2Engineering osteoarthritic cartilage model through differentiating senescent human mesenchymal stem cells for testing disease-modifying drugs显示文摘Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis(OA).In this study,we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs(DMOADs).Spe-cifically,human bone marrow-derived mesenchymal stromal cells(MSCs)were expanded in vitro up to passage 10(P10-MSCs).Following their senescent phenotype formation,P10-MSCs were subjected to pellet culture in chondrogenic medium.Results from qRT-PCR,histology,and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents,when compared to that from normal passage 4(P4)-MSCs.Interestingly,the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples,as demonstrated by RNA sequencing data and other analysis methods.Lastly,the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics.The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage.The P4-and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans.Ning Wang Yuchen He Silvia Liu Meagan J.Makarcyzk Guanghua Lei Alexander Chang Peter G Alexander Tingjun Hao Anne-Marie Padget Nuria de Pedro Tsapekos Menelaos Hang Lin 2022Science China(Life Sciences)2022,65,2:5
3Inducing Flow Velocities to Manage Fish Reproduction in Regulated Rivers显示文摘Conservation of endangered or economic fish and control of invasive fish is a great challenge of hydraulic engineering worldwide.Flow velocity has been recognized to affect the spawning of fishes delivering drifting eggs in rivers.However,solid scientific supports and associated mechanisms to establish quantitative relations between flow velocity and fish reproduction,taking into account spawning,fertilizing,hatching,as well as surviving,are lacking.In this paper,we quantified the relationship between flow velocity and reproduction of Chinese carps through both lab and field experiments.The results showed that a minimum velocity was required to trigger Hypophthalmichthys molitrix(H.molitrix)releasing eggs,and a velocity range was preferred to sustain spawning activity.However,the embryo incubation and larvae development of H.molitrix were found to be inhibited with the increase in flow velocity.Considering the requirements of spawning and hatching,as well as larvae surviving,an optimized flow velocity processes was identified for the reproduction of H.molitrix in rivers.These findings were of great significance to the adaptive operation of reservoirs to create reasonable and precise ecological flows for managing fish reproduction,as shown by the promising results in the engineering application to the Three Gorges Reservoir.Qiuwen Chen Jianyun Zhang Yuchen Chen Kangle Mo Jun Wang Lei Tang Yuqing Lin Lei Chen Yong Gao Wei Jiang Yuxin Zhang 2021Engineering2021,7,2:4
4Analog Optical Computing for Artificial Intelligence显示文摘The rapid development of artificial intelligence(AI)facilitates various applications from all areas but also poses great challenges in its hardware implementation in terms of speed and energy because of the explosive growth of data.Optical computing provides a distinctive perspective to address this bottleneck by harnessing the unique properties of photons including broad bandwidth,low latency,and high energy efficiency.In this review,we introduce the latest developments of optical computing for different AI models,including feedforward neural networks,reservoir computing,and spiking neural networks(SNNs).Recent progress in integrated photonic devices,combined with the rise of AI,provides a great opportunity for the renaissance of optical computing in practical applications.This effort requires multidisciplinary efforts from a broad community.This review provides an overview of the state-of-the-art accomplishments in recent years,discusses the availability of current technologies,and points out various remaining challenges in different aspects to push the frontier.We anticipate that the era of large-scale integrated photonics processors will soon arrive for practical AI applications in the form of hybrid optoelectronic frameworks.Jiamin Wu Xing Lin Yuchen Guo Junwei Liu Lu Fang Shuming Jiao Qionghai Dai 2022Engineering2022,8,3:4

