2Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

4Histological Subtypes of Lung Cancer in Chinese Males from 2000 to 2012显示文摘Objective To characterize the histological and epidemiological features of male lung cancer patients in China. Methods The demographic and histological information about male lung cancer patients identified from 2000-01-01 to 2012-12-31,was collected from the Cancer Hospital of the Chinese Academy of Medical Sciences. Relative frequencies(RF) were estimated for major histological subtypes and compared according to the years of diagnosis and birth. Results The RF of adenocarcinoma(ADC) increased from 21.96% to 43.36% and the RF of squamous cell carcinoma(SCC) decreased from 39.11% to 32.23% from 2000 to 2012 in the 15 427 male lung cancer patients included in this study(Z=17.909,P<0.0001; Z=-6.117,P<0.0001). The RF of ADC increased from 28.72% in 2000-2004,36.88% in 2005-2008 to 48.61% in 2009-2012 in patients born after 1960. The age-adjusted RF of ADC in 2007-2012 increased consistently in all the investigated areas. Conclusion The increased RF of ADC in male lung cancer patients highlights the need for further investigation of the etiologic factors of these tumors. 5Preliminary results of environmental monitoring of the natural gas hydrate production test in the South China Sea显示文摘Natural gas hydrate (NGH)is considered as one of the new clean energy sources of the 21st century with the highest potential.The environmental issues of NGH production have attracted the close attention of scientists in various countries.From May 10 to July 9,2017,the first offshore NGH production test in the South China Sea (SCS)was conducted by the China Geological Survey.In addition,environmental security has also been effectively guaranteed via a comprehensive environmental monitoring system built during the NGH production test.The monitoring system considered sea-surface atmosphere methane and carbon dioxide concentrations,dissolved methane in the sea water column,and the seafloor physical oceanography and marine chemistry environment.The whole process was monitored via multiple means, in multiple layers,in all domains,and in real time.After the production test,an environmental investigation was promptly conducted to evaluate the environmental impact of the NGH production test. 8Ginsenoside Rg3 inhibits colon cancer cell migration by suppressing nuclear factor kappa B activity显示文摘OBJECTIVE: To study the mechanism of the inhibitory effect of ginsenoside Rg3 on colon cancer cell migration.METHODS: Transwell migration assays were performed to investigate the inhibitory effect of ginsenoside Rg3 on SW480 cell migration. Electrophoretic mobility shift assays(EMSAs) and dual luciferase reporter assays were used to study the suppression capability of Rg3 on nuclear factor kappa B(NF-κB) activity. Western blotting was adopted to determine protein levels.RESULTS: Two-hundred micromolar ginsenoside Rg3 significantly inhibited SW480 cell migration(P < 0.05). EMSA showed that Rg3 suppressed the DNA binding ability of NF-κB. Dual luciferase reporter assay showed that Rg3 decreased NF-κB-regulated gene transcription(P < 0.01). Western blots indicated that Rg3 down-regulated expression of the NF-κB-regulated matrix metalloproteinase 9,cyclooxygenase-2 and C-Myc. An NF-κB inhibitor,pyrrolidine dithiocarbamate,enhanced the inhibitory effect of Rg3 on SW480 cell migration.CONCLUSION: Ginsenoside Rg3 has a strong antitumor migration capability by suppressing NF-κB activity and expression of NF-κB-regulated gene products. 10Multiscale carbon foam confining single iron atoms for efficient electrocatalytic CO2 reduction to CO显示文摘Electrocatalytic CO2 reduction to CO is a sustainable process for energy conversion.However,this process is still hindered by the diffusi limited mass transfer,low electrical conductivity and catalytic activity.Therefore,new strategies for catalyst design should be adopted to solve these problems and improve the electrocatalytic performa nee for CO production.Herein,we report a multiscale carb on foam confining〔single iron atoms prepared with the assistant of S1O2 template.The pore-enriched environment at the macro-scale facilitates the diffusion of reacta nts and products.The graphe ne nano sheets at the nano-scale promote the charge tran sfer duri ng the reaction.The single iron atoms con fined in carb on matrix at the atomic-scale provide the active sites for electrocatalytic CO2 reductio n to CO.The optimized catalyst achieves a CO Faradaic efficiency of 94.9%at a moderate potential of-0.5 V vs.RHE.Furthermore,the performance can be maintained over 60 hours due to the stable single iron atoms coordi nated with four n itroge n atoms in the carb on matrix.This work provides a promising strategy to improve both the activity and stability of single atom catalysts for electrocatalytic CO2 reduction to CO.Zheng Zhang Chao Ma Yunchuan Tu Rui Si Jie Wei Shuhong Zhang Zhen Wang Jian-Feng Li Ye Wang Dehui Deng 2019Nano Research2019,12,9:11
