1Pancreatic stellate cell: Pandora's box for pancreatic disease biology显示文摘Pancreatic stellate cells(PSCs) were identified in the early 1980 s, but received much attention after 1998 when the methods to isolate and culture them from murine and human sources were developed. PSCs contribute to a small proportion of all pancreatic cells under physiological condition, but are essential for maintaining the normal pancreatic architecture. Quiescent PSCs are characterized by the presence of vitamin A laden lipid droplets. Upon PSC activation, these perinuclear lipid droplets disappear from the cytosol, attain a myofibroblast like phenotype and expresses the activation marker, alpha smooth muscle actin. PSCs maintain their activated phenotype via an autocrine loop involving different cytokines and contribute to progressive fibrosis in chronic pancreatitis(CP) and pancreatic ductal adenocarcinoma(PDAC). Several pathways(e.g., JAK-STAT, Smad, Wnt signaling, Hedgehog etc.), transcription factors and mi RNAs have been implicated in the inflammatory and profibrogenic function of PSCs. The role of PSCs goes much beyond fibrosis/desmoplasia in PDAC. It is now shown that PSCs are involved in significant crosstalk between the pancreatic cancer cells and the cancer stroma. These interactions result in tumour progression, metastasis, tumour hypoxia, immune evasion and drug resistance. 2Systematic review: Preventive and therapeutic applications of metformin in liver disease显示文摘Metformin, a biguanide derivative, is the most commonly prescribed medication in the treatment of type 2 diabetes mellitus. More recently, the use of metformin has shown potential as a preventive and therapeutic agent for a broad spectrum of conditions, including liver disease and hepatic malignancies. In this systematic review,we critically analyze the literature behind the potential use of metformin across the spectrum of liver disease and malignancies. The Pub Med and Ovid MEDLINE databases were searched from 2000 to March 2015, using a combination of relevant text words and MeS H terms: metformin and mammalian target of rapamycin, hepatitis B virus(HBV), hepatitis B virus(HCV), nonalcoholic fatty liver disease(NAFLD), hepatocellular carcinoma(HCC) or cholangiocarcinoma. The search results were evaluated for pertinence to the issue of metformin in liver disease as well as for quality of study design. Metformin has a number of biochemical effects that would suggest a benefit in treating chronic liver diseases, particularly in the context of insulin resistance and inflammation. However, the literature thus far does not support any independent therapeutic role in NAFLD or HCV. Nonetheless, there is Level Ⅲ evidence for a chemopreventive role in patients with diabetes and chronic liver disease, with decreased incidence of HCC and cholangiocarcinoma. The use of metformin seems to be safe in patients with cirrhosis, and provides a survival benefit. Once hepatic malignancies are already established, metformin does not offer any therapeutic potential. In conclusion, there is insufficient evidence to recommend use of metformin in the adjunctive treatment of chronic liver diseases, including NAFLD and HCV. 3Mortality and morbidity in necrotizing pancreatitis managed on principles of step-up approach:7 years experience from a single surgical unit显示文摘AIM To determine percentage of patients of necrotizing pancreatitis(NP) requiring intervention and the types of interventions performed. Outcomes of patients of step up necrosectomy to those of direct necrosectomy were compared. Operative mortality, overall mortality, morbidity and overall length of stay were determined. METHODS After institutional ethics committee clearance and waiver of consent, records of patients of pancreatitis were reviewed. After excluding patients as per criteria, epidemiologic and clinical data of patients of NP was noted. Treatment protocol was reviewed. Data of patients in whom stepup approach was used was compared to those in whom it was not used. RESULTS A total of 41 interventions were required in 39% patients. About 60% interventions targeted the pancreatic necrosis while the rest were required to deal with the complications of the necrosis. Image guided percutaneous catheter drainage was done in 9 patients for infected necrosis all of whom required further necrosectomy and in 3 patients with sterile necrosis. Direct retroperitoneal or anterior necrosectomy was performed in 15 patients. The average time to first intervention was 19.6 d in the non step-up group(range 11-36) vs 18.22 d