| 1 | Identify lymphatic metastasis-associated genes in mouse hepatocarcinoma cell lines using gene chip显示文摘AIM: In order to obtain lymphogenous metastasisassociated genes, we compared the transcriptional profiles of mouse hepatocarcinoma cell lines Hca-F with highly lymphatic metastasis potential and Hca-P with low lymphatic metastasis potential.METHODS: Total RNA was isolated from Hca-F and Hca-P cells and synthesized into double-stranded cDNA. In vitro transcription double-stranded cDNA was labeled with biotin (i.e. biotin-labeled cRNA, used as the probe). The cRNA probes hybridized with Affymetrix GeneChip() MOE430A (containing 22 690 transcripts, including 14 500 known mouse genes and 4 371 ESTs) respectively and the signals were scanned by the GeneArray Scanner. The results were then analyzed by bioinformatics.RESULTS: Out of the 14 500 known genes investigated,110 (0.8%) were up regulated at least 23 fold. Among the total 4 371 ESTs, 17 ESTs (0.4%) (data were not presented) were up regulated at least 23 fold. According to the Gene Ontology and TreeView analysis, the 110genes were further classified into two groups: differential biological process profile and molecular function profile.CONCLUSION: Using high-throughput gene chip method,a large number of genes and their cellular functions about angiogenesis, cell adhesion, signal transduction, cell motility, transport, microtubule-based process, cytoskeleton organization and biogenesis, cell cycle, transcription,chaperone activity, motor activity, protein kinase activity,receptor binding and protein binding might be involved in the process of lymphatic metastasis and deserve to be used as potential candidates for further investigation.Cyclin D1, Fosl1, Hsp47, EGFR and AR, and Cav-1 are selected as the possible candidate genes of the metastatic phenotype, which need to be validated in later experiments.ESTs (data were not presented) might indicate novel genes associated with lymphatic metastasis. | 2 | Competitive binding of postsynaptic density 95 and Ca^(2+)-calmodulin dependent protein kinase II to N-methyl-D-aspartate receptor subunit 2B in rat brain显示文摘AIM: To investigate the interactions among postsynaptic density 95 (PSD-95), Ca2+-calmodulin dependent protein kinase IIα (CaMKIIα), and N-methyl-D-aspartate receptor subunit 2B (NR2B) during ischemia and reperfusion in hippocampus of rats. METHODS: Brain ischemia was induced by four-vessel occlusion procedure in rats. Immu- noprecipitation and immunoblotting were performed to study the interactions and phosphorylation of proteins. The association-dissociation of PSD-95 and CaMKIIα to and from N-methyl-D-aspartate (NMDA) receptor induced by ischemia and reperfusion and the effects of 1-[N,O-bis-(5-isoquinolinesulfonyl)-N-methyl-L-tyrosyl]-4-phenyl-pip- erazine (KN-62, a selective inhibitor of CaMKII) on these protein interactions were investigated. Coimmuno- precipitation and immunoblotting were performed for the studies of interactions among proteins. RESULTS: The alternations of the binding level of PSD-95 and CaMKIIα to NR2B during ischemia and reperfusion demonstrated the negative correlation to each other. Pre-administration of KN62 through both cerebral ventricles inhibited the 10 min ischemia-induced increase of the binding of PSD-95 to NR2B and, on the contrary, promoted the binding of CaMKIIα to NR2B. | 3 | Construction and selection of subtracted cDNA library of mouse hepatocarcinoma cell lines with different lymphatic metastasis potential显示文摘AIM: In order to elucidate the molecular mechanism of lymphatic metastasis of hepatocarcinoma, we detected the difference of gene expression between mouse hepatocarcinoma cell lines Hca-F and Hca-P with different lymphatic metastasis potential.METHODS: cDNA of Hca-F cells was used as a tester and cDNA of Hca-P cells was used as a driver, cDNAs highly expressed in Hca-F cells were isolated by the suppression subtractive hybridization (SSH) method. The isolated cDNA was cloned into T/A cloning vector. The ligation products were transformed into DH5 α competent cells. Individual dories were randomly selected and used for PCR amplification.Vector DNA from positive clones was isolated for sequencing.RESULTS: There were 800 positive clones in amplified subtracted cDNA library. Random analysis of 160 clones with PCR showed that 95% of the clones contained 100-700 bp inserts. Analysis of 20 sequenced cDNA clones randomly picked from the SSH library revealed 4 known genes(mouse heat shock protein 84 ku, DNA helicase, ribosomal protein S13 ,ethanol induced 6 gene) and 3 expressed sequence tags (ESTs). Four cDNAs showed no homology and presumably represent novel genes.CONCLUSION: A subtracted cDNA library of differentially expressed genes in mouse heptocarcinoma cell lines with different lymphatic metastasis potential was successfully constructed with SSH and T/A cloning techniques. The library is efficient and lays a solid foundation for searching new lymphatic metastasis related genes. The morphology, chemical composition and size distribution of the inclusions in deoxidized liquid ironhave been investigated by using of SEM and image-analysis techniques. The investigated results show that the deoxidation productsin molten iron are complex inclusions of rare earth oxide or Al2O3 combined with FeO finely distributed. The proper holding timeand rapid cooling rate can make inclusions finer. | 5 | Brain ischemia induces serine phosphorylation of neuronal nitric oxide synthase by Ca^(2+)/calmodulin-dependent protein kinase II in rat hippocampus显示文摘AIM: To investigate whether brain ischemia induces serine phosphorylation of neuronal nitric oxide synthase(nNOS) by Ca2+/calmodulin-dependent protein kinase II (CaMKII) and the interaction between CaMKIIa and nNOSin rat hippocampus. METHODS: Brain ischemia was induced by bilateral carotid artery occlusion procedure.Phosphorylation and the interaction of proteins were studied by