1Plecanatide-mediated activation of guanylate cyclase-C suppresses inflammation-induced colorectal carcinogenesis in Apc+/Min-FCCC mice显示文摘AIM To evaluate the effect of orally administered plecanatide on colorectal dysplasia in Apc^(+/Min-FCCC) mice with dextran sodium sulfate(DSS)-induced inflammation. METHODS Inflammation driven colorectal carcinogenesis was induced in Apc^(+/Min-FCCC) mice by administering DSS in their drinking water. Mice were fed a diet supplemented with plecanatide(0-20 ppm) and its effect on the multiplicity of histopathologically confirmed polypoid,flat and indeterminate dysplasia was evaluated. Plecanatide-mediated activation of guanylate cyclase-C(GC-C) signaling was assessed in colon tissues by measuring cyclic guanosine monophosphate(cG MP) by ELISA, protein kinase G-II and vasodilator stimulated phosphoprotein by immunoblotting. Ki-67, c-myc and cyclin D1 were used as markers of proliferation. Cellular levels and localization of b-catenin in colon tissues were assessed by immunoblotting and immunohistochemistry, respectively. Uroguanylin(UG) and GC-C transcript levels were measured by quantitative reverse transcription polymerase chain reaction(RT-PCR). A mouse cytokine array panel was used to detect cytokines in the supernatant of colon explant cultures. RESULTS Oral treatment of Apc^(+/Min-FCCC) mice with plecanatide produced a statistically significant reduction in the formation of inflammation-driven polypoid, flat and indeterminate dysplasias. This anti-carcinogenic activity of plecanatide was accompanied by activation of cG MP/GC-C signaling mediated inhibition of Wnt/b-catenin signaling and reduced proliferation. Plecanatide also decreased secretion of pro-inflammatory cytokines(IL-6, IL-1 TNF), chemokines(MIP-1, IP-10) and growth factors(GCSF and GMCSF) from colon explants derived from mice with acute DSS-induced inflammation. The effect of plecanatidemediated inhibition of inflammation/dysplasia on endogenous expression of UG and GC-C transcripts was measured in intestinal tissues. Although GC-C expression was not altered appreciably, a statistically significant increase in the level of UG transcripts was detected in the proximal small intestine and colon, potentially due to a reduction in intestinal inflammation and/or neoplasia. 2Oral treatment with plecanatide or dolcanatide attenuates visceral hypersensitivity via activation of guanylate cyclase-C in rat models显示文摘AIM To investigate the effects of plecanatide and dolcanatide on maintenance of paracellular permeability, integrity of tight junctions and on suppression of visceral hypersensitivity. METHODS Transport of fluorescein isothiocyanate(FITC)-dextran was measured to assess permeability across cell monolayers and rat colon tissues. Effects of plecanatide and dolcanatide on the integrity of tight junctions in Caco-2 and T84 monolayers and on the expression and localization of occludin and zonula occludens-1(ZO-1) were examined by immunofluorescence microscopy. Anti-nociceptive activity of these agonists was evaluated in trinitrobenzene sulfonic acid(TNBS)-induced inflammatory as well as in non-inflammatory partial restraint stress(PRS) rat models. Statistical significance between the treatment groups in the permeability studies were evaluated using unpaired t-tests.RESULTS Treatment of T84 and Caco-2 monolayers with lipopolysaccharide(LPS) rapidly increased permeability, which was effectively suppressed when monolayers were also treated with plecanatide or dolcanatide. Similarly, when T84 and Caco-2 monolayers were treated with LPS, cell surface localization of tight junction proteins occludin and ZO-1 was severely disrupted. When cell monolayers were treated with LPS in the presence of plecanatide or dolcanatide, occludin and ZO-1 were localized at the cell surface of adjoining cells, similar to that observed for vehicle treated cells. Treatment of cell monolayers with plecanatide or dolcanatide without LPS did not alter permeability, integrity of tight junctions and cell surface localization of either of the tight junction proteins. In rat visceral hypersensitivity models, both agonists suppressed the TNBS-induced increase in abdominal contractions in response to colorectal distension without affecting the colonic wall elasticity, and both agonists also reduced colonic hypersensitivity in the PRS model. CONCLUSION Our results suggest that activation of GC-C signaling might be involved in maintenance of barrier function, possibly through regulating normal localization of tight junction proteins. Consistent with these findings, plecanatide and dolcanatide showed potent antinociceptive activity in rat visceral hypersensitivity models. 4Outline of Ascomycota: 2017显示文摘Taxonomic placement of genera have been changing rapidly as taxonomists widely use DNA sequence data in phylogenetic and evolutionary studies. It is essential to update existing databases/outlines based on recent studies, since thesesources are widely used as a foundation for other research. In this outline, we merge both asexual and sexual genera intoone outline. The phylum Ascomycota comprises of three subphyla viz. Pezizomycotina (including 13 classes, 124 ordersand 507 families), Saccharomycotina (including one class, one order and 13 families) and Taphrinomycotina (five classes,five orders and six families). 