1Fabrication of gelatin methacryloyl hydrogel microneedles for transdermal delivery of metformin in diabetic rats显示文摘Injection therapy for diabetes has poor patient compliance,and the pain occurring at the site of subcutaneous injections causes significant inconvenience to diabetic patients.In this work,to demonstrate the benefits of an alternative drug delivery technique that overcomes these issues,methacrylated gelatin hydrogel-forming microneedles integrated with metformin were developed to adjust blood glucose levels in diabetic rats.Gelatin methacryloyl microneedles(GelMA-MNs)with different degrees of substitution were successfully prepared by a micro-molding method.The resultant GelMA-MNs exhibited excellent mechanical properties and moisture resistance.Metformin,an anti-diabetic drug,was further encapsulated into the GelMA-MNs,and its release rate could be controlled by the three-dimensional cross-linked network of microneedles,thereby exhibiting sustained drug release behaviors in vitro and implying a better therapeutic effect compared with that of subcutaneous injection in diabetic rats.The drug release period could be significantly prolonged by improving the cross-link density of GelMA-MNs.The results of hypoglycemic effect evaluation show that the application of GelMA-MNs for transdermal delivery in diabetic rats has promising benefits for diabetes treatment.Zhiyong Zeng Guohua Jiang Tianqi Liu Gao Song Yanfang Sun Xueya Zhang Yanting Jing Mingjia Feng Yufei Shi 2021Bio-Design and Manufacturing2021,4,4:4

5Genome degradation promotes Salmonella pathoadaptation by remodeling fimbriae-mediated proinflammatory response显示文摘Understanding changes in pathogen behavior(e.g.increased virulence,a shift in transmission channel)is critical for the public health management of emerging infectious diseases.Genome degradation via gene depletion or inactivation is recognized as a pathoadaptive feature of the pathogen evolving with the host.However,little is known about the exact role of genome degradation in affecting pathogenic behavior,and the underlying molecular detail has yet to be examined.Using large-scale global avian-restricted Salmonella genomes spanning more than a century,we projected the genetic diversity of Salmonella Pullorum(bvSP)by showing increasingly antimicrobial-resistant ST92 prevalent in Chinese flocks.The phylogenomic analysis identified three lineages in bvSP,with an enhancement of virulence in the two recently emerged lineages(L2/L3),as evidenced in chicken and embryo infection assays.Notably,the ancestor L1 lineage resembles the Salmonella serovars with higher metabolic flexibilities and more robust environmental tolerance,indicating stepwise evolutionary trajectories towards avian-restricted lineages.Pan-genome analysis pinpointed fimbrial degradation from a virulent lineage.The later engineeredfim-deletion mutant,and all other five fimbrial systems,revealed behavior switching that restricted horizontal fecal-oral transmission but boosted virulence in chicks.By depleting fimbrial appendages,bvSP established persistent replication with less proinflammation in chick macrophages and adopted vertical transovarial transmission,accompanied by ever-increasing intensification in the poultry industry.Together,we uncovered a previously unseen paradigm for remodeling bacterial surface appendages that supplements virulence-enhanced evolution with increased vertical transmission.Xiao Zhou Xiamei Kang Jiaqi Chen Yan Song Chenghao Jia Lin Teng Yanting Tang Zhijie Jiang Xianqi Peng Xiaoxi Tao Yiwei Xu Linlin Huang Xuebin Xu Yaohui Xu Tengfei Zhang Shenye Yu Jiansen Gong Shaohui Wang Yuqing Liu Guoqiang Zhu Corinna Kehrenberg Franois-Xavier Weill Paul Barrow Yan Li Guoping Zhao Min Yue 2023National Science Review2023,10,10:0
