1Outcomes in refractory diffuse large B-cell lymphoma:results fromamulticenter real-world study in China显示文摘Background:Diffuse large B-cell lymphoma(DLBCL)patients refractory to rituximab-based immunochemotherapy have a dismal prognosis.However,the definition of refractory DLBCL remains inconsistent and no large cohort study data is available from Asian countries.To validate the definition and outcomes of refractory DLBCL in China,we conducted a multicenter,retrospective cohort study.Methods:The REtrospective AnaLysis of Treatment REspoNse of refractory DLBCL(REAL-TREND)study was performed using real-world data from 8 centers in China.DLBCL patients with curative intent were included in the REAL-TREND dataset.Overall survival(OS)was estimated using the Kaplan-Meier method and compared by the log-rank test.Due to heterogeneity in response rates among different centers,the response rates of refractory patients were pooled using random-effect models.Multivariate survival analysis was performed using the Cox regression model.Results:A total of 2778 DLBCL patients diagnosed between January,2010 and December,2015 were enrolled to this study.After validating previous definitions,the SCHOLAR-1 study was most suitable to define refractory DLBCL.The estimated 5-year cumulative incidence of refractory patients was 20%(95% confidence Interval[CI]=18%-22%).After the determination of refractory disease,overall response rate and complete remission rate were 30%(95%CI=22%-38%)and 9%(95%CI=4%-15%),respectively.Patients with either no response to immunochemotherapy or relapse within 12 months after stem-cell transplantation had inferior survival with a median OS of 5.9 months(95%CI=5.5-7.1 months)and 2-year OS rate of 16%(95%CI=12%-20%).International prognostic index score 4-5(hazard ratio[HR]=2.22;95%CI=1.47-3.35),central nervous systemrelapse(HR=1.43;95%CI=1.04-1.97),and best response status(HR=2.68;95%CI=1.42-5.03 for partial remission.HR=5.97,95%CI=3.21-11.11 for stable disease/progressive disease)were independent unfavorable prognostic factors.Conclusions:This is the first large-scale Asian cohort study focusing on outcomes of refractory DLBCL.The definition of the SCHOLAR-1 study identifies patients with homogenously inferior survival,thus is appropriate to select refractory DLBCL.Due to poor clinical outcomes in the rituximab era,patients with refractory DLBCL may be potential candidates for novel treatment modalities.Shuo Wang Li Wang Jianda Hu Wenbin Qian Xi Zhang Yu Hu Qi Zhu Bobin Chen Depei Wu Chung-Chou H.Chang Pengpeng Xu Xiaoyun Zheng Juying Wei Yao Liu Guohui Cui Yong Tang Yan Ma Haiwen Huang Hongmei Yi Weili Zhao 2021Cancer Communications2021,41,3:10

3Study on the dynamic characteristics of flow over building cluster at high Reynolds number by large eddy simulation显示文摘In this paper,the dynamic characteristics of building clusters are simulated by large eddy simulation at high Reynolds number for both homogeneous and heterogeneous building clusters.To save the computational cost a channel-like flow model is applied to the urban canopy with free slip condition at the upper boundary.The results show that the domain height is an important parameter for correct evaluation of the dynamic characteristics.The domain height must be greater than 8h(h is the average building height)in order to obtain correct roughness height while displacement height and roughness sublayer are less sensitive to the domain height.The Reynolds number effects on the dynamic characteristics and flow patterns are investigated.The turbulence intensity is stronger inside building cluster at high Reynolds number while turbulence intensity is almost unchanged with Reynolds number above the building cluster.Roughness height increases monotonously with Reynolds number by 20%from Re*=103 to Re*=105 but displacement height is almost unchanged.Within the canopy layer of heterogeneous building clusters,flow structures vary between buildings and turbulence is more active at high Reynolds number.WANG BoBin WANG ZhiShi CUI GuiXiang ZHANG ZhaoShun 2014Science China(Physics,Mechanics & Astronomy)2014,57,6:5
4AOD estimation in WSN localization system with synthetic aperture technique显示文摘Angel of departure(AOD) based distributed localization scheme in wireless sensor networks allows the single antenna sensor node to estimate its own AOD information by an equivalent antenna array for further position calculation.However,the condition that the number of antennas at the multi-antenna agent/anchor node should be larger than the number of multipath signal components(MPCs) constrains its actual application.In this paper,we provide a more practical approach to achieve AOD information estimation under multipath environment.Given that the sensor node can not identify the line of sight(LOS) signal component due to the insufficient degree of freedom,which is determined by the above limiting condition,we introduce a synthetic aperture technique with the aid of a fast moving multi-antenna agent/anchor node.This operation greatly enlarges the degree of freedom of the equivalent antenna array constructed at sensor node end but without requiring other extra physical antennas equipped at agent/anchor node or sensor node end,which means that it relaxes the limiting condition.Therefore,the AOD information of LOS signal component can be estimated and extracted successfully in complex multipath environment.Theoretical analysis proves the effectiveness of our proposed synthetic aperture technique and numerical simulations show great performance improvement.WANG WenJie YAO BoBin YIN QinYe 2012Science China(Information Sciences)2012,55,10:4
