| 1 | Quantitation of Genital Herpes Virus DNA by Polymerase Chain Reaction and ELISA显示文摘Objective:To detect and quantitate genital herpes simplex virus (HSV) DNA in specimens from 100 patients clinically diagnosed with genital herpes. Methods: Polymerase Chain Reaction (PCR) and enzyme-linked immunosorbent assay (ELISA) were used with a standard curve of DNA copies of HSV as quantitative contrast. Results: Ninety-three cases were confirmed HSV positive and 7 cases were found to be negative. There were 58 cases of HSV-2 (62.4%) and 35 cases of HSV-1 (37.6%) among the 93 positive cases. The number of DNA plasmids ranged from 115 to 1.1×l0^5 per 250pL among the 93 positive samples (mean =7.1×10^4/250μL). The number of HSV DNA plasmids ranged from 136 to 1.1×l0^5 copies per 250pL (mean =7.6×10^4) among those with HSV-2, and 115 to 9.4×10^4 per 250pL (mean =6.3×10^4) among those with HSV-1. Meanwhile 10μL of extracted and dissolved DNA randomly taken from 8 each of HSV-2 and HSV-1 samples were tested. The number of HSV-2 DNA plasmids ranged from 35 copies to 2.7×10^4 (Mean=l.8×10^4) and the number of HSV-1 DNA ranged from 29 to 2.5×10^4 (Mean = 1.6×10^4). In the 7 negative cases, the quantity of HSV plasmids was zero. Conclusion: The sensitivity of ELISA quantitation (93%) is equal to that of Southern blot. | 3 | AIDS-Related High-Risk Behaviors and Affecting Factors of Men Who Have Sex with Men (MSM) in China's Mainland显示文摘Objectives: To describe and analyze HIV/AIDS/STD-related high risk behaviors and affecting factors of men who have sex with men (MSM) in China's Mainland~ Methods: 426 self-administered or interview-based questionnaires collected from MSM were analyzed with SPSS or SAS Systems. Results: Of the MSM, four-fifths had engaged in oral-genital intercourse, about half had engaged in anal intercourse, and in the past year more than two-fifths had casual sexual partner(s).Very few maintained monogamous sexual relationships, and nearly half also had intercourse with women. One fourth had previously been diagnosed with a venereal diseases. One HIV+individual was found among the 40 who reported having been tested for HIV. Only two men consistently used condoms among the one-third who had ever used condoms, despite almost universal knowledge of HIV and its transmission. Conclusions: MSM in China's Mainland are very sexually active, highlighting the possibility of a serious HIV epidemic that may be directly spread to the female population. | 4 | Low Frequencies of CCR5-Δ32 and CCR5-m303,but High Frequencies of CCR2-641 and SDF1-3'A Alleles in Indigenous Ethnic Groups in China's Mainland显示文摘Objective:The aim in this study was to identify the allelic frequencies of the chemokine (SDF1-3'A) and chemokine receptor (CCR5△32, CCR5m303 and CCR2-64I) genes resistant to HIV-1 infection and/or disease progression in indigenous Chinese populations. Methods: By using QIAamp DNA Blood Mini Kit, the genomic DNA samples were purified from whole peripheral blood of healthy individuals (n=2067) from Han, Uygur, Mongolian and Tibetan ethnic groups, as well as Han patients including HIV-1 carriers (n=330), patients with other sexually transmitted diseases (STDs, n=259) and intravenous drug users (IVDUs, n=125). The allelic polymorphisms were identified by means of PCR or PCR-RFLP analyses. The sequences of randomly selected amplified PCR products were further confirmed by direct DNA sequencing. Results: The mutant frequencies were identified to be 0%-3.48% for CCR5△32, 0% for CCR5m303,19.15%-28.79% for CCR2-64 and 19.10%-28.73% for SDF1-3'A alleles, respectively, in Chinese healthy individuals from four ethnic groups. Our findings indicated the allelic frequencies vary among the different ethnic groups. Furthermore, the HIV-1 carriers, STD cases and IVDUs (all of Han ethnicity) were found to have the allelic frequencies of 0%-0.19% (CCR5A32), 0% (CCR5m303), 19.31%-20.45% (CCR2-64) and 25.61%-26.83% (SDF1-3'A) with minor variations in their frequencies between the patients and healthy Han groups. There was no CCR5-m303 mutation found in any subject in this study. Conclusion: The examined subjects of four Chinese ethnic origins showed lower frequencies of CCR5A32 and CCR5m303 alleles, but higher frequencies of mutant CCR264I and SDF1-3'A alleles compared to those identified in northern-European and American Caucasians. Methods: Data were collected from 240 Chinese gays whowere attracted by their same gender. Results: The extent of sex among China's Mainland gays issimilar to that of industrialized countries, and behaviors that exacerbate the HIV epidemic are universal.Perceptions “being a girl is better”, “ Dislike toys likeswords or pistols in early childhood were predictors topassive anal intercourse in adulthood. Those gay men whobegan sexual intercourse younger than 16 years old tendedto have more sex partners and suffered more pain. Thosedisliked gender of male or self-regarded as female tended tohave more passive anal intercourse and related to childhood cross-gender experience/psychology while those were raised as girls by their parentsdid not take on the above characters. Individuals who experienced sex coercion and drug abuse were the twosubgroups with the greatest high-risk behaviors. Individualswho had sex with females had the greatest number of malesex partners and were more prone to group sex. Individualswith pedophilia or gerontophilia had more casual sexpartners. Psychological problems encountered by Chinesegays related to suicide attempts and commitment. Conclusions : The prospect of an extensive AIDSepidemic among Chinese gays does exist and somesubgroups of gays play more important roles in theepidemic.
