| 1 | Isolation of the promoter of a cotton β-galactosidase gene (GhGal1) and its expression in transgenic tobacco plants显示文摘β-galactosidases (EC 3.2.1.23) constitute a widespread family of glycosyl hydrolases in plants and are thought to be involved in metabolism of cell wall polysaccharides. A cDNA of the cotton (Gossypium hirsutum) β-galactosidase gene, designated GhGal1, has previously been identified and its transcripts are highly abundant at the elongation stage of the cotton fiber. To examine the temporal and spatial control of GhGal1 expression, a transcriptional fusion of the GhGal1 promoter region (1770 bp) with the β-glucuronidase (GUS) reporter gene was introduced into tobacco plants by the Agrobacterium infection method. The resulting transgenic plants showed higher GUS activity of fruit in the transgenic plants than that in the negative and positive controls. Histochemical localization of GUS activity demonstrated that the expression of the GUS gene could be found in the meristem zones of roots, cotyledons, vascular tissues, fruit and trichomes in transgenic tobacco plants. Additionally, se-quence analysis of the regulatory region also revealed several conserved motifs among which some were shared with previously reported fruit/seed-specific elements and the others were related with trichome expression. | 3 | The SOC1-like gene BoMADS50 is associated with the flowering of Bambusa oldhamii显示文摘Bamboo is known for its edible shoots and beautiful texture and has considerable economic and ornamental value.Unique among traditional flowering plants,many bamboo plants undergo extensive synchronized flowering followed by large-scale death,seriously affecting the productivity and application of bamboo forests.To date,the molecular mechanism of bamboo flowering characteristics has remained unknown.In this study,a SUPPRESSOR OF OVEREXPRESSION OF CONSTANS1(SOC1)-like gene,BoMADS50,was identified from Bambusa oldhamii.BoMADS50 was highly expressed in mature leaves and the floral primordium formation period during B.oldhamii flowering and overexpression of BoMADS50 caused early fl owering in transgenic rice.Moreover,BoMADS50 could interact with APETALA1/FRUITFULL(AP1/FUL)-like proteins(BoMADS 14-1/2,BoMADS15-1/2)in vivo,and the expression of BoMADS50 was signi ficantly promoted by BoMADS14-1,further indicating a synergistic effect between BoMADS50 and BoAP1/FUL-like proteins in regulating B.oldhamii flowering.We also identi fi ed four additional transcripts of BoMADS50(BoMADS50-1/2/3/4)with different nucleotide variations.Although the protein-CDS were polymorphic,they had flowering activation functions similar to those of BoMADS50.Yeast one-hybrid and transient expression assays subsequently showed that both BoMADS50 and BoMADS50-1 bind to the promoter fragment of itself and the SHORT VEGETATIVE PHASE(SVP)-like gene BoSVP,but only BoMADS50-1 can positively induce their transcription.Therefore,nucleotide variations likely endow BoMADS50-1 with strong regulatory activity.Thus,BoMADS50 and BoMADS50-1/2/3/4 are probably important positive flowering regulators in B.oldhamii.Moreover,the functional conservatism and speci ficity of BoMADS50 and BoMADS50-1 might be related to the synchronized and sporadic flowering characteristics of B.oldhamii. | Dan Hou Ling Li Tengfei Ma Jialong Pei Zhongyu Zhao Mengzhu Lu Aimin Wu Xinchun Lin | 2021 | Horticulture Research2021,8,1: | 4 |
| 4 | Influence of boron contents on properties of AlMgB films prepared by RF magnetron sputtering显示文摘Ternary AlMgB thin films were synthesized on silicon (100) substrate at 573 K by radio frequency (RF) magnetron sputtering method using one Al/Mg co-target and one boron target. The thickness of the as-deposited thin film was controlled to 500 nm by adjusting deposition time. The influences of sputtering powers on the elemental contents and structural and mechanical properties were investigated by electron probe microanalysis (EPMA), X-ray diffraction (XRD), high-resolution transmission electron microscopy (HR-TEM), and nanoindentation system. At the same time, the ball-on-disk tribometer was used to measure the friction behavior of the films. Experimental results indicate that the as-deposited boron-rich films are primarily amorphous structure and possess a dramatic high hardness up to 39 GPa with 99.03 at.