2miR-21 promotes the differentiation of hair folliclederived neural crest stem cells into Schwann cells显示文摘Hair follicle-derived neural crest stem cells can be induced to differentiate into Schwann cells in vivo and in vitro. However, the underlying regulatory mechanism during cell differentiation remains poorly understood. This study isolated neural crest stem cells from human hair follicles and induced them to differentiate into Schwann cells. Quantitative RT-PCR showed that microRNA(miR)-21 expression was gradually increased during the differentiation of neural crest stem cells into Schwann cells. After transfection with the miR-21 agonist(agomir-21), the differentiation capacity of neural crest stem cells was enhanced. By contrast, after transfection with the miR-21 antagonist(antagomir-21), the differentiation capacity was attenuated. Further study results showed that SOX-2 was an effective target of miR-21. Without compromising SOX2 mRNA expression, miR-21 can down-regulate SOX protein expression by binding to the 3′-UTR of miR-21 mRNA. Knocking out the SOX2 gene from the neural crest stem cells significantly reversed the antagomir-21 inhibition of neural crest stem cells differentiating into Schwann cells. 4Apparent diffusion coefficient evaluation for secondary changes in the cerebellum of rats after middle cerebral artery occlusion显示文摘Supratentorial cerebral infarction can cause functional inhibition of remote regions such as the cerebellum,which may be relevant to diaschisis.This phenomenon is often analyzed using positron emission tomography and single photon emission CT.However,these methods are expensive and radioactive.Thus,the present study quantified the changes of infarction core and remote regions after unilateral middle cerebral artery occlusion using apparent diffusion coefficient values.Diffusion-weighted imaging showed that the area of infarction core gradually increased to involve the cerebral cortex with increasing infarction time.Diffusion weighted imaging signals were initially increased and then stabilized by 24 hours.With increasing infarction time,the apparent diffusion coefficient value in the infarction core and remote bilateral cerebellum both gradually decreased,and then slightly increased 3–24 hours after infarction.Apparent diffusion coefficient values at remote regions(cerebellum)varied along with the change of supratentorial infarction core,suggesting that the phenomenon of diaschisis existed at the remote regions.Thus,apparent diffusion coefficient values and diffusion weighted imaging can be used to detect early diaschisis.Yunjun Yang Lingyun Gao Jun Fu Jun Zhang Yuxin Li Bo Yin Weijian Chen Daoying Geng 2013Neural Regeneration Research2013,8,31:3
5Effect of volatile oil from Blumea Balsamifera(L.) DC. leaves on wound healing in mice显示文摘OBJECTIVE: To assess the effectiveness of violateoil from Blumea Balsamifera(L.) DC. leaves(BB oil)on wound healing in mice.METHODS: Undiluted BB oil and its diluted solutions with olive oil to 1/5 and 1/10 to yield BB oil1/5and BB oil1/10 were applied to the wounded skin before wound healing conditions were assessed by healing rate, histopathology, and contents of collagen, hydroxyproline, and Neuropeptide Substance P(SP). All above results were compared with the efficacies of the control, pure olive oil, basic fibroblast growth factor(BFGF), and cream of Jing Wan Hong(JWH).RESULTS: BB oil1/5and BB oil1/10 improved wound contraction and closure. Histopathology study further confirmed a desirable histological organization of wound tissues. BB oil1/5and BB oil1/10 reduced the number of inflammatory cells, increased wound-healing rates, and significantly increased the hydroxyproline content. Both BB oil1/5and BB oil1/10 improved formation of collagen, and reduced the frequency of fibroblasts. Moreover, BB oil1/5and BB oil1/10 markedly promoted SP expression. However, undiluted BB oil may induce skin thickening and hardening, inhibite collagen synthesis and delay complete skin wound healing.CONCLUSION: The BB oil1/5and BB oil1/10 promoted capillary regeneration, blood circulation, collagen deposition, granular tissue formation, epithelial deposition, and wound contraction. 