1Oncogenic induction of cellular high CpG methylation by Epstein-Barr Barr virus in malignant epithelial cells显示文摘Epstein-Barr virus(EBV)is a well-known human herpesvirus associated with virtually all nasopharyngeal carcinoma(NPC)and^10%of gastric cancer(GC)worldwide.Increasing evidence shows that acquired genetic and epigenetic alterations lead to the initiation and progression of NPC and GC.However,even deep whole exome sequencing studies showed a relatively low frequency of gene mutations in NPC and EBV-associated GC(EBVa GC),suggesting a predominant role of epigenetic abnormities,especially promoter Cp G methylation,in the pathogenesis of NPC and EBVa GC.High frequencies of promoter methylation of tumor suppressor genes(TSGs)have been frequently reported in NPC and EBVa GC,with several EBV-induced methylated TSGs identified.Further characterization of the epigenomes(genome-wide Cp G methylation profile—methylome)of NPC and EBVa GC shows that these EBV-associated tumors display a unique high Cp G methylation epigenotype with more extensive gene methylation accumulation,indicating that EBV acts as a direct epigenetic driver for these cancers.Mechanistically,oncogenic modulation of cellular Cp G methylation machinery,such as DNA methyltransferases(DNMTs),by EBV-encoded viral proteins accounts for the EBV-induced high Cp G methylation epigenotype in NPC and EBVa GC.Thus,uncovering the EBV-associated unique epigenotype of NPC and EBVa GC would provide new insight into the molecular pathogenesis of these unique EBVassociated tumors and further help to develop pharmacologic strategies targeting cellular methylation machinery in these malignancies.Lili Li Yuan Zhang Bing-Bing Guo Francis K.L.Chan Qian Tao 2014Chinese Journal of Cancer2014,33,12:6
2A metal-free composite photocatalyst of graphene quantum dots deposited on red phosphorus显示文摘A simple approach to enhance the photocatalytic activity of red phosphorus(P) was developed.A mechanical ball milling method was applied to reduce the size of red P and to deposit graphene quantum dots onto red P. The product was characterized by scanning electron microscopy, transmission electron microscopy, contact angle measurements, zeta-potential measurements, X-ray diffraction and UV–vis absorption spectroscopy. 4Assembly of IMPDH2-Based,CTPS-Based,and Mixed Rod/Ring Structures Is Dependent on Cell Type and Conditions of Induction显示文摘Inhibition of guanosine triphosphate(GTP)and cytidine triphosphate(CTP)biosynthetic pathways induces cells to assemble rod/ring(RR)structures,also named cytoophidia,which consist of the enzymes cytidine triphosphate synthase(CTPS)and inosine-50-monophosphate dehydrogenase 2(IMPDH2).We aim to explore the interaction of CTPS and IMPDH2 in the generation of RR structures.He La and COS-7 cells were cultured in normal conditions or in the presence of 6-diazo-5-oxo-L-norleucine(DON),ribavirin,or mycophenolic acid(MPA).Over 90%of DON-treated cells presented RR structures.In He La cells,35%of the RR structures were positive for IMPDH2alone,26%were CTPS alone,and 31%were IMPDH2/CTPS mixed,while in COS-7 cells,42%of RR were IMPDH2 alone,41%were CTPS alone,and 10%were IMPDH2/CTPS mixed.Ribavirin and MPA treatments induced only IMPDH2-based RR.Cells were also transfected with an N-terminal hemagglutinin(NHA)-tagged CTPS1 construct.Over 95%of NHA-CTPS1 transfected cells with DON treatment presented IMPDH2-based RR and almost 100%presented CTPS1-based RR;when treated with ribavirin,over 94%of transfected cells presented IMPDH2-based RR and 37%presented CTPS1-based RR,whereas 2%of untreated transfected cells presented IMPDH2-based RR and 28%presented CTPS1-based RR.These results may help in understanding the relationship between CTP and GTP biosynthetic pathways,especially concerning the formation of filamentous RR structures.Gerson Dierley Keppeke S.John Calise Edward K.L.Chan Luis Eduardo C.Andrade 2015Journal of Genetics and Genomics2015,42,6:2

