1ACPAs promote IL-1β production in rheumatoid arthritis by activating the NLRP3 inflammasome显示文摘OBJECTIVES:Anti-citrullinated protein antibodies(ACPAs)are a group of autoantibodies targeted against citrullinated proteins/peptides and are informative rheumatoid arthritis(RA)biomarkers.ACPAs also play a crucial role in RA pathogenesis,and their underlying mechanism merits investigation.METHODS:Immunohistochemical(IHC)assays were carried out to determine IL-1βlevels in ACPA+and ACPA−RA patients.PBMCderived monocytes were differentiated into macrophages before stimulation with ACPAs purified from RA patients.The localization and interaction of molecules were analyzed by confocal microscopy,co-IP,and surface plasmon resonance.RESULTS:In our study,we found that IL-1βlevels were elevated in ACPA+RA patients and that ACPAs promoted IL-1βproduction by PBMC-derived macrophages.ACPAs interacted with CD147 to enhance the interaction between CD147 and integrinβ1 and,in turn,activate the Akt/NF-κB signaling pathway.The nuclear localization of p65 promoted the expression of NLRP3 and pro-IL-1β,resulting in priming.Moreover,ACPA stimulation activated pannexin channels,leading to ATP release.The accumulated ATP bound to the P2X7 receptor,leading to NLRP3 inflammasome activation.CONCLUSIONS:Our study suggests a new hypothesis regarding IL-1βproduction in RA involving ACPAs,which may be a potential therapeutic target in RA treatment.Xiwen Dong Zhaohui Zheng Peng Lin Xianghui Fu Fanni Li Jianli Jiang Ping Zhu 2020Cellular & Molecular Immunology2020,17,3:11
2Analysis of DNA methylation variation in wheat genetic background after alien chromatin introduction based on methylation-sensitive amplification polymorphism显示文摘During the process of alien germplasm introduced into wheat genome by chromosome engineering, extensive genetic variations of genome structure and gene expression in recipient could be induced. In this study, we performed GISH (genome in situ hybridization) and AFLP (amplified fragment length po-lymorphism) on wheat-rye chromosome translocation lines and their parents to detect the identity in genomic structure of different translocation lines. The results showed that the genome primary struc-ture variations were not obviously detected in different translocation lines except the same 1RS chromosome translocation. Methylation sensitive amplification polymorphism (MSAP) analyses on genomic DNA showed that the ratios of fully-methylated sites were significantly increased in translo-cation lines (CN12, 20.15%; CN17, 20.91%; CN18, 22.42%), but the ratios of hemimethylated sites were significantly lowered (CN12, 21.41%; CN17, 23.43%; CN18, 22.42%), whereas 16.37% were fully-me-thylated and 25.44% were hemimethylated in case of their wheat parent. Twenty-nine classes of me-thylation patterns were identified in a comparative assay of cytosine methylation patterns between wheat-rye translocation lines and their wheat parent, including 13 hypermethylation patterns (33.74%), 9 demethylation patterns (22.76%) and 7 uncertain patterns (4.07%). 3Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 4Experimental analysis on air-to-water sound transmission loss in shallow water显示文摘The research of propagation characteristics of air-to-water sound transmission is of great importance to the detection of aerial targets from underwater.In order to study the propagation characteristics of air-to-water sound transmission in shallow water,State Key Laboratory of Acoustics,Institute of Acoustics,conducted an experiment in the South China Sea in March,2013.During the experiment,multi-frequency signals transmitted by a hooter hung on a research ship were received by an underwater hydrophone,and the distance between the hooter and the hydrophone was from 2.4 km to 9.8 km approximately.Through analyzing experimental data in this work,the experimental air-to-water transmission loss at frequencies128 Hz and 256 Hz is estimated up to 9.8 km in range,and its oscillation structure is evident.The wave-number integration approach is used to simulate theoretical air-to-water transmission losses,which are in good agreement with experimental values and to explain the experimental air-to-water sound transmission characteristics.ZHANG Lingshan PENG Zhaohui WANG Guangxu LUO Wenyu 2017Chinese Journal of Acoustics2017,36,2:7