6Occurrence, variations, and risk assessment of neonicotinoid insecticides in Harbin section of the Songhua River, northeast China显示文摘Neonicotinoid insecticides(NNIs)have been intensively used and exploited,resulting in their presence and accumulation in multiple environmental media.We herein investigated the current levels of eight major NNIs in the Harbin section of the Songhua River in northeast China,providing the first systematic report on NNIs in this region.At least four NNIs in water and three in sediment were detected,with total concentrations ranging from 30.8 to 135 ng L^(-1) and from 0.61 to 14.7 ng g^(-1) dw,respectively.Larger spatial variations in surface water NNIs concentrations were observed in tributary than mainstream(p<0.05)due to the intensive human activities(e.g.,horticulture,urban landscaping,and household pet flea control)and the discharge of wastewater from many treatment plants.There was a significant positive correlation(p<0.05)between the concentrations of residual imidacloprid(IMI),clothianidin(CLO),and S4NNIs in the sediment and total organic carbon(TOC).Due to its high solubility and low octanol-water partition coefficient(Kow),the sediment-water exchange behavior shows that NNIs in sediments can re-enter into the water body.Human exposure risk was assessed using the relative potency factor(RPF),which showed that infants have the highest exposure risk(estimated daily intake(SIMIeq EDI):31.9 ng kg^(-1) bw$d^(-1)).The concentration thresholds of NNIs for aquatic organisms in the Harbin section of the Songhua River were determined using the species sensitivity distribution(SSD)approach,resulting in a value of 355 ng L^(-1) for acute hazardous concentration for 5%of species(HC5)and 165 ng L^(-1) for chronic HC5.Aquatic organisms at low trophic levels were more vulnerable to potential harm from NNIs.Zhikun Liu Song Cui Leiming Zhang Zulin Zhang Rupert Hough Qiang Fu Yi-Fan Li Lihui An Mingzhi Huang Kunyang Li Yuxin Ke Fuxiang Zhang 2021Environmental Science and Ecotechnology2021,,4:2
7Time-sensitivity mechanism of rock stress memory properties under tensile stress显示文摘In deep underground engineering,understanding of time-related stress memory properties is critical to evaluate the in situ stress conditions of a rock mass.In this study,the time-sensitivity mechanism of the rock stress memory properties under tensile stress was investigated.It was found that the material property(Poisson’s ratio)and crack angle were the controlling factors of the Kaiser effect(KE)under tensile stress.In particular,the time-sensitivity of the stress memory properties was closely related to the crack growth path.When the failure of the rock specimen was dominated by tensile microcracks and the crack development direction was deflected by up to 30°in the successive loading process,the stress memory capacity was likely to be time-independent for a sandstone specimen.The distribution of the Felicity ratio in a Brazilian test was more discrete than that in a three-point bending test It also showed that the changes in the crack path,rather than the time interval between successive loading cycles,led to inaccuracy of the detected KE.This study provides insights into stress memory-related issues under uniaxial or more complex stress conditions and thus facilitates development of methods for testing in situ mechanical behaviors of rocks with acoustic emission(AE)technology.Yuxin Ban Xiang Fu Qiang Xie Jun Duan 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,3:1

