2NOD2 3020insC frameshift mutation is not associated with inflammatory bowel disease in Chinese patients of Hart nationality显示文摘AIM: An insertion mutation at nucleotide 3020 (3020insC) in the Caspase recruitment domain gene (CARD15), originally reported as NOD2, is strongly associated with Crohn's disease. The C-insertion mutation at nucleotide 3020 (3020inC) in the leucine-rich repeat (LRR) region results in a frameshift in the 10^th LRR followed by a premature stop codon. This truncation mutation is responsible for the inability to activate nuclear factor (NF)-κB in response to bacterial lipopolysaccharide (LPS). The present study aimed to genotype NOD2/CARD15 gene 3020insC frameshift mutation in Chinese patients with inflammatory bowel disease. METHODS: We genotyped an insertion polymorphism affecting the leucine-rich region of the protein product by the allele specific PCR in 74 unrelated patients with ulcerative colitis of Han nationality in Hubei Province of China, 15 patients with Crohn's disease and 172 healthy individuals. RESULTS: No significant differences were found in the genotype and allele frequendes of the C-insertion mutation of NOD2 gene among patients with Crohn's disease and ulcerative colitis and healthy controls. 3Association of Fas-670 gene polymorphism with inflammatory bowel disease in Chinese patients显示文摘AIM: Recent studies suggest that Fas-mediated apoptosis is involved in the pathogenesis of inflammatory bowel disease (IBD). It has been hypothesized that either increased apoptosis of intestinal epithelium or decreased apoptosis of lamina propria lymphocytes may induce inflammation of gut. The aim of this study was to determine whether the Fas gene promoter polymorphism at position-670 was associated with IBD in Chinese patients.METHODS: Fifty unrelated Chinese patients with IBD (38patients with ulcerative colitis and 12 with Crohn's disease)and 124 healthy controls were genotyped for the Fas-670polymorphism by PCR-restriction fragment length polymorphism method. The PCR product was digested by Mva I restriction enzyme.RESULTS: Distribution of the Fas-670 gene polymorphism was 33% for the AA genotype, 52% for the AG genotype and 15% for the GG genotype in 124 healthy subjects. In patients with IBD, 30% was for the AA genotype, 42% for the AG genotype and 28% for the GG genotype respectively. However, there was no significant difference in the genotype (P= 0.1498), allele frequencies (P= 0.3198)and carriage frequencies (P = 0.4133) between healthy controls and IBD patients. 4Response of soil water dynamics to precipitation years under different vegetation types on the northern Loess Plateau, China显示文摘Implementation of the Grain-for-Green project has led to rapid land cover changes and resulted in a significantly increased vegetation cover on the Loess Plateau of China during the past few decades. The main objective of this study was to examine the responses of soil water dynamics under four typical vegetation types against precipitation years. Soil water contents(SWCs) were measured in 0–4.0 m profiles on a hillslope under the four vegetation types of shrub, pasture, natural fallow and crop in a re-vegetated catchment area from April to October in normal(2010), dry(2011), wet(2014) and extremely wet(2013) years. The results indicated that precipitation and vegetation types jointly controlled the soil water temporal dynamics and profile characteristics in the study region. SWCs in 0–4.0 m profiles of the four vegetation types were ranked from high to low as crop>fallow>pasture>shrub and this pattern displayed a temporal stability over the four years. In the extremely wet year, SWC changes occurred in the 0–2.0 m layer under shrub and pasture while the changes further extended to the depth of 4.0-m deep layers under fallow and crop. In the other three years, SWCs changes mainly occurred in the 0–1.0 m layer and kept relatively stable in the layers deeper than 1.0 m for all the four vegetation types. The interannual variation in soil depth of SWCs was about 0–2.0 m for shrub and pasture, about 0–3.4 m for fallow