| 1 | Efficient elimination of multidrug-resistant bacteria using copper sulfide nanozymes anchored to graphene oxide nanosheets显示文摘Antibacterial nanomaterials have attracted growing interest for bacterial infection therapy.However,most nanomaterials eliminate bacteria either physically or chemically,which hampers their efficacy when dealing with multidrug-resistant bacteria.To overcome this,we integrated copper sulfide(CuS)nanoparticles with active graphene oxide nanosheets(GO NSs)to synthesize a superior nanocomposite(CuS/GO NC)that acts both physically and chemically on the bacteria.CuS/GO NC was produced using a facile hydrothermal method,whereby the CuS nanoparticles grew and were uniformly dispersed on the GO NSs in situ.We found that the CuS/GO NC possesses a unique needle-like morphology that physically damages the bacterial cell membrane.CuS/GO NC also exhibits high oxidase-and peroxidase-like activity,ensuring efficient generation of the reactive oxygen species•OH from H2O2,which kills bacteria chemically.These features endow the CuS/GO NC with excellent antibacterial capabilities to kill multidrug-resistant bacteria such as methicillin-resistant Staphylococcus aureus(MRSA)with only a single dose.Additionally,it was found that the CuS/GO NC accelerated the healing of infected wounds in vivo owing to its good biocompatibility as well as facilitation of cell migration and collagen secretion.This study provides a new strategy to combine the physical and chemical antibacterial modes of nanomaterials to develop more effective therapies to combat multidrug-resistant bacterial infections. | Wanshun Wang Binglin Li Huili Yang Zefeng Lin Lingling Chen Zhan Li Jiayuan Ge Tao Zhang Hong Xia Lihua Li Yao Lu | 2020 | Nano Research2020,13,8: | 8 |

| 7 | Micheliolide exerts effects in myeloproliferative neoplasms through inhibiting STAT3/5 phosphorylation via covalent binding to STAT3/5 proteins显示文摘Ruxolitinib is a cornerstone of management for some subsets of myeloproliferative neoplasms(MPNs);however,a considerable number of patients respond suboptimally.Here,we evaluated the efficacy of micheliolide(MCL),a natural guaianolide sesquiterpene lactone,alone or in combination with ruxolitinib in samples from patients with MPNs,JAK2V617F-mutated MPN cell lines,and a Jak2V617F knock-in mouse model.MCL effectively suppressed colony formation of hematopoietic progenitors in samples from patients with MPNs and inhibited cell growth and survival of MPN cell lines in vitro.Co-treatment with MCL and ruxolitinib resulted in greater inhibitory effects compared with treatment with ruxolitinib alone.Moreover,dimethylaminomicheliolide(DMAMCL),an orally available derivative of MCL,significantly increased the efficacy of ruxolitinib in reducing splenomegaly and cytokine production in Jak2V617F knock-in mice without evident effects on normal hematopoiesis.Importantly,MCL could target the Jak2V617F clone and reduce mutant allele burden in vivo.Mechanistically,MCL can form a stable covalent bond with cysteine residues of STAT3/5 to suppress their phosphorylation,thus inhibiting JAK/STAT signaling.Overall,these findings suggest that MCL is a promising drug in combination with ruxolitinib in the setting of suboptimal response to ruxolitinib. | Huijun Huang Jinqin Liu Lin Yang Yiru Yan Meng Chen Bing Li Zefeng Xu Tiejun Qin Shiqiang Qu Liang Wang Gang Huang Yue Chen Zhijian Xiao | 2023 | Blood Science2023,5,4: | 0 |

| 9 | Biomimetic three-layered membranes comprising(poly)-e-caprolactone,collagen and mineralized collagen for guided bone regeneration显示文摘The distinct structural properties and osteogenic capacity are important aspects to be taken into account when developing guided bone regeneration membranes.Herein,inspired by the structure and function of natural periosteum,we designed and fabricated using electrospinning a fibrous membrane comprising(poly)-e-caprolactone(PCL),collagen-I(Col)and mineralized Col(MC).The three-layer membranes,having PCL as the outer layer,PCL/Col as the middle layer and PCL/Col/MC in different ratios(5/2.5/2.5(PCM-1);3.3/3.3/3.3(PCM-2);4/4/4(PCM-3)(%,w/w/w))as the inner layer,were produced.The physiochemical properties of the different layers were investigated and a good integration between the layers was observed.The three-layeredmembranes showed tensile properties in the range of those of natural periosteum.Moreover,the membranes exhibited excellent water absorption capability without changes of the thickness.In vitro experiments showed that the inner layer of the membranes supported attachment,proliferation,ingrowth and osteogenic differentiation of human bone marrowderived stromal cells.In particular cells cultured on PCM-2 exhibited a significantly higher expression of osteogenesis-related proteins.The three-layered membranes successfully supported new bone formation inside a critical-size cranial defect in rats,with PCM-3 being the most efficient.The membranes developed here are promising candidates for guided bone regeneration applications. | Jingjing Wu Mengyu Yao Yonggang Zhang Zefeng Lin Wenwu Zou Jiaping Li Pamela Habibovic Chang Du | 2021 | Regenerative Biomaterials2021,8,6: | 0 |
| 10 | Genetic analysis of the seed dehydration process in maize based on a logistic model显示文摘Seed moisture at harvest is a critical trait affecting maize quality and mechanized production,and is directly determined by the dehydration process after physiological maturity.However,the dynamic nature of seed dehydration leads to inaccurate evaluation of the dehydration process by conventional determination methods.Seed dry weight and fresh weight were recorded at 14 time points after pollination in a recombinant inbred line(RIL)population derived from two inbred lines with contrasting seed dehydration dynamics.The dehydration curves of RILs were determined by fitting trajectories of dry weight accumulation and dry weight/fresh weight ratio change based on a logistic model,allowing the estimation of eight characteristic parameters that can be used to describe dehydration features.Quantitative trait locus(QTL)mapping,taking these parameters as traits,was performed using multiple methods.Single-trait QTL mapping revealed 76 QTL associated with dehydration characteristic parameters,of which the phenotypic variation explained(PVE)was 1.03%to 15.24%.Multipleenvironment QTL analysis revealed 21 related QTL with PVE ranging from 4.23%to 11.83%.Multiple-trait QTL analysis revealed 58 QTL,including 51 pleiotropic QTL.Combining these mapping results revealed 12 co-located QTL and the dehydration process of RILs was divided into three patterns with clear differences in dehydration features.These results not only deepen general understanding of the genetic characteristics of seed dehydration but also suggest that this approach can efficiently identify associated genetic loci in maize. | Shuangyi Yin Jun Liu Tiantian Yang Pengcheng Li Yang Xu Huimin Fang Shuhui Xu Jie Wei Lin Xue Derong Hao Zefeng Yang Chenwu Xu | 2020 | The Crop Journal2020,8,2: | 0 |
