| 1 | Features of colorectal cancer in China stratified by anatomic sites:A hospital-based study conducted in university-affiliated hospitals from 2014 to 2018显示文摘Objective:The clinical and biological characteristics of colorectal cancer have been found to differ depending on the anatomic site of the cancer.However,for Chinese patients,there is limited information on the proportion of cases at each site and the related features.In this study,we explored the location,distribution and other features of colorectal cancers at each anatomic site in Chinese patients.Methods:We conducted a hospital-based study using hospitalization summary reports from 10 Peking University-affiliated hospitals from 2014 to 2018;the reports covered a total of 2,097,347 hospitalizations.Incident cases were chosen as the study population,and their epidemiological features were further analyzed.Results:A total of 20,739 colorectal cancer patients were identified.Rectum was the most common location(48.3%)of the cancer,whereas the proportions of patients with distal and proximal colon cancer were 24.5%and18.6%,respectively.Patients with rectal cancer were predominantly male and were the youngest for all anatomical sites(each P<0.001).The highest proportion of emergency admissions,the longest hospital stays and the highest hospitalization costs were found in patients with proximal colon cancer(each P<0.001).The proximal colon cancer subgroup included the highest proportions of patients with medical histories of cholecystectomy,cholecystolithiasis and/or gallbladder polyps and appendectomy(P=0.009,P<0.001 and P<0.001,respectively).The distal colon cancer subgroup included the highest proportions of patients with medical histories of diabetes and hypertension(P<0.001,respectively).Conclusions:The patterns of colorectal cancer observed in this study differ from those reported for Western patients and show a significantly higher proportion of patients with rectal cancer.Different epidemiological features were also found based on anatomic sites.Further studies based on tumor location should be conducted to facilitate more accurate screening and treatment. | Ruize Qu Yanpeng Ma Liyuan Tao Xiaoyuan Bao Xin Zhou Bingyan Wang Fei Li Siyi Lu Lin Tuo Siyan Zhan Zhipeng Zhang Wei Fu | 2021 | Chinese Journal of Cancer Research2021,33,4: | 10 |

| 3 | Hippocampal insulin resistance and the Sirtuin 1 signaling pathway in diabetes-induced cognitive dysfunction显示文摘In the peripheral nervous system,the activation of Sirtuin 1 can improve insulin resistance;however,the role played by Sirtuin 1 in the central nervous system remains unknown.In this study,rat models of diabetes mellitus were generated by a single injection of streptozotocin.At 8 weeks after streptozotocin injection,the Morris water maze test and western blot assays confirmed that the diabetic model rats had learning and memory deficits,insulin resistance,and Sirtuin 1 expression could be detected in the hippocampus.Insulin and the insulin receptor inhibitor S961 were intranasally administered to investigate the regulatory effects of insulin signaling on Sirtuin 1.The results showed that insulin administration improved the impaired cognitive function of diabetic model rats and increased the expression levels of phosphorylated insulin receptor,phosphorylated insulin receptor substrate 1,and Sirtuin 1 in the hippocampus.Conversely,S961 administration resulted in more severe cognitive dysfunction and reduced the expression levels of phosphorylated insulin receptor,phosphorylated insulin receptor substrate 1,and Sirtuin 1.The Sirtuin 1 activator SRT2104 and the inhibitor Sirtinol were injected into the lateral ventricle,which revealed that the activation of Sirtuin 1 increased the expression levels of target of rapamycin complex 1,phosphorylated cAMP-response elementbinding protein,and brain-derived neurotrophic factor.Hippocampal dendritic length and spine density also increased in response to Sirtuin 1 activation.In contrast,Sirtinol decreased the expression levels of target of rapamycin complex 1,phosphorylated cAMP-response elementbinding protein,and brain-derived neurotrophic factor and damaged the dendritic structure.These findings suggest that the Sirtuin 1 signaling pathway plays an important role in the development of insulin resistance-related cognitive deficits in diabetic rats.This study was approved by the Animal Ethics Welfare Committee of the First Affiliated Hospital of Hunan University of Chinese Medicine(approval No.ZYFY201811207)in November 2018. | Hui Yang Lin Tang Zhan Qu Shi-Hui Lei Wei Li Yu-Hong Wang | 2021 | Neural Regeneration Research2021,16,12: | 6 |
