| 1 | Rapeseed research and production in China显示文摘Rapeseed(Brassica napus L.) is the largest oilseed crop in China and accounts for about 20% of world production.For the last 10 years,the production,planting area,and yield of rapeseed have been stable,with improvement of seed quality and especially seed oil content.China is among the leading countries in rapeseed genomic research internationally,having jointly with other countries accomplished the whole genome sequencing of rapeseed and its two parental species,Brassica oleracea and Brassica rapa.Progress on functional genomics including the identification of QTL governing important agronomic traits such as yield,seed oil content,fertility regulation,disease and insect resistance,abiotic stress,nutrition use efficiency,and pod shattering resistance has been achieved.As a consequence,molecular markers have been developed and used in breeding programs.During 2005–2014,215 rapeseed varieties were registered nationally,including 210 winter-and 5 spring-type varieties.Mechanization across the whole process of rapeseed production was investigated and operating instructions for all relevant techniques were published.Modern techniques for rapeseed field management such as high-density planting,controlled-release fertilizer,and biocontrol of disease and pests combined with precision tools such as drones have been developed and are being adopted in China.With the application of advanced breeding and production technologies,in the near future,the oil yield and quality of rapeseed varieties will be greatly increased,and more varieties with desirable traits,especially early maturation,high yield,high resistance to biotic and abiotic stress,and suitability for mechanized harvesting will be developed.Application of modern technologies on the mechanized management of rapeseed will greatly increase grower profit. | Qiong Hu Wei Hua Yan Yin Xuekun Zhang Lijiang Liu Jiaqin Shi Yongguo Zhao Lu Qin Chang Chen Hanzhong Wang | 2017 | The Crop Journal2017,5,2: | 25 |
| 2 | Expression of survivin in primary and metastatic gastric cancer cells obtained by laser capture microdissection显示文摘AIM:Survivin, a recently identified member of the inhibitor of apoptosis protein family, is expressed during development and in various human cancers. However, its expression in normal tissues and clinical relevance in cancers are still debated. In the present study, we analyzed the expression of the survivin gene in human primary and metastatic gastric cancer cells as well as in paired epithelial cells from normal gastric mucosa by means of a novel laser capture microdissection (LCM) technique coupled with reverse transcription -polymerase chain reaction (RT-PCR).METHODS: Thirty patients who had undergone gastrectomy with lymph node dissection for gastric cancer without preoperative treatments were included. Neoplastic tissue, metastatic lymph nodes, and apparently uninvolved normal tissue were collected from each patient. LCM-captured 'pure' cell groups were respectively subjected to RT-PCR analysis with primers specific for the survivin gene.RESULTS: Of the paired samples from 30 gastric cancer patients studied, 24 (80%) primary gastric cancer cell groups and 7 (23%) adjacent morphologically 'normal' gastric epithelial cell groups were shown to have a detectable survivin expression. There was a statistically significant difference in suvivin expression between these two groups (P<0.01). Meanwhile, 95% (19/20) of the metastatic gastric cancer cell groups from lymph nodes had a clear expression of the survivin gene. However, no significant correlation between survivin expression and clinicopathological features of gastric cancer was observed in the present study. CONCLUSION: Survivin expression is present in the majority of gastric cancer cell groups obtained by LCM techniques. The high expression rate in metastatic lesions suggests a possible role of survivin in cancer invasiveness and metastasis. It may contribute to the detection of gastric cancer micrometastasis as a potential molecular marker. | 3 | Characterization of the Rice Floral Organ Number Mutantfon3显示文摘A spontaneous rice mutant named floral organ number 3 (fon3) had major mutations in floral organ numbers. Genetic analysis indicated thatfon3 acted as a single recessive gene. Microscopic observation showed that the number of floral organs infon3 increased centripetally. For example, the number of pistils was the more frequently increased than organs in the outer whorls. Homeotic conversion of lodicules and glumes into palea/lemma-like organs was observed in some flowers. Scanning electron microscopy observation showed that the size of flower meristems was maintained the same or similar until the lemma primordium started to differentiate, at which time the floral meristem became enlarged, suggesting abnormal development of the inner whorls of rice florets. | 5 | Association of Fas-670 gene polymorphism with inflammatory bowel disease in Chinese patients显示文摘AIM: Recent studies suggest that Fas-mediated apoptosis is involved in the pathogenesis of inflammatory bowel disease (IBD). It has been hypothesized that either increased apoptosis of intestinal epithelium or decreased apoptosis of lamina propria lymphocytes may induce inflammation of gut. The