| 1 | Crosstalk of liver immune cells and cell death mechanisms in different murine models of liver injury and its clinical relevance显示文摘BACKGROUND: Liver inflammation or hepatitis is a result of pluripotent interactions of cell death molecules, cytokines, chemokines and the resident immune cells collectively called as microenvironment. The interplay of these inflammatory mediators and switching of immune responses during hepatotoxic, viral, drug-induced and immune cell-mediated hepatitis decide the fate of liver pathology. The present review aimed to describe the mechanisms of liver injury, its relevance to human liver pathology and insights for the future therapeutic interventions.DATA SOURCES: The data of mouse hepatic models and relevant human liver diseases presented in this review are systematically collected from Pub Med, Science Direct and the Web of Science databases published in English. RESULTS: The hepatotoxic liver injury in mice induced by the metabolites of CCl4, acetaminophen or alcohol represent necrotic cell death with activation of cytochrome pathway, formation of reactive oxygen species(ROS) and mitochondrial damage. The Fas or TNF-α induced apoptotic liver injury was dependent on activation of caspases, release of cytochrome c and apoptosome formation. The Con A-hepatitis demonstrated the involvement of TRAIL-dependent necrotic/necroptotic cell death with activation of RIPK1/3. The α-Gal Cer-induced liver injury was mediated by TNF-α. The LPS-induced hepatitis involved TNF-α, Fas/Fas L, and perforin/granzyme cell death pathways. The MHV3 or Poly(I:C) induced liver injury was mediated by natural killer cells and TNF-α signaling. The necrotic ischemia-reperfusion liver injury was mediated byhypoxia, ROS, and pro-inflammatory cytokines; however, necroptotic cell death was found in partial hepatectomy. The crucial role of immune cells and cell death mediators in viral hepatitis(HBV, HCV), drug-induced liver injury, non-alcoholic fatty liver disease and alcoholic liver disease in human were discussed.CONCLUSIONS: The mouse animal models of hepatitis provide a parallel approach for the study of human liver pathology. | 2 | NS1 antigen: A new beam of light in the early diagnosis of dengue infection显示文摘Objective: To evaluate NS1 antigen detection ELISA for the early laboratory diagnosis of dengue virus infection. Methods: The present study was conducted to evaluate the overall positivity of NS1 antigen detection ELISA and its comparison with viral RNA detection via real time PCR and Ig M antibodies detection by ELISA. Results: A total of 1 270 serum samples were tested 86%(1 097/1 270) were detected positive by one or more than one diagnostic test. | 3 | Snowmelt runoff prediction under changing climate in the Himalayan cryosphere: A case of Gilgit River Basin显示文摘There are serious concerns of rise in temperatures over snowy and glacierized Himalayan region that may eventually affect future river flows of Indus river system. It is therefore necessary to predict snow and glacier melt runoff to manage future water resource of Upper Indus Basin(UIB). The snowmelt runoff model(SRM) coupled with MODIS remote sensing data was employed in this study to predict daily discharges of Gilgit River in the Karakoram Range. The SRM was calibrated successfully and then simulation was made over four years i.e. 2007, 2008, 2009 and 2010 achieving coefficient of model efficiency of 0.96, 0.86, 0.9 and 0.94 respectively. The scenarios of precipitation and mean temperature developed from regional climate model PRECIS were used in SRM model to predict future flows of Gilgit River. The increase of 3 C in mean annual temperature by the end of 21 th century may result in increase of 35-40% in Gilgit River flows. | 4 | Transcranial magnetic stimulation in animal models of neurodegeneration显示文摘Brain stimulation techniques offer powerful means of modulating the physiology of specific neural structures. In recent years, non-invasive brain stimulation techniques, such as transcranial magnetic stimulation(TMS) and transcranial direct current stimulation, have emerged as therapeutic tools for neurology and neuroscience. However, the possible repercussions of these techniques remain unclear, and there are few reports on the incisive recovery mechanisms through brain stimulation. Although several studies have recommended the use of non-invasive brain stimulation in clinical neuroscience, with a special emphasis on TMS, the suggested mechanisms of action have not been confirmed directly at the neural level. Insights into the neural mechanisms of non-invasive brain stimulation would unveil