| 2 | Schistosoma mansoni proteins attenuate gastrointestinal motility disturbances during experimental colitis in mice显示文摘AIM:To investigate the therapeutic effect of Schistosoma mansoni(S.mansoni) soluble worm proteins on gastrointestinal motility disturbances during experimental colitis in mice. METHODS:Colitis was induced by intrarectal injection of trinitrobenzene sulphate(TNBS) and 6 h later,mice were treated ip with S.mansoni proteins.Experiments were performed 5 d after TNBS injection.Inflammationwas quantified using validated inflammation parameters. Gastric emptying and geometric center were measured to assess in vivo gastrointestinal motility.Peristaltic activity of distal colonic segments was studied in vitro using a modified Trendelenburg set-up.Cytokine profiles of T-lymphocytes isolated from the colon were determined by real time reverse transcriptase-polymerase chain reaction. RESULTS:Intracolonic injection of TNBS caused severe colitis.Treatment with S.mansoni proteins significantly ameliorated colonic inflammation after 5 d.TNBS did not affect gastric emptying but significantly decreased the geometric center and impaired colonic peristaltic activity 5 d after the induction of colitis.Treatment with S.mansoni proteins ameliorated these in vivo and in vitro motility disturbances.In addition,TNBS injection caused a downregulation of effector T cell cytokines after 5 d,whereas a S.mansoni protein effect was no longer observed at this time point. | 3 | Phosphorus mobility in dam reservoir affected by redox oscillations:An experimental study显示文摘The internal sedimentary phosphorus(P) load of aquatic systems is able to support eutrophication, especially in dam–reservoir systems where sedimentary P stock is high and where temporary anaerobic conditions occur. The aim of this study therefore is to examine the response of sedimentary P exposed to redox oscillations. Surface sediments collected in the Champsanglard dam–reservoir(on the Creuse River, France) were subjected to two aerobic phases(10 and 12 days) alternated with two anaerobic periods(21 and 27 days)through batch incubations. The studied sediment contained 77 ± 3 μmol/g DW of P, mainly associated with the ascorbate fraction(amorphous Fe/Mn oxyhydroxides). The used sediment was rich in organic matter(OM)(21% ± 1%) with primarily allochthone signature.Our results showed that redox oscillations enhance dissolved inorganic phosphorus release at sediment/water interface. During the first anaerobic stage, the P release was mainly controlled by the dissolution/precipitation of iron minerals. The more pronounced increase of P release during the second anaerobic stage(44%) was due to various mechanisms related to the change in quality of dissolved organic matter(DOM), namely a higher SUVA254 and humification indices. The release of more refractory DOM(rDOM) served to lower the microbial metabolism activity, possibly favored iron oxyhydroxide aggregation and thus limiting iron reduction. In addition, rDOM is able to compete for mineral P sorption sites,leading to a greater P release. | 4 | 从单核细胞分化的树突状细胞的低温保存及其临床应用(英文)显示文摘树突状细胞 (dendriticcells,DCs)作为专职的抗原递呈细胞 ,已被广泛地应用于临床的肿瘤疫苗治疗。目前的临床方案大多为分次给病人注射 >10 6 个细胞 /次 ,不同批次培养的DCs,连续 4 - 6周。为了提高疗效 ,简化治疗程序及规范疗程 ,有必要将DCs低温保存 ,使患者在治疗过程中得到同批次的培养的DCs。本实验从患者外周血采取单个核细胞 ,经细胞淘洗分离单核细胞 ,在 80 0U/mlGM CSF + 10 0ng/mlIL 13的培养条件下 ,将单核细胞于Teflon疏水袋中诱导产生DCs。将DCs以 - 1℃梯度降温及不控温两种方式将DCs冻存于 - 80℃及液氮中。 1个月后 ,4 2℃快速复温。检测其免疫表型 (CD1a ,CD14,CD4 0 ,CD80 ,CD83,CD86,CD5 4 ,CD5 8,CD16,CD32 ,CD64,HLA DR)及其对自体及异体淋巴细胞的刺激作用。结果表明 ,2种方式的低温保存及复温过程不改变DCs的免疫表型及对淋巴细胞的刺激功能。结论 :应用Teflon袋 ,能够从外周血单核细胞中诱导出大批量的DCs ,这些DCs可被低温保存而不改变其功能 。 | VERG Véronique MARTINACHE Chantal LEON Anne GORIN Norbert-Claude BERNARD Jacky LOPEZ Manuel | 2000 | 中国实验血液学杂志2000，8，4： | 6 |

| 8 | Liver myofibroblasts activate protein C and respond to activated protein C显示文摘AIM:To study the protein C activation system in human liver myofibroblasts,and the effects of activated protein C(APC)on these cells.METHODS:Human liver myofibroblasts were obtained by outgrowth.Expression of protease activated receptor 1(PAR-1),endothelial protein C receptor(EPCR) and thrombomodulin(TM)was analyzed by flow cytometry.Extracellular signal-regulated kinase(ERK)1/2 activation was assessed by Western blotting using anti-phospho-ERK antibodies.Collagen synthesis was studied with real-time reverse transcription-polymerase chain reaction(RT-PCR).Activation of protein C was studied by incubating liver myofibroblasts with zymogen protein C in the presence of thrombin and detecting the generation of APC with a colorimetric assay using a peptide substrate. RESULTS:Primary cultures of human liver myofibroblasts expressed EPCR on their surface,together with PAR-1 and TM.This receptor system was functional since exposure of myofibroblasts to APC inducedERK1/2 phosphorylation in a dose-and time-dependent manner.Furthermore,APC significantly upregulated the expression of collagen mRNA,as shown by real-time RT-PCR.Collagen upregulation was controlled through the ERK pathway as it was inhibited when using the mitogen-activated protein/extracellular signal-regulated kinase kinase inhibitor PD98059.Finally,using a cell-based colorimetric assay,we showed that intact myofibroblasts converted protein C into APC in the presence of thrombin.CONCLUSION:These data suggest that APC is a new modulator of liver myofibroblast activity and contributes to the pathophysiology of chronic liver diseases. | Jennifer Gillibert-Duplantier Anne Rullier Véronique Neaud Walter Kisiel Jean Rosenbaum | 2010 | World Journal of Gastroenterology2010，16，2： | 2 |
