3D printing of bone tissue engineering scaffolds显示文摘Tissue engineering is promising in realizing successful treatments of human body tissue loss that current methods cannot treat well or achieve satisfactory clinical outcomes. In scaffold-based bone tissue engineering,a high performance scaffold underpins the success of a bone tissue engineering strategy and a major direction in the field is to produce bone tissue engineering scaffolds with desirable shape, structural, physical, chemical and biological features for enhanced biological performance and for regenerating complex bone tissues.Three-dimensional(3D)printing can produce customized scaffolds that are highly desirable for bone tissue engineering. The enormous interest in 3D printing and 3D printed objects by the science, engineering and medical communities has led to various developments of the 3D printing technology and wide investigations of 3D printed products in many industries, including biomedical engineering, over the past decade. It is now possible to create novel bone tissue engineering scaffolds with customized shape, architecture, favorable macro-micro structure, wettability, mechanical strength and cellular responses. This article provides a concise review of recent advances in the R&D of 3D printing of bone tissue engineering scaffolds. It also presents our philosophy and research in the designing and fabrication of bone tissue engineering scaffolds through 3D printing. | Chong Wang Wei Huang Yu Zhou Libing He Zhi He Ziling Chen Xiao He Shuo Tian Jiaming Liao Bingheng Lu Yen Wei Min Wang | 2020 | Bioactive Materials2020,5,1: | 20
Global antimicrobial resistance:a system-wide comprehensive investigation using the Global One Health Index显示文摘Background:Antimicrobial resistance(AMR)is one of the top ten global public health challenges. However, given the lack of a comprehensive assessment of worldwide AMR status, our objective is to develop a One Health-based system-wide evaluation tool on global AMR.Methods:We have further developed the three-hierarchical Global One Health Index(GOHI)-AMR indicator scheme, which consists of five key indicators,17 indicators, and 49 sub-indicators,by incorporating 146 countries'data from diverse authoritative databases, including WHO's Global Antimicrobial Resistance and Use Surveillance System(GLASS)and the European CDC.We investigated the overall-or sub-rankings of GOHI-AMR at the international/regional/national levels for data preprocessing and score calculation utilizing the existing GOHI methodology.Additionally,a correlation analysis was conducted between the GOHI-AMR and other socioeconomic factors. Results:The average GOHI-AMR score for 146 countries is 38.45.As expected,high-income countries(HICs)outperform the other three income groups on overall rankings and all five key indicators of GOHI-AMR,whereas lowincome countries unexpectedly outperform upper-middle-income countries and lower-middle-income countries on the antibiotics-resistant key indicator(ARR)and ARR-subordinate indicators, including carbapenem-,β-lactam-,and quinolone resistance, and even HICs on aminoglycoside resistance. There were no significant differences among the four groups on the environmental-monitoring indicator(P>0.05).GOHI-AMR was positively correlated with gross domestic product, life expectancy, and AMR-related publications, but negatively with natural growth rate and chronic respiratory disease. In contrast to Cyprus, the remarkably lower prevalence of'ESKAPE pathogens'in high-scoring Sweden and Denmark highlights Europe's huge gaps.China and Russia outperformed the other three BRICS countries on all key indicators, particularly India's ARR and Brazil's AMR laboratory network and coordination capacity.Furthermore, significant internal disparities in carbapenem-resistant Klebsiella pneumoniae(CRKP)and methicillin-resistant Staphylococcus aureus(MRSA)prevalence were observed between China and the USA,with MRSA prevalence both gradually declining, whereas CRKP prevalence has been declining in the USA but increasing in China, consistent with higher carbapenems-related indicator'performance in USA.Conclusions:GOHI-AMR is the most comprehensive tool currently available for the assessment of AMR status worldwide. We discovered unique features impacting AMR in each country and offered precise recommendations to improve the capacity to tackle AMR in low-ranking countries. | Nan Zhou Zile Cheng Xiaoxi Zhang Chao Lv Chaoyi Guo Haodong Liu Ke Dong Yan Zhang Chang Liu Yunfu Chang Sheng Chen Xiaokui Guo Xiao-Nong Zhou Min Li Yongzhang Zhu | 2022 | Infectious Diseases of Poverty2022,11,4: | 2

Chaperone-mediated Autophagy Regulates Cell Growth by Targeting SMAD3 in Glioma显示文摘Previous studies suggest that the reduction of SMAD3(mothers against decapentaplegic homolog 3)has a great impact on tumor development, but its exact pathological function remains unclear. In this study,we found that the protein level of SMAD3 was greatly reduced in human-grade IV glioblastoma tissues,in which LAMP2A(lysosome-associated membrane protein type 2A)was significantly up-regulated.LAMP2A is a key ratelimiting protein of chaperone-mediated autophagy(CMA),a lysosome pathway of protein degradation that is activated in glioma. We carefully analyzed the amino-acid sequence of SMAD3 and found that it contained a pentapeptide motif biochemically related to KFERQ,which has been proposed to be a targeting sequence for CMA.In vitro,we confirmed that SMAD3 was degraded in either serum-free or KFERQ motif deleted condition, which was regulated by LAMP2A and interacted with HSC70(heat shock cognate 71 kDa protein).Using isolated lysosomes,amino-acid residues 75 and 128 of SMAD3 were found to be of importance for this process, which affected the CMA pathway in which SMAD3 was involved.Similarly,down-regulating SMAD3 or up-regulating LAMP2A in cultured glioma cells enhanced their proliferation and invasion.Taken together, these results suggest that excessive activation of CMA regulates glioma cell growth by promoting the degradation of SMAD3.Therefore,targeting the SMAD3-LAMP2Amediated CMA-lysosome pathway may be a promising approach in anti-cancer therapy. | Hanqun Liu Yuxuan Yong Xingjian Li Panghai Ye Kai Tao Guoyou Peng Mingshu Mo Wenyuan Guo Xiang Chen Yangfu Luo Yuwan Lin Jiewen Qiu Ziling Zhang Liuyan Ding Miaomiao Zhou Xinling Yang Lin Lu Qian Yang Pingyi Xu | 2022 | Neuroscience Bulletin2022,38,6: | 0
