| 1 | Bone morphogenetic protein-2 gene controls tooth root development in coordination with formation of the periodontium显示文摘Formation of the periodontium begins following onset of tooth-root formation in a coordinated manner after birth. Dental follicle progenitor cells are thought to form the cementum, alveolar bone and Sharpey's fibers of the periodontal ligament (PDL). However, little is known about the regulatory morphogens that control differentiation and function of these progenitor cells, as well as the progenitor cells involved in crown and root formation. We investigated the role of bone morphogenetic protein-2 (Bmp2) in these processes by the conditional removal of the Bmp2 gene using the Sp7-Cre-EGFP mouse model. Sp7-Cre-EGFP first becomes active at E18 in the first molar, with robust Cre activity at postnatal day 0 (P0), followed by Cre activity in the second molar, which occurs after P0. There is robust Cre activity in the periodontium and third molars by 2 weeks of age. When the Bmp2 gene is removed from Sp7+ (Osterix+) cells, major defects are noted in root, cellular cementum and periodontium formation. First, there are major cell autonomous defects in root-odontoblast terminal differentiation. Second, there are major alterations in formation of the PDLs and cellular cementum, correlated with decreased nuclear factor IC (Nfic), periostin and α-SMA+ cells. Third, there is a failure to produce vascular endothelial growth factor A (VEGF-A) in the periodontium and the pulp leading to decreased formation of the microvascular and associated candidate stem cells in the Bmp2-cKOSp7-Cre-EGFP . Fourth, ameloblast function and enamel formation are indirectly altered in the Bmp2-cKOSp7-Cre-EGFP . | 4 | A review of UHMWPE wear-induced osteolysis:the role for early detection of the immune response显示文摘In a world where increasing joint arthroplasties are being performed on increasingly younger patients,osteolysis as the leading cause of failure after total joint arthroplasty(TJA) has gained considerable attention.Ultra-high molecular weight polyethylene wear-induced osteolysis is the process by which prosthetic debris mechanically released from the surface of prosthetic joints induces an immune response that favors bone catabolism, resulting in loosening of prostheses with eventual failure or fracture. The immune response initiated is innate in that it is nonspecific and self-propagating, with monocytic cells and osteoclasts being the main effectors. To date, detecting disease early enough to implement effective intervention without unwanted systemic side effects has been a major barrier. These barriers can be overcome using newer in vivo imaging techniques and modules linked with fluorescence and/or chemotherapies. We discuss the pathogenesis of osteolysis, and provide discussion of the challenges with imaging and therapeutics. | 6 | Age and menopausal status are important factors influencing the serum human epididymis secretory protein 4 level:a prospective cross-sectional study in healthy Chinese people显示文摘Background::Human epididymis secretory protein 4(HE4)is a new ovarian cancer biomarker.The factors influencing HE4 levels are not clear,and the reference data in China are limited.Here,we aim to evaluate the effects of menopause and age on HE4 levels and to provide a possible reference value for HE4 in healthy Chinese people.Methods::A total of 2493 healthy females aged 40 years or older were recruited from March 2013 to March 2017 with the cooperation of four medical institutions across Beijing,China.The serum levels of HE4 and cancer antigen 125(CA125)were measured by enzyme-linked immunosorbent assay.The Wilcoxon rank-sum test of variance and a stratified analysis were used to analyze the relationships among age,menopausal status,and levels of HE4 or CA125.Confidence intervals(5%-95%)were determined for reference ranges in different populations.Results::There was a statistically significant difference in median HE4 levels between the post-menopausal(n=2168)and premenopausal groups(n=325)(36.46 vs.24.04 pmol/L,Z=-14.41,P<0.001).HE4 increased significantly with age in the post-menopausal groups(H=408.18,P<0.001)but not in the pre-menopausal subjects(Z=-0.43,P=0.67).The upper 95th percentile of HE4 levels were 44.63 pmol/L for pre-menopausal women,78.17 pmol/L for post-menopausal women,and 73.3 pmol/L for all women.In the post-menopausal population,the HE4 reference ranges were 13.15 to 47.31,14.31 to 58.04,17.06 to 73.51,24.50 to 115.25,and 35.71 to 212.37 pmol/L for different age groups from forty divided by decade.The CA125 level was affected mainly by menopausal status and not age.Conclusions::Menopausal status and age were both important factors influencing the level of HE4,and age affected HE4 levels mainly in post-menopausal women.The HE4 level was higher in the post-menopausal population than in the pre-menopausal population and increased with age. | Hong-Yan Cheng Lin Zeng Xue Ye Rui-Qiong M a Zhi-Jian Tang Hong-Ling Chu Yi-M ing Zhao Li-Rong Zhu Yu-Nong Gao Xiao-Hong Chang Heng Cui | 2020 | Chinese Medical Journal2020,,11: | 6 |

| 12 | Sand dunes as potential sources of dust in northern China显示文摘While saltation bombardment of sand grains on a fine substrate can produce considerable dust, the well-sorted nature of sand dunes tends to preclude them from consideration as major dust sources. Recent research, however, has revealed that sand dunes can, in some cases, be large sources of dust. We used the PI-SWERL(Portable In-Situ Wind Erosion Laboratory) to measure in the field the potential of sand dunes and other desert landforms to emit particulate matter <10 μm(PM-10) dust in the Tengger, Ulan Buh, and Mu Us deserts of northern China. Combined with high resolution particle size measurements of the dune sand, an assessment of sand dunes as a dust source can be made. Large active transverse dunes tend to contain little to no stored PM-10, yet they produce a low dust flux. Coppice dunes stabilized by vegetation contain appreciable PM-10 and have very high dust emission potential. There is a positive correlation between the amount of PM-10 stored in a dune and its potential dust flux. Saltation liberates loose fines stored in dunes, making them very efficient dust emitters compared to landforms such as dry lake beds and washes where dust particles are unavailable for aeolian transport due to protective crusts or sediment cohesion. In cases where large dunes do not store PM-10 yet emit dust when active, two hypotheses can be considered:(1) iron-oxide grain coatings are removed during saltation, creating dust, and(2) sand grains collide during saltation, abrading grains to create dust. Observations reveal that iron oxide coatings are present on some dune sands. PI-SWERL data suggests that low dust fluxes from dunes containing no stored dust may represent an estimate for the amount of PM-10 dust produced by removal of iron oxide coatings. These results are similar to results from dunes in the United States.
