| 1 | Mechanisms of Huangqi Decoction Granules (黄芪汤颗粒剂) on Hepatitis B Cirrhosis Patients Based on RNA-Sequencing显示文摘Objective: To explore the action mechanisms of Huangqi Decoction Granules(黄芪汤颗粒剂, HQDG) on hepatitis B cirrhosis. Methods: A total of 85 patients with hepatitis B cirrhosis were randomly divided into HQDG group(42 cases) and control group(43 cases) by a random number table and were treated with HQDG or placebo for 48 weeks(6 g per times and orally for 3 times a day), respectively. After RNA-sequencing of serum samples extracted from the patients, the differentially expressed genes(DEGs) in HQDG and control groups before and after treatment were separately screened. The DEGs were then performed pathway enrichment analysis and proteinprotein interaction(PPI) network analysis. The expression levels of key genes were detected by quantitative realtime polymerase chain reaction(qRT-PCR). Results: After the investigation, 4 and 3 cases were respectively excluded from HQD and control groups because of the incomplete data. Additionally, 3 and 5 cases were lost to follow up in HQD and control groups, respectively. Finally, a total of 70 cases with good compliance were included for further DEGs analysis. A total of 1,070 DEGs(including 455 up-regulated genes and 615 down-regulated genes) in HQDG group and 227 DEGs(including 164 up-regulated genes and 63 down-regulated genes) in the control group were identified after treatment. Compared with the control group, 1,043 DEGs were specific in HQDG group. Besides, 1 up-regulated transcription factor(TF, such as GLI family zinc finger 1, GLI1) and 25 down-regulated TFs(such as drosophila mothers against decapentaplegic proteinfamily member 2, SMAD2) were identified. Pathway enrichment analysis showed that down-regulated Ras homolog gene family member A(RHOA) was enriched in pathogenic Escherichia coli infection. In the PPI network, up-regulated epidermal growth factor receptor(EGFR), and down-regulated cell division cycle 42(CDC42) as well as v-akt murine thymoma viral oncogene homolog 1(AKT1) had higher degrees. Moreover, long non-coding RNAs(lncR NA) growth arrest-specific 5(GAS5) was involved in the lncR NA-target regulatory network. Furthermore, qR T-PCR revealed that expression levels of CDC42 and GLI1 had significant differences in HQDG group before and after treatment(P<0.05). Conclusions: CDC42 and GLI1 may be the targets of HQDG in patients with hepatitis B cirrhosis. | 2 | An all-silk-derived functional nanosphere matrix for sequential biomolecule delivery and in situ osteochondral regeneration显示文摘Endogenous repair of osteochondral defect is usually limited by the insufficient number of cells in the early stage and incomplete cell differentiation in the later stage.The development of drug delivery systems for sequential release of pro-migratory and pro-chondrogenic molecules to induce endogenous bone marrow-derived mesenchymal stem cells(BMSCs)recruitment and chondrogenic differentiation is highly desirable for in situ osteochondral regeneration.In this study,a novel,all-silk-derived sequential delivery system was fabricated by incorporating the tunable drug-loaded silk fibroin(SF)nanospheres into a SF porous matrix.The loading efficiency and release kinetics of biomolecules depended on the initial SF/polyvinyl alcohol(PVA)concentrations(0.2%,1%and 5%)of the nanospheres,as well as the hydrophobicity of the loaded molecules,resulting in controllable and programmed delivery profiles.Our findings indicated that the 5%nanosphere-incorporated matrix showed a rapid release of E7 peptide during the first 120 h,whereas the 0.2%nanosphere-incorporated matrix provided a slow and sustained release of Kartogenin(KGN)longer than 30 days.During in vitro culture of BMSCs,this functional SF matrix incorporated with E7/KGN nanospheres showed good biocompatibility,as well as enhanced BMSCs migration and chondrogenic differentiation through the release of E7 and KGN.Furthermore,when implanted into rabbit osteochondral defect,the SF nanosphere matrix with sequential E7/KGN release promoted the regeneration of both cartilage and subchondral bone.This work not only provided a novel all-silk-derived drug delivery system for sequential release of molecules,but also a functional tissueengineered scaffold for osteochondral regeneration. | Wei Zhang Chen Ling Aini Zhang Haoyang Liu Yujie Jiang ,Xiaolong Li Renwang Sheng Qingqiang Yao Jialin Chen | 2020 | Bioactive Materials2020,5,4: | 5 |

| 4 | Reconciliation of research on forest carbon sequestration and water conservation显示文摘Carbon sequestration and water conservation are two of the key ecosystem services that forests provide for societal need to address environmental issues.Optimization of the dual services is the ultimate goal in forest management for mitigating global climate change and safeguarding terrestrial water balance.However,there are some tradeoff s between gain in forest productivity and ecosystem water balance.We conducted literature review based on published articles for learned knowledge on forest carbon fi xation and hydrological regulations.Some knowledge gaps and research needs are identifi ed by examining the inter-connections between forest carbon sequestration and water conservation.Past researches have helped gain basic understanding