| 2 | Mitotic motor CENP-E cooperates with PRC1 in temporal control of central spindle assembly显示文摘Error-free cell division depends on the accurate assembly of the spindle midzone from dynamic spindle microtubules to ensure chromatid segregation during metaphase-anaphase transition.However,the mechanism underlying the key transition from the mitotic spindle to central spindle before anaphase onset remains elusive.Given the prevalence of chromosome instability phenotype in gastric tumorigenesis,we developed a strategy to model context-dependent cell division using a combination of light sheet microscope and 3D gastric organoids.Light sheet microscopic image analyses of 3D organoids showed that CENP-E inhibited cells undergoing aberrant metaphase-anaphase transition and exhibiting chromosome segregation errors during mitosis.Highresolution real-time imaging analyses of 2D cell culture revealed that CENP-E inhibited cells undergoing central spindle splitting and chromosome instability phenotype.Using biotinylated syntelin as an affinity matrix,we found that CENP-E forms a complex with PRC1 in mitotic cells.Chemical inhibition of CENP-E in metaphase by syntelin prevented accurate central spindle assembly by perturbing temporal assembly of PRC1 to the midzone.Thus,CENP-E-mediated PRC1 assembly to the central spindle constitutes a temporal switch to organize dynamic kinetochore microtubules into stable midzone arrays.These findings reveal a previously uncharacterized role of CENP-E in temporal control of central spindle assembly.Since CENP-E is absent from yeast,we reasoned that metazoans evolved an elaborate central spindle organization machinery to ensure accurate sister chromatid segregation during anaphase and cytokinesis. | Xu Liu Leilei Xu Junying Li Phil Y.Yao Wanjuan Wang Hazrat Ismail Haowei Wang Bryce Liao Zhihong Yang Tarsha Ward Ke Ruan Jianchun Zhang Quan Wu Ping He Xia Ding Dongmei Wang Chuanhai Fu Zhen Dou Feng Yan Wenwen Wang Xing Liu Xuebiao Yao | 2020 | Journal of Molecular Cell Biology2020,12,8: | 3 |
| 3 | The precise multimode GNSS positioning for UAV and its application in large scale photogrammetry显示文摘In this paper,we use multimode GNSS instead of single GPS constellation to resolve the three exterior line elements of each image.The principles of differential GNSS positioning and GNSSsupported aerial triangulation(AT)are presented and an implementation case is demonstrated.With multi-constellation system,the number of visible satellites is significantly increased and the geometry distribution of the satellites is well improved.The positioning accuracy and robustness are therefore getting better compared to GPS positioning.Experimental results show that differential GNSS has remarkable increment on the integer rate of ambiguity solution when GPS has few number and low elevation angle of satellites.The combined AT adjustment of GNSS resolution and 10 ground control points(GCPs)achieve the horizontal accuracy of±18 cm and vertical accuracy of±23 cm for the check points,which are comparable with traditional bundle adjustment with dense GCPs and better than GPS-supported AT.The achieved accuracy also satisfies the requirement for 1:500 topographic maps with the bonus of 84%GCPs eliminated.In conclusion,GNSS-supported AT is of feasibility and superiority for large scale Unmanned Aerial Vehicle-based photogrammetry. | Hongxing Sun Leilei Li Xuewen Ding Bingxuan Guo | 2016 | Geo-Spatial Information Science2016,19,3: | 3 |

| 6 | Stress rupture properties and deformation mechanisms of K4750 alloy at the range of 650℃ to 800℃显示文摘The stress rupture properties and deform ation mechanism s of K4750 alloy at 650 ℃, 700 ℃, 750 ℃ and 800 ℃ were investigated. As the decrease of tem perature and stress, the stress rupture life gradually increased. A Larson-Miller Parameter (LMP) method was used for analyzing the stress rupture life under different conditions. The linear fitting formula between stress (σ) and LMP was derived as σ= 3166.455-119.969 ×LMP and the fitting coefficient w as 0.98. After testing, the dislocation configurations of all stress rupture samples w ere investigated by transm ission electron microscopy (TEM). The tem perature and stress had a significant impact on the deform ation m echanism, thereby affected the stress rupture life of K4750 alloy. As the increasing stress at a given tem perature, the deform ation m echanism gradually transform ed from Orowan looping to stacking fault shearing. Based on experim ental results, the threshold stress at 650 ℃, 700 ℃, 750 ℃ and 