| 4 | A Comprehensive Overview of Skeletal Phenotypes Associated with Alterations in Wnt/β-catenin Signaling in Humans and Mice显示文摘The Wnt signaling pathway plays key roles in differentiation and development and alterations in this signaling pathway are causally associated with numerous human diseases.While several laboratories were examining roles for Wnt signaling in skeletal development during the 1990s,interest in the pathway rose exponentially when three key papers were published in 2001-2002.One report found that loss of the Wnt co-receptor,Low-density lipoprotein related protein-5(LRP5),was the underlying genetic cause of the syndrome Osteoporosis pseudoglioma(OPPG).OPPG is characterized by early-onset osteoporosis causing increased susceptibility to debilitating fractures.Shortly thereafter,two groups reported that individuals carrying a specific point mutation in LRP5(G171V)develop high-bone mass.Subsequent to this,the causative mechanisms for these observations heightened the need to understand the mechanisms by which Wnt signaling controlled bone development and homeostasis and encouraged significant investment from biotechnology and pharmaceutical companies to develop methods to activate Wnt signaling to increase bone mass to treat osteoporosis and other bone disease.In this review,we will briefly summarize the cellular mechanisms underlying Wnt signaling and discuss the observations related to OPPG and the high-bone mass disorders that heightened the appreciation of the role of Wnt signaling in normal bone development and homeostasis.We will then present a comprehensive overview of the core components of the pathway with an emphasis on the phenotypes associated with mice carrying genetically engineered mutations in these genes and clinical observations that further link alterations in the pathway to changes in human bone. | Kevin A.Maupin Casey J.Droscha Bart O.Williams | 2013 | Bone Research2013,1,1: | 18 |
| 5 | No association between cyclooxygenase-2 and uridine diphosphate glucuronosyltransferase 1A6 genetic polymorphisms and colon cancer risk显示文摘AIM:To investigate the association of variations in the cyclooxygenase-2(COX2) and uridine diphosphate glucuronosyltransferase 1A6(UGT1A6) genes and non-steroidal anti-inflammatory drugs(NSAIDs) use with risk of colon cancer.METHODS:NSAIDs,which are known to reduce the risk of colon cancer,act directly on COX2 and reduce its activity.Epidemiological studies have associated variations in the COX2 gene with colon cancer risk,but others were unable to replicate this finding.Similarly,enzymes in the UGT1A6 gene have been demonstrated to modify the therapeutic effect of NSAIDs on colon adenomas.Polymorphisms in the UGT1A6 gene have been statistically shown to interact with NSAID intake to influence risk of developing colon adenomas,but not colon cancer.Here we examined the association of tagging single nucleotide polymorphisms(SNPs) in the COX2 and UGT1A6 genes,and their interaction with NSAID consumption,on risk of colon cancer in a population of 422 colon cancer cases and 481 population controls.RESULTS:No SNP in either gene was individually statistically significantly associated with colon cancer,nor did they statistically significantly change the protective effect of NSAID consumption in our sample.Like others,we were unable to replicate the association of variants in the COX2 gene with colon cancer risk(P > 0.05),and we did not observe that these variants modify the protective effect of NSAIDs(P > 0.05).We were able to confirm the lack of association of variants in UGT1A6 with colon cancer risk,although further studies will have to be conducted to confirm the association of these variants with colon adenomas.CONCLUSION:Our study does not support a role of COX2 and UGT1A6 genetic variations in the development of colon cancer. | Cheryl L Thompson Sarah J Plummer Alona Merkulova Iona Cheng Thomas C Tucker Graham Casey Li Li | 2009 | World Journal of Gastroenterology2009,15,18: | 11 |
