,1:17

20Long-term thermal regimes of the Qinghai-Tibet Railway embankments in plateau permafrost regions显示文摘Ten years of ground temperature data(2003–2013) indicate that the long-term thermal regimes within embankments of the Qinghai-Tibet Railway(QTR) vary significantly with different embankment structures. Obvious asymmetries exist in the ground temperature fields within the traditional embankment(TE) and the crushed-rock basement embankment(CRBE). Measurements indicate that the TE and CRBE are not conducive to maintaining thermal stability. In contrast, the ground temperature fields of both the crushed-rock sloped embankment(CRSE) and the U-shaped crushed-rock embankment(UCRE) were symmetrical. However, the UCRE gave better thermal stability than the CRSE because slow warming of deep permafrost was observed under the CRSE. Therefore, the UCRE has the best long-term effect of decreasing ground temperature and improving the symmetry of the temperature field. More generally, it is concluded that construction using the cooling-roadbed principle meets the design requirements for long-term stability of the railway and for train transport speeds of 100 km h?1.