800 ℃ for the transition of deformation mechanism was estimated to be about 650 MPa, 530 MPa, 430 MPa and 350 MPa, respectively. Below the threshold stress,γ phase effectively hindered dislocation motion by Orowan looping mechanism, K4750 alloy had along stress rupture life. Slightly above the threshold stress, Orowan looping combining stacking fault shearing was the dom inant mechanism ,the stress rupture life decreased. | 7 | Influence of yttrium on purification and carbide precipitation of superalloy K4169显示文摘The effect of reactive element yttrium on purification and carbide precipitation of superalloy K4169 has been studied through vacuum induction melting.The results show that yttrium has a strong deoxidizing and desulfurizing ability,and the impurity elements of sulfur and oxygen in melting K4169 are greatly reduced to 2-5 ppm and 4-5 ppm,respectively.The metallurgical reaction product is mainly Y_(2)O_(3)and yttrium containing sulfides have not been observed.With the increase of the content of yttrium from0.005 wt.%to 0.033 wt.%,the morphology and size of MC carbides in casting are significantly influenced due to that the yttrium changes the diffusion of the carbide forming elements and solid fraction during solidification.In the alloys without yttrium,only isolated blocky or strip carbides exist,while skeletonlike carbides are observed in the alloys with yttrium.Both the average size and the number density of skeleton carbides decrease monotonously with yttrium content increasing.After yttrium increasing to 0.033 wt.%,a phase of hexagonal Ni17 Y2 is observed to precipitate in the form of symbiosis with MC carbide and matrix.The addition of yttrium is not beneficial for the properties of K4169 alloys at the room temperature,which is quite related to the MC carbides with a large skeleton-like morphology through the analysis of tensile fracture. | Shuting Cao Yaqian Yang Bo Chen Kui Liu Yingche Ma Leilei Ding Junjie Shi | 2021 | Journal of Materials Science & Technology2021,,27: | 1 |

| 9 | The Relationship Between Vegetation Characteristics and Altitudes in Transitional Permafrost Zone in Xidatan, Qinghai-Tibetan Plateau显示文摘Many studies showed that permafrost has profound influence on alpine ecosystem. However, former researches were mainly focused on typical points by temporal scales. There were few studies about the correlation between vegetation characteristics and different altitudes covering a large region in spatial pattern, especially in transitional permafrost(TP). There were continuous permafrost(CP) discontinuous permafrost(DCP) and seasonal frozen ground(SFG) in this study region. The types of permafrost changed from SFG to DCP, and finally become CP as the altitudes of Xidatan increase. In this paper, 112 845 points interpreted by HJ1-B(environment and disaster monitoring and prediction small satellite constellation), vegetation investigation points, thawing layer thickness research sites, ground temperature and water content observation plots were used to examine the spatial pattern of vegetation which were located in different altitudes in Xidatan, a typical TP region, in Qinghai-Tibetan Plateau. Vegetation characteristics, soil moisture content(SMC) and thaw depths were collected in 15 August to 25 August2012. Characteristics of vegetation were mainly represented by fractional vegetation cover(FVC) derived from the normalized difference vegetation index(NDVI), as well as above ground biomass(AGB). In this paper, we analyzed that the distinction of vegetation characteristics in each range through statistics data. These ranges were divided by varied altitudes. For examples, the ranges were divided into 50 m or 100 m. In this study we use a large area plots method to further discuss the relationship between the features of vegetation and the different regions of permafrost based on altitudes shifts in Xidatan. A diagram described the vegetation characteristics variability with rising altitudes in transitional permafrost region was drawn in this paper. Our results illustrated the FVCs first increased in SFG region and then decreased in DCP zone slowly, and in CP region FVCs soared then dropped dramatically. | 11 | Reduced phosphatidylcholine synthesis suppresses the embryonic lethality of seipin deficiency显示文摘Seipin plays a vital role in lipid droplet homeostasis,and its deficiency causes congenital generalized lipodystrophy type II in humans.It is not known whether the physiological defects are all caused by cellular lipid droplet defects.Loss-of-function mutation of seip-1,the