1HER2 in gastric cancer: Comparative analysis of three different antibodies using whole-tissue sections and tissue microarrays显示文摘AIM:To compare the performance of three commercially available anti-human epidermalgrowth factor receptor 2(HER2)antibodies in whole-tissue sections and tissue microarrays(TMAs)of a series of gastric tumors.METHODS:We present a comparative analysis of three anti-HER2 antibodies(HercepTest,4B5 and SP3)using TMA and whole-tissue sections prepared from the same paraffin blocks of 199 gastric adenocarcinomas operated upon between January 2004 and December2008 at a Brazilian cancer hospital.The data on the patients’age,sex,the anatomical location of the tumor and the Lauren’s histological classification were collected from clinical and pathological records.The immunohistochemical(IHC)results were examined by two pathologists and the cases were classified as positive(3+),equivocal(2+)and negative(0 or 1+),according to the criteria of the IHC scoring system of gastric cancer.TMAs and whole-tissue sections were evaluated separately and independently.All cases yielding discordant IHC results and/or scored as 2+were subjected to dual-color in situ hybridization in order to determine the final HER2 status.Besides determining the sensitivity and predictive value for HER2-positive status,we measured the accuracy of each antibody by calculating the area under the receiver operating characteristic(ROC)curve.The agreement between the results obtained using the TMAs and those obtained using the whole-tissue sections was assessed by means of Kappa coefficient.RESULTS:Intratumoral heterogeneity of HER2 expression was observed with all antibodies.HER2-positive expression(3+)in the whole-tissue sections was observed in 23 cases(11.6%)using the 4B5 antibody,in 18 cases(9.1%)using the SP3 antibody and in 10 cases(5.1%)using the HercepTest antibody.In the TMAs,11 positive cases(5.6%)were identified using SP3 antibody,9(4.6%)using the 4B5 antibody and 6(3%)using the HercepTest antibody.The sensitivity using whole-tissue sections and TMA,respectively,was 95.2%and 42.9%with 4B5,90.5%and 66.7%with SP3 and 47.6%and42.9%with HercepTest.The accuracy,calculated from the area under the ROC curve,using whole-tissue sections and TMA,respectively,was 0.91 and 0.79 by 4B5,0.86 and 0.80 by SP3 and 0.73 and 0.71 by HercepTest.The concordance of the results obtained using wholetissue sections and TMA was 97.4%(Kappa 0.75)using HercepTest,85.6%(Kappa 0.56)using SP3 and 84.1%(Kappa 0.38)using 4B5.CONCLUSION:The use of the 4B5 antibody on wholetissue sections was the most accurate IHC method for evaluating HER2 expression in gastric adenocarcinoma.Lucas Faria Abraho-Machado Alexandre Andrade dos Anjos Jácome Durval Renato Wohnrath José Sebastio dos Santos Estela Cristina Carneseca José Humberto Tavares Guerreiro Fregnani Cristovam Scapulatempo-Neto 2013World Journal of Gastroenterology2013,19,38:12
2Application of light detection and ranging and ultrasonic sensors to high-throughput phenotyping and precision horticulture: current status and challenges显示文摘Ultrasonic and light detection and ranging(LiDAR)sensors have been some of the most deeply investigated sensing technologies within the scope of digital horticulture.They can accurately estimate geometrical and structural parameters of the tree canopies providing input information for high-throughput phenotyping and precision horticulture.A review was conducted in order to describe how these technologies evolved and identify the main investigated topics,applications,and key points for future investigations in horticulture science.Most research efforts have been focused on the development of data acquisition systems,data processing,and high-resolution 3D modeling to derive structural tree parameters such as canopy volume and leaf area.Reported applications of such sensors for precision horticulture were restricted to real-time variable-rate solutions where ultrasonic or LiDAR sensors were tested to adjust plant protection product or fertilizer dose rates according to the tree volume variability.More studies exploring other applications in site-specific management are encouraged;some that integrates canopy sensing data with other sources of information collected at the within-grove scale(e.g.,digital elevation models,soil type maps,historical yield maps,etc.).Highly accurate 3D tree models derived from LiDAR scanning demonstrate their great potential for tree phenotyping.However,the technology has not been widely adopted by researchers to evaluate the performance of new plant varieties or the outcomes from different management practices.Commercial solutions for tree scanning of whole groves,orchards,and nurseries would promote such adoption and facilitate more applied research in plant phenotyping and precision horticulture.AndréF.Colaço JoséP.Molin Joan R.Rosell-Polo Alexandre Escolà 2018Horticulture Research2018,5,1:4

4Personalized medicine in gastric cancer:Where are we andwhere are we going?显示文摘Despite improvements in adjuvant therapies for gastric cancer in recent years, the disease is characterized by high recurrence rates and a dismal prognosis. The major improvement in the treatment of recurrent or metastatic gastric cancer in recent years has been the incorporation of trastuzumab, a monoclonal antibody that inhibits human epidermal growth factor receptor 2(HER2) heterodimerization, after the demonstrated predictive value of the overexpression and/or amplification of this receptor. Beyond HER2, other genetic abnormalities have been identified, and these mutations may be targetable by tyrosine kinase inhibitors or monoclonal antibodies. The demonstration of four distinct molecular subtypes of gastric cancer by the Cancer Genome Atlas study highlight the enormous heterogeneity of the disease and its complex interplay between genetic and epigenetic alterations and provide a roadmap to implement genome-guided personalized therapy in gastric cancer. 11New design solutions for low-power energy production in water pipe systems显示文摘This study is the result of ongoing research for a European Union 7th Framework Program Project regarding energy converters for very low heads,and aims to analyze optimization of new cost-effective hydraulic turbine designs for possible implementation in water supply systems(WSSs) or in other pressurized water pipe infrastructures,such as irrigation,wastewater,or drainage systems.A new methodology is presented based on a theoretical,technical and economic analysis.Viability studies focused on small power values for different pipe systems were investigated.Detailed analyses of alternative typical volumetric energy converters were conducted on the basis of mathematical and physical fundamentals as well as computational fluid dynamics(CFD) associated with the interaction between the flow conditions and the system operation.Important constraints(e.g.,size,stability,efficiency,and continuous steady flow conditions) can be identified and a search for alternative rotary volumetric converters is being conducted.As promising cost-effective solutions for the coming years,adapted rotor-dynamic turbomachines and non-conventional axial propeller devices were analyzed based on the basic principles of pumps operating as turbines,as well as through an extensive comparison between simulations and experimental tests.Helena M.RAMOS Alexandre BORGA Mariana SIMO 2009Water Science and Engineering2009,2,4:1