13Taurocholic acid inhibits the response to interferon-αtherapy in patients with HBeAg-positive chronic hepatitis B by impairing CD8^(+)T and NK cell function显示文摘Pegylated interferon-alpha (PegIFNα) therapy has limited effectiveness in hepatitis B e-antigen (HBeAg)-positive chronic hepatitis B (CHB) patients. However, the mechanism underlying this failure is poorly understood. We aimed to investigate the influence of bile acids (BAs), especially taurocholic acid (TCA), on the response to PegIFNα therapy in CHB patients. Here, we used mass spectrometry to determine serum BA profiles in 110 patients with chronic HBV infection and 20 healthy controls (HCs). We found that serum BAs, especially TCA, were significantly elevated in HBeAg-positive CHB patients compared with those in HCs and patients in other phases of chronic HBV infection. Moreover, serum BAs, particularly TCA, inhibited the response to PegIFNα therapy in HBeAg-positive CHB patients. Mechanistically, the expression levels of IFN-γ, TNF-α, granzyme B, and perforin were measured using flow cytometry to assess the effector functions of immune cells in patients with low or high BA levels. We found that BAs reduced the number and proportion and impaired the effector functions of CD3^(+)CD8^(+) T cells and natural killer (NK) cells in HBeAg-positive CHB patients. TCA in particular reduced the frequency and impaired the effector functions of CD3^(+)CD8^(+) T and NK cells in vitro and in vivo and inhibited the immunoregulatory activity of IFN-α in vitro. Thus, our results show that BAs, especially TCA, inhibit the response to PegIFNα therapy by impairing the effector functions of CD3^(+)CD8^(+) T and NK cells in HBeAg-positive CHB patients. 16A new strategy for fabrication of unique heterostructured titanium laminates and visually tracking their synchronous evolution of strain partitions versus microstructure显示文摘Heterostructured(HS)material with extraordinary mechanical properties has been regarded as one of the most promising structural materials.Here,we reported a new strategy for preparing heterostructured pure titanium laminates that possess a good combination of strength and ductility by combining gradient structure(GS)and heterogeneous lamella structure(HLS).The deformation characteristic versus microstructure evolution of GS/HLS titanium laminates,namely the strain partitions between different-sized grains(480–25μm)was visualized using a scanning electron microscope(SEM)equipped with electron backscattered diffraction(EBSD)mode combined with the digital image correlation(SEM-DIC)with an ultrahigh spatial resolution for the first time.As a result,the hetero-deformation of unique GS/HLS structure by the characteristic of strain partitions could be accurately captured.While the hetero-deformation could result in the hetero-deformation induced(HDI)stress strengthening and HDI hardening,which were regarded as the key reason that the resulting GS/HLS Ti laminates showed a superior combination of strength and ductility.This could promote a more in-depth understanding of the strengtheningtoughening mechanism of heterostructured material.Hao Ding Xiping Cui Zhiqi Wang Tao Zhao Yuchen Wang Yuanyuan Zhang Hongtao Chen Lujun Huang Lin Geng Junfeng Chen 2022Journal of Materials Science & Technology2022,,12:1

18Catalyst for Increasing Ethylene and Propylene Production and Its Industrial Application in a Catalytic Pyrolysis Unit显示文摘Light olefins,particularly ethylene and propylene,are the most important building blocks for the petrochemical industry,and demand for their production has been increasing.The catalytic pyrolysis process(CPP)and the corresponding catalyst,developed by SINOPEC Research Institute of Petroleum Processing Co.,Ltd.,are designed to maximize the light olefin yield from catalytic cracking of heavy feedstocks.However,owing to the continuing degradation of feedstocks,the original catalyst can no longer maintain its activity.Herein,we describe the rational design of the new catalyst,Epylene,from a new metal-modified hierarchical ZSM-5 zeolite and matrix.Epylene was tested in the CPP unit of Shaanxi Yanchang Coal Yulin Energy and Chemical Company.A test run and base run were conducted to demonstrate the better performance of Epylene compared with the original catalyst.The properties of the feedstocks and the operating conditions in both runs were similar.The light olefin yield was increased from 33.95%to 36.50%and the coke yield was only 9.58%in the test run,which was lower than that in the base run.Sha Yuchen Wang Peng Ouyang Ying Zhu Genquan Lu Lijun Song Haitao Lin Wei Luo Yibin 2023China Petroleum Processing & Petrochemical Technology2023,25,2:1