6Synergistic effects of AAGL and anti-PD-1 on hepatocellular carcinoma through lymphocyte recruitment to the liver显示文摘Objective:Therapy for hepatocellular carcinoma(HCC)is a major challenge,and targeted therapies provide only a modest benefit in terms of overall survival.Treatment with antibodies to programmed cell death protein 1(PD-1)/PD-L1 can restore the functions of tumor-infiltrating T cells in HCC and has shown clinical efficacy in 20%of patients with advanced HCC.Novel approaches are urgently needed to treat HCC and to augment the efficacy of immunotherapy.Methods:Tumor-bearing mice were treated with Agrocybe aegerita galectin(AAGL)alone or in combination with anti-PD-1,and the tumor sizes and lifespans of mice were determined.Transcriptome analysis,cytokine analysis,flow cytometry analysis of the number and proportion of immune cell subsets in the liver and spleen,and molecular and cellular analyses of tumors were used to define the underlying mechanisms.Results:AAGL significantly inhibited the growth of liver tumors in a dose-dependent manner.Furthermore,AAGL increased the expression of multiple cytokines and chemokines in tumor-bearing mouse livers;this effect was associated with the activation and migration of T cells and macrophages,in agreement with the in vitro results.Importantly,the aggregation of T cells and macrophages induced by AAGL in tumor-bearing mouse livers clearly enhanced the response to PD-1 blockade immunotherapy.Conclusions:The results showed that AAGL induced the activation and migration of lymphocytes to the liver,and that the combination of AAGL and anti-PD-1 may be a promising strategy for HCC treatment.Xiangdong Ye Xueqing Wang Wenhui Yu Qing Yang Yan Li Yanxia Jin Yanting Su Jiaqi Song Bo Xu Hui Sun 2021Cancer Biology & Medicine2021,18,4:0
7Myeloid peroxisome proliferator-activated receptor α deficiency accelerates liver regeneration via IL-6/STAT3 pathway after 2/3 partial hepatectomy in mice显示文摘Background:Liver regeneration is a fundamental process for sustained body homeostasis and liver function recovery after injury.Emerging evidence demonstrates that myeloid cells play a critical role in liver regeneration by secreting cytokines and growth factors.Peroxisome proliferator-activated receptorα(PPARα),the target of clinical lipid-lowering fibrate drugs,regulates cell metabolism,proliferation,and survival.However,the role of myeloid PPARαin partial hepatectomy(PHx)-induced liver regeneration remains unknown.Methods:Myeloid-specific PPARa-deficient(Ppara^(Mye−/−))mice and the littermate controls(Ppara^(fl/fl))were subjected to sham or 2/3 PHx to induce liver regeneration.Hepatocyte proliferation and mitosis were assessed by immunohistochemical(IHC)staining for 5-bromo-2'-deoxyuridine(BrdU)and Ki67 as well as hematoxylin and eosin(H&E)staining.Macrophage and neutrophil infiltration into livers were reflected by IHC staining for galectin-3 and myeloperoxidase(MPO)as well as flow cytometry analysis.Macrophage migration ability was evaluated by transwell assay.The mRNA levels for cell cycle or inflammation-related genes were measured by quantitative real-time RT-PCR(qPCR).The protein levels of cell proliferation related protein and phosphorylated signal transducer and activator of transcription 3(STAT3)were detected by Western blotting.Results:Ppara^(Mye−/−)mice showed enhanced hepatocyte proliferation and mitosis at 32 h after PHx compared with Ppara^(fl/fl)mice,which was consistent with increased proliferating cell nuclear antigen(Pcna)mRNA and cyclinD1(CYCD1)protein levels in Ppara^(Mye−/−)mice at 32 h after PHx,indicating an accelerated liver regeneration in Ppara^(Mye−/−)mice.IHC staining showed that macrophages and neutrophils were increased in Ppara^(Mye−/−)liver at 32 h after PHx.Livers of Ppara^(Mye−/−)mice also showed an enhanced infiltration of M1 macrophages at 32 h after PHx.In vitro,Ppara-deficient bone marrow-derived macrophages(BMDMs)exhibited markedly enhanced migratory capacity and upregulated M1 genes Il6 and Tnfa but downregulated M2 gene Arg1 expressions.Furthermore,the phosphorylation of STAT3,a key transcript factor mediating IL6-promoted hepatocyte survival and proliferation,was reinforced in the liver of Ppara^(Mye−/−)mice after PHx.Conclusions:This study provides evidence that myeloid PPARαdeficiency accelerates PHx-induced liver regeneration via macrophage polarization and consequent IL-6/STAT3 activation,thus providing a potential target for manipulating liver regeneration.Guomin Xie Yanting Song Na Li Zhenzhen Zhang Xia Wang Ye Liu Shiyu Jiao Ming Wei Baoqi Yu Yan Wang Hua Wang Aijuan Qu 2022Hepatobiliary Surgery and Nutrition2022,11,2:0

10Four temporin-derived peptides exhibit antimicrobial and antibiofilm activities against methicillin-resistant Staphylococcus aureus显示文摘Temporin-GHa(GHa)was cloned from Hylarana guentheri,showing a weak antimicrobial activity.In order to improve its bactericidal efficacy,GHaR6R,GHaR7R,GHaR8R and GHaR9W were designed and synthesized.Compared to the parent peptide,the GHