| 1 | Expression and significance of heat shock protein 70 and glucose-regulated protein 94 in human esophageal carcinoma显示文摘AIM: To investigate the expression and significance of heat shock protein 70 (HSP70) and glucose-regulated protein 94 (grp94) in human esophageal carcinoma and adjacent normal tissues.METHODS: The expression of HSP70 and grp94 in 78human esophageal cancer and adjacent normal tissues was studied by immunohistochemistry and pathology photograph analysis.RESULTS: Both esophageal cancer and adjacent normal tissues could express HSP70 and grp94. Of the 78 cases of esophageal carcinoma, 95.0%(72/78) showed positive HSP70, mainly stained in nuclei, while grp94 was mainly stained in cell plasma, and the positive rate was 71.8%(56/78).There was a significant difference in the expression of HSP70 and grp94 between esophageal cancer and adjacent normal tissues (P<0.01). Compared with adjacent normal tissues, there was a significant difference between differential types and HSP70 expression (P<0.01).CONCLUSION: HSP70 and grp94 express differently in cell plasma and nuclei. | 2 | Loss of clusterin both in serum and tissue correlates with the tumorigenesis of esophageal squamous cell carcinoma via proteomics approaches显示文摘AIM: To identify the differentially secreted proteins or polypeptides associated with tumorigenesis of esophageal squamous cell carcinoma (ESCC) from serum and to find potential tumor secreted biomarkers.METHODS: Proteins from human ESCC tissue and its matched adjacent normal tissue; pre-surgery and postsurgery serum; and pre-surgery and normal control serum were separated by two-dimensional electrophoresis (2-DE)to identify differentially expressed proteins. The silverstained 2-DE were scanned with digital ImageScanner and analyzed with ImageMaster 2D Elite 3.10 software. A cluster of protein spots differentially expressed were selected and identified with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). One of the differentially expressed proteins, clusterin, was downregulated in cancer tissue and pre-surgery serum, but it was reversed in post-surgery serum. The results were confirmed by semi-quantitative reverse-transcription (RT)-PCR and western blot.RESULTS: Comparisons of the protein spots identified on the 2-DE maps from human matched sera showed that some proteins were differentially expressed, with most of them showing no differences in composition, shape or density.Being analyzed by MALDI-TOF-MS and database searching,clusterin was differentially expressed and down-regulated in both cancer tissue and pre-surgery serum compared with their counterparts. The results were also validated by RTPCR and western blot.CONCLUSION: The differentially expressed clusterin may play a key role during tumorigenesis of ESCC. | 3 | Translocation of annexin I from cellular membrane to the nuclear membrane in human esophageal squamous cell carcinoma显示文摘AIM: To investigate the alteration of the annexin I subcellular localization in esophageal squarnous cell carcinoma (ESCC)and the correlation between the translocation and the tumorigenesis of ESCC.METHODS: The protein localization of annexin I was detected in both human ESCC tissues and cell line via the indirect immunofluorescence strategy.RESULTS: In the normal esophageal epithelia the annex in I was mainly located on the plasma membrane and formed a consecutive typical trammels net. Annexin I protein also expressed dispersively in cytoplasm and the nuclei without specific localization on the nuclear membrane. In esophageal cancer annexin I decreased very sharply with scattered disappearance on the cellular membrane, however it translocated and highly expressed on the nuclear membrane,which was never found in normal esophageal epithelia. | 4 | Antitumor immunity induced by DNA vaccine encoding alpha-fetoprotein/heat shock protein 70显示文摘AIM:To construct a DNA vaccine encoding human alpha-fetoprotein (hAFP)/heat shock protein 70 (HSP70), and to study its ability to induce specific CTL response and its protective effect against AFP-expressing tumor.METHODS: A DNA vaccine was constructed by combiningh AFP gene with HSP70 gene. SP2/0 cells were stably transfected with pBBS212-hAFP and pBBS212-hAFP/HSP70 eukaryotic expression vectors. Mice were primed and boosted with DNA vaccine hAFP/HSP70 by intramuscular injection, whereas plasmid with hAFP or HSP70 was used as controls. ELISPOT and ELISA were used to detect IFN-γ-producing splenocytes and the level of serum anti-AFP antibody from immunized mice respectively. In vivo tumor challenge was measured to assess the immune effect of the DNA vaccine.RESULTS: By DNA vaccine immunization, the results of ELISPOT and ELISA showed that the number of