3Natural continuous influent nitrifier immigration effects on nitrification and the microbial community of activated sludge systems显示文摘Two sequencing batch reactors(SBRs) were operated for 100 days under aerobic conditions,with one being fed with unsterilized municipal wastewater(USBR), and the other fed with sterilized municipal wastewater(SSBR). Respirometric assays and fluorescence in situ hybridization(FISH) results show that active nitrifiers were present in the unsterilized influent municipal wastewater. The maximum ammonia utilization rate(AUR) and nitrite utilization rate(NUR) of the unsterilized influent were 0.32 ± 0.12 mg NH4+-N/(L·hr) and0.71 ± 0.18 mg NO2--N/(L·hr). Based on the maximum utilization rates, the estimated seeding intensity for the ammonia oxidizing bacteria(AOB) and nitrite oxidizing bacteria(NOB) of the USBR was 0.08 g AOB/(g AOB·day) and 0.20 g NOB/(g NOB·day) respectively. The fraction of nitrifiers/total bacteria in the influent was 5.35% ± 2.1%, the dominant AOB was Nitrosomonas spp., Nitrosococcus mobilis hybridizated with Nsm156, and the dominant NOB was Nitrospira hybridizated with Ntspa662. The influent nitrifiers potentially seeded the activated sludge of the bioreactor and hence demonstrated a mitigation of the acclimatization times and instability during start-up and early operation. The AUR and NUR in the USBR was 15% and 13% higher than the SSBR respectively during the stable stage, FISH results showed that nitrifiers population especially the Nitrospira in the USBR was higher than that in the SSBR. 5Overexpression of cytoglobin gene inhibits hypoxic injury to SH-SY5Y neuroblastoma cells显示文摘A plasmid for cytoglobin expression, pAcGFP1-C1-cytoglobin, was transfected into SH-SY5Y cells. Cobalt chloride was used to establish a model of hypoxia. Western blotting indicated that cytoglobin was overexpressed and there was low expression of hypoxia-inducible factor-1α in SH-SY5Y cells after transfection. Following cobalt chloride-induced hypoxia, cytoglobin and hypoxia-inducible fac-tor-1α expression gradually increased in SH-SY5Y cells. Flow cytometry showed that with increas-ing duration of hypoxia, the proportion of normal cells significantly diminished in the transfected and non-transfected groups. The proportion of cells in the early stages of apoptosis increased. However, the proportion of apoptotic cells was significantly lower in the transfected group compared with the non-transfected group. These results demonstrate that cytoglobin and hypoxia-inducible factor-1α are strongly up-regulated by hypoxia, and that there is a strong relationship between hy-poxia-inducible factor-1α and cytoglobin during hypoxic injury.Xiuling Yu Dianwen Gao 2013Neural Regeneration Research2013,8,23:3

7Ancient DNA sequences of rice from the low Yangtze reveal significant genotypic divergence显示文摘Rice (Oryza sativa) was first domesticated in the lower and middle Yangtze regions of China, and rice remains have been found in many Chinese archaeological sites. Until now, only phenotypic archeobotanical evidence, such as the spikelet bases of ancient grains, has been used to speculate on the domestication process and domestication rate of rice. In this study, we sequenced 4 genomic segments from rice remains in Tianluoshan, a site of the local Hemudu Neolithic culture in the low Yangtze and two other archaeological sites (~2400 and 1200 BC, respectively). We compared our sequences with those of the current domesticated and wild rice (O. rufipogon) populations. At least two genotypes were found in the remains from each site, suggesting a heterozygotic state of the rice seeds. One ancient genotype was not found in the current domesticated population and might have been lost. The rice remains belonged to the japonica group, and most if not all were japonica-type, suggesting that the remains might be at an early stage of indica-japonica divergence or an indica-japonica mixture. 16Development and validation of a machine learning-based nomogram for prediction of intrahepatic cholangiocarcinoma in patients with intrahepatic lithiasis显示文摘Background:Accurate diagnosis of intrahepatic cholangiocarcinoma(ICC)caused by intrahepatic lithiasis(IHL)is crucial for timely and effective surgical intervention.The aim of the present study was to develop a nomogram to identify ICC associated with IHL(IHL-ICC).Methods:The study included 2,269 patients with IHL,who received pathological diagnosis after hepatectomy or diagnostic biopsy.Machine learning algorithms including Lasso regression and random forest were used to identify important features out of the available features.Univariate and multivariate logistic regression analyses were used to reconfirm the features and develop the nomogram.The nomogram was externally validated in two independent cohorts.Results:The seven potential predictors were revealed for IHL-ICC,including age,abdominal pain,vomiting,comprehensive radiological diagnosis,alkaline phosphatase(ALK),carcinoembryonic antigen(CEA),and cancer antigen(CA)19-9.The optimal cutoff value was 2.05μg/L for serum CEA and 133.65 U/mL for serum CA 19-9.The accuracy of the nomogram in predicting ICC was 82.6%.The area under the curve(AUC)of nomogram in training cohort was 0.867.The AUC for the validation set was 0.881 from The Second Affiliated Hospital of Wenzhou Medical University,and 0.938 from The First Affiliated Hospital of Fujian Medical University.Conclusions:The nomogram holds promise as a novel and accurate tool to predict IHL-ICC,which can identify lesions in IHL in time for hepatectomy or avoid unnecessary surgical resection.Xian Shen Huanhu Zhao Xing Jin Junyu Chen Zhengping Yu Kuvaneshan Ramen Xiangwu Zheng Xiuling Wu Yunfeng Shan Jianling Bai Qiyu Zhang Qiqiang Zeng 2021Hepatobiliary Surgery and Nutrition2021,10,6:1