| 4 | Effects of preventive acupuncture or moxibustion at the Guanyuan acupoint on expression of interleukin-2 and its receptors in hypothalamus, serum, and uterus of menopausal rats显示文摘BACKGROUND: Previous studies have indicated preventive acupuncture and moxibustion at the Guanyuan point have positive effects on the immune system of menopausal rats. OBJECTIVE: To observe the effects of preventive acupuncture or moxibustion at the Guanyuan point on interleukin-2 (IL-2) and its receptor (IL-2R mRNA) levels in hypothalamus, serum, and uterus of menopausal rats. DESIGN: Randomized control animal experiment. SETTING: School of Acupuncture and Moxibustion, Beijing University of Chinese Medicine. MATERIALS: This study was performed at the Laboratory of Beijing University of Chinese Medicine from November 2004 to July 2005. The vaginal smear method was used to determine the estrus cycle of 9.5-month-old Sprague Dawley (SD) female rats. Three continuous estrus cycles were measured, and 160 rats with estrus cycle disorder, weighing (300 ± 20) g, were included in this study. The rats were randomly divided into four groups: preventive acupuncture group (n =48), preventive moxibustion group (n =48), model groups (n =48), and 10-month-old group (n =16). Sixteen 3.5-month-old healthy SD female rats, weighing (240 ± 20) g, were also assigned to the young group. All rats were clean grade and were purchased from the Laboratory Animal Center, Beijing University of Chinese Medicine. The experiment was in accordance with animal ethics standards. METHODS: Acupuncture was applied to the preventive acupuncture group at 10 months of age. The needle was inserted upward and perpendicularly two fen to a point that was two fen below the Guanyuan point. The rats freely moved around during needle retention (20 minutes). Acupuncture was administered twice a week and continued for eight weeks. A moxa cone (small) moxibustion was applied to the preventive moxibustion group at 10 months of age. The rats were caught gently and kept at the same position as the preventive acupuncture group. The moxa cone was ignited and held to the Guanyuan point directly. Only one moxa cone was used each time. After moxibustion, the local skin turned slightly red. Moxibustion was used twice a week and continued for eight weeks. Nothing except gently catching and lying was administered to the model group and the young group. MAIN OUTCOME MEASURES: Radioimmunoassay, in situ hybridization, and similar methods were employed to detect IL-2 in serum and the uterus, as well as IL-2R mRNA expression in the hypothalamus and uterus arcuate nuclei, of the preventive acupuncture group, preventive moxibustion group, model groups (at 12, 14, and 16 months), 10 month old group (at 10 months), and the young group (at 4 months). RESULTS: All 176 rats were included in the final analysis. ① IL-2R mRNA: IL-2R mRNA expression decreased significantly in the uterus of 12-, 14-, and 16-month-old model groups and in the hypothalamus of 14- and 16-month-old model groups (P < 0.05-0.01). Compared to the same month-aged model groups, the expression of IL-2R mRNA increased significantly in the hypothalamus and uterus of 16-month-old preventive acupuncture or moxibustion groups (P < 0.05-0.01).