| 1 | MiR-181b suppresses proliferation of and reduces chemoresistance to temozolomide in U87 glioma stem cells显示文摘MicroRNAs regulate self renewal and differentiation of cancer stem cells.There,we sought to identify the expression of miR-181b in glioma stem cells and investigate the biological effect of miR-181b on glioma stem cells in this study.MiR-181b expression was measured by real-time PCR in glioma stem cells isolated from U87 cells by FACS sorting.After miR-181b was overexpressed in U87 glioma stem cells by miR-181b lentiviral expression vector and/or treatment of temozolomide,secondary neurosphere assay,soft agar colony assay and MTT assay were performed.Compared with U87 cells,the expression of miR-181b was significantly decreased in U87 glioma stem cells.Overexpression of miR-181b decreased neurosphere formation by U87 glioma stem cells in vitro and suppressed colony formation in soft agar,and the cell growth inhibition rates increased in a time-dependent manner in U87 glioma stem cells infected with miR-181b lentivirus.Furthermore,miR-181b had a synergistic effect on temozolomide-induced inhibition of secondary neurosphere and soft agar colony,and on cell growth inhibition rates.MiR-181b functions as a tumor suppressor that suppresses proliferation and reduces chemoresistance to temozolomide in glioma stem cells. | Ping Li Xiaoming Lu Yingyi Wang Lihua Sun Chunfa Qian Wei Yan Ning Liu Yongping You Zhen Fu | 2010 | The Journal of Biomedical Research2010,24,6: | 14 |

| 3 | Growth monitoring of greenhouse lettuce based on a convolutional neural network显示文摘Growth-related traits,such as aboveground biomass and leaf area,are critical indicators to characterize the growth of greenhouse lettuce.Currently,nondestructive methods for estimating growth-related traits are subject to limitations in that the methods are susceptible to noise and heavily rely on manually designed features.In this study,a method for monitoring the growth of greenhouse lettuce was proposed by using digital images and a convolutional neural network(CNN).Taking lettuce images as the input,a CNN model was trained to learn the relationship between images and the corresponding growth-related traits,i.e.,leaf fresh weight(LFW),leaf dry weight(LDW),and leaf area(LA).To compare the results of the CNN model,widely adopted methods were also used.The results showed that the values estimated by CNN had good agreement with the actual measurements,with R^(2) values of 0.8938,0.8910,and 0.9156 and normalized root mean square error(NRMSE)values of 26.00,22.07,and 19.94%,outperforming the compared methods for all three growth-related traits.The obtained results showed that the CNN demonstrated superior estimation performance for the flat-type cultivars of Flandria and Tiberius compared with the curled-type cultivar of Locarno.Generalization tests were conducted by using images of Tiberius from another growing season.The results showed that the CNN was still capable of achieving accurate estimation of the growth-related traits,with R2 values of 0.9277,0.9126,and 0.9251 and NRMSE values of 22.96,37.29,and 27.60%.The results indicated that a CNN with digital images is a robust tool for the monitoring of the growth of greenhouse lettuce. | Lingxian Zhang Zanyu Xu Dan Xu Juncheng Ma Yingyi Chen Zetian Fu | 2020 | Horticulture Research2020,7,1: | 2 |

| 5 | Influencing factors of colostrum exposure to low-level lead and their relationship with early neural development of infants显示文摘BACKGROUND: Exposure to low-level lead has a toxic effect on the development of neonates, which has attracted wide attention. Colostrum lead level can be used as the indication of lead exposure. OBJECTIVE: To observe the relationship of colostrum lead level and the neurobehavioral development of infants. DESIGN: A prospective control observation. SETTING: Center for Maternal and Child Health, Shanxi Provincial Children's Hospital. PARTICIPANTS: Totally 128 neonates of full-term normal delivery, 76 male and 52 female, from Shanxi Provincial Maternal and Child Health Center and Jiexiu Maternal and Child Health Center were involved in this study. All the involved neonates had no peripartal ischemic/hypoxic history or fetus intrauterine developmental lag. Pregnant women had no various acute and chronic diseases in pregnancy, family history of neurological disease or occupational lead exposure. 128 portions of colostrum sample of full-term normal delivery were collected. Informed consents of detected items were obtained from the puerperants and their relatives. METHODS: ① Experimental grouping: Lead level in the colostrum was determined by atomic absorption spectrometry. According to lead level in the colostrum, the neonates were classified into two exposure groups of greater than or equal to 0.24 μmol/L in a high-level lead group and less than 0.24 μ mol/L in a low-level lead group. ② Experimental evaluation: Mental developmental index (MDI) and psychological developmental index (PDI) of 3-month-old infants were evaluated with Bayley Scales of Infant Development (BSID). The relationships of MDI, PDI and colostrum lead level were performed correlation regression analysis; The relationship of colostrum lead level and development was performed multi-factor analysis with family environment and health questionnaires. MAIN OUTCOME MEASURES: ① Evaluation results of MDI and PDI. ② Multi-factor analysis results. RESULTS: Totally 128 neonates were involved in the study. Ten and eleven neonates were lost due to emigration in the high-level lead group and low-level lead group respectively, and the other 107 neonates participated in the final analysis. ① MDI and PDI in the high-level lead group were significantly lower than those in the low-level lead group, respectively (P < 0.01); Regression analysis results showed that two developmental indexes were statistically negatively correlated with colostrum lead level (regression equation y = 1.9+0.01x1,-0.04x2,+0.04x3,+0.03x4). ② Four variables of the factors included by family environment and health questionnaires were taken into equation. Large maternal age, irrational dietary pattern in pregnancy and pollution degree of habitation environment in pregnancy were the risk factors of colostrum lead level (partial regression coefficien t =0.598 4,0.426 8,0.306 7,P < 0.05-0.01), and calcium supplementation in pregnancy was a protective factor (partial regression coefficien t =-0.455 8, P < 0.01). | 8 | Analysis of amino acid enantiomers in ambient aerosols:Effects and removal of coexistent aerosol matrix显示文摘Amino acids(AAs)including D-and L-enantiomers are a group of organic nitrogen species in ambient aerosol.Due to the low abundances of AAs(level of ng/m^(3))and the matrix effects by coexistent components,it is challenging to quantify AA enantiomers in ambient aerosols especially under pollution conditions.In this study,we present an optimized method for analyzing AA enantiomers in atmospheric aerosol samples including a pretreatment process and the detection by high performance liquid chromatography coupled to a fuorescence detector(HPLC-FLD).Matrix effects caused by coexistent chemicals on AA enantiomers analysis in ambient aerosol samples were investigated especially for those collected in pollution episodes.The results revealed that the determination of AA enantiomers is significantly affected by the coexistent organic carbon(as a proxy of organic matter)and water-soluble ion of NH_(4)^(+).To remove the matrix effects,we applied a pretreatment using the solid phase extraction column coupled with alkaline adjustment to sample extract.After pretreatment,18 AAs including 6 pairs of D-and L-enantiomers(i.e.,leucine,isoleucine,valine,alanine,serine,and aspartic acid)can be successfully separated and quantified in aerosol samples by HPLC-FLD.The recoveries are in the range of 67%-106%.This method was successfully applied to the urban aerosol samples from pollution and non-pollution periods for AA enantiomers determination.We suggest that the concentrations of D-AAs and the ratio of D-AA/L-AA are indicative of the contribution of bacterial sources and the infuence of biomass burning. | Ying Li Xiaoying Li Libin Wu Luhan Shi ShanWang Pingqing Fu Yingyi Zhang Senchao Lai | 2024 | Journal of Environmental Sciences2024,,3: | 0 |
