2Activation of the cold-sensing TRPM8 channel triggers UCP1-dependent thermogenesis and prevents obesity显示文摘Brown adipose tissue(BAT)is an energy-expending organ that produces heat.Expansion or activation of BAT prevents obesity and diabetes.Chronic cold exposure enhances thermogenesis in BAT through uncoupling protein 1(UCP1)activation triggered via a b-adrenergic pathway.Here,we report that the cold-sensing transient receptor potential melastatin 8(TRPM8)is functionally present in mouse BAT.Challenging brown adipocytes with menthol,a TRPM8 agonist,up-regulates UCP1 expression and requires protein kinase A activation.Upon mimicking long-term cold exposure with chronic dietary menthol application,menthol significantly increased the core temperatures and locomotor activity in wild-type mice;these effects were absent in both TRPM82/2 and UCP12/2 mice.Dietary obesity and glucose abnormalities were also prevented by menthol treatment.Our results reveal a previously unrecognized role for TRPM8,suggesting that stimulation of this channel mediates BAT thermogenesis,which could constitute a promising way to treat obesity.Shuangtao Ma Hao Yu Zhigang Zhao Zhidan Luo Jing Chen Yinxing Ni Rongbing Jin Liqun Ma Peijian Wang Zhenyu Zhu Li Li Jian Zhong Daoyan Liu Bernd Nilius Zhiming Zhu 2012Journal of Molecular Cell Biology2012,4,2:2
3Rim101-upregulated Fets contribute to dark pigment formation in gray cells of Candida albicans显示文摘Candida albicans has long been known to switch between white and opaque phases;however,a third cell type,referred to as the‘gray’phenotype,was recently characterized.The three phenotypes have different colonial morphologies,with white cells forming white-colored colonies and opaque and gray cells forming dark-colored colonies.We previously showed that Wor1-upregulated ferroxidases(Fets)function as pigment multicopper oxidases that regulate the production of dark-pigmented melanin in opaque cells.In this study,we demonstrated that Fets also contributed to dark pigment formation in gray colonies but in a Wor1-independent manner.Deletion of both WOR1 and EFG1 locked cells in the gray phenotype in some rich media.However,the efg1/efg1 wor1/wor1 mutant could switch between white&gray in minimal media depending on the ambient pH.Specifically,mutant cells exhibited the white phenotype at pH 4.5 but switched to gray at pH 7.5.Consistent with phenotype switching,Fets expressions&melanin production were also regulated by ambient pH.Ectopic expression of the Rim101-405 allele in the mutant enabled the pH restriction to be bypassed&promoted gray cell formation in acidic media.Our data suggest that Rim101-upregulated Fets contribute to dark pigment formation in the gray cells.Baodi Dai Yinxing Xu Hongyu Wu Jiangye Chen 2021Acta Biochimica et Biophysica Sinica2021,53,12:1