11How to identify dislocations in molecular dynamics simulations?显示文摘Dislocations are of great importance in revealing the underlying mechanisms of deformed solid crystals.With the development of computational facilities and technologies,the observations of dislocations at atomic level through numerical simulations are permitted.Molecular dynamics(MD)simulation suggests itself as a powerful tool for understanding and visualizing the creation of dislocations as well as the evolution of crystal defects.However,the numerical results from the large-scale MD simulations are not very illuminating by themselves and there exist various techniques for analyzing dislocations and the deformed crystal structures.Thus,it is a big challenge for the beginners in this community to choose a proper method to start their investigations.In this review,we summarized and discussed up to twelve existing structure characterization methods in MD simulations of deformed crystal solids.A comprehensive comparison was made between the advantages and disadvantages of these typical techniques.We also examined some of the recent advances in the dynamics of dislocations related to the hydraulic fracturing.It was found that the dislocation emission has a significant effect on the propagation and bifurcation of the crack tip in the hydraulic fracturing.LI Duo WANG Feng Chao YANG Zhen Yu ZHAO Ya Pu 2014Science China(Physics,Mechanics & Astronomy)2014,57,12:11
12Antibiotic growth promoter olaquindox increases pathogen susceptibility in fish by inducing gut microbiota dysbiosis显示文摘Low dose antibiotics have been used as growth promoters in livestock and fish. The use of antibiotics has been associated with reduced pathogen infections in livestock. In contrast, antibiotic growth promoter has been suspected of leading to disease outbreaks in aquaculture. However, this phenomenon is circumstantial and has not been confirmed in experimental conditions. In this study,we showed that antibiotic olaquindox increased the susceptibility of zebrafish to A. hydrophila infection. Olaquindox led to profound alterations in the intestinal microbiota of zebrafish, with a drastic bloom of Enterobacter and diminishing of Cetobacterium. Moreover, the innate immune responses of zebrafish were compromised by olaquindox(P<0.05). Transfer of microbiota to GF zebrafish indicated that while the immuo-suppression effect of olaquindox is a combined effect mediated by both OLA-altered micro biota and direct action of the antibiotic(P<0.05), the increased pathogen susceptibility was driven by the OLA-altered microbiota and was not dependent on direct antibiotic effect. 13Toxic Effects of Atrazine on Reproductive System of Male Rats显示文摘Objective This study was designed to evaluate the toxic effects of Atrazine(ATZ) on the reproductive system of male rats. Methods Male Sprague-Dawley rats were exposed to ATZ by gavage at dosages of 0, 38.5, 77, and 154 mg/kg bw/day for 30 d. The toxic effects of ATZ to rats were assessed through histopathologcal observation, spermatozoa quality evaluation, testicular marker enzyme indicators, antioxidant capacity and reproductive hormone levels. Results Significant adverse effects on reproductive system were observed in rats exposed to ATZ at different dosages compared with 0 mg/kg group, including an irregular and disordered arrangement of the seminiferous epithelium in 154 mg/kg group; a decreased spermatozoa number and an increased spermatozoa abnormality rate in 77 and 154 mg/kg groups; decreased levels of acid phosphatase(ACP), alkaline phosphatase(AKP), lactic dehydrogenase(LDH), and succinate dehydrogenase(SDH) with the increasing of ATZ concentration; a decreased level of total antioxidant capacity(TAC) in a dose-dependent manner, and a decreased reduced glutathione(GSH) level and an increased malondialdehyde(MDA) content in 154 mg/kg group; and decreased serum levels of testosterone(T) and inhibin-B(INH-B) and an increased serum level of follicle stimulating hormone(FSH) in 77 and 154 mg/kg groups, and an increased serum level of luteinizing hormone(LH) in 154 mg/kg group. 