% boron. Obviously, it has exceeded the hardness value of 32 GPa of pure AlMgB 14 bulk material prepared by sintering method. | 5 | Fabrication and its characteristics of hard coating Ti-Al-N system prepared by DC magnetron sputtering显示文摘Pseudobinary Ti 1 x Al x N films were synthesized on Si (100) wafer by DC magnetron sputtering method using Ti 1 x Al x alloy targets with different Al contents. The composition of the Ti 1 x Al x N films was determined by electron probe microanalysis (EPMA). Structural characteristic was performed by X-ray diffraction (XRD), transmission electron microscopy (TEM), and high-resolution TEM (HRTEM). First principles virtual crystal calculations for the Ti 1 x Al x N disordered alloys were used for the XRD simulations. The crystalline structure of the Ti 0.61 Al 0.39 N film was found to be a metastable single phase with NaCl (B1) structure. Its lattice constant, determined by XRD, was less than that of pure TiN. With the increase of Al content, the lattice constant of B1 phase was continually decreased, while würtzite (B4) structure was observed in the Ti 0.40 Al 0.60 N film. When x reached 0.75, the B1 phase disappeared, and only B4 phase was remained. The critical Al content for the phase transition from NaCl to würtzite structure in this paper was about 0.60, which could be explained by both the thermodynamic model and the electron theory. As-deposited Ti 1 x Al x N films exhibited excellent mechanical properties. | 6 | Human hepatopoietin augmenter of liver regeneration──A hepatotrophic factor for liver regeneration, and its potential antihepatitis effect in vivo显示文摘The effect of human hapatopoietin i.e. augmenter of liver regeneration (hALR) was determined on hepatocyte DNA synthesis, and on CCl 4-induced hepatitis in animal in vitro and in vivo. It is found that ALR could directly stimulate DNA synthesis of hepatocytes in primary culture in a dose-dependent manner. In 30% hepatectomied rats, significant DNA synthesis occurred in control rats ; even so, exogenous hALR gene encoding protein increased DNA synthesis of regeneration liver by 2.3 fold compared with control rats. A lower dose of CCl 4 was administrated in rats and the effect of ALR on DNA synthesis of regenerating liver 48 h after CCl 4 administration was analyzed. Few Budr-labeled hepatocytes were visible in control rats. However, exogenous hALR markedly increased the number of labeled cells in a dose-dependent manner. Statistical analysis showed that 10 or 40 μg·kg -1 ALR protein stimulated DNA synthesis by 1.7 and 4.8 fold respectively. In addition to enhancing cell growth, adiministration of hALR achieved a significant improvement in reversing the lethality of rat hepatic failure when compared with that of control group; the elevation of cytosolic enzymes was dramatically suppressed by exogenous hALR in CCl 4-treated mice in vivo and in vitro; histologically, hepatocytes around the central vein were necrotic, and the degree of hepatocyte necrosis in control mice was more prominent than that in the mice given 40 μg·kg-1 hALR 48 h after CCl 4 administration. We also noted that hALR had a strong antihepatitis effect in vitro which was determined with primary cultured rat hepatocytes. These findings suggest that hALR protects the integrity of hepatocytes against severe hepatitis, and indicate that ALR may be an important regulator of liver regeneration and play a major role in liver injury repair. | 7 | Methylesterification of cell-wall pectin controls the diurnal flower-opening times in rice显示文摘Flowers are the core reproductive organ of plants, and flowering is essential for cross-pollination. Diurnal flower-opening time is thus a key trait influencing reproductive isolation, hybrid breeding, and thermostability in plants. However, the molecular mechanisms controlling this trait remain unknown. Here, we report that rice Diurnal Flower Opening Time 1 (DFOT1) modulates pectin methylesterase (PME) activity to regulate pectin methylesterification levels of the lodicule cell walls, which affect