aim of this study was to determine whether the Fas gene promoter polymorphism at position-670 was associated with IBD in Chinese patients.METHODS: Fifty unrelated Chinese patients with IBD (38patients with ulcerative colitis and 12 with Crohn's disease)and 124 healthy controls were genotyped for the Fas-670polymorphism by PCR-restriction fragment length polymorphism method. The PCR product was digested by Mva I restriction enzyme.RESULTS: Distribution of the Fas-670 gene polymorphism was 33% for the AA genotype, 52% for the AG genotype and 15% for the GG genotype in 124 healthy subjects. In patients with IBD, 30% was for the AA genotype, 42% for the AG genotype and 28% for the GG genotype respectively. However, there was no significant difference in the genotype (P= 0.1498), allele frequencies (P= 0.3198)and carriage frequencies (P = 0.4133) between healthy controls and IBD patients. | 6 | Resistance to rice blast (Pyricularia oryzae) caused by the expression of trichosanthin gene in transgenic rice plants transferred through agrobacterium method显示文摘The gene of trichosanthin has been transferred into rice plants through agrobacterium method. The single copy insertion and the expression of foreign gene have been proved in regenerated plants. In antifungal assay the degrees of rice blast (Pyricularia oryzae) infection of the transgenic plants expressing trichosanthin and expressing GUS gene as control have been evaluated. The differences such as the time of disease symptom observed, the number of infected plants and damaged leaves, the growth of infected plants of the two transgenic plants after being inoculated by rice blast (Pyricularia oryzae) are significant. The transgenic plants with trichosanthin gene grew faster than the plants with GUS gene, even when humidity environment was removed. The results show that the transgenic plants that expressed trichosanthin are able to delay the infection of rice blast compared with the plants as control. | 10 | An assessment of arctic sea ice concentration retrieval based on “HY-2” scanning radiometer data using field observations during CHINARE-2012 and other satellite instruments显示文摘A retrieval algorithm of arctic sea ice concentration(SIC) based on the brightness temperature data of 'HY-2' scanning microwave radiometer has been constructed. The tie points of the brightness temperature were selected based on the statistical analysis of a polarization gradient ratio and a spectral gradient ratio over open water(OW), first-year ice(FYI), and multiyear ice(MYI) in arctic. The thresholds from two weather filters were used to reduce atmospheric effects over the open ocean. SIC retrievals from the 'HY-2' radiometer data for idealized OW, FYI, and MYI agreed well with theoretical values. The 2012 annual SIC was calculated and compared with two reference operational products from the National Snow and Ice Data Center(NSIDC) and the University of Bremen. The total ice-covered area yielded by the 'HY-2' SIC was consistent with the results from the reference products. The assessment of SIC with the aerial photography from the fifth Chinese national arctic research expedition(CHINARE) and six synthetic aperture radar(SAR) images from the National Ice Service was carried out. The 'HY-2' SIC product was 16% higher than the values derived from the aerial photography in the central arctic. The root-mean-square(RMS) values of SIC between 'HY-2' and SAR were comparable with those between the reference products and SAR, varying from 8.57% to 12.34%. | 13 | A quantum-chemical study on the discharge reaction mechanism of lithium-sulfur batteries显示文摘Lithium-sulfur batteries have attracted a great interest in electrochemical energy conversion and storage, but their discharge mechanism remains not well understood up to now. Here, we report density functional theory (DFT) calculation study of the discharge mechanism for lithium-sulfur batteries which are based on the structure of S8 and Li2S x (1≤x≤8) clusters. The results show that for Li2S x (1≤x≤8) clusters, the most stable geometry is chainlike when x = 1 and 6, while the minimal-energy structure is found to be cyclic when x = 2-5, 7,8. The stability of Li2S x (1≤x≤8) clusters increases with the decreasing x value, indicating a favorable thermodynamic tendency of transition from S8 to Li2S. A three-step reaction route has been proposed during the discharge process, that is, S8 →Li2S4 at about 2.30 V, Li2S4 →Li2S2 at around 2.22 V, and Li2S2 → Li2S at 2.18 V. Furthermore, the effect of the electrolyte on the potential platform has been also investigated. The discharge potential is found to increase with the decrease of dielectric constant of the electrolyte. | 14 | Somatic mutation analysis of p53 and ST7 tumor suppressor genes in gastric carcinoma by DHPLC显示文摘AIM: To verify the effectiveness of denaturing highperformance liquid chromatography (DHPLC) in detecting somatic mutation of p53 gene in gastric carcinoma tissues.The superiority of this method has been proved in the detection of germline mutations, but it was not very affirmative with respect to somatic mutations in tumor specimens. ST7 gene, a candidate tumor suppressor gene identified recently at human chromosome 7q31.1, was also detected because LOH at this site has also been widely reported