IFN-γ-producing splenocytes and the level of serum anti-AFP antibody were significantly higher in rhAFP/HSP70 group than in hAFP and empty plasmid groups (95.50±10.90 IFN-γ spots/10^6 cells vs 23.60±11.80 IFN-γ spots/10^6 cells,7.17±4.24 IFN-γ spots/106 cells, P<0.01; 126.50±8.22μg/mL vs 51.72±3.40μg/mL, 5.83±3.79μg/mL, P<0.01). The tumor volume in rhAFP/HSP70 group was significantly smaller than that in pBBS212-hAFP and empty plasmid groups (37.41±7.34 mm^3 vs 381.13±15.48 mm^3, 817.51±16.25 mm^3,P<0.01). | 5 | Molecular self-assembly behavior of mono[6-O-6-(4-carboxyl-phenyl)]-β-CD in solution and solid state显示文摘A novel modified cyclodextrin, mono[6-O-6-(4- carboxyl-phenyl)]-b-CD (1), has been synthesized by the reaction of mono[6-(p-toluenesulfonyl)]- b-CD with 4-hydro- xybenzoate, and its molecular self-assembly behavior in both solution and solid state was studied by means of crystallography, NMR spectroscopy and microcalorimetry. The results indicate that the bezoic acid groups are successively penetrated intermolecularly into the adjacent b-CD cavities to form helical columnar supramolecules in the solid state. As compared with crystal, the similar self-assembly behavior of 1 in aqueous solution has also been confirmed by the 1H ROESY spectroscopy. | 6 | A time series of filament eruptions observed by three eyes from space:from failed to successful eruptions显示文摘We present stereoscopic observations of six sequential eruptions of a filament in the active region NOAA 11045 on 2010 Feb 8,with the advantage of the STEREO twin viewpoints in combination with Earth's viewpoint from SOHO instruments and ground-based telescopes.The last one of the six eruptions is a coronal mass ejection,but the others are not.The flare in this successful one is more intense than in the others.Moreover,the velocity of filament material in the successful one is also the largest among them.Interestingly,all the filament velocities are found to be proportional to the power of their flares.We calculate magnetic field intensity at low altitude,the decay indexes of the external field above the filament,and the asymmetry properties of the overlying fields before and after the failed eruptions and find little difference between them,indicating the same coronal confinement exists for both the failed and successful eruptions.The results suggest that,besides the confinement of the coronal magnetic field,the energy released in the low corona should be another crucial element affecting a failed or successful filament eruption.That is,a coronal mass ejection can only be launched if the energy released exceeds some critical value,given the same initial coronal conditions. | Yuan-DengShen YuLiu RuiLiu | 2011 | Research in Astronomy and Astrophysics2011,11,5: | 2 |

| 13 | Cx31 is assembled and trafficked to cell surface by ER-Golgi pathway and degraded by proteasomal or lysosomal pathways显示文摘Gap junctions, consisting of connexins, allow the exchange of small molecules (<1 kD) between adjacent cells, thusproviding a mechanism for synchronizing the responses of groups of cells to environmental stimuli. Connexin 31 is amember of the connexin family. Mutations on connexin 31 are associated with erythrokeratodermia variabilis, hearingimpairment and peripheral neuropathy. However, the pathological mechanism for connexin 31 mutants in these diseasesare still unknown. In this study, we analyzed the assembly, trafficking and metabolism of connexin 31 in HeLa cellsstably expressing connexin 31. Calcein transfer assay showed that calcein transfer was inhibited when cells weretreated with Brefeldin A or cytochalasin D, but not when treated with nocodazole or α-glycyrrhetinic acid, suggestingthat Golgi apparatus and actin filaments, but not microtubules, are crucial to the trafficking and assembly of connexin31, as well as the formation of gap junction intercellular communication by connexin 31. Additionally, α-glycyrrhetinicacid did not effectively inhibit gap junctional intercellular communication formed by connexin 31. Pulse-chase assayrevealed that connexin 31 had a half-life of about 6 h. Moreover, Western blotting and fluorescent staining demonstratedthat in HeLa cells stably expressing connexin 31, the amount of connexin 31 was significantly increased after these cellswere treated with proteasomal or lysosomal inhibitors. | 18 | Hydrocarbon accumulation history in Lower Cretaceous in northern slope of Bongor Basin in Chad,Central Africa显示文摘Based on the analysis of the fluid inclusion homogenization temperature and apatite fission track on the northern slope zone of the Bongor Basin in Chad,this paper studied the time and stages of hydrocarbon accumulation in the study area.The results show that:(1)The brine