10The O2-ZmGRAS11 transcriptional regulatory network orchestrates the coordination of endosperm cell expansion and grain filling in maize显示文摘Maize(Zea mays)endosperm filling is coordinated with cell expansion to enlarge the grain size,but the mechanism coupling the two processes is poorly understood.Starchy endosperm cells basically contain no visible vacuoles for cell expansion.During grain filling,efficient synthesis of storage compounds leads to reduced cytoplasm and thus lowered cell turgor pressure.Although bioactive gibberellins(GAs)are essential for cell expansion,they accumulate at a low level at this stage.In this study,we identified an endosperm-specific GRAS domain-containing protein(ZmGRAS11)that lacks the DELLA domain and promotes cell expansion in the filling endosperm.The zmgras11 loss-of-function mutants showed normal grain filling but delayed cell expansion,thereby resulting in reduced kernel size and weight.Overexpression of ZmGRAS11 led to larger endosperm cells and therefore increased kernel size and weight.Consistent with this,ZmGRAS11 positively regulates the expression of ZmEXPB12,which is essential for cell expansion,at the endosperm filling stage.Moreover,we found that Opaque2(O2),a central transcription factor that regulates endosperm filling,could directly bind to the promoter of ZmGRAS11 and activate its expression.Taken together,these results suggest that endosperm cell expansion is coupled with endosperm filling,which is orchestrated by the O2-ZmGRAS11-centered transcriptional regulatory network.Our findings also provide potential targets for maize yield improvement by increasing the storage capacity of endosperm cells.Chen Ji Lina Xu Yujie Li Yuxin Fu Shuai Li Qiong Wang Xing Zeng Zhongqin Zhang Zhiyong Zhang Wenqin Wang Jiechen Wang Yongrui Wu 2022Molecular Plant2022,15,3:1
11Heat wave mitigation of ecosystems in mountain areas-a case study of the Upper Yangtze River basin显示文摘Background:Natural ecosystems,such as forests and grasslands,can mitigate heat waves,but research on heat wave mitigation in mountain ecosystems is lacking.Considering the upper reaches of the Yangtze River basin(URYB)as the study area,we first divided the URYB into mountainous and non-mountainous areas based on a digital elevation model(DEM).Then,we used temperature to identify heat waves and used the temperature and humidity index to identify suitable days.Finally,the differences between mountainous and non-mountainous areas were compared,and regional development strategies are proposed.Result:(1)The frequency and duration of heat waves increased by 0.433/y(P<0.1),0.07/y(P<0.05),while the number of suitable days in June,July,and August gradually decreased slightly by 0.0096/y(P<0.1),0.0125/y(P<0.1),0.004/y(P<0.1),respectively from 1986 to 2015.(2)The average monthly number of suitable days was the highest in mountainous areas during the summer from 1986 to 2015.Mountainous areas have an advantage in mitigating heat waves owing to landform-based changes in the local climate.(3)In addition to the landform,altitude was the main factor responsible for mitigating heat waves in mountainous areas.Conclusion:Parts of the plateau and whole mountain areas were found to be best for avoiding extremely high temperatures based on the number of suitable summer days from 1986 to 2015.We suggest formulating tourism promotion strategies,and strengthening tourism infrastructure,considering landscape protection and creation to promote sustainable regional development.Yuxin Zhang Bin Fu Juying Sun 2022Ecosystem Health and Sustainability2022,8,1:0
12ZnO Films on Transferable and Low Thermal Resistance Graphite Substrate Grown by Ultrasonic Spray Pyrolysis显示文摘ZnO thin films were deposited on graphite substrates by ultrasonic spray pyrolysis method with Zn(CH 3 COO) 2 ·2H 2 O aqueous solution as precursor. The crystalline structure, morphology, and optical properties of the as-grown ZnO films were investigated systematically as a function of deposition temperature and growth time. Near-band edge ultraviolet(UV) emission was observed in room temperature photoluminescence spectra for the optimized samples, yet the usually observed defect related deep level emissions were nearly undetectable, indicating that high optical quality ZnO thin fi lms could be achieved via this ultrasonic spray pyrolysis method. 