7Electron transport in solution-grown TIPS-pentacene single crystals:Effects of gate dielectrics and polar impurities显示文摘The n-channel behavior has been occasionally reported in the organic field-effect transistors(OFETs) that usually exhibit p-channel transport only.Reconfirmation and further examination of these unusual device performances should deepen the understanding on the electron transport in organic semiconductors.6,13-bis(triisopropyl-silylethynyl) pentacene(TIPS-pentacene),a widely examined p-channel material as Au is used for source-drain electrodes,has recently been reported to exhibit electron transport when grown from non-polar solvent on divinyltetramethyldisiloxanebis(benzocyclobutene)(BCB) dielectric,spurring the study on this unusual electron transport.This paper describes FET characteristics of solution-grown TIPS-pentacene single crystals on five polymer gate dielectrics including polystyrene(PS),poly(methyl methacrylate)(PMMA),poly(4-vinyl phenol)(PVP),poly(vinyl alcohol)(PVA) and poly(vinylidene fluoride-trifluoroethylene-chlorofluoroethylene)(P(VDF-TrFE-CFE)).In addition to the p-channel behavior,electron transport occurs in the crystals on PMMA,PS,thick PVA(40 nm) and a bilayer dielectric of PMMA on P(VDF-TrFE-CFE),while does not on PVP and thin PVA(2 nm).The two distinct FET characteristics are consistent with the previous reported trap effect of hydroxyl groups(in PVP and PVA) and reduced injection barrier by Na+ ions(as impurity in PVA).The highest electron mobility of 0.48 cm^2 V^1 s^1 has been achieved in the crystals on PMMA.Furthermore,the electron transport is greatly attenuated after the crystals are exposed to the vapor of a variety of polar solvents and the attenuated electron transport partially recovers if the crystals are heated,indicating the adverse effect of polar impurities on electron transport.By reconfirming the n-channel behavior in the OFETs based on TIPS-pentacene,this work has implications for the design of n-channel and ambipolar OFETs.Zhuo-Ting Huang Guo-Biao Xue Jia-Ke Wu Shuang Liu Huan-Bin Li Yu-Hui Yang Feng Yan Paddy K.L.Chan Hong-Zheng Chen Han-Ying Li 2016Chinese Chemical Letters2016,27,12:0
8A patient with relapsed high-grade serous ovarian carcinoma with somatic RAD51C mutations treated with PARPi monotherapy:a case report显示文摘We report our experience in the management of a relapsed ovarian cancer patient with somatic RAD51C mutation,treated with olaparib monotherapy.The patient was diagnosed with stage 4 high-grade serous ovarian carcinoma and was treated with neoadjuvant chemotherapy,cytoreductive surgery,and postoperative chemotherapy.After a second cancer recurrence,she underwent FoundationOne CDx testing following disease progression on multiple lines of chemotherapy.Based on the FoundationOne CDx results,olaparib monotherapy was started.After 13 months of therapy,all lesions responded to the treatment,and she achieved complete response as demonstrated by normalization of the levels of CA125 and positron emission tomography-computed tomography(PET-CT).We plan to continue olaparib monotherapy until disease progression.Siew-Fei Ngu Hextan Y.S.Ngan Karen K.L.Chan 2022Cancer Drug Resistance2022,5,3:0
9Multi-omic analyses identify mucosa bacteria and fecal metabolites associated with weight loss after fecal microbiota transplantation显示文摘Fecal microbiota transplantation(FMT)has shown promising results in animal models of obesity,while results in human studies are inconsistent.We aimed to determine factors associated with weight loss after FMT in nine obese subjects using serial multi-omics analysis of the fecal and mucosal microbiome.The mucosal microbiome,fecal microbiome,and fecal metabolome showed individual clustering in each subject after FMT.The colonic microbiome in patients showed more marked variance after FMT compared with the duodenal microbiome,characterized by an increased relative abundance of Bacteroides.Subjects who lost weight after FMT sustained enrichment of Bifidobacterium bifidum and Alistipes onderdonkii in the duodenal,colonic mucosal,and fecal microbiome and increased levels of phosphopantothenate biosynthesis and fecal metabolite eicosapentaenoic acid(EPA),compared with those without weight loss.Fecal levels of amino acid metabolism-associated were positively correlated with the fecal abundance of B.bifidum,and fatty acid metabolism-associated metabolites showed positive correlations with A.onderdonkii.Fen Zhang Tao Zuo Yating Wan Zhilu Xu Chunpan Cheung Amy YLi Wenyi Zhu Whitney Tang Paul K.S.Chan Francis K.L.Chan Siew C.Ng 2022The Innovation2022,3,5:0