and about 0–4.0 m for crop, respectively. The dried soil layers formed at the depths of 1.0, 0.6, 1.6 and 0.7 m under shrub, and 1.0, 1.0, 2.0 and 0.9 m under pasture, respectively in 2010, 2011, 2013 and 2014. The infiltrated rainwater mostly stayed in the 0–1.0 m layer and hardly supplied to soil depth >1.0 m in normal, dry and wet years. Even in the extremely wet year of 2013, rainwater recharge depth did not exceed 2.0 m under shrub and pasture. This implied that soil desiccation was difficult to remove in normal, dry and wet years, and soil desiccation could be removed in 1.0–2.0 m soil layers even in the extremely wet year under shrub and pasture. 5Association of tumor necrosis factor genetic polymorphism with chronic atrophic gastritis and gastric adenocarcinoma in Chinese Han population显示文摘AIM:To investigate the association of TNF polymorphisms with chronic atrophic gastritis (CAG) and gastric adenocarcin-oma in Chinese Han patients.METHODS:The TNFa-e 5 microsatellites and 3 RFLP sites were typed using PCR technique,followed by high-voltage denaturing PAGE with silver staining and restriction enzyme digestion respectively in specimens from 53 patients with CAG and 56 patients with agstric daenocarcinoma and 164 healthy controls.The PCR products were cloned and sequenced.RESULTS:The frequency of TNF-β Ncol*1/2 genotype was higher in patients with chronic atrophic gastritis than in healthy controls,but no significant difference was observed(60.38% vs 46.34%,p=0.076).The frequency of TNa10 allele was significantly higher in patients with chronic atrophic gastritis than in healthy controls(19.81% vs 11.89%,p=0.04).However,it did not relate to age,gender,atrophic degree or intestinal metaplasin in patients with chronic atrophic gastritis,The frequency of TNF-β Ncol*1/2 and d2/d6 genotypes were significantly higher in patients with gastric adenocarcinoma than in healthy indiveduals(p>0.05).However,TNF-β Ncol*1/2 and d2/d6 genotypes did not relate to age,gender,grade of differentiation and clinicopathologic state in patients with gastric adenocarcinoma.The frequency of TNFa6b5c1 hapolotype homoaygote was significantly lower in patients with gastric adenocarcinoma than in healthy controls (1.79% vs 15.85%,p=0.006).CONCLUSION:TNFa10 allele may be a risk factor for chronic atrophic gastritis ,TNF-β Ncol*1/2 and d2/d6 genotypes are associated with the suscepotibility to gastric adenocarcinoma,whereas TNFa6b5c1 haplotype homozygote may contribute to the resistance against gastric adenocarcinoma.Bao-YingFei BingXia Chang-ShengDeng Xian-QingXia MinXie JBartACrusius ASalvadorPena 2004World Journal of Gastroenterology2004,10,9:9
6Effects of low molecular weight heparin on platelet surface P-selectin expression and serum interleukin-8 production in rats with trinitrobenzene sulphonic acid-induced colitis显示文摘AIM:To observe the effects of low molecular weight heparin(LMWH) on platelet surface P-selectin expression and serum interleukin-8 production in rats with trinitrobenzene sulphonic acid (TNBS) induced colitis.METHODS:Colitis was induced in female Sprauge-Dawley rats by colonic administration of 2, 4, 6-TNBS. LMWH, a dalteparin (150U/kg, 300U/kg) was subcutaneously administrated one hour before induction of colitis and went on once a day for 6 days. Then a half dose was given for the next 7 days. Control animals received the same volume of normal saline once a day for 14 days after treated by TNBS.Animals were sacrificed at 24h, days 7 and 14 after induction of colitis. The colon was excised for the evaluation of macroscopic and histological findings and TNF-α immunohistochemical assay. Platelet surface P-selectin expression was determined by radioimmunoassay and serum IL-8 production was assayed by ELISA method.RESULTS:LMWH treatment in a dose of 300U/kg for 14 days significantly improved colonic inflammation by histological examination. Serum IL-8 production in the 300U/kg treatment group was more significantly decreased at day 14 than that at 24h (P<0.05). However, platelet surface P-selectin expression and TNF-α staining in colonic tissue were not significantly different among the three groups.CONCLUSION:LMWH has an anti-infiammatory effect on TNBS induced colitis in rats. 7Tumor microenvironments self-activated nanoscale metal-organic frameworks for ferroptosis based cancer chemodynamic/photothermal/chemo therapy显示文摘Ferroptosis,as a newly discovered cell death form,has become an attractive target for precision cancer therapy.Several ferroptosis therapy strategies based on nanotechnology have been reported by either increasing intracellular iron levels or by inhibition of glutathione(GSH)-dependent lipid hydroperoxidase glutathione peroxidase 4(GPX4).However,the strategy by simultaneous iron delivery and GPX4 inhibition has rarely been reported.Herein,novel tumor microenvironments(TME)-activated metal-organic frameworks involving Fe&Cu ions bridged by disulfide bonds with PEGylation(FCSP MOFs)were developed,which would be degraded specifically under the redox TME,simultaneously achieving GSH-depletion induced GPX4 inactivation and releasing Fe ions to produce ROS via Fenton reaction,therefore causing ferroptosis.More ROS could be generated by the acceleration of Fenton reaction due to the released Cu ions and the intrinsic photothermal capability of FCSP MOFs.The overexpressed GSH and H2O2 in TME could ensure the specific TME self-activated therapy.Better tumor therapeutic efficiency could be achieved by doxorubicin(DOX)loading since it can not only cause apoptosis,but also indirectly produce H2O2 to amplify Fenton reaction.Remarkable anti-tumor effect of obtained FCSP@DOX MOFs was verified via both in vitro and in vivo assays.Yu Liang Li Zhang Chao Peng Shiyu Zhang Siwen Chen Xin Qian Wanxian Luo Qing Dan Yongyan Ren Yingjia Li Bingxia Zhao 2021Acta Pharmaceutica Sinica B2021,11,10:7
8Detection and Analysis of Routine Blood Indexes in Guike Pig显示文摘[Objectives] In this study,routine blood indexes of Guike pig were detected to clarify quantity variation and morphology distribution of different functional cells( red blood cells,white blood cells and platelets),thus providing reference data for immunological evaluation and disease resistance research of Guike pig. [Methods] Blood samples were collected from forty-four 49-day-old healthy Guike pigs( 22 males and 22 females) once a week for seven consecutive weeks to detect routine blood indexes,and the results were compared with routine blood indexes of Landrace pig and Enshi black pig. [Results]Among red blood cell indexes of Guike pig at different day ages,red blood cell count( RBC) and hemoglobin count( HGB) both reached the lowest levels( 6. 85 × 1012/L and 97. 1 g/L)at 70 days of age,which were significantly lower than the corresponding indexes at 49,56,63,84 and 91 days of age( P < 0. 05,similarly hereinafter); mean corpuscular hemoglobin( MCH) and mean corpuscular hemoglobin concentration( MCHC) demonstrated basically the same variation pattern,which both reached the lowest levels( 15. 11 pg and 275. 70 g/L) at 70 days of age and reached the highest levels( 16. 40 pg and 314. 30 g/L) at 91 days of age; hematocrit( HCT),mean corpuscular volume( MCV) and red blood cell distribution width( RDW) exhibited relatively stable changes with no significant differences among different day ages( P > 0. 05,similarly hereinafter). Among white blood cell indexes of Guike pig at different day ages,white blood cell count( WBC) reached the lowest level( 13. 71 × 109/L) at 63 days of age and reached the highest level( 17. 52 × 109/L) at 84 days of age,with significant differences; neutrophil count( NEUT)reached the lowest level( 3. 