| 3 | Culturing adequate CAR-T cells from less peripheral blood to treat B-cell malignancies显示文摘Objective:Chimeric antigen receptor-modified T(CAR-T)cells have shown impressive results against relapsed/refractory B cell malignancies.However,the traditional manufacture of CAR-T cells requires leukapheresis to isolate large amounts of peripheral blood T cells,thus making some patients ineligible for the procedure.Methods:We developed a simple method for CAR-T cell preparation requiring small volumes of peripheral blood.First,CD3+T cells isolated from 50 mL peripheral blood from patients(B-cell malignancies)were stimulated with immobilized anti-CD3/RetroNectin in 6-well plates and then transduced with CAR-expressing lentiviral vector.After 4 d,the T cells were transferred to culture bags for large-scale CAR-T cell expansion.In vitro and animal experiments were performed to evaluate the activity of the manufactured CAR-T cells.Finally,29 patients with B-cell acute lymphoblastic leukemia(B-ALL)and 9 patients with B-cell lymphoma were treated with the CAR-T cells.Results:The CAR-T cells were expanded to 1–3×10^(8) cells in 8–10 d and successfully killed B cell-derived malignant tumor cells in vitro and in vivo.For patients with B-ALL,the complete remission rate was 93%1 month after CAR-T cell infusion;after 12 months,the overall survival(OS)and leukemia-free survival rates were 69%and 31%,respectively.For patients with lymphoma,the objective response rate(including complete and partial remission)was 78%2 months after CAR-T cell infusion,and after 12 months,the OS and progression-free survival rates were 71%and 43%,respectively.Cytokine-release syndrome(CRS)occurred in 65.51%and 55.56%of patients with B-ALL and B-cell lymphoma,respectively;severe CRS developed in 20.69%of patients with B-ALL and in no patients with lymphoma.Conclusions:Our novel method can generate sufficient numbers of CAR-T cells for clinical use from 50–100 mL peripheral blood,thus providing an alternative means of CAR-T cell generation for patients ineligible for leukapheresis. | Lu Han Jian Zhou Linlin Li Keshu Zhou Lingdi Zhao Xinghu Zhu Qingsong Yin Yufu Li Hongqin You Jishuai Zhang Yongping Song Quanli Gao | 2021 | Cancer Biology & Medicine2021,18,4: | 2 |

| 6 | Fabrication and characterization of Ba Ce_(0.8)Y_(0.2)O_(2.9)-Ce_(0.85)Sm_(0.15)O_(1.925) composite electrolytes for IT-SOFCs显示文摘The Ba Ce0.8Y0.2O2.9-Ce0.85Sm0.15O1.925 composite electrolytes were prepared with Ba Ce0.8Y0.2O2.9(BCY) and Ce0.85Sm0.15O1.925(SDC). The SDC and BCY powders were mixed in the weight ratio of 95:5, 85:15, and 75:25, respectively(named as BS95, BS85, and BS75). Because of the composite effect between the SDC and BCY phases, the BS95 and BS85 exhibit improved conductivity compared with the pure SDC and BCY. The conductivity of BS95 is higher than that of BS85, indicating that the composite effect of BS95 is greater than that of BS85. Nevertheless, the composite effect in BS75 does not exist. Hence, we conclude that the composite effect in the BCY-SDC composites will decrease with the increase of the amount of BCY and even disappear when the amount of BCY exceeds a certain value. In our case, the optimum composition of the composite electrolyte is 95 wt% SDC and 5 wt% BCY. The BS95 has the highest conductivity(σ1t=0.07808 S cm-1, at 800 °C) and the fuel cell based on the BS95 shows the best performance(the maximum power density reaches as high as 526 mw cm-2 at 750 °C). | 8 | Non-conjugated polymers as thickness-insensitive electron transport materials in high-performance inverted organic solar cells显示文摘Two non-conjugated polymers PEIE-DBO and PEIE-DCO, prepared by quaternization of polyethyleneimine ethoxylate by 1,8-dibromooctane and 1,8-dichlorooctane respectively, are developed as electron transport layer(ETL) in high-performance inverted organic solar cells(OSCs), and the effects of halide ions on polymeric photoelectric performance are fully investigated. PEIE-DBO possesses higher electron mobility(3.68×10-4 cm2 V-1s-1), higher conductivity and more efficient exciton dissociation and electron extraction, attributed to its lower work function(3.94 eV) than that of PEIE-DCO, which results in better photovoltaic performance in OSCs. The inverted OSCs with PTB7-Th: PC71BM as photoactive layer and PEIE-DBO as ETL exhibit higher PCE of 10.52%, 9.45% and 9.09% at the thickness of 9, 35 and 50 nm,respectively. To our knowledge, PEIE-DBO possesses the best thickness-insensitive performance in polymeric ETLs of inverted fullerene-based OSCs. | 11 | Driving mechanisms of desertification process in the Horqin Sandy Land—a case study in Zhalute Banner,Inner Mongolia of China显示文摘Both natural and human factors contributing to desertification were examined to understand the driving mechanisms of the desertification process in Zhalute Banner,Inner Mongolia of China.The coefficient of variation(CV)and climate departure index(Z)were calculated to examine the fluctuations and trends of interannual variations of temperature and precipitation;TM remote sensing data was extracted to obtain the sandy land area;linear regression analysis was used to analyze climate changes and the socio-economic evolution over the years,and it was also used to standardize the variables,which included annual temperature,annual precipitation,human population,and livestock number,in order to measure the difference in the rate of change between climate and anthropogenic factors.The results showed that there was a rise of about 1.6uC in temperature but no significant change in precipitation from 1961 to 2000,which indicated a short-term climatic trend toward aridity in this area,a condition necessary for desertification.The fraction of precipitation in spring tended to increase whilst the fraction in autumn and winter decreased.Both the human population and livestock population had tripled and the cultivated area had doubled from 1961 to 2000,suggesting that socio-economic factors might have contributed more significantly to the desertification.Between 1988 and 1997,the sandy land area increased by 12.5%,nearly 2.4 times in the farming section.It could be concluded that the driving mechanisms of the desertification processes in Zhalute banner are mainly the policy of cropland expansion and the rising populations of humans and their livestock,which has affected the land use pattern in the past decades. | Haiping TANG Yufu CHEN Xinyu LI | 2008 | Frontiers of Environmental Science & Engineering2008,2,4: | 1 |

Such objects are only represented by relatively small coverage of pixels, and the features are degraded severely after being extracted by a deep convolutional neural network, which is detrimental to the detection performance for small objects. Therefore, an intuitive solution is to increase the resolution of small objects by cropping the original image. In this paper, we propose a simple but effective object density map guided region localization module (DMGRL) to locate and crop the regions of interest where small objects may exist. Firstly, the density map of the objects is estimated by object density map estimation network, and then the coordinates of the small object regions are calculated;Secondly, the continuous differentiable affine transformation is utilized to crop these regions so that the detector with DMGRL can be trained end-to-end instead of two-stage training. Finally, the all prediction results of input image and cropped region images are merged together to output the final detection results by non maximum suppression (NMS).