16Role of hydrogen sulfide in portal hypertension and esophagogastric junction vascular disease显示文摘AIM:To investigate the association between endogenous hydrogen sulfide(H2S)and portal hypertension as well as its effect on vascular smooth muscle cells.METHODS:Portal hypertension patients were categorized by Child-Pugh score based on bilirubin and albumin levels,prothrombin time,ascites and hepatic encephalopathy.Plasma H2S concentrations and portal vein diameters(PVDs)were compared between portal hypertension patients and control participants,as well as between portal hypertension patients with varying degrees of severity.In addition,we established a rabbit hepatic schistosomiasis portal hypertension(SPH)model and analyzed liver morphology,fibrosis grade,plasma and liver tissue H2S concentrations,as well as cystathionineγ-lyase(CSE)activity and phosphorylated extracellular signal-regulated kinase(pERK)1/2,B cell lymphoma(Bcl)-2 and Bcl-XL expression in portal vein smooth muscle cells,in addition to their H2S-induced apoptosis rates.RESULTS:In portal hypertension patients,endogenous H2S levels were significantly lower than those in healthy controls.The more severe the disease was,the lower were the H2S plasma levels,which were inversely correlated with PVD and Child-Pugh score.Liver tissue H2S concentrations and CSE expression were significantly lower in the SPH rabbit livers compared with the control animals,starting at 3 wk,whereas pERK 1/2expressions gradually increased 12-20 wk after SPH model establishment.In portal vein smooth muscle cells,increasing H2S levels led to increased apoptosis,while Bcl-2 and Bcl-XL expression decreased.CONCLUSION:H2S prevents vascular restructuring caused by excessive proliferation of smooth muscle cells via apoptosis induction,which helps to maintain normal vascular structures.Chao Wang Juan Han Liang Xiao Chang-E Jin Dong-Jian Li Zhen Yang 2014World Journal of Gastroenterology2014,20,4:8
17Geometrical feature analysis and disaster assessment of the Xinmo landslide based on remote sensing data显示文摘At 5:39 am on June 24, 2017, a landslide occurred in the village of Xinmo in Maoxian County, Aba Tibet and Qiang Autonomous Prefecture(Sichuan Province, Southwest China). On June 25, aerial images were acquired from an unmanned aerial vehicle(UAV), and a digital elevation model(DEM) was processed. Landslide geometrical features were then analyzed. These are the front and rear edge elevation, accumulation area and horizontal sliding distance. Then, the volume and the spatial distribution of the thickness of the deposit were calculated from the difference between the DEM available before the landslide, and the UAV-derived DEM collected after the landslide. Also, the disaster was assessed using high-resolution satellite images acquired before the landslide. These include Quick Bird, Pleiades-1 and GF-2 images with spatial resolutions of 0.65 m, 0.70 m, and 0.80 m, respectively, and the aerial images acquired from the UAV after the landslide with a spatial resolution of 0.1 m. According to the analysis, the area of the landslide was 1.62 km2, and the volume of the landslide was 7.70 ± 1.46 million m3. The average thickness of the landslide accumulation was approximately 8 m. The landslide destroyed a total of 103 buildings. The area of destroyed farmlands was 2.53 ha, and the orchard area was reduced by 28.67 ha. A 2-km section of Songpinggou River was blocked and a 2.1-km section of township road No. 104 was buried. Constrained by the terrain conditions, densely populated and more economically developed areas in the upper reaches of the Minjiang River basin are mainly located in the bottom of the valleys. 18Synthesis of a Novel Core-shell Type Acrylic-polyurethane Hybrid Emulsion Containing Siloxane and Fluorine as well as Water and the Oil Resistances of Cured Film显示文摘Siliconated polyurethane (Si-PU) was synthesized using isophorone diisocyanate (IPDI), hydroxybutyl-terminated polydimethylsiloxane (PDMS), polytetramethylene ether glycol (PTMG), polypropylene glycol (PPG), 1,6-hexanediol (HDO), dimethylol propionic acid (DMPA) and triethylamine (TEA). Based on butyl acrylate (BA), 2, 2, 2-trifluoroethylmethacrylate (TFEMA) and Si-PU as a seed emulsion, a novel core-shell type acrylic-polyurethane hybrid emulsion, containing siloxane and fluorine (F-Si-PU), was prepared by seeded emulsion polymerization. The contents of siloxane and fluorine were determined according to the feed ratio. Fourier transform infrared spectroscopy (FTIR) was used to identify the chain structures of Si-PU and F-Si-PU. Investigation of transmission electron microscopy (TEM) confirmed the core-shell structure of F-Si-PU emulsion.