immunoprecipitation and immunoblotting. Weinvestigated during brain ischemia serine phosphorylation and amount of nNOS in crude membranes fraction (P)and cytosolic fraction (S), interaction between CaMKIIa and nNOS, and the effects of 1-[N,O-bis-(5-isoquinolinesulfonyl)-N-methyl-L-tyrosyl]-4-phenylpiperazine (KN-62, a selective inhibitor of CaMKII) on phos-phorylation and the interaction of proteins in P. RESULTS: Serine phosphorylation of nNOS in P increased persis-tently during brain ischemia, and 15 min ischemia-induced serine phosphorylation of nNOS was attenuated signifi-cantly by KN-62. But there was no serine phosphorylation of nNOS in S. The distributions of nNOS were notaffected by ischemia and KN-62. However, the binding levels of both CaMKIIa with nNOS and Thr286autophosphorylated CaMKIIa with nNOS increased after ischemia, and were diminished by KN-62. | 6 | Mapping Toll-Like Receptor Signaling Pathway Genes of Zhikong Scallop(Chlamys farreri) with FISH显示文摘Toll-like receptor(TLR) signaling pathway plays a pivotal role in the innate immune system. Studies on TLR signaling pathway genes in Zhikong scallop(Chlamys farreri) have mainly focused on sequence analysis and expression profiling, no research has been carried out on their localization. The chromosomal position of TLR signaling pathway genes can be valuable for assemblying scallop genome and analysizing gene regulatory networks. In the present study, five key TLR signaling pathway genes(Cf TLR, Cf Myd88, Cf TRAF6, Cf NFκB, and Cf IκB) containing bacterial artificial chromosomes(BACs) were isolated and physically mapped through fluorescence in situ hybridization on five non-homologous chromosome pairs, showing a similar distribution to another five model species. | 7 | Effect of Wood Vinegar on Seed Germination and Water Implantation of Corn显示文摘To analyze the effect of wood vinegar on seed germination and seedling growth, the seeds of corn are dipped in wood vinegar of different densities. The results showed that signifi cant effects were found through all the treatments on the seed germination rate, the seed germinating energy and the germinating index. The regress CUBICS curves were developed to describe the relation. The reasonable parameters range was obtained. At the same time, the corn was cultivated by wood vinegar of different densities, aiming to study the effect of wood vinegar on biomass. It showed that all treatments had obvious effects on the seedling length and dry weight aboveground, the chlorophyll and dry weight underground were not included. | 11 | Biomass Catalytic Pyrolysis with Ni Based Catalyst to Produce Hydrogen Rich Gas显示文摘Hydrogen rich gas was produced using rice husk as biomass material on the continuous biomass pyrolysis apparatus which consisted of continuous pyrolysis reactor and secondary catalytic cracking reactor. Ni based catalysts of different Ni/Al mass ratio and calcined temperature were prepared by impregnating method. The catalysts were characterized by X-ray diffraction (XRD), scan electron microscope (SEM) and FT-IR Spectrometer (FT-IR). Ni based catalyst showed good selectivity for H2 production from biomass. Catalysts prepared under different conditions had little influence on the yields of three states products when used at the same cracking temperature. Ni/Al mass ratio played an important role in products selectivity. However, the content of NiO increased further when Ni/Al mass ratio values reached 0.7 : 10, and the yield of H2 slightly increased. Hydrogen yield was greatly impacted by calcined temperature. Catalyst calcined at 550℃ performed best. When the catalyst was calcined at high temperature, NiO in the catalyst transformed into NiAl2O4, and the acid site also changed, which caused the deactivation of the catalyst. The hydrogen yield increased with the cracking temperature. | 20 | Decoding the spatiotemporal regulation of transcription factors during human spinal cord development显示文摘The spinal cord is a crucial component of the central nervous system that facilitates sensory processing and motor performance.Despite its importance,the spatiotemporal codes underlying human spinal cord development have remained elusive.In this study,we have introduced an image-based single-cell transcription factor(TF)expression decoding spatial transcriptome method(TF-seqFISH)to investigate the spatial expression and regulation of TFs during human spinal cord development.By combining spatial transcriptomic data from TF-seqFISH and single-cell RNA-sequencing data,we uncovered the spatial distribution of neural progenitor cells characterized by combinatorial TFs along the dorsoventral axis,as well as the molecular and spatial features governing neuronal generation,migration,and differentiation along the mediolateral axis.Notably,we observed a sandwich-like organization of excitatory and inhibitory interneurons transiently appearing in the dorsal horns of the developing human spinal cord.In addition,we integrated data from 10×Visium to identify early and late waves of neurogenesis in the dorsal horn,revealing the formation of laminas in the dorsal horns.Our study also illuminated the spatial differences and molecular cues underlying motor neuron(MN)diversification,and the enrichment of Amyotrophic Lateral Sclerosis(ALS)risk genes in MNs and microglia.Interestingly,we detected disease-associated microglia(DAM)-like microglia groups in the developing human spinal cord,which are predicted to be vulnerable to ALS and engaged in the TYROBP causal network and response to unfolded proteins.These findings provide spatiotemporal transcriptomic resources on the developing human spinal cord and potential strategies for spinal cord injury repair and ALS treatment. | Yingchao Shi Luwei Huang Hao Dong Meng Yang Wenyu Ding Xiang Zhou Tian Lu Zeyuan Liu Xin Zhou Mengdi Wang Bo Zeng Yinuo Sun Suijuan Zhong Bosong Wang Wei Wang Chonghai Yin Xiaoqun Wang Qian Wu | 2024 | Cell Research2024,34,3: | 0 |