② IL-2: IL-2 in serum of 12- and 14-month-old model groups and in the uterus of 12-, 14-, and 16-month-old model groups decreased significantly compared to the young group (P < 0.05-0.01). IL-2 serum levels increased significantly in the 16-month-old group with preventive moxibustion; in addition, IL-2 expression levels increased significantly in the uterus of the 12-month-old group with preventive moxibustion, as well as the 12- and 16-month-old groups with preventive acupuncture (P < 0.05-0.01). CONCLUSION: As a rat ages and becomes menopausal, IL-2R mRNA expression in the hypothalamus anduterus, as well as IL-2 levels in the serum and uterus, decrease. However, preventive acupuncture and preventive moxibustion can repress the decrease to some extent. | 5 | Antibody Cocktail Exhibits Broad Neutralization Activity Against SARS-CoV-2 and SARS-CoV-2 Variants显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)has precipitated multiple variants resistant to therapeutic antibodies.In this study,12 high-affinity antibodies were generated from convalescent donors in early outbreaks using immune antibody phage display libraries.Of them,two RBD-binding antibodies(F61 and H121)showed high-affinity neutralization against SARS-Co V-2,whereas three S2-target antibodies failed to neutralize SARS-Co V-2.Following structure analysis,F61 identified a linear epitope located in residues G446–S494,which overlapped with angiotensinconverting enzyme 2(ACE2)binding sites,while H121 recognized a conformational epitope located on the side face of RBD,outside from ACE2 binding domain.Hence the cocktail of the two antibodies achieved better performance of neutralization to SARS-Co V-2.Importantly,these two antibodies also showed efficient neutralizing activities to the variants including B.1.1.7 and B.1.351,and reacted with mutations of N501 Y,E484 K,and L452 R,indicated that it may also neutralize the recent India endemic strain B.1.617.The unchanged binding activity of F61 and H121 to RBD with multiple mutations revealed a broad neutralizing activity against variants,which mitigated the risk of viral escape.Our findings revealed the therapeutic basis of cocktail antibodies against constantly emerging SARS-Co V-2 variants and provided promising candidate antibodies to clinical treatment of COVID-19 patients infected with broad SARS-Co V-2 variants. | Yuanyuan Qu Xueyan Zhang Meiyu Wang Lina Sun Yongzhong Jiang Cheng Li Wei Wu Zhen Chen Qiangling Yin Xiaolin Jiang Yang Liu Chuan Li Jiandong Li Tianlei Ying Dexin Li Faxian Zhan Youchun Wang Wuxiang Guan Shiwen Wang Mifang Liang | 2021 | Virologica Sinica2021,36,5: | 3 |
| 6 | Green Synthesis of Magnetic Adsorbent Using Groundwater Treatment Sludge for Tetracycline Adsorption显示文摘Groundwater treatment sludge is an industrial waste that is massively produced from groundwater treatment plants.Conventional methods for treatment of this sludge,such as discharge into deep wells or the sea,or disposal at landfills,are not environmentally sustainable.Here,we demonstrate an alternative strategy to recycle the sludge by preparing a magnetic maghemite adsorbent via a one-step hydrothermal method with NaOH solution as the only solvent.With this method,the weakly magnetized sludge,which contained 33.2%iron(Fe)and other impurities(e.g.,silicon(Si),aluminum(Al),and manganese(Mn)),was converted to magnetic adsorbent(MA)with the dissolution of Si/Al oxides(e.g.,quartz and albite)into the liquid fraction.At a NaOH concentration of 2 mol L^-1,approximately 18.1%of the ferrihydrite in the Fe oxides of the sludge was converted into 11.2%maghemite and 6.9%hematite after the hydrothermal treatment.MA2(i.e.,MA produced by a 2 mol L^-1 NaOH concentration)exhibited a good magnetic response of 8.2 emu g^-1(1 emu=10^-3 A m^2),and a desirable surface site concentration of 0.75 mmol g^-1.The synthesized MA2 was used to adsorb the cationic pollutant tetracycline(TC).The adsorption kinetics of TC onto MA2 fitted well with a pseudo-second-order model,and the adsorption isotherms complied well with the Langmuir model.The maximum adsorption capacity of MA2 for TC was 362.3 mg g^-1,and the main mechanism for TC adsorption was cationic exchange.This study is the first to demonstrate the preparation of an MA from recycled sludge without a reductant and/or exogenous Fe source.The prepared adsorbent can be used as a low-cost adsorbent with high capacity for TC sorption in the treatment of TC-containing wastewater. | Zhan Qu Yaqiong Wu Suiyi Zhu Yang Yu Mingxin Huo Leilei Zhang Jiakuan Yang Dejun Bian Yi Wang | 2019 | Engineering2019,5,5: | 2 |