lodicule swelling to control diurnal flower-opening time. DFOT1 is specifically expressed in the lodicules, and its expression gradually increases with the approach to flowering but decreases with flowering. Importantly, a knockout of DFOT1 showed earlier diurnal flower opening. We demonstrate that DFOT1 interacts directly with multiple PMEs to promote their activity. Knockout of PME40 also resulted in early diurnal flower opening, whereas overexpression of PME42 delayed diurnal flower opening. Lower PME activity was observed to be associated with higher levels of pectin methylesterification and the softening of cell walls in lodicules, which contribute to the absorption of water by lodicules and cause them to swell, thus promoting early diurnal flower opening. Higher PME activity had the opposite effect. | 11 | Generation of transgenic cucumbers with expression of a ten-tandem repeat long-acting GLP-1 analogue and their biological function on diabetic rats显示文摘Glucagon-like peptide-1 (GLP-1) is a 30-amino acid peptide hormone, which has the regulatory function of stimulating the secretion of insulin to balance blood glucose levels. In this study, marker-free expression vector pX6 with a fusion gene of ten tandem repeated GLP-1 analog ([Ser8, Gln26 and Asp34]-GLP-1) genes was constructed and transformed into cucumbers by Agrobacterium tumefaciens-mediated transformation method. Four transgenic lines were obtained by PCR and Southern blotting analysis and two transgenic lines successfully expressed the fusion protein (named GLP-T), which was revealed by Western blotting analysis. The biological activity test results showed that the serum glucose level of diabetic rats was significantly decreased after oral administration of the fusion protein GLP-T extracted from the transgenic cucumber fruitage. | 12 | The effect of continuous Ni(Ⅱ) exposure on the organic degradation and soluble microbial product(SMP) formation in two-phase anaerobic reactor显示文摘A two-phase anaerobic reactor fed with glucose substrate(3 g chemical oxygen demand(COD)/L) was used to investigate the effects of toxic metals on the degradation of organics and the soluble microbial product(SMP) formation. Low concentrations of Ni(II)(5 and10 mg/L) promoted the acid phase, whereas high concentrations(15, 20, and 25 mg/L)exhibited an inhibitory effect on, but did not alter the fermentative method, which mainly involved the fermentation of propionic acid. The methanogenic microorganism exhibited a strong capability adapting constantly increased Ni(II) levels. The acid phase was an accumulation stage of SMP. In the absence of Ni(II), the high-molecular-weight material in the effluent SMP mainly contained polysaccharide, tryptophan, and casein. Methanogens metabolized most of the polysaccharide, the whole tryptophan content, and part of the casein, leading to the presence of humic acid and protein in effluent. After Ni(II) dosage, the protein and polysaccharide of the acid phase increased, and tryptophan changed, while casein remained stable. More protein than polysaccharide was produced, suggesting the prominent function of protein when addressing the negative effect of toxic metals. | 20 | Reprogramming mature terminally differentiated adipocytes to induced pluripotent stem cells显示文摘Mature adipocytes are terminally differentiated somatic cells. Here, we report the successful generation of induced pluripotent stem(i PS) cells from mouse mature adipocytes by forced expression of six transcription factors(Oct4, Sox2, c-Myc, Klf4, Rarc, and Lrh1) with a piggy Bac transposon-based strategy. The resulting i PS cells were pluripotent as evidenced by the fact that they stained positive for alkaline phosphatase, expressed high levels of key pluripotency markers including Oct4, Nanog, and SSEA1,and remained pluripotent on a 2i media. In vitro differentiation of the i PS cells showed that the cell derivatives of all three germ layers could be readily obtained through formation of embryoid bodies. Most importantly, these adipocytederived i PS cells were capable of producing chimera with high frequencies when reintroduced into early-stage embryos and transmitted through the germ line.