in the Step-up group(range 13-25). The average hospital stay in non step-up group was 33.3 d vs 38 d in step up group. The mortality in the step-up group was 0%(0/9) vs 13%(2/15) in the non step up group. Overall mortality was 10.3% while post-operative mortality was 8.3%. Average hospital stay was 22.25 d.CONCLUSION Early conservative management plays an important role in management of NP. In patients who require intervention, the approach used and the timing of intervention should be based upon the clinical condition and local expertise available. Delaying intervention and use of minimal invasive means when intervention is necessary is desirable. The step-up approach should be used whenever possible. Even when the classical retroperitoneal catheter drainage is not feasible, there should be an attempt to follow principles of step-up technique to buy time. The outcome of patients in the step-up group compared to the non stepup group is comparable in our series. 4Targeted therapy in chronic diseases using nanomaterial-based drug delivery vehicles显示文摘The application of nanomedicines is increasing rapidly with the promise of targeted and efficient drug delivery.Nanomedicines address the shortcomings of conventional therapy,as evidenced by several preclinical and clinical investigations indicating site-specific drug delivery,reduced side effects,and better treatment outcome.The development of suitable and biocompatible drug delivery vehicles is a prerequisite that has been successfully achieved by using simple and functionalized liposomes,nanoparticles,hydrogels,micelles,dendrimers,and mesoporous particles.A variety of drug delivery vehicles have been established for the targeted and controlled delivery of therapeutic agents in a wide range of chronic diseases,such as diabetes,cancer,atherosclerosis,myocardial ischemia,asthma,pulmonary tuberculosis,Parkinson’s disease,and Alzheimer’s disease.After successful outcomes in preclinical and clinical trials,many of these drugs have been marketed for human use,such as Abraxane®,Caelyx®,Mepact®,Myocet®,Emend®,and Rapamune®.Apart from drugs/compounds,novel therapeutic agents,such as peptides,nucleic acids(DNA and RNA),and genes have also shown potential to be used as nanomedicines for the treatment of several chronic ailments.However,a large number of extensive clinical trials are still needed to ensure the short-term and long-term effects of nanomedicines in humans.This review discusses the advantages of various drug delivery vehicles for better understanding of their utility in terms of current medical needs.Furthermore,the application of a wide range of nanomedicines is also described in the context of major chronic diseases.Akhand Pratap Singh Arpan Biswas Aparna Shukla Pralay Maiti 2019Signal Transduction and Targeted Therapy2019,4,1:6
5A GFP-based bacterial biosensor with chromosomally integrated sensing cassette for quantitative detection of Hg(Ⅱ) in environment显示文摘A mercury biosensor was constructed by integrating biosensor genetic elements into E. coli JM109 chromosome in a single copy number, using the attP/attB recombination mechanism of λ phage. The genetic elements used include a regulatory protein gene (merR) along with operator/promoter (O/P) derived from the mercury resistance operon from pDU1358 plasmid of Serratia marcescens. The expression of reporter gene gfp is also controlled by merR/O/P. Integration of the construct into the chromosome was done to increase the stability and precision of the biosensor. This biosensor could detect Hg(Ⅱ) ions in the concentration range of 100–1700 nmol/L, and manifest the result as the expression of GFP. The GFP expression was significantly different (P 0.05) for each concentration of inducing Hg(Ⅱ) ions in the detection range, which reduces the chances of misinterpretation of results. 6Imaging of the temporomandibular joint: An update显示文摘Imaging of the temporomandibular joint(TMJ) is continuously evolving with advancement of imaging technologies. Many different imaging modalities are currently used to evaluate the TMJ. Magnetic resonance imaging is commonly used for evaluation of the TMJ due to its superior contrast resolution and its ability to acquire dynamic imaging for demonstration of the functionality of the joint. Computed tomography and ultrasound imaging have specific indication in imaging of the TMJ. This article focuses on state of the art imaging of the temporomandibular joint. Relevant normal anatomy and biomechanics of movement of the TMJ are discussed for better understanding of many TMJ pathologies. Imaging of internal derangements is discussed in detail. 