1Adsorbents in blood purification: From lab search to clinical therapy显示文摘Various types of porous resins adsorbents based on polystyrene,polyvinyl alcohol,cellulose as matrixes and DNA,amino acids with other biological active molecules as ligands were extensively studied at Nankai University.Molecular recognition between the ligand and pathogenic molecule was investigated.Several commercialized products are now widely used in hospitals all over China.Whole blood hemoperfusion are used to treat patients suffering from autoimmune diseases,uremia acute intoxication and hyperbilirubinemia.Clinical performances of hundreds and thousands of patients,treated by whole blood sorption therapy are safe,efficient(90%)and cost-effective.YU YaoTing 2013Chinese Science Bulletin2013,58,35:4
2High performance of a unique mesoporous polystyrene-based adsorbent for blood purification显示文摘A multi-functional polystyrene based adsorbent(NKU-9)with a unique mesoporous and a high surface area was prepared by suspension polymerization for removal of therapeutic toxins in blood purification.The adsorbent produced had an almost equal amount of mesopore distribution in the range from 2 to 50 nm.The adsorption of serum toxins with different molecular weights were examined by in vitro adsorption assays and compared with some clinical currently used adsorbents such as HA-330,Cytosorb and BL-300 which are produced by China,America and Japan,respectively.Test results indicated that the adsorption rate for pentobarbital by NKU-9 was 81.24%which is nearly as high as HA-330(81.44%).The latter adsorbent is currently used for acute detoxification treatment in China.To reach adsorption equilibrium,NKU-9 was faster than HA-330,which implies short treatment time.For the removal of middle molecular toxins such as b2-microglobulin(98.88%),NKU-9 performed better adsorptive selectivity than Cytosorb(92.80%).In addition,NKU-9 showed high performance for the removal of albumin-bound toxins(e.g.,bilirubin),and its adsorption rate for total bilirubin(80.79%)in plasma was 8.4%higher than that of anion exchange resin BL-300 which is currently used to eliminate bilirubin in clinic.Therefore,our results indicate that the newly developed adsorbent with a wide distribution and almost equal amount of mesopores is a multifunctional adsorbent for high efficient removal of serum toxins with different molecular weights which might be an excellent blood purification adsorbent especially to treat diseases that conventional medical methods are low or not efficient.Jian Chen Wenyan Han Jie Chen Wenhui Zong Weichao Wang Yue Wang Guanghui Cheng Chunran Li Lailiang Ou Yaoting Yu 2017Regenerative Biomaterials2017,4,1:3
3New method for the preparation of adsorbent with high adsorption capacity显示文摘Polyamidoamine (PAMAM) is a type of macro- molecule having a definite chemical structure and a large number of terminal functional groups. The synthetic ap-proach of the adsorbent with high adsorption capacity in-volved arching of PAMAM onto cellulose beads by reaction with epichlorohydrin. The product thus prepared had a large number of amino groups of 2.94 mmol/g derived from 0.23 mmol/g epoxy groups on cellulose beads. 4Non-viral gene carrier mediated short hairpin RNA interference for inhibition of tumor cells growth显示文摘A tumor-targeting gene vector G250mAb-PEI-PEG has been prepared by modification of polyethylenimine (PEI) with polyethyleneglycol (PEG) and G250, a monoclonal antibody against the G250 antigen on tumor cell surface. The transfection efficiency was as high as 70% in G250 positive HeLa cells, whereas the transfection efficiency was relatively low (30%) in normal NIH3T3 cells. A plasmid encoding the short hairpin RNA (shRNA) specific for nucleostemin gene (NS) was efficiently transfected into the HeLa cells with this nonviral gene vector. RNA interference down-regulated the expression of NS gene in HeLa cells, inhibited cells proliferation and induced apoptosis. However, the growth and activity of the NIH3T3 cells were not affected under the same treatment. 5Interaction between poly(amidoamine) dendrimers and DNA studied by spectroscopic methods显示文摘The interaction between amino-terminated, and ethylenediamine core poly(amidoamine) (PAMAM) den-drimers and herring sperm DNA was investigated by various spectroscopic methods including UV spectroscopy, fluores-cence spectroscopy, microscopic FTIR- and circular dichro-ism (CD-) spectroscopy. Ethidium bromide (EB) is used as a nucleic acid probe for this study. Experimental results show that PAMAM dendrimers can form stable complexes with DNA and the dendrimers bind to DNA sufficiently strong which cannot be displaced by EB, and we also found that the formation of the complexes can cause the conformation change of the DNA secondary structure. 9Fluorescence study on the interactions of PAMAM dendrimers and their derivatives with bovine serum albumin显示文摘The interactions of amino-terminated, and ethylenediamine core poly(amidoamine) (PAMAM) den-drimers and their derivatives with bovine serum albumin (BSA) were investigated by fluorescence spectroscopy. Ex-perimental results showed that the fluorescence intensity of BSA decreased after the addition of different modified den-drimers, and the extent of the fluorescence quenching caused by various modified dendrimers strongly depends upon the different functional groups on their surfaces. We also inves-tigated the influence of pH and ionic strength on the interac-tion between various modified dendrimers and BSA. 18ERCP combined with colonoscopy in the treatment of biliary-colonic fistula:case report and literature review显示文摘Introduction Internal biliary fistula is a potentially serious complication of laparoscopic cholecystectomy[1].Biliary-colonic fistula is a rare type of internal biliary fistula.Formation of a biliary-colonic fistula can occur as the result of surgical injury,stone obstruction,and biliary hypertension.More infrequent causes include intestinal ulcer and malignant tumor.The formation of a fistula also has the effect of infection and inflammation.While very few cases of biliary-colonic fistulas have been reported,these cases typically present with the aforementioned clinical presentations[2,3].In the present case,a biliary-colonic fistula resulting from bile-duct injury after laparoscopic cholecystectomy was identified by endoscopic retrograde cholangiopancreatography(ERCP).We innovatively adopted ERCP combined with colonoscopy to clamp the sinus tract for the patient.Hongfei He Tingting Yu Yaoting Li Senlin Hou Lichao Zhang 2023Gastroenterology Report2023,11,1:0
19STRUCTURAL AND ADSORPTION RELATIONSHIP OF MACROPOROUS R ESIN IN HEMOPERFUSION显示文摘In order to achieve high adsorption capacity of adsorbents in hemoperfusion,macroporous resins were tailor made with appropriate pore sizes and specific area. This allows the toxin molecules to diffuse freely within the channels ofthe porous resins and rinally reach the adsorption sites.Macroporous resin NK - 107 having small pore size and large Surface area(1086cm2) were synthesized which showed a high adsorption capacity of 82 - 94%for small molecules such as hypnotic and sedative drugs. For the adsorption of middle molecules i. e. Vit B12 or cytochrome C adsorbents having large pore sizes weretailor made. In the case of the adsorption of unconjugated bilirubin which is a verybig molecule, the pore size of the adsorbent should be at least>160A. NK - 110 istailor made with a very big pore size,which have a high adsorption capacity of 87%for bilirubin. DNA immobilized on carbonized resins,which have a porous structurewith a large surface area of 1000m2/g,can adsorb DNA antibody effectively.