inclusions of the samples from the Kubla and Prosopis formations in the Lower Cretaceous coexisting with the hydrocarbon generally present two sets of peak ranges of homogenization temperature,with the peak ranges of low temperature and high temperature being 75–105℃ and 115–135℃,respectively;(2)The samples from the Kubla and Prosopis formations have experienced five tectonic evolution stages,i.e.,rapid subsidence in the Early Cretaceous,tectonic inversion in the Late Cretaceous,small subsidence in the Paleogene,uplift at the turn of the Paleogene and Neogene,and subsidence since the Miocene,in which the denudation thickness of the Late Cretaceous and after the turn of the Paleogene and Neogene are~1.8 km and~0.5 km,respectively.The cumulative denudation thickness of the two periods is about 2.3 km;(3)Using the brine inclusion homogenization temperature coexisting with the hydrocarbon as the capture temperature of the hydrocarbon,and combining with the apatite fission track thermal history modeling,the result shows that the Kubla and Prosopis formations in the Lower Cretaceous on the northern slope of the Bongor Basin have the same hydrocarbon accumulation time and stages,both of which have undergone two stages of hydrocarbon charging at 80–95 Ma and 65–80 Ma.The first stage of charging corresponds to the initial migration of hydrocarbon at the end of the Early Cretaceous rapid sedimentation,while the second stage of charging is in the stage of intense tectonic inversion in the Late Cretaceous. | WANG Li NIE Zhiquan DU Yebo WANG Lin MENG Fanchao CHEN Yuliu HU Jie DING Ruxin | 2024 | Petroleum Exploration and Development2024,51,1: | 0 |
| 19 | Characterization of rtSH3p13 gene encoding a development protein involved in vesicular traffic in spermiogenesis显示文摘A cDNA, designated as rtSH3p13, was isolated from a rat testis cDNA library. It consists of 1463 bp nuclear acids,which encodes a protein of 312 amino acids and was assigned the GenBank accession number AF227439. The deduced rtSH3p13 protein is a truncated isoform of SH3p13 as a result of mRNA alternative splicing. It is mainly expressed in the rat testis, detected in spermatids at the steps 8-19 of spermiogenesis, and found around the acrosome. During postnatal development, rtSH3p13 appears on day 18 and reaches maximum on day 60. Further experimental results suggested that rtSH3p13 forms a complex with activated epidermal growth factor receptor (EGFR) and interacts with synaptojanin I. | 20 | Liberica coffee as an alternative cropping system for sustainable farming on Indonesian Peatlands显示文摘Indonesian peatlands are a fragile ecosystem,and to protect it,growing Liberica coffee is a promising way for both the environment and the economy.This study aimed to evaluate the performance of the liberica coffee on peatland with different water tables and develop an improved cultivation system in this ecosystem.The study area was in Tanjung Jabung Barat District,Jambi Province,Indonesia.The plant age is greater than 15 years and the average plant density was about 1000 trees/hm2.The study was conducted in two stages.The 1st stage was a survey to identify and characterize smallholder liberica coffee farming at three peatland zones,namely 0-100 m,200-300 m,and>400 m from the principal drainage canal,and the 2nd stage was a field experiment to test the effectiveness of amendments in improving liberica coffee growth and improving degraded peatland.The treatments were arranged in a randomized complete block design with four replications,including manure(M),at the rate of 10 t/hm^(2);lime(L),3 t/hm^(2);peat surface elevation(P),and a farmer’s practice(Control)without manure and/or lime as a control.This research revealed that liberica coffee can not tolerate high water table as it may stimulate disease incidence and cause low-quality yield.However,recycling of organic C of about 17.14 t/(hm^(2)·a)through cherry residue application and litter compensated part of the loss of organic C through CO_(2)emissions,coffee bean removal,and maintaining peat fertility for sustainable farming.The low yield of(0.70±0.12)t/(hm^(2)∙a)coffee bean could be increased to(0.87±0.24)and(0.94±0.14)t/(hm^(2)∙a)by adding 3 t/(hm^(2)∙3a)of lime or 10 t/(hm^(2)∙a)of manure,respectively.This research revealed that water table management and amendments are two main factors in liberica coffee farming on peatlands.It is of great significance to study the cultivation technology of coffee in peatland. | Bariot Hafif Yulius Ferry Budi Martono Rita Harni Kurnia Dewi Sasmita Busyra B.Saidi Maswar Maswar R.Bambang Heryanto Novizar Nazir Fahmuddin Agus Meynarti Sari Dewi Ibrahim | 2024 | International Journal of Agricultural and Biological Engineering2024,17,1: | 0 |