of the mechanisms and controls of forest carbon fi xation and hydrological regulations as two separate issues.Tools and approaches are well established for quantifying and monitoring forest carbon and hydrological issues,operating at diff erent spatial and temporal scales.There are knowledge gaps on how to design aff orestation schemes facilitating enhanced ecosystem services in forest carbon sequestration and water conservation.For the top-down planning of aff orestation in regions where water availability is anticipated to be problematic,the questions of how much and where to plant for given land availability,known environmental implications,and sustained regional development and livelihood need to be addressed.For local management considerations,the questions of what and how to plant prevail.Eff orts are needed in joint studies of forest carbon sequestration and water conservation functionalities,specifi cally in relation to establishment and management of planted forests aiming for delivering regulatory ecosystem services in carbon sequestration,water conservation and other social values.We propose an integrated framework with dual consideration of carbon sequestration and water conservation in forest management for future research pursue. | Tanzeel Javaid Aini Farooqi Xuhua Li Zhen Yu Shirong Liu Osbert Jianxin Sun | 2021 | Journal of Forestry Research2021,32,1: | 4 |
| 5 | Determination of calorie and protein intake among acute and sub-acute traumatic brain injury patients显示文摘Purpose:Malnutrition is a common problem among hospitalized patients,especially among traumatic brain injury(TBI)patients.It is developed from hypermetabolism and the condition may worsen under the circumstance of underfeeding or incompatible dietary management.However,the data of nutrient intake especially calorie and protein among TBI patients were scarce.Hence,this study aimed to determine the calorie and protein intake among acute and sub-acute TBI patients receiving medical nutrition therapy in hospital Sultanah Nur Zahirah,Terengganu.Methods:This observational study involved 50 patients recruited from the neurosurgical ward.Method of 24 h dietary recall was utilized and combined with self-administered food diaries for 2-8 days.Food consumptions including calorie intake and protein intake were analyzed using Nutritionist PRO™(Woodinville,USA)and manual calculation based on the Malaysian food composition database(2015).Results:Patients consisted of 56%males and 44%females with the median age of 28.0(IQR=22.8-36.5)years,of which 92%were diagnosed as mild TBI and the remaining(8%)as moderate TBI.The Glasgow coma scale(GCS)was adopted to classify TBI severity with the score 13-15 being mild and 9-12 being moderate.The median length of hospital stay was 2(IQR=2.0e3.3)days.Calorie and protein intake improved significantly from day 1 to discharge day.However,the intake during discharge day was still considered as suboptimal,i.e.75%of calorie requirement,whilst the median protein intake was only 61.3%relative to protein requirement.Moreover,the average percentages of calorie and protein intakes throughout hospitalization were remarkably lower,i.e.52.2%and 41.0%,respectively.Conclusion:Although the calorie and protein intakes had increased from baseline,hospitalized TBI patients were still at a risk to develop malnutrition as the average intakes were considerably low as compared to their requirements.Optimum nutrient intakes especially calorie and protein are crucial to ensure optimum recovery process as well as to minimize risks of infection and complications. | Mohd Ibrahim Abdullah Aryati Ahmad Sharifah Wajihah Wafa Syed Saadun Tarek Wafa Ahmad Zubaidi Abdul Latif Noor Aini Mohd Yusoff Muhammad Khalis Jasmiad Nujaimin Udin Kartini Abdul Karim | 2020 | Chinese Journal of Traumatology2020,23,5: | 3 |

| 8 | Cytokine regulation of immune tolerance显示文摘The immune system provides defenses against invading pathogens while maintaining immune tolerance to self-antigens.This immune homeostasis is harmonized by the direct interactions between immune cells and the cytokine environment in which immune cells develop and function.Herein,we discuss three non-redundant paradigms by which cytokines maintain or break immune tolerance.We firstly describe how anti-inflammatory cytokines exert direct inhibitory effects on immune cells to enforce immune tolerance,followed by discussing other cytokines that maintain immune tolerance through inducing CD_(4)^(+) Foxp_(3)^(+) regulatory T cells(Tregs),which negatively control immune cells.Interleukin(IL)-2 is the most potent cytokine in promoting the development and survival of Tregs,thereby mediating immune tolerance.IL-35 is mainly produced by Tregs,but its biology function remains to be defi ned.Finally,we discuss the actions of proinflammatory cytokines that breach immune tolerance and induce autoimmunity,which include IL-7,IL-12,IL-21,and IL-23.Recent genetic studies have revealed the role of these cytokines(or their cognate receptors)in susceptibility to autoimmune diseases.Taken together,we highlight in this review the cytokine regulation of immune tolerance,which will help in further understanding of human diseases that are caused by dysregulated immune system. | Jie Wu Aini Xie Wenhao Chen | 2014 | Burns & Trauma2014,2,1: | 2 |