14Genome editing of PAR2 through targeted delivery of CRISPR-Cas9 system for alleviating acute lung inflammation via ERK/NLRP3/IL-1β and NO/iNOS signalling显示文摘Excessive and uncontrollable inflammatory responses in alveoli can dramatically exacerbate pulmonary disease progressions through vigorous cytokine releases,immune cell infiltration and protease-driven tissue damages.It is an urgent need to explore potential drug strategies for mitigating lung inflammation.Protease-activated receptor 2(PAR2)as a vital molecular target principally participates in various inflammatory diseases via intracellular signal transduction.However,it has been rarely reported about the role of PAR2 in lung inflammation.This study applied CRISPR-Cas9 system encoding Cas9 and sg RNA(p Cas9-PAR2)for PAR2 knockout and fabricated an anionic human serum albuminbased nanoparticles to deliver p Cas9-PAR2 with superior inflammation-targeting efficiency and stability(TAP/p Cas9-PAR2).TAP/p Cas9-PAR2 robustly facilitated p Cas9-PAR2 to enter and transfect inflammatory cells,eliciting precise gene editing of PAR2 in vitro and in vivo.Importantly,PAR2 deficiency by TAP/p Cas9-PAR2 effectively and safely promoted macrophage polarization,suppressed proinflammatory cytokine releases and alleviated acute lung inflammation,uncovering a novel value of PAR2.It also revealed that PAR2-mediated pulmonary inflammation prevented by TAP/p Cas9-PAR2was mainly dependent on ERK-mediated NLRP3/IL-1β and NO/i NOS signalling.Therefore,this work indicated PAR2 as a novel target for lung inflammation and provided a potential nanodrug strategy for PAR2 deficiency in treating inflammatory diseases.Xin Zhuo Yue Wu Xiujuan Fu Jianbin Li Yuxin Xiang Xiaoyu Liang Canquan Mao Yuhong Jiang 2024Acta Pharmaceutica Sinica B2024,14,3:0
15Aldolase A promotes proliferation and G1/S transition via the EGFR/MAPK pathway in non-small cell lung cancer显示文摘Background:Our previous study demonstrated that aldolase A(ALDOA)is overexpressed in clinical human lung squamous cell carcinoma and that ALDOA promotes epithelial-mesenchymal transition and tumorigenesis.The pre-sent study aimed to explore the function of ALDOA in the modulation of non-small cell lung cancer(NSCLC)prolifera-tion and cell cycle progression and the potential mechanism.Methods:ALDOA was knocked down by short hairpin RNA in H520 and H1299 cells.ALDOA was overexpressed with vectors carrying the full-length ALDOA sequence in H1299 and H157 cells.The proliferation capacities were assessed with immunohistochemical staining,Cell Counting Kit-8 and colony formation assays.The cell cycle distribution was examined by flow cytometry,and molecular alterations were determined by western blotting.Cell synchronization was induced with nocodazole.The stability of cyclin D1 mRNA was tested.The pyruvate kinase M2 and ALDOA protein distributions were examined.Aerobic glycolysis was evaluated with Cell Titer-Glo assay,glucose colorimetric assay and lactate colorimetric assay.Results:ALDOA knockdown inhibited the proliferation and G1/S transition in H520 cells.Conversely,ALDOA over-expression promoted the proliferation and G1/S transition in H157 cells.The cell cycle synchronization assay showed that ALDOA expression increased in the G1 phase and G1/S transition.Furthermore,ALDOA knockdown reduced cyclin D1 expression by regulating epidermal growth factor receptor/mitogen-activated protein kinase(EGFR/MAPK)pathway.Similar results were found in H1299 and H157 cells.The inhibition of mitogen-activated protein kinase kinase 1/2 prompted the nuclear distribution of ALDOA.Additionally,ALDOA knockdown reduced nuclear distribution of PKM2,the extracellular lactate and intracellular adenosine triphosphate concentrations and elevated the extracellular glucose concentration.Conclusions:ALDOA contributed to activation of the EGFR/MAPK pathway,thus promoting cyclin D1 expression and enhancing proliferation and G1/S transition in NSCLC.Additionally,ALDOA facilitated NSCLC aerobic glycolysis.Hailu Fu Huijun Gao Xiaoyu Qi Lei Zhao Donghua Wu Yuxin Bai Huimin Li Xuan Liu Jun Hu Shujuan Shao 2018Cancer Communications2018,38,1:0