61 × 109/L) at 70 days of age,which was significantly lower than that at 49,56 and 77 days of age; lymphocyte count( LYMPH) exhibited relatively stable changes and was not significantly affected by the increase in day age. Among platelet indexes of Guike pig at different day ages,platelet count( PLT),thrombocytocrit( PCT) and platelet volume distribution width( PDW) exhibited relatively stable changes among different day ages,which were not significantly affected by the increase in day age. Compared to 35-day-old Landrace pig and 5-month-old Enshi black pig,most routine blood indexes of Guike pig were similar to or ranged between those of Landrace pig and Enshi black pig except HCT,mean platelet volume( MPV),PCT and PDW. 9GNP6,a novel allele of MOC1,regulates panicle and tiller development in rice显示文摘The yield of rice is mostly affected by three factors,namely,panicle number,grain number and grain weight.Variation in panicle and grain numbers is mainly caused by tiller and panicle branches generated from axillary meristems(AMs).MOC1 encodes a putative GRAS family nuclear protein that regulates AM formation.Although several alleles of MOC1 have been identified,its variation in germplasm resources remains unclear.In the present study we characterized a novel mocl allele named gnp6 which has a thymine insertion in the coding sequence of the SAW motif in the GRAS domain.This mutation causes arrested branch formation.The SAW motif is necessary for nuclear localization of GNP6/MOC1 where it functions as a transcription factor or co-regulator.Haplotype analysis showed that the coding region of GNP6/MOC1 was conserved without any non-synonymous mutations in 240 rice accessions.However,variation in the promoter region might affect the expression of it and its downstream genes.Joint haplotype analysis of GNP6/MOC1 and MOC3 showed that haplotype combinations H9,H10 and H11,namely MOC1-Hap1 in combination with MOC3-Hap3,MOC3-Hap4 or MOC3-Hap5 could be bred to promote branch formation.These findings will enrich the genetic resources available for rice breeders.Zhanying Zhang Xingming Sun Xiaoqian Ma Bingxia Xu Yong Zhao Zhiqi Ma Gangling Li Najeeb Ullah Khan Yinghua Pan Yuntao Liang Hongliang Zhang Jinjie Li Zichao Li 2021The Crop Journal2021,9,1:6
10Detection of Porcine Epidemic Diarrhea Virus in Guangxi Province from 2011 to 2014 and Sequence Analysis of Its M Gene显示文摘Detection of pigs epidemic diarrhea virus( PEDV) was conducted on 331 piglets diarrhea fecal samples collected in Nanning,Yulin and other 12 areas of Guangxi Province from January of 2011 to April of 2014 by the method of reverse transcription-polymerase chain reaction( RT-PCR). The results showed that the positive samples of PEDV were 210 and the positive rate was 63. 44%. The clone and sequencing of M gene was carried out on 25 positive samples. PEDV reference strains were selected from Gene Bank to conduct the sequence homology alignment analysis and the phylogenetic tree of M gene. The M gene homology and amino acid sequence identity between 25 isolated strains and 51 reference strains were 96. 0%- 99. 6% and 94. 3%- 99. 6%,respectively. The genetic variation analysis of M gene showed that the genetic relationship of PEDV prevalent strains in Guangxi Province from 2013 to 2014 was close to that of the prevalent strains in Beijing,Anhui,Wuhan,Hebei and Guangdong from 2010 to 2013,and which were far from that of the Chinese early isolates CH / S( Gen Bank number: JN547228),vaccine strain CV777( Gen Bank number: AF353511) and Attenuated DR13( Gen Bank number: JQ023162). 11Relationship between glutathione and malonaldehyde levels in erythrocytes and the deformation index of erythrocyte in patients at various periods following acute cerebral infarction显示文摘BACKGROUND: Glutathione, as an in vivo free radical scavenger, plays an important role in the anti-oxidation defense mechanism in patients with acute cerebral infarction. OBJECTIVE: To observe the relationship between the levels of glutathione (GSH) and malonaldehyde (MDA), the metabolite of lipid peroxidation, in erythrocyte and deformation index of erythrocyte in patients at various periods following acute cerebral infarction. DESIGN: Controlled observation. SETTING: Qilu Hospital of Shandong University and Institute of Cerebrovascular Disease of Qingdao Medical College. PARTICIPANTS: We chose 98 inpatients with acute cerebral infarction from Department of Neurology, Qilu Hospital of Shandong University from January to December 2000 , serving as cerebral infarction group, including 50 male and 48 female , with mean age of (62±7)years. There were 23 cases found on the 1st day after onset; 25 cases on the 3rd day after onset; 25 cases on the 7th day after onset; 25 cases on the 14th days after onset, and they were all confirmed by craniocerebral CT or MRI. Another 30 homeochronous inpatients with neurosis, cervical syndrome, lumbar intervertebral disc herniation and motor neuron disease were chosen as control group, including 20 male and 10 female, with mean age of (52±8)years . There was no significant difference in age and gender distribution between two groups (P > 0.05). Patients in the two groups were informed of detected index. METHODS: ①Ulnar venous blood was chosen from the patients who were fasted on the 1st, 3rd, 7th and 14th days after onset. Deformation index of erythrocyte was measured with BL88-CKX laser diffraction erythrocyte deformeter and photographing was performed. GSH level in erythrocyte was measured with DTNB assay introduced by Beu-tler. MDA level in erythrocyte was measured with modified thiobarbituric acid colorimetric method. ②At each sample collecting , according to the criteria accepted by the Fourth National Conference of Cerebrovascular Disease, intergrant of neurologic impairment < 15 points was regarded as mild (n=46),15 to 30 points as moderate (n=40)and, > 30 points as severe (n=12). ③ t test was used to compare data between two groups , and linear correlation analysis was used in relationship analysis among indexes. MAIN OUTCOME MEASURES: ① Comparison of erythrocyte GSH and MDA levels and deformation index of erythrocyte at various periods between patients with acute cerebral infarction and controls. ②Correlation of erythrocyte GSH level with erythrocyte MDA level and with deformation index of erythrocyte in patients with cerebral infarction. ③ Relationship between erythrocyte GSH level and severity of disease in patients with acute cerebral infarction. RESULTS: Totally 98 patients with acute cerebral infarction and 30 controls all entered the stage of result analysis. ① Erythrocyte GSH level and deformation index of erythrocyte were lower on the 1st, 3rd, 7th and 14th days after onset in cerebral infarction group than in control group (P < 0.05-0.01), and erythrocyte MDA level was significantly higher in cerebral infarction group than in control group (P < 0.05-0.01). The three indexes changed most significantly on day 3 after onset in patients, and began to recover or decrease on day 7 after onset and inclined to be normal on day 14 after onset. ② Erythrocyte GSH level was significantly negatively correlated with erythrocyte MDA level in patients with acute cerebral infarction on the 1st, 3rd, 7th and 14th days after onset (r=-0.534, -0.713, -0.645, -0.656, respectively, P < 0.05-0.01),and significantly positively correlated with erythrocyte deformation index (r=0.502, 0.560, 0.455, 0.504, respectively, P < 0.05). ③Erythrocyte GSH level was significantly lower in moderate or severe patients with acute cerebral infarction than in mild patients[(0.215±0.088),(0.192±0.102),(0.281±0.090) g/L, P < 0.05]. CONCLUSION: ①Erythrocyte GSH and MDA levels and deformation index of erythrocyte change significantly on the 3rd day following acute cerebral infarction. Index detection results gradually tended to be normal on from the 7th day to 14th days day after onset. ② Change of erythrocyte GSH level can reflect the severity of disease of patients with acute cerebral infarction. ③Decrease of erythrocyte GSH level in patients with acute cerebral infarction is one of reasons that result in the decrease of deformation ability of erythrocyte.Dongjun Zhang Guangrun Xu Zhaofu Chi Bingxia Shi 2006Neural Regeneration Research2006,1,1:2