5Exosomal PD-L1 induces osteogenic differentiation and promotes fracture healing by acting as an immunosuppressant显示文摘A moderate inflammatory response at the early stages of fracture healing is necessary for callus formation.Over-active and continuous inflammation,however,impairs fracture healing and leads to excessive tissue damage.Adequate fracture healing could be promoted through suppression of local over-active immune cells in the fracture site.In the present study,we achieved an enriched concentration of PD-L1 from exosomes(Exos)of a genetically engineered Human Umbilical Vein Endothelial Cell(HUVECs),and demonstrated that exosomes overexpressing PD-L1 specifically bind to PD-1 on the T cell surface,suppressing the activation of T cells.Furthermore,exosomal PD-L1 induced Mesenchymal Stem Cells(MSCs)towards osteogenic differentiation when pre-cultured with T cells.Moreover,embedding of Exos into an injectable hydrogel allowed Exos delivery to the surrounding microenvironment in a time-released manner.Additionally,exosomal PD-L1,embedded in a hydrogel,markedly promoted callus formation and fracture healing in a murine model at the early over-active inflammation phase.Importantly,our results suggested that activation of T cells in the peripheral lymphatic tissues was inhibited after local administration of PD-L1-enriched Exos to the fracture sites,while T cells in distant immune organs such as the spleen were not affected.In summary,this study provides the first example of using PD-L1-enriched Exos for bone fracture repair,and highlights the potential of Hydrogel@Exos systems for bone fracture therapy through immune inhibitory effects.Ze Lin Yuan Xiong Weilin Meng Yiqiang Hu Lili Chen Lang Chen Hang Xue Adriana CPanayi Wu Zhou Yun Sun Faqi Cao Guodong Liu Liangcong Hu Chenchen Yan Xudong Xie Chuanchuan Lin Kaiyong Cai Qian Feng Bobin Mi Guohui Liu 2022Bioactive Materials2022,7,7:4
6Progress in octahedral spherical hohlraum study显示文摘In this paper,we give a review of our theoretical and experimental progress in octahedral spherical hohlraum study.From our theoretical study,the octahedral spherical hohlraums with 6 Laser Entrance Holes(LEHs)of octahedral symmetry have robust high symmetry during the capsule implosion at hohlraum-to-capsule radius ratio larger than 3.7.In addition,the octahedral spherical hohlraums also have potential superiority on low backscattering without supplementary technology.We studied the laser arrangement and constraints of the octahedral spherical hohlraums,and gave a design on the laser arrangement for ignition octahedral hohlraums.As a result,the injection angle of laser beams of 50°-60°was proposed as the optimum candidate range for the octahedral spherical hohlraums.We proposed a novel octahedral spherical hohlraum with cylindrical LEHs and LEH shields,in order to increase the laser coupling efficiency and improve the capsule symmetry and to mitigate the influence of the wall blowoff on laser transport.We studied on the sensitivity of the octahedral spherical hohlraums to random errors and compared the sensitivity among the octahedral spherical hohlraums,the rugby hohlraums and the cylindrical hohlraums,and the results show that the octahedral spherical hohlraums are robust to these random errors while the cylindrical hohlraums are the most sensitive.Up till to now,we have carried out three experiments on the spherical hohlraum with 2 LEHs on Shenguang(SG)laser facilities,including demonstration of improving laser transport by using the cylindrical LEHs in the spherical hohlraums,spherical hohlraum energetics on the SGIII prototype laser facility,and comparisons of laser plasma instabilities between the spherical hohlraums and the cylindrical hohlraums on the SGIII laser facility.Ke Lan Jie Liu Zhichao Li Xufei Xie Wenyi Huo Yaohua Chen Guoli Ren Chunyang Zheng Dong Yang Sanwei Li Zhiwen Yang Liang Guo Shu Li Mingyu Zhang Xiaoying Han Chuanlei Zhai Lifei Hou Yukun Li Keli Deng Zheng Yuan Xiayu Zhan Feng Wang Guanghui Yuan Haijun Zhang Bobin Jiang Lizhen Huang Wei Zhang Kai Du Runchang Zhao Ping Li Wei Wang Jingqin Su Xuewei Deng Dongxia Hu Wei Zhou Huaiting Jia Yongkun Ding Wanguo Zheng Xiantu He 2016Matter and Radiation at Extremes2016,1,1:2
7Advances and perspective on animal models and hydrogel biomaterials for diabetic wound healing显示文摘Diabetic wounds are a common complication in diabetes patients.Due to peripheral nerve damage and vascular dysfunction,diabetic wounds are prone to progress to local ulcers,wound gangrene and even to require amputation,bringing huge psychological and economic burdens to patients.However,the current treatment methods for diabetic wounds mainly include wound accessories,negative pressure drainage,skin grafting and surgery;there is still no ideal treatment to promote diabetic wound healing at present.Appropriate animal models can simulate the physiological mechanism of diabetic wounds,providing a basis for translational research in treating diabetic wound healing.Although there are no animal models that can fully mimic the pathophysiological mechanisms of diabetic wounds in humans,it is vital to explore animal simulation models used in basic research and preclinical studies of diabetic wounds.In addition,hydrogel materials are regarded as a promising treatment for diabetic wounds because of their good antimicrobial activity,biocompatibility,biodegradation and appropriate mechanical properties.Herein,we review and discuss the different animal models used to investigate the pathological mechanisms of diabetic wounds.We further discuss the promising future application of hydrogel biomaterials in diabetic wound healing.Yiqiang Hu Yuan Xiong Ranyang Tao Hang Xue Lang Chen Ze Lin Adriana C.Panayi Bobin Mi Guohui Liu 2022Biomaterials Translational2022,3,3:2