16Large scalable,ultrathin and self-cleaning cellulose aerogel film for daytime radiative cooling显示文摘Passive cooling strategy shows great potential in mitigating global warming and reducing energy consumption.Because of the high emissivity in the atmospheric transparency window(λ≈8–13μm),cellulose is considered as a good candidate for radiative cooling.However,traditional cellulose coolers generally show poor solar reflection and can be polluted by dust outside,thereby resulting in poor daytime cooling efficiency.To address these drawbacks,we developed sustainable cellulose nanowhiskers(CNWs)/ZnO composite aerogel films with favorable optical performance,mechanical robustness,and self-cleaning function for efficient daytime radiative cooling,which can be achieved via freeze casting and hot-pressing process.Due to formation of multi-level porous structure and chemical bonds(Si-O-C/Si-O-Si),such aerogel film exhibited high solar reflectance(97%)and high infrared emittance(92.5%).It achieved a sub-ambient temperature drop of 6.9℃under direct sunlight in hot weather.Most importantly,the surface roughness and low surface energy enable cellulose aerogel film hydrophobicity(contact angle=133°),thereby resulting in an anti-dust function.This work provides insight into the design of sustainable thermal regulating materials to realize carbon neutrality.Chenyang Cai Yuanbo Sun Yi Chen Zechang Wei Yibo Wang Fuling Chen Wanquan Cai Jiawen Ji Yuxin Ji Yu Fu 2023Journal of Bioresources and Bioproducts2023,8,4:0
17Lamellar-assembled PdNi super-nanosheets as effective oxygen redox dual-electrocatalysts for rechargeable Zn-air batteries显示文摘Exploration of bifunctional electrocatalysts toward effective oxygen reduction reaction(ORR)and oxygen evolution reaction(OER)is pivotal for developing high-efficiency and rechargeable metal-air batteries but remains great challenging.Here we elaborately synthesize lamellar-assembled PdNi super-nanosheets(SNSs)with an optimized Pd/Ni molar ratio to serve as attractive ORR and OER bifunctional electrocatalysts for rechargeable high-powered Zn-air batteries(ZABs).The products are layer-by-layer stackings of ultrathin PdNi nanosheet motifs.On the drastically extended two-dimensional(2D)surface,the inserted Ni atoms can substantially lower down the d-band center of surface Pd toward improved ORR kinetics and concurrently create oxytropic NiOx sites to adsorb–OH groups for promoting the reverse OER electrocatalysis.Specifically,the ORR mass activity and specific activity of the primary Pd_(92)Ni_(8)SNSs attain 2.5 A·mg^(−1)and 3.15 mA·cm^(−2),which are respectively 14 and 9 times those of commercial Pt/C.Meanwhile,the OER activity and stability of Pd_(92)Ni_(8)SNSs/C distinctly outperform those of the RuO_(2)benchmark,suggesting excellent reversible oxygen electrocatalysis.The power density of the ZAB with Pd_(92)Ni_(8)SNSs/C as the air cathode is 2.7 times higher than that by Pt/C benchmark.Significantly,it can last for over 150 h without significant performance degradation during the charge–discharge cycle test.This work showcases a feasible strategy for developing self-assembled multimetallic 2D nanomaterials with excellent bifunctional catalytic performances toward energy conversion applications.Luhong Fu Kai Liu Chao Zhen Yuxin Zhu Zixi Lyu Guifen Du Shuifen Xie 2023Nano Research2023,16,2:0
18Recent development of chemical components in propolis显示文摘Propolis is a resinous substance collected by honeybees from various plant sources.On account of its chemical composition,propolis possesses several biologi-cal and pharmacological properties.In recent years,many papers have provided information concerning its composi-tion.This review compiles data from most studies of propolis,focusing on the chemical composition of ethanol extracts of propolis(EEP),water extracts of propolis(WEP),and volatile oils from propolis(VOP).The characteristic compounds of EEP are polyphenols includ-ing flavonoids and related phenolic acids,and flavonoids are the most abundant and effective parts.They are considered to contribute more to the antibacterial,antiviral,and antioxidant