the strategies necessary to enhance the safety and efficacy of this progressive approach. Therefore, animal studies investigating the mechanisms of TMSinduced recovery at the neural level are crucial for the elaboration of non-invasive brain stimulation. Translational research done using animal models has several advantages and is able to investigate knowledge gaps by directly targeting neuronal levels. | 5 | Ethnobotanical inventory and medicinal uses of some important woody plant species of Kotli,Azad Kashmir,Pakistan显示文摘Objective:To document ethnobotanical informations of useful woody plant species in the region of Kotli,Azad Kashmir.Methods:An ethnobotanical survey was conducted in Kotli.Data were collected by interview and semi structured questionnaire from selected local informants and traditional practitioners as well as by field assessment.Results:The present study documented the etnobotanical uses of 33 woody plant species.Most of the species have been used for dual purpose.Only 5 species are used for one purpose.Study revealed all species have medicinal value,among which 21 were used as fuel wood species,16 as fodder species,4 as timber wood species,12 as edible fruit species,6 as fence or hedge plant,7 as ornamental species and 12 species had other uses.Conclusions:Medicinal plants are still widely used for health care by locals of Kotli.Some species of woodlands seem to be vulnerable to overcollection and deforestation.As the young generation is diverted toward allelopathic medicines,ethnobotanical knowledges of important medicinal plants are restricted to the old people only.It is suggested to close the forest of district Kotli for next two to three decades for the conservation of plant biodiversity. | Muhammad Shoaib Amjad Muhammad Arshad | 2014 | Asian Pacific Journal of Tropical Biomedicine2014,4,12: | 3 |
| 6 | RNF13通过IRE1/XBP-1通路调控内质网应激介导的细胞凋亡显示文摘目的:在研究内质网应激介导的细胞凋亡过程中,我们发现Ring finger protein13(RNF13)具有促进细胞凋亡的功能。我们拟研究沉默RNF13后细胞对Tunicamycin等引起的细胞凋亡的影响,以及RNF13对活性形式的caspase3,XBP1(X-box binding protein 1)的剪切以及IRE1(Endoplasmic reticulum to nucleus signaling 1)磷酸化的影响以有助于了解RNF13促进细胞凋亡的信号通路的研究。方法:基因沉默RNF13,利用MTT方法研究RNF13沉默后对细胞增殖的影响,RNF13基因沉默后对XBP1剪切的影响,免疫印迹观察RNF13对IRE1磷酸化的影响。结果:RNF13基因沉默效率在80%以上。RNF13基因沉默后明显抑制细胞凋亡;敲低RNF13的细胞可抵抗衣霉素以及毒胡萝卜素的诱导的细胞凋亡。Caspase-3是细胞凋亡的关键蛋白。敲低RNF13后caspase-3的活性形式明显降低(降低70%,P<0.001)。在加入衣霉素引起内质网应激的情况下,敲除RNF13的细胞XBP1的切割活性明显降低。敲除RNF13的细胞中IREl的磷酸化明显降低(降低90%,P<0.001)。结论:RNF13通过IRE1-XBP1信号通路调节细胞凋亡。 | 谷晓峰 邓学峰 马群风 郁苗 Muhammad Arshad 周林康 | 2016 | 现代生物医学进展2016,16,5: | 3 |
| 7 | Synthesis, Crystal Structure and Antimicrobial Activity of Ethyl 2-(1-cyclohexyl-4-phenyl-1H-1,2,3-triazol-5-yl)-2-oxoacetate显示文摘A novel 1,4,5-trisubstituted 1,2,3-triazole(C18H21N3O3) was synthesized by a one-pot three component reaction of 1-azidocyclohexane, 1-copper(I) phenylethyne and ethoxalyl chloride at room temperature. The molecular structure was determined by single-crystal X-ray analysis. The compound crystallizes in the monoclinic system, space group P21/n with a = 12.8167(9), b = 8.0966(6), c = 16.7079(9) , β = 98.716(2)o, Z = 4 and V = 1713.8(2). In the crystal, the molecules are related by inversion and paired into dimers via C–H···O and C–O···C interactions involving(oxo) acetate groups. Furthermore, X-ray analysis results are compared with the optimized structure computed by using B3 LYP method with 6-311 G basis set. The calculated results showed that optimized geometry can well reproduce the crystal structure parameters. | 9 | Structural and optical properties of thermally reduced graphene oxide for energy devices显示文摘Natural intercalation of the graphite oxide, obtained as a product of Hummer's method, via ultra-sonication of water dispersed graphite oxide has been carried out to obtain graphene oxide(GO) and thermally reduced graphene oxide(RGO).Here we report the effect of metallic nitrate on the oxidation properties of graphite and then formation of metallic oxide(MO) composites with GO and RGO for the first time. We observed a change in the efficiency of the oxidation process as we replaced the conventionally used sodium nitrate with that of nickel nitrate Ni(NO_3)_2, cadmium nitrate Cd(NO_3)_2,and zinc nitrate Zn(NO_3)_2. The structural properties were investigated by x-ray diffraction and observed the successful formation of composite of MO–GO and MO–RGO(M = Zn, Cd, Ni). We sought to study the effect on the oxidation process through optical characterization via UV-Vis spectroscopy and Fourier Transform Infrared(FTIR) spectroscopy.Moreover, Thermo Gravimetric Analysis(TGA) was carried out to confirm > 90% weight loss in each process thus proving