in stomach cancer.METHODS: DNA was extracted from 39 cases of surgical gastric carcinoma specimen and their correspondent normal mucosa. Seven fragments spanning the 11 exons were used to detect the mutation of p53 gene and the four exons reported to have mutations in ST7 gene were amplified by PCR and directly analyzed by DHPLC without mixing with wild-type allele.RESULTS: In the analysis of p53 gene mutation, 9 aberrant DHPLC chromatographies were found in tumor tissues, while their normal-adjacent counterparts running in parallel showed a normal shape. Subsequent sequencing revealed nine sequence variations, 1 polymorphism and 8 mutations including 3 mutations not reported before. The mutation rate of p53 gene (21%) was consistent with that previously reported. Furthermore, no additional aberrant chromatography was found when wild-type DNA was added into the DNA of other 30 tumor samples that showed normal shapes previously. The positivity of p53 mutations was significantly higher in intestinal-type carcinomas (40 %) than that in diffuse-type (8.33 %) carcinomas of the stomach. No mutation of ST7 gene was found.CONCLUSION: DHPLC is a very convenient method for the detection of somatic mutations in gastric carcinoma. The amount of wild type alleles supplied by the non-tumorous cells in gastric tumor specimens is enough to form heteroduplex with mutant alleles for DHPLC detection. Many inner Au nanoparticles were immobilized onto the thioled surface of the Au electrode, then more specific thiolated sin- gle-stranded DNA (ssDNA) probes could be fixed through Au-SH bonding. The hybridization was in- duced by exposing the ssDNA probe to the complementary target DNA of E. coli O157:H7 gene eaeA, then resulted in a mass change and corresponding frequency shifts ( △f ) of the QCM. The outer avidin-coated Au nanoparticles could combine with the target DNA to increase the mass. The electro- chemical techniques, cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were adopted to manifest and character each step. The target DNA corresponding to 2.0×103 colony forming unit (CFU)/mL E. coli O157:H7 cells can be detected by this biosensor, so it is practical to develop a sensitive and effective QCM biosensor for pathogenic bacteria detection based on specific DNA analy- sis. | 16 | Methane adsorption-induced coal swelling measured with an optical method显示文摘In order to quantify the effect of matrix shrinkage on reservoir permeability during coalbed methane production, coal samples from Huozhou, Changzhi and Jincheng areas in Shanxi province(classified as high-volatile bituminous coal, low-volatile bituminous coal and anthracite, respectively) were collected,and adsorption-induced coal swelling in methane were determined by an optical method at 40 °C and pressure up to 12 MPa. All three coals showed similar behavior-that swelling increased as a function of pressure up to about 10 MPa but thereafter no further increase in swelling was observed. Swelling in the direction perpendicular to the bedding plane is greater than that parallel to the bedding plane,and the differences are about 7.77–8.33%. The maximum volumetric swelling ranges from 2.73% to3.21%-increasing with increasing coal rank. The swelling data can be described by a modified DR model.In addition, swelling increases with the amount of adsorption. However, the increase shows a relatively slower stage followed by a relatively faster stage instead of a linear increase. Positive clones were observed and counted using Giemsa staining.Cell proliferative ability was assayed by Ml-r.RESULTS: (1) The done number of pIRES-G2 group decreased,clone volume reduced. The number of cell clones in pIRESneo group was 87+3, that of pIRES-G2 group was 53+4,occupying 60.1% of pIRESneo group, there was significant difference obviously (P<0.01, t=15.45). | 20 | Antibiotics-mediated intestinal microbiome perturbation aggravates tacrolimus-induced glucose disorders in mice显示文摘Both immunosuppressants and antibiotics (ABX) are indispensable for transplant patients. However, the former increases the risk of new-onset diabetes, whereas the latter impacts intestinal microbiota (IM). It is still unclear whether and how the interaction between immunosuppressants and ABX alters the IM and thus leads to glucose metabolism disorders. This study examined the alterations of glucose and lipid metabolism and IM in mice exposed to tacrolimus (TAC) with or without ABX. We found that ABX further aggravated TAC-induced glucose tolerance and increased insulin secretion. Combined treatment resulted in exacerbated lipid accumulation in the liver. TAC-altered microbial community was further amplified by ABX administration, as characterized by reductions in phylum Firmicutes, family Lachnospiraceae, and genus Copyococcus. Analyses based on the metagenomic profiles revealed that ABX augmented the effect of TAC on microbial metabolic function mostly related to lipid metabolism. The altered components of gut microbiome and predicted microbial functional profiles showed significant correlation with hepatic lipid accumulation and glucose disorders. In conclusion, ABX aggravated the effect of TAC on the microbiome and its metabolic capacities, which might contribute to hepatic lipid accumulation and glucose disorders.