Caenorhabditis elegans seipin ortholog,causes embryonic lethality and lipid droplet abnormality.We uncover nhr-114 and spin-4 as two suppressors of seip-1 embryonic lethality.Mechanistically,nhr-114 and spin-4 act in the“B12-one-carbon cycle-phosphatidylcholine(PC)”axis,and reducing PC synthesis suppresses the embryonic lethality of seip-1 mutants.Conversely,PC deficiency enhances the lipid droplet abnormality of seip-1 mutants.The suppression of seip-1 embryonic lethality by PC reduction requires polyunsaturated fatty acid.In addition,the suppression is enhanced by the knockdown of phospholipid scramblase epg-3.Therefore,seipin and PC exhibit opposite actions in embryogenesis,while they function similarly in lipid droplet homeostasis.Our results demonstrate that seipin-mediated embryogenesis is independent of lipid droplet homeostasis. | Jinglin Zhu Sin Man Lam Leilei Yang Jingjing Liang Mei Ding Guanghou Shui Xun Huang | 2022 | Life Metabolism2022,1,2: | 0 |

| 13 | Modeling the effects of extreme high-temperature stress at anthesis and grain filling on grain protein in winter wheat显示文摘Extreme high-temperature stress(HTS) associated with climate change poses potential threats to wheat grain yield and quality. Wheat grain protein concentration(GPC) is a determinant of wheat quality for human nutrition and is often neglected in attempts to assess climate change impacts on wheat production. Crop models are useful tools for quantification of temperature impacts on grain yield and quality.Current crop models either cannot simulate or can simulate only partially the effects of HTS on crop N dynamics and grain N accumulation. There is a paucity of observational data on crop N and grain quality collected under systematic HTS scenarios to develop algorithms for model improvement as well as evaluate crop models. Two-year phytotron experiments were conducted with two wheat cultivars under HTS at anthesis, grain filling, and both stages. HTS significantly reduced total aboveground N and increased the rate of grain N accumulation, while total aboveground N and the rate of grain N accumulation were more sensitive to HTS at anthesis than at grain filling. The observed relationships between total aboveground N, rate of grain N accumulation, and HTS were quantified and incorporated into the WheatGrow model. The new HTS routines improved simulation of the dynamics of total aboveground N, grain N accumulation, and GPC by the model. The improved model provided better estimates of total aboveground N, grain N accumulation, and GPC under HTS(the normalized root mean square error was reduced by 40%, 85%, and 80%, respectively) than the original WheatGrow model. | 14 | Integrated Transcriptome and Small RNA Sequencing Analyses Reveals Insights into the Molecular Mechanism of Seed Germination in Mung Bean显示文摘During the life cycle of a plant,seed germination is crucial.Upon ingestion of water,the dry seeds resumed energy metabolism and cellular repair.To dissect the complex mechanisms at the very beginning of seed germination,two approaches including transcriptome and small RNA sequencing were conducted during the water imbibition process of mung bean seeds compared with dry seed.The transcriptome sequencing analysis identified 10,108 differentially expressed genes(DEGs)between dry and imbibed mung bean seeds.KEGG enrichment analyses demonstrated numerous DEGs involved in hormone signaling pathways,carbohydrate,and energy metabolism.Out of the total DEGs,129 genes were investigated to involve in abscisic acid,gibberellin,and ethylene pathways,with 56 genes involved in glycolysis and TCA cycle.Through the analysis of small RNA sequencing among different samples,there were 284 miRNAs found in the imbibed and dried seeds of mung bean containing 213 known and 71 novel miRNAs.From comparison between the two types of seeds,51 miRNAs were differentially expressed.The integrated analyses of transcriptome and miRNAome data showed that 23 DEGs are the putative target genes of 8 miRNAs including miR156,miR171b-3p,miR166e-3p,miR169-1,etc.QRT-PCR was used to validate several DEGs and miRNAs.The data suggested that the seed imbibition process is regulated by a variety of DEGs and microRNAs.Meanwhile,we hope to gain a deeper insight into how mung bean seeds germinate in order to optimize seed growth. | Yanyan Pu Liwen Wang Leilei Li Yujun Si Shubin Xie Yunzhe Cong Dong Wang Yongchao Gong Rumei Tian Xue Chen Xiaoyan Zhang Min Liu Hanfeng Ding Nana Li | 2023 | Phyton-International Journal of Experimental Botany2023,92,6: | 0 |