the reliability of the oxidation cycles. We have found that the nature of the oxidation process of graphite powder and its optical and electrochemical characteristics can be tuned by replacing the sodium nitrate(NaNO_3) by other metallic nitrates as Cd(NO_3)_2, Ni(NO_3)_2, and Zn(NO_3)_2. The growing aging population is a significant healthcare problem globally that researchers must investigate to better understand the underlying aging processes. Advances in microarrays and sequencing techniques have resulted in deeper analyses of diverse essential genomes(e.g., mouse, human, and rat) and their corresponding cell types, their organ-specific transcriptomes, and the tissue involved in aging. Traditional gene controllers such as DNA-and RNA-binding proteins significantly influence such programs, causing the need to sort out long non-coding RNAs, a new class of powerful gene regulatory elements. However, their functional significance in the aging process and senescence has yet to be investigated and identified. Several recent researchers have associated the initiation and development of senescence and aging in mammals with several well-reported and novel long non-coding RNAs. | 14 | The electronic and magnetic properties of wurtzite Mn:CdS, Cr:CdS and Mn:Cr:CdS: first principles calculations显示文摘In this article, density functional theory(DFT) based on generalized gradient approximation(GGA)and GGACU, U is Hubbard term, is used to study the electronic properties of CdS doped with different dopants(Cr, Mn). The calculations are carried out for Mn-doped CdS, Cr-doped CdS, and co-doping of Mn/Cr in CdS simultaneously. It is found that hopping of electrons is possible with Cr:CdS and Mn:Cr:CdS while Mn:CdS does not allow the hopping of electrons. Moreover, double exchange interactions are observed in Cr:CdS and d-d superexchange interactions are observed in Mn:CdS. Now the problem becomes interesting when one magnetic ion(Cr) supporting double exchange interactions and another ion(Mn) supporting d-d super-exchange interactions are doped simultaneously in the same system(CdS). The co-doped CdS is more stable even at high Curie temperature due to p-d double exchange interactions and d-d super exchange interactions. Compound 1a was synthesized by the interaction of 6-nitro-tetrazolo[1.5-a]-pyridine with ethyl propynoate at room temperature in the presence of Cu(OAc)2 as a catalyst and THF as solvent. Compound 1b was also synthesized by the same manner except that tert-butyl propionate, instead of ethyl propynoate, was used. The compounds were characterized by IR, 1H-NMR, 13C-NMR and single-crystal X-ray diffraction analysis. Compound 1a(C10H9N5O4) crystallizes in the triclinic system, space group P1 with a = 5.0894(9), b = 8.9834(13), c = 13.089(2) ?, α= 83.041(7), β= 80.256(7), γ=87.296(8)°, V = 585.24(16)?3, Z = 2, Mr = 263.22, crystal size(mm) = 0.37 × 0.20 ×0.18,(I > 2σ(I)) = 8557, 2493, 1229, Rint = 0.057. | 19 | Hydrological Response to Environment Change in Himalayan Watersheds: Assessment from Integrated Modeling Approach显示文摘Land use changes such as deforestation,increase in cropping or grazing areas and built-up land, likely modify the water balance and land surface behavior in the Himalayan watersheds.An integrated approach of hydrological and hydraulic modeling was adopted for comparative analysis of hydrological pattern in three Himalayan watersheds i.e.Khanpur,Rawal and Simly situated in the Northern territory of Pakistan.The rainfall-runoff model SWAT- Soil and water assessment tool and Hydro CAD were calibrated for the selected watersheds.The correlation analysis of the precipitation data of two climate stations i.e.Murree and Islamabad, with the discharge data of three rivers was utilized to select best suitable input precipitation data for Hydro CAD rainfall-runoff modeling.The peak flood hydrograph were generated using Hydro CAD runoff to optimize the basin parameters like CN, runoff volume, peak flows of the three watersheds.The hydrological response of the Rawal watershed was studied as a case study to different scenarios of land use change using SWAT model.The scenario of high deforestation indicated a decline of about 6.3% in the groundwater recharge tostream while increase of 7.1% in the surface runoff has been observed under the scenario of growth in urbanization in the recent decades.The integrated modeling approach proved helpful in investigating the hydrological behavior under changing environment at watershed level in the Himalayan region. | Zulfiqar AHMAD Arshad ASHRAF Muhammad ZAHEER Humaira BASHIR | 2015 | Journal of Mountain Science2015,12,4: | 1 |