Sodium carbonate-assisted synthesis of hierarchically porous single-crystalline nanosized zeolites显示文摘Hierarchically porous single-crystalline nanosized zeolites as heterogeneous catalysts show great potential in fine chemistry because they offer more rich hierarchically porous channels for the mass transfer and molecular diffusion. However, the synthesis of hierarchically porous nanosized zeolites generally requires the assistance of templates acting as the mesoporogens, which limits its popularity. Herein,we report a one-pot and template-free synthesis of hierarchically porous single-crystalline nanosized zeolite beta only by introducing sodium carbonate in precursor solution. The resulted sample features the extraordinary properties, including the uniform nanocrystal(200–300 nm), high pore volume(0.65 cm^3g^(-1)) and the hierarchical pore-size distribution(e.g. 2–8 and 90–150 nm). After slicing processing, it is interestingly found that a large number of interconnected mesopores penetrate throughout whole material, which enables the hierarchically porous nanosized zeolite beta remarkably superior catalytic activity than the conventional zeolite beta in condensation of benzaldehyde with ethanol at room temperature. More importantly, this one-pot sodium carbonate-assisted synthetic strategy is highly versatile, which has also been successfully developed to synthesize hierarchically porous nanosized singlecrystalline zeolites ZSM-5 and TS. | Xiaoxia Zhou Yu Chen Tongguang Ge Zile Hua Hangrong Chen Jianlin Shi | 2017 | Science Bulletin2017,62,14: | 0
Cleaner geopolymer prepared by co-activation of gasification coal fly ash and steel slag:durability properties and economic assessment显示文摘Geopolymer is a material with high early strength. However, the insufficient durability properties, such as long-term strength,acid-base resistance,freeze-thaw resistance, leaching toxicity, thermal stability, sulfate resistance and carbonation resistance, restrain its practical application.Herein,a longterm stable geopolymer composite with high final strength(ASK1)was synthesized from shell coal gasification fly ash(SFA)and steel slag(SS).Additionally,a geopolymer composite with high early strength(ASK2)was also synthesized for comparison. The results showed that ASK1 exhibited better performance on freezing-thawing resistance, carbonization resistance and heavy metals stabilization compared to the ASK2 at long-term curing.Raising the curing temperature could accelerate the unconfined compressive strength(UCS)development at initial curing ages of 3 to 7 d.Both ASK1 and ASK2 exhibited excellent acid-base and sulfate corrosion resistance. An increase for UCS was seen under KOH solution and MgSO_(4)solution corrosion for ASK1.All leaching concentrations of heavy metals out of the two geopolymers were below the standard threshold, even after 50 freezingthawing cycles. Both ASK1 and ASK2 geopolymer concrete exhibited higher sustainability and economic efficiency than Portland cement concrete. The result of this study not only provides a suitable way for the utilization of industrial solid waste in civil and environmental engineering, but also opens a new approach to improve the long-term stabilities of the geopolymers. | Xian Zhou Xia Chen Ziling Peng Yongmen Zhou Yan Li Wang Jian Zeyu Fan Yuchi Chen | 2023 | Frontiers of Environmental Science & Engineering2023,17,12: | 0
Decellularized Extracellular Matrix Containing Electrospun Fibers for Nerve Regeneration:A Comparison Between Core–Shell Structured and Preblended Composites显示文摘Advanced biomaterial-based strategies for treatment of peripheral nerve injury require precise control over both topological and biological cues for facilitating rapid and directed nerve regeneration. As a highly bioactive and tissue-specifc natural material, decellularized extracellular matrix(dECM)derived from peripheral nerves(decellularized nerve matrix,DNM)has drawn increasing attention in the feld of regenerative medicine, due to its outstanding capabilities in facilitating neurite outgrowth and remyelination. To induce and maintain sufcient topological guidance, electrospinning was conducted for fabrication of axially aligned nanofbers consisting of DNM and poly(ε-caprolactone)(PCL).Core–shell structured fbers were prepared by coaxial electrospinning using DNM as the shell and PCL as the core.Compared to the aligned electrospun fbers using preblended DNM/PCL,the core–shell structured fbers exhibited lower tensile strength, faster degradation, but considerable toughness for nerve guidance conduit preparation and relatively intact fbrous structure after long-term degradation.More importantly, the full DNM surface coverage of the aligned core–shell fbers efectively promoted axonal extension and Schwann cells migration. The DNM contents further triggered neurite bundling and myelin formation toward nerve fber maturation and functionalization.Herein,we not only pursue a multi-functional scafold design for nerve regeneration,a detailed comparison between core–shell structured and preblended electrospinning of DNM/PCL composites was also provided as an applicable paradigm for advanced tissue-engineered strategies using dECM-based biomaterials. | Rongli Deng Ziling Luo Zilong Rao Zudong Lin Shihao Chen Jing Zhou Qingtang Zhu Xiaolin Liu Ying Bai Daping Quan | 2022 | Advanced Fiber Materials2022,4,3: | 0