7Neurological consequences of systemic inflammation in the premature neonate显示文摘Despite substantial progress in neonatal care over the past two decades leading to improved survival of extremely premature infants, extreme prematurity continues to be associated with long term neurodevelopmental impairments. Cerebral white matter injury is the predominant form of insult in preterm brain leading to adverse neurological consequences. Such brain injury pattern and unfavorable neurologic sequelae is commonly encountered in premature infants exposed to systemic inflammatory states such as clinical or culture proven sepsis with or without evidence of meningitis, prolonged mechanical ventilation, bronchopulmonary dysplasia, necrotizing enterocolitis and chorioamnionitis. Underlying mechanisms may include cytokine mediated processes without direct entry of pathogens into the brain, developmental differences in immune response and complex neurovascular barrier system that play a critical role in regulating the cerebral response to various systemic inflammatory insults in premature infants. 9Screening Helicobacter pylori genes induced during infection of mouse stomachs显示文摘AIM:To investigate the effect of in vivo environment on gene expression in Helicobacter pylori(H.pylori) as it relates to its survival in the host.METHODS:In vivo expression technology(IVET) systems are used to identify microbial virulence genes.We modified the IVET-transcriptional fusion vector,pIVET8,which uses antibiotic resistance as the basis for selection of candidate genes in host tissues to develop two unique IVET-promoter-screening vectors,pIVET11 and pIVET12.Our novel IVET systems were developed by the fusion of random Sau3A DNA fragments of H.pylori and a tandem-reporter system of chloramphenicol acetyltransferase and beta-galactosidase.Additionally,each vector contains a kanamycin resistance gene.We used a mouse macrophage cell line,RAW 264.7 and mice,as selective media to identify specific genes that H.pylori expresses in vivo.Gene expression studies were conducted by infecting RAW 264.7 cells with H.pylori.This was followed by real time polymerase chain reaction(PCR) analysis to determine the relative expression levels of in vivo induced genes.RESULTS:In this study,we have identified 31 in vivo induced(ivi) genes in the initial screens.These 31 genes belong to several functional gene families,including several well-known virulence factors that are expressed by the bacterium in infected mouse stomachs.Virulence factors,vacA and cagA,were found in this screen and are known to play important roles in H.pylori infection,colonization and pathogenesis.Their detection validates the efficacy of these screening systems.Some of the identified ivi genes have already been implicated to play an important role in the pathogenesis of H.pylori and other bacterial pathogens such as Escherichia coli and Vibrio cholerae.Transcription profiles of allivi genes were confirmed by real time PCR analysis of H.pylori RNA isolated from H.pylori infected RAW 264.7 macrophages.We compared the expression profile of H.pylori and RAW 264.7 coculture with that of H.pylori only.Some genes such as cag A,vac A,lpx C,mur I,tlp C,trx B,sod B,tnp B,pgi,rbf A and inf B showed a 2-20 fold upregulation.Statistically significant upregulation was obtained for all the above mentioned genes(P < 0.05).tlp C,cag A,vac A,sod B,rbf A,inf B,tnp B,lpx C and mur I were also significantly upregulated(P < 0.01).These data suggest a strong correlation between results obtained in vitro in the macrophage cell line and in the intact animal.CONCLUSION:The positive identification of these genes demonstrates that our IVET systems are powerful tools for studying H.pylori gene expression in the host environment.Aparna Singh Nathaniel Hodgson Ming Yan Jungsoo Joo Lei Gu Hong Sang Emmalena Gregory-Bryson William G Wood Yisheng Ni Kimberly Smith Sharon H Jackson William G Coleman 2012World Journal of Gastroenterology2012,18,32:5