5Fungal diversity notes 367-490:taxonomic and phylogenetic contributions to fungal taxa显示文摘This is a continuity of a series of taxonomic papers where materials are examined,described and novel combinations are proposed where necessary to improve our traditional species concepts and provide updates on their classification.In addition to extensive morphological descriptions and appropriate asexual and sexual connections,DNA sequence data are also analysed from concatenated datasets(rDNA,TEF-a,RBP2 and b-Tubulin)to infer phylogenetic relationships and substantiate systematic position of taxa within appropriate ranks.Wherever new species or combinations are being proposed,we apply an integrative approach(morphological and molecular data as well as ecological features wherever applicable).Notes on 125 fungal taxa are compiled in this paper,including eight new genera,101 new species,two new combinations,one neotype,four reference specimens,new host or distribution records 7Diffusion tensor imaging pipeline measures of cerebral white matter integrity: An overview of recent advances and prospects显示文摘Cerebral small vessel disease(CSVD)is a leading cause of age-related microvascular cognitive decline,resulting in significant morbidity and decreased quality of life.Despite a progress on its key pathophysiological bases and general acceptance of key terms from neuroimaging findings as observed on the magnetic resonance imaging(MRI),key questions on CSVD remain elusive.Enhanced relationships and reliable lesion studies,such as white matter tractography using diffusion-based MRI(dMRI)are necessary in order to improve the assessment of white matter architecture and connectivity in CSVD.Diffusion tensor imaging(DTI)and tractography is an application of dMRI that provides data that can be used to non-invasively appraise the brain white matter connections via fiber tracking and enable visualization of individual patient-specific white matter fiber tracts to reflect the extent of CSVD-associated white matter damage.However,due to a lack of standardization on various sets of software or image pipeline processing utilized in this technique that driven mostly from research setting,interpreting the findings remain contentious,especially to inform an improved diagnosis and/or prognosis of CSVD for routine clinical use.In this minireview,we highlight the advances in DTI pipeline processing and the prospect of this DTI metrics as potential imaging biomarker for CSVD,even for subclinical CSVD in at-risk individuals.Amanina Ahmad Safri Che Mohd Nasril Che Mohd Nassir Ismail Nurul Iman Nur Hartini Mohd Taib Anusha Achuthan Muzaimi Mustapha 2022World Journal of Clinical Cases2022,10,24:2

15Predicting global numbers of teleomorphic ascomycetes显示文摘Sexual reproduction is the basic way to form high genetic diversity and it is beneficial in evolution and speciation of fungi.The global diversity of teleomorphic species in Ascomycota has not been estimated.This paper estimates the species number for sexual ascomycetes based on five different estimation approaches,viz.by numbers of described fungi,by fungus:substrate ratio,by ecological distribution,by meta-DNA barcoding or culture-independent studies and by previous estimates of species in Ascomycota.The assumptions were made with the currently most accepted,“2.2–3.8 million”species estimate and results of previous studies concluding that 90%of the described ascomycetes reproduce sexually.The Catalogue of Life,Species Fungorum and published research were used for data procurement.The average value of teleomorphic species in Ascomycota from all methods is 1.86 million,ranging from 1.37 to 2.56 million.However,only around 83,000 teleomorphic species have been described in Ascomycota and deposited in data repositories.The ratio between described teleomorphic ascomycetes to predicted teleomorphic ascomycetes is 1:22.Therefore,where are the undiscovered teleomorphic ascomycetes?The undescribed species are no doubt to be found in biodiversity hot spots,poorly-studied areas and species complexes.Other poorly studied niches include extremophiles,lichenicolous fungi,human pathogens,marine fungi,and fungicolous fungi.Undescribed species are present in unexamined collections in specimen repositories or incompletely described earlier spe-cies.Nomenclatural issues,such as the use of separate names for teleomorph and anamorphs,synonyms,conspecific names,illegitimate and invalid names also affect the number of described species.Interspecies introgression results in new species,while species numbers are reduced by extinctions.Indunil C.Senanayake Dhandevi Pem Achala R.Rathnayaka Subodini N.Wijesinghe Saowaluck Tibpromma Dhanushka N.Wanasinghe Rungtiwa Phookamsak Nuwan D.Kularathnage Deecksha Gomdola Dulanjalee Harishchandra Lakmali S.Dissanayake Mei-mei Xiang Anusha H.Ekanayaka Eric H.C.McKenzie Kevin D.Hyde Hao-xing Zhang Ning Xie 2022Fungal Diversity2022,,3:1
