1Regulation of the inflammatory cycle by a controllable release hydrogel for eliminating postoperative inflammation after discectomy显示文摘Surgery is the final choice for most patients with intervertebral disc degeneration(IDD).Operation-caused trauma will cause inflammation in the intervertebral disc.Serious inflammation will cause tissue defects and induce tissue degeneration,IDD recurrence and the occurrence of other diseases.Therefore,we proposed a scheme to treat recurrence after discectomy by inhibiting inflammation with an aspirin(ASP)-loaded hydrogel to restore the mechanical stability of the spine and relieve local inflammation.ASP-liposomes(ASP-Lips)were incorporated into a photocrosslinkable gelatin-methacryloyl(GelMA)via mixing.This material can effectively alleviate inflammation by inhibiting the release of high mobility group box 1(HMGB1)from the nucleus to the cytoplasm.We further assessed the expression of inflammatory cytokines,such as interleukin 6(IL-6)and tumor necrosis factor-α(TNF-α),and degeneration-related factors,such as type II collagen(COL-2),Aggrecan,matrix metallopeptidases-3(MMP-3),MMP-13,a disintegrin and metalloproteinase with thrombospondin motifs-4(ADAMTS-4)and ADAMTS-5 in rat nucleus pulpous cells.The level of IDD was analyzed through H&E,safranin-O staining and immunohistochemistry in rabbit samples.In vitro,we found that ASP-Lip@GelMA treatment significantly decreased inflammatory cytokines,MMP-3 and-13,and ADAMTS-4 and-5 and up-regulated COL-2 and Aggrecan via the inhibited release of HMGB-1 from the nucleus.In vivo,ASP-Lip@GelMA can effectively inhibit inflammation of local tissue after disc surgery and fill local tissue defects.This composite hydrogel system is a promising way to treat the recurrence of IDD after surgery without persistent complications.Yu Liu Jiacheng Du Peng Peng Ruoyu Cheng Jiayi Lin Congxin Xu Huilin Yang Wenguo Cui Haiqing Mao Yuling Li Dechun Geng 2021Bioactive Materials2021,6,1:6

4Multi-Agent System for Electric Vehicle Charging Scheduling in Parking Lots显示文摘As the number of electric vehicles(EVs)increases,massive numbers of EVs have started to gather in commercial parking lots to charge and discharge,which may significantly impact the operation of the grid.There may also be a deviation in the departure time of charged and discharged EVs in commercial parking lots.This deviation can lead to insufficient battery energy when the EVs leave the parking lot.This study uses the simulation software AnyLogic to build a commercial parking lot multi-agent simulation model,and the agent-based model can fully reflect the autonomy of individual EVs.Based on this simulation model,we propose an EV scheduling algorithm.The algorithm contains two main agents.The first is the power distribution center agent(PDCA),which is used to coordinate the energy output of photovoltaic(PV),energy storage system(ESS),and distribution station(DS)to solve the problem of grid overload.The second is the scheduling center agent(SCA),which is used to solve the insufficient battery energy problem due to EVs’random departures.The SCA includes two stages.In the first stage,a priority scheduling algorithm is proposed to emphasize the fairness of EV charging.In the second stage,a genetic algorithm is used to accurately determine the time interval between charging and discharging to ensure the maximum benefit of EV owner.Finally,simulation experiments are conducted in AnyLogic,and the results demonstrate the superiority of the algorithm over the classical algorithm.Mao Tan Zhonglin Zhang Yuling Ren Irampaye Richard Yuzhou Zhang 2023Complex System Modeling and Simulation2023,3,2:0
5MSNCs and MgO-MSNCs as drug delivery systems to control the adsorption kinetics and release rate of indometacin显示文摘Mesoporous silica cocoon materials(MSNCs) and MgO doped mesoporous silica cocoons(MgO-MSNCs) with the cocoon-like hierarchical morphology and different alkalinities were synthesized as carriers for acidic drugs. Indomethacin(IMC) was selected as a model drug and loaded into carriers. All materials and the drug-loaded samples were characterized by nitrogen adsorption, FTIR spectroscopy, transmission electron microscopy(TEM), powder X-Ray diffraction(XRD) and differential scanning calorimetry(DSC). The effect of the Mg/Si molar ratio on the kinetics and equilibrium of IMC adsorption on MgO-MSNCs was thoroughly examined, and it was found that the increase in the Mg/Si molar ratio resulted in an increasing IMC adsorption rate due to the increased affinity between alkaline MgO-MSNCs and weak acid IMC. The adsorption kinetics fitted a pseudo second-order model well. The Freundlich isotherm showed a better fit, indicating that the coverage of IMC on the surface of MgO-MSNCs was heterogeneous. The maximum adsorption capacity of adsorbent was calculated by the Langmuir isotherm equation. The Temkin equation provided further support that the IMC adsorption on MgO-MSNCs was dominated by a chemisorption process. MgO-MSNCs also have the advantage of allowing an adjustment of the drug release rate of weak acid drug. The cytotoxicity assay indicated good biocompatibility of MgO-MSNCs. 7Immune-awakening Saccharomyces-inspired nanocarrier for oral target delivery to lymph and tumors显示文摘Utilization of the intestinal lymphatic pathway will allow extraordinary gains in lymph and tumors cascade-targeted delivery of oral drugs and awakening the innate/adaptive immunity of the body and the lesion microenvironment,in addition to improving oral bioavailability relative to other means of delivery of oral drugs.Here,inspired by the specific invasion route of intestinal microorganisms,we pioneered an immune-awakening Saccharomyces-inspired mesoporous silicon nanoparticle(yMSN)for the ingenious cascade-targeted delivery of therapeutic cancer vaccines and antitumor drugs to lymph and tumors via the intestinal lymphatic pathway.Encouragingly,yMSN high-loaded tumor-specific antigens(OVA,11.9%)and anti-tumor drugs(Len,28.6%)with high stability,namely Len/OVA/yMSN,efficiently co-delivered OVA and Len to their desired target sites.Moreover,yMSN concomitantly awakened the innate antitumor immunity of dendritic cells and macrophages,strengthening vaccine-induced adaptive immune responses and reversing macrophage-associated immunosuppression in the tumor microenvironment.Surprisingly,Len/OVA/yMSN treatment resulted in excellent synergistic antitumor efficacy and long-term antitumor memory in OVA-Hepa1-6-bearing mice.This high-performance nanocarrier provides a novel approach for lesion-targeting delivery of oral drugs accompanied with awakening of the innate/adaptive immunity of the lesion environment,and also represents a novel path for the oral delivery of diverse therapeutic agents targeting other lymph-mediated diseases.Yuling Mao Xiudan Wang Caishun Chen Qinfu Zhao Yanfeng Liu Jinghai Zhang Siling Wang 2022Acta Pharmaceutica Sinica B2022,12,12:0