11Murine hepatocellular carcinoma derived stem cells reveal epithelial-to-mesenchymal plasticity显示文摘AIM To establish a model to enrich and characterize stemlike cells from murine normal liver and hepatocellular carcinoma(HCC) cell lines and to further investigate stem-like cell association with epithelial-to-mesenchymal transition(EMT).METHODS In this study,we utilized a stem cell conditioned serumfree medium to enrich stem-like cells from mouse HCC and normal liver cell lines,Hepa 1-6 and AML12,respectively.We isolated the 3-dimensional spheres and assessed their stemness characteristics by evaluating theRNA levels of stemness genes and a cell surface stem cell marker by quantitative reverse transcriptase-PCR(q RTPCR).Next,we examined the relationship between stem cells and EMT using q RT-PCR.RESULTS Three-dimensional spheres were enriched by culturing murine HCC and normal hepatocyte cell lines in stem cell conditioned serum-free medium supplemented with epidermal growth factor,basic fibroblast growth factor and heparin sulfate.The 3-dimensional spheres had enhanced stemness markers such as Klf4 and Bmi1 and hepatic cancer stem cell(CSC) marker Cd44 compared to parental cells grown as adherent cultures.We report that epithelial markers E-cadherin and ZO-1 were downregulated,while mesenchymal markers Vimentin and Fibronectin were upregulated in 3-dimensional spheres.The 3-dimensional spheres also exhibited changes in expression of Snai,Zeb and Twist family of EMT transcription factors.CONCLUSION Our novel method successfully enriched stem-like cells which possessed an EMT phenotype.The isolation and characterization of murine hepatic CSCs could establish a precise target for the development of more effective therapies for HCC.Aparna Jayachandran Ritu Shrestha Bijay Dhungel I-Tao Huang Marianna Yumi Kawashima Vasconcelos Brian J Morrison Charmaine A Ramlogan-Steel Jason C Steel 2017World Journal of Stem Cells2017,9,9:4

13Imaging of pancreatic ductal adenocarcinoma: State of the art显示文摘Significant advances in imaging technology have changed the management of pancreatic cancer. In computed tomography (CT), this has included development of multidetector row, rapid, thin-section imaging that has also facilitated the advent of advanced reconstructions, which in turn has offered new perspectives from which to evaluate this disease. In magnetic resonance imaging, advances including higher field strengths, thin-section volumetric acquisitions, diffusion weighted imaging, and liver specific contrast agents have also resulted in new tools for diagnosis and staging. Endoscopic ultrasound has resulted in the ability to provide high-resolution imaging rivaling intraoperative ultrasound, along with the ability to biopsy via real time imaging suspected pancreatic lesions. Positron emission tomography with CT, while still evolving in its role, provides whole body staging as well as the unique imaging characteristic of metabolic activity to aid disease management. 14Starch based nanofibrous scaffolds for wound healing applications显示文摘Starch is an attractive polymer for wound healing applications because of its wide availability,low cost,biocompatibility,biodegradability and wound-healing property.Here,we have fabricated starch-based nanofibrous scaffolds by electrospinning for wound healing applications.The diameter of the optimized nanofibers was determined by field emission scanning electron microscopy(FE-SEM)and was found to be in the range of 110e300 nm.The mechanical strength(0.5e0.8 MPa)of the nanofibrous scaffolds was attuned using polyvinyl alcohol(plasticizer)and glutaraldehyde(crosslinking agent),to impart them with sufficient durability for skin tissue engineering.Absence of negative interactions between the polymers was confirmed by Attenuated Total Reflectance-Fourier Transform Infrared Spectroscopy(ATR-FTIR),differential scanning microscopy(DSC)and thermal gravimetric analysis(TGA).Cellular assays with L929 mouse fibroblast cells indicated the ability of the scaffolds to promote cellular proliferation,without exhibiting any toxic effect to the cells.Thus,the nanofibrous scaffolds demonstrated potential for wound healing applications.Vijaya Sadashiv Waghmare Pallavi Ravindra Wadke Sathish Dyawanapelly Aparna Deshpande Ratnesh Jain Prajakta Dandekar 2018Bioactive Materials2018,3,3:3
15Synthesis,optimization and characterization of biochar based catalyst from sawdust for simultaneous esterification and transesterification显示文摘The development of a benign environmental catalyst for the generation of biodiesel is an area of importance to reduce the overall usage of fossil fuels. In the current work, biochar was produced by slow pyrolysis of Irul wood sawdust. The optimization for biochar generation was carried out for different reaction temperatures and heating durations. The biochar was used to prepare solid acid catalysts by sulfonation process. The characterization of biochar and the sulfonated catalyst were carried out using Elemental analysis, Fourier Transform Infrared spectroscopy(FTIR), Powder X-ray Diffraction(XRD), Thermo