13Cis- and Trans-Cinnamic Acids Have Different Effects on the Catalytic Properties of Arabidopsis Phenylalanine Ammonia Lyases PAL1, PAL2, and PAL4显示文摘Cis-cinnamic acid (CA) is a naturally occurring compound, presumably converted from transCA in higher plants. To investigate the effect of cis-CA on the activity of Arabidopsis phenylalanine ammonia lyase (PAL), AtPAL1, AtPAL2, and AtPAL4 genes were isolated using reverse transcription polymerase chain reaction. These genes were fused to a glutathione S-transferase gene and overexpressed in a heterologous prokaryotic system of Escherichia coli. The purified PAL1, PAL2 and PAL4 enzymes were characterized biochemically to determine the effects of cis-CA on the kinetic parameter Km. 17Chemo-drugs in cell microparticles reset antitumor activity of macrophages by activating lysosomal P450 and nuclear hnRNPA2B1显示文摘Macrophages in tumors(tumor-associated macrophages,TAMs),a major population within most tumors,play key homeostatic functions by stimulating angiogenesis,enhancing tumor cell growth,and suppressing antitumor immunity.Resetting TAMs by simple,efficacious and safe approach(s)is highly desirable to enhance antitumor immunity and attenuate tumor cell malignancy.Previously,we used tumor cell-derived microparticles to package chemotherapeutic drugs(drug-MPs),which resulted in a significant treatment outcome in human malignant pleural effusions via neutrophil recruitments,implicating that drug-MPs might reset TAMs,considering the inhibitory effects of M2 macrophages on neutrophil recruitment and activation.Here,we show that drug-MPs can function as an antitumor immunomodulator by resetting TAMs with M1 phenotype and IFN-βrelease.Mechanistically,drug molecules in tumor MPs activate macrophage lysosomal P450 monooxygenases,resulting in superoxide anion formation,which further amplifies lysosomal ROS production and pH value by activating lysosomal NOX2.Consequently,lysosomal Ca^(2+)signaling is activated,thus polarizing macrophages towards M1.Meanwhile,the drug molecules are delivered from lysosomes into the nucleus where they activate DNA sensor hnRNPA2B1 for IFN-βproduction.This lysosomal-nuclear machinery fully arouses the antitumor activity of macrophages by targeting both lysosomal pH and the nuclear innate immunity.These findings highlight that drug-MPs can act as a new immunotherapeutic approach by revitalizing antitumor activity of macrophages.This mechanistic elucidation can be translated to treat malignant ascites by drug-MPs combined with PD-1 blockade.Keke Wei Huafeng Zhang Shuaishuai Yang Yuxiao Cui Bingxia Zhang Jincheng Liu Liang Tang Yaoyao Tan Simin Liu Shiqi Chen Wu Yuan Xiao Luo Chen Chen Fei Li Junwei Liu Jie Chen Pingwei Xu Jiadi Lv Ke Tang Yi Zhang Jingwei Ma Bo Huang 2023Signal Transduction and Targeted Therapy2023,8,2:0

19Isolation of Swine Corynebacteria and Analysis of 16S rDNA Gene显示文摘A strain of gram-positive bacillus was isolated from suppurative lung organs of nursery pigs in a pig farm,which was further characterized by morphological observation,cultivation test,biochemical test,drug sensitivity test,pathogenicity test and 16 S rDNA gene cloning and sequence analysis. The results showed that the isolate grew well in rabbit blood agar plate and horse serum tryptone soybean agar( TSA) plate under aerobic condition,which could lead to mortality of mice and were susceptible to cephalosporin antibiotics and fluoroquinolone antibiotics. 20Construction and Preliminary Application of RT-LAMP Detection Method for Swine Japanese B Encephalitis Virus显示文摘[Objective] This study aimed to establish a rapid,sensitive and specific method using reverse transcription loop-mediated isothermal amplification(RT-LAMP) technology to detect swine Japanese B encephalitis virus(JEV).[Method] Four specific LAMP primers were designed according to six loci the conservative region of JEV E gene sequence. Positive JEV RNA sample was used as a template for one-step amplification,and the reaction conditions and reaction system were optimized.[Result] Experimental results showed that the established method had high sensitivity,with the detection limit of 0. 5pg; specificity experiments indicated that the method had high specificity and there was no amplification reaction for other viral pathogens. The coincidence rate between detection results of RT-LAMP and RT-Nested-PCR was 90. 9%. After RT-LAMP reaction,a chemiluminescent agent was added for visual observation,which greatly reduced the detection time.