6Anomalous sound propagation due to the horizontal variation of seabed acoustic properties显示文摘The sound propagation in shallow water is greatly influenced by the acoustic properties of seabed. An anomalous transmission loss was observed in an experiment, and a range dependent bottom model with horizontal variation of seabed acoustic property is proposed and could be well used to explain the anomalous phenomena. 7Sound propagation from the shelfbreak to deep water显示文摘Motivated by a phenomenon in an experiment conducted in the Northwestern Pacific indicating that the energy of the received signal around the sound channel axis is much greater than that at shallower depths,we study sound propagation from the transitional area(shelfbreak)to deep water.Numerical simulations with different source depths are first performed,from which we reach the following conclusions.When the source is located near the sea surface,sound will be strongly attenuated by bottom losses in a range-independent oceanic environment,whereas it can propagate to a very long range because of the continental slope.When the source is mounted on the bottom in shallow water,acoustic energy will be trapped near the sound channel axis,and it converges more evidently than the case where the source is located near the sea surface.Then,numerical simulations with different source ranges are performed.By comparing the relative energy level in the vertical direction between the numerical simulations and the experimental data,the range of the air-gun source can be approximated.QIN JiXing ZHANG RenHe LUO WenYu PENG ZhaoHui LIU JianJun WANG DeJun 2014Science China(Physics,Mechanics & Astronomy)2014,57,6:3
8Moment tensor analysis of transversely isotropic shale based on the discrete element method显示文摘In this study,the moment tensor of transversely isotropic shale was analyzed using a discrete element method-acoustic emission model(DEM-AE model).Firstly,the failure modes of the shale obtained from the acoustic emission(AE) events and physical experiments were compared.Secondly,the relationships between AE events and seismic magnitudes,and AE events and the resulting cracks were analyzed.Finally,a moment tensor T-k chart describing the seismic source was introduced to demonstrate the differences in the transversely isotropic shale.The results showed that,for different anisotropy angles,a linear logarithmic relationship existed between the cumulative AE events and the seismic magnitude in the concentration area of the AE events.A normal distribution was observed for the number of AE events as the seismic magnitude changed from small to large.The moment tensor T-k chart indicated that the number and proportion of linear tension cracks in the shale were highest.When θ = 30°,the peak seismic magnitude was at a minimum.The average seismic magnitude in the concentration area of the AE events was also relatively small.Points close to the U=-1/3V line and the number of cracks included in a single AE event were at a minimum,and the corresponding peak stress also reached its lowest level.In contrast,when θ=90°,all related parameters were contrary to the above θ = 30° case.The DEM-AE model and the moment tensor T-k chart are suitable for analyzing the distribution of shale cracks appearing during the loading process.This study can provide constructive references for future research on the fracturing treatment of shale.Chong Zhaohui Li Xuehua Hou Peng Chen Xiangyu Wu Yuechao 2017International Journal of Mining Science and Technology2017,27,3:3