Gravimetric Analysis(TGA), Scanning Electron Microscopy(SEM) and Surface area analyzer(BET). The characterization results showed that sulfonation of biochar resulted in biochar based solid acid catalyst containing various functional acidic groups like weak acidic\\OH groups, strong acidic\\COOH and SO3 H groups. The total acid density and sulfonic acid group density of catalyst were estimated and showed excellent acidic sites concentration which gives a good catalytic activity for biodiesel production through simultaneous esterification and transesterification. The enhanced catalytic activity is due to the high acid density of SO3 H groups and the reactant accessibility towards acidic sites as well as the strong affinity between the hydrophilic reactants and the neutral OH groups which are bonded with the polycyclic aromatic carbon rings. The performance of biochar catalyst for the production of biodiesel was evaluated by comparing the yield obtained. 17Transcriptomics-based identification and characterization of 11 CYP450 genes of Panax ginseng responsive to MeJA显示文摘Cytochromes P450 (CYP450s),a superfamily of mono-oxygenases,are essential to generate highly functionalized secondary metabolites in plants and contribute to the diversification of specialized triterpenoid biosynthesis in eudicots.However,screening and identifying the exact CYP450 genes in ginsenoside biosynthesis is extremely challenging due to existence of large quantities of members in CYP450 superfamily.Therefore,to screen the CYP450 genes involved in ginsenoside bio- synthesis,transcriptome dataset of Panax ginseng was created in our previous work using the technique of the next-generation sequencing.On the basis of bioinformatics analysis,16 putative CYP450 genes with significant differential expression were screened from the dataset and submit- ted to GenBank,in which 11 of them have been cloned.Methyl jasmonate (MeJA)was used as an elicitor to analyze the expression profiles of candidate CYP450 genes by quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR).The results of qRT-PCR analysis revealed that the expression of some CYP450 genes were strongly induced by MeJA and showed different transcription levels at different treatment time points.Homology analysis indicated that each putative CYP450 protein of P.ginseng has a conserved domain consisting of E-E-R-F-P-R-G. 19An alternative,zeaxanthin epoxidase-independent abscisic acid biosynthetic pathway in plants显示文摘Abscisic acid(ABA)is an important carotenoid-derived phytohormone that plays essential roles in plant response to biotic and abiotic stresses as well as in various physiological and developmental processes.In Arabidopsis,ABA biosynthesis starts with the epoxidation of zeaxanthin by the ABA DEFICIENT 1(ABA1)enzyme,leading to epoxycarotenoids;e.g.,violaxanthin.The oxidative cleavage of 9-cis-epoxycaro-tenoids,a key regulatory step catalyzed by 9-C/S-EPOXYCAROTENOID DIOXYGENASE,forms xanthoxin,which is converted in further rea.ctions mediated by ABA DEFICIENT 2(ABA2),ABA DEFICIENT 3(ABA3),and ABSCISIC ALDEHYDE OXIDASE 3(AAO3)into ABA.By combining genetic and biochemical approaches,we unravel here an ABA1-independent ABA biosynthetic pathway starting upstream of zeaxanthin.We iden-tified the carotenoid cleavage products(i.e.,apocarotenoids,β-apo-11-carotenal,9-cis-β-apo-11-carotenal,3-OH-β-apo-11-carotenal,and 9-cis-3-OH-β-apo-11-carotenal)as intermediates of this ABA1-independent ABA biosynthetic pathway.Using labeled compounds,we showed thatβ-apo-11-carotenal,9-cis-β-apo-11-carotenal,and 3-OH-β-apo-11-carotenal are successively converted into 9-cis-3-OH-β-apo-11-carotenal,xanthoxin,and finally into ABA in both Arabidopsis and rice.When applied to Arabidopsis,theseβ-apo-11-carotenoids exert ABA biological functions,such as maintaining seed dormancy and inducing the expression of ABA-responsive genes.Moreover,the transcdptomic analysis revealed a high overlap of differentially expressed genes regulated byβ-apo-11-carotenoids and ABA,suggesting thatβ-apo-11-carot-enoids exert ABA-independent regulatory activities.Taken together,our study identifies a biological function for the common plant metabolites,β-apo-11-carotenoids,extends our knowledge about ABA biosynthesis,and provides new insights into plant apocarotenoid metabolic networks.Kun-Peng Jia Jianing Mi Shawkat Ali Hajime Ohyanagi Juan C.Moreno Abdugaffor Ablazov Aparna Balakrishna Lamis Berqdar Alessia Fiore Gianfranco Diretto Claudio Martínez Angel R.de Lera Takashi Gojobori Salim Al-Babili 2022Molecular Plant2022,15,1:2