| 6 | Pancreatic injury in patients with septic shock: A literature review显示文摘Sepsis and septic shock are life threatening condition associated with high mortality rate in critically-ill patients. This high mortality is mainly related to the inadequacy between oxygen delivery and cellular demand leading to the onset of multiorgan dysfunction. Whether this multiorgan failure affect the pancreas is not fully investigated. In fact, pancreatic injury may occur because of ischemia, overwhelming inflammatory response, oxidative stress, cellular apoptosis and/or metabolic derangement. Increased serum amylase and/or lipase levels are common in patients with septic shock. However, imaging test rarely reveal significant pancreatic damage. Whether pancreatic dysfunction does affect the prognosis of patients with septic shock or not is still a matter of debate. In fact, only few studies with limited sample size assessed the clinical relevance of the pancreatic injury in this group of patients. In this review, we aimed to describe the epidemiology and the physiopathology of pancreatic injury in septic shock patients, to clarify whether it requires specific management and to assess its prognostic value. Our main finding is that pancreatic injury does not significantly affect the outcome in septic shock patients. | 7 | Water vapor δD dynamics over China derived from SCIAMACHY satellite measurements显示文摘This study investigates water vapor isotopic patterns and controls over China using high-quality water vapor δD data retrieved from the Scanning Imaging Absorption Spectrometer for Atmospheric Chartography(SCIAMACHY) observations.The results show that water vapor δD values on both annual and seasonal time-scales broadly exhibit a continental effect,with values largely decreasing northwestward from coastal lowlands to high-elevation mountainous regions.However,region-specific analysis reveals spatially distinct patterns of water vapor δD between seasons.In the monsoon domain(e.g.,China south of 35°N),depletion in D in the summer and fall seasons is closely tied to monsoon moisture sources(the Indian and Pacific oceans) and subsequent amount effect,but higher δD values in winter and spring are a result of isotopically-enriched continental-sourced moisture proceeded by less rainout.In contrast,farther inland in China(non-monsoon domain),moisture is derived overwhelmingly from the dry continental air masses and local evaporation,and δD values are largely controlled by the temperature effect,exhibiting a seasonality with isotopically enriched summer and depleted winter/spring.The observation that the spatial pattern of water vapor δD is the opposite to that of precipitation δD in the summer season also suggests that partial evaporation of falling raindrops is a key driver of water vapor isotope in the non-monsoon domain.This study highlights the importance of non-Rayleigh factors in governing water vapor isotope,and provides constraints on precipitation isotope interpretation and modern isotope hydrological processes over China. | LIU ZhongFang Kei YOSHIMURA Casey D.KENNEDY WANG XinHui PANG ShuoGuang | 2014 | Science China Earth Sciences2014,57,4: | 7 |
| 8 | Fibronectin: Functional character and role in alcoholic liver disease显示文摘Fibronectins are adhesive glycoproteins that can be found in tissue matrices and circulating in various fluids of the body. The variable composition of fibronectin molecules facilitates a diversity of interactions with cell surface receptors that suggest a role for these proteins beyond the structural considerations of the extracellular matrix. These interactions implicate fibronectin in the regulation of mechanisms that also determine cell behavior and activity. The two major forms, plasma fibronectin (pFn) and cellular fibronectin (cFn), exist as balanced amounts under normal physiological conditions. However, during injury and/or disease, tissue and circulating levels of cFn become disproportionately elevated. The accumulating cFn, in addition to being a consequence of prolonged tissue damage, may in factstimulate cellular events that promote further damage. In this review, we summarize what is known regarding such interactions between fibronectin and cells that may influence the biological response to injury. We elaborate on the effects of cFn in the liver, specifically under a condition of chronic alcohol-induced injury. Studies have revealed that chronic alcohol consumption stimulates excess production of cFn by sinusoidal endothelial cells and hepatic stellate cells while impairing its clearance by other cell types resulting in the build up of this glycoprotein throughout the liver and its consequent increased availability to influence cellular activity that could promote the development of alcoholic liver disease. We describe recent findings by our laboratory that support a plausible role for cFn in the promotion of liver injury under a condition of chronic alcohol abuse and the implications of cFn stimulation on the pathogenesis of alcoholic liver disease.