13Effects of exogenic chloride on oxidative degradation of chlorinated azo dye by UV-activated peroxodisulfate显示文摘Recently,the degradation of organic compounds in saline dye wastewater by sulfate radicals(SO4·-)-based advanced oxidation processes(AOPs) have attracted much attention.However,previous studies on these systems have selected non-chlorinated dyes as model compounds,and little is known about the transformation of chlorinated dyes in such systems.In this study,acid yellow 17(AY-17) was selected as a model of chlorinated contaminants,and the degradation kinetics and evolution of oxidation byproducts were investigated in the UV/PDS system.AY-17 can be efficiently degraded(over 98% decolorization)under 90 min irradiation at pH 2.0-3.0,and the reaction follows pseudo-first order kinetics.Cl-accelerated the degradation of AY-17,but simultaneously led to an undesirable increase of absorbable organic halogen(AOX).Several chlorinated byproducts were identified by liquid chromatography-mass spectrometry(LC-MS/MS) in the UV/PDS system.It indicates that endogenic chlorine and exogenic Cl-reacted with SO_(4)·- to form chloride radicals,which are involved in the dechlorination and rechlorination of AY-17 and intermediates.The possible degradation mechanisms of AY-17 photooxidative degradation are proposed.This work provides valuable information for further studies on the role of exogenic chloride in the degradation of chlorinated azo dyes and the kinetic parameters in the PDS-based oxidation process.Wenya Peng Yu Fu Lingli Wang Yifan Wang Yongxia Dong Ying Huang Zhaohui Wang 2021Chinese Chemical Letters2021,32,8:1

16Numerical modeling of the dynamic variation in multiphase CH_(4) during CO_(2) enhanced gas recovery from depleted shale reservoirs显示文摘Regarding CO_(2)enhanced shale gas recovery,this work focuses on changes in the multiphase(free/adsorbed)CH_(4)in the process of CO_(2)enhanced shale gas recovery,by utilizing a rigorous numerical model with real geological parameters.This work studies nine injection well(IW)and CH_(4)production well(PW)combinations of CO_(2)to determine the influence of IW and PW locations on the dynamic interaction of multiphase CH_(4)during 10000 d of CO_(2)injection.The results indicate that the content of both the adsorbed CH_(4)and free CH_(4)is strongly variable before(and during)the CO_(2)-CH_(4)displacement.In addition,during the simulation process,the proportion of the adsorbed CH_(4)among all extracted CH_(4)phases dynamically increases first and then tends to stabilize at 70%-80%.Moreover,the IW-PWs combinations signifi-cantly affect the outcomes of CO_(2)enhanced shale gas recovery-for both the proportion of adsorbed/free CH_(4)and the recovery efficiency.A longer IW-PW distance enables more adsorbed CH_(4)to be recovered but results in a lower efficiency of shale gas recovery.Basically,a shorter IW-PWs distance helps recover CH_(4)via CO_(2)injection if the IW targets the bottom layer of the Wufeng-Longmaxi shale formation.This numerical work expands the knowl-edge of CO_(2)enhanced gas recovery from depleted shale reservoirs.Lijun CHENG Ye ZHANG Jun LIU Zhaohui LU Chunlin ZENG Peng ZHAO 2021Frontiers of Earth Science2021,15,4:1

19A lipid-based LMP2-mRNA vaccine to treat nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is a serious and highly invasive epithelial malignancy that is closely associated with Epstein‒Barr virus(EBV).Due to the lack of therapeutic vaccines for NPC,we selected EBV latent membrane protein 2(LMP2)as a preferable targeting antigen to develop a lipid-based LMP2-mRNA(mLMP2)vaccine.Full-length mLMP2 expressing LMP2 was first synthesized using an in vitro transcription method and then encapsulated into(2,3-dioleacyl propyl)trimethylammonium chloride(DOTAP)-based cationic liposomes to obtain the mRNA vaccine(LPX-mLMP2).The cell assays showed that the antigenpresenting cells were capable of highly efficient uptake of LPX-mLMP2 and expression of LMP2.LMP2 could subsequently be presented to form the peptide-major histocompatibility complex(pMHC).Furthermore,LPX-mLMP2 could accumulate in the spleen,express antigens,promote the maturation of dendritic cells and stimulate antigen-specific T-cell responses in vivo.It dramatically inhibited the tumor growth of the LMP2-expressing tumor model after three doses of vaccination.Additionally,the proliferation of antigen-specific T cells in the tumor site made a good sign for the promise of mRNA vaccines in virus-induced cancer.Overall,we provided a newly developed antigen-encoding mRNA vaccine with advantages against NPC.We also demonstrated that mRNA vaccines are attractive candidates for cancer immunotherapy.Mengran Guo Xing Duan Xingchen Peng Zhaohui Jin Hai Huang Wen Xiao Qian Zheng Yongqi Deng Na Fan Kepan Chen Xiangrong Song 2023Nano Research2023,16,4:1