effects than the other constituents.The main flavonoids in EEP are pinocembrin,galangin,chrysin,quercetin,kaempferol,and naringenin.The constituents reported to be in WEP include phenolic acids,caffeoylquinic acid,3-mono-O-caffeoylquinic acid,caffeic acid,flavonoids,etc.The propolis volatile compounds are benzyl alcohol,benzyl acetate,cinnamic alcohol,vanillin,eudensmol,cyclohexyl benzoate,and benzyl benzoate,which are responsible for several biological properties.As a natural mixture,propolis is widely used in medicine and cosmetics,as well as being a constituent of health foods.Since propolis has been used extensively,information on its composition is not only of interest to the academic field,but also to propolis users.Yuanjun XU Liping LUO Bin CHEN Yuxin FU 2009Frontiers in Biology2009,4,4:0
19Sedimentary characteristics and model of gravity flow channel-lobe complex in a depression lake basin:A case study of Chang 7 Member of Triassic Yanchang Formation in southwestern Ordos Basin,NW China显示文摘To reveal the development characteristics and distribution of gravity flow sedimentary system under micro-paleogeomorphologic units of the Chang 7 Member of Triassic Yanchang Formation in the southwestern Ordos Basin,on the basis of the restoration of the paleogeomorphological form of the Chang 7 depositional period by the impression method,each micro-paleogeomorphologic unit was depicted in-depth,and the characteristics and development models of gravity flow deposits in the study area were studied in combination with outcrop,core,mud logging and log data.The results show that:(1)The paleogeomorphology in the Chang 7 depositional period was an asymmetrical depression,wide and gentle in the northeast and steep and narrow in the southwest.Three sub-paleogeomorphologic units were developed in the basin,including gentle paleo-slope,paleo-slope and paleo-depression,and they can be further subdivided into eight micro-paleogeomorphologic units:bulge,groove,slope break belt,plain of lake bottom,deep depression of lake bottom,paleo-channel,paleo-ridge of lake bottom,and paleo-uplift of lake bottom.(2)There are 9 types of lithofacies and 4 types of lithofacies assemblages of Chang 7 Member.According to lithofacies composition and lithofacies vertical combination,the gravity flow deposit is further divided into 5 types of microfacies:restricted channel,unrestricted channel,natural levee,inter-channel,lobe.(3)Paleogeomorphology plays an important role in controlling sediment source direction,type and spatial distribution of sedimentary microfacies,genetic types and distribution of sand bodies in Chang 7 Member.LYU Qiqi FU Jinhua LUO Shunshe LI Shixiang ZHOU Xinping PU Yuxin YAN Hongguo 2022Petroleum Exploration and Development2022,49,6:0
20Self-assembly multifunctional DNA tetrahedron for efficient elimination of antibiotic-resistant bacteria显示文摘Antibiotic resistance is a major challenge in the clinical treatment of bacterial infectious diseases.Herein,we constructed a multifunctional DNA nanoplatform as a versatile carrier for bacteria-specific delivery of clinical antibiotic ciprofloxacin(CIP)and classic nanoantibiotic silver nanoparticles(AgNP).In our rational design,CIP was efficiently loaded in the self-assembly double-bundle DNA tetrahedron through intercalation with DNA duplex,and single-strand DNA-modified AgNP was embedded in the cavity of the DNA tetrahedron through hybridization.With the site-specific assembly of targeting aptamer in the well-defined DNA tetrahedron,the bacteria-specific dual-antibiotic delivery system exhibited excellent combined bactericidal properties.With enhanced antibiotic accumulation through breaking the out membrane of bacteria,the antibiotic delivery system effectively inhibited biofilm formation and promoted the healing of infected wounds in vivo.This DNAbased antibiotic delivery system provides a promising strategy for the treatment of antibiotic-resistant infections.Tiantian Wu Yu Fu Shuang Guo Yanqiang Shi Yuxin Zhang Zhijin Fan Bin Yang Baoquan Ding Yuhui Liao 2024Aggregate2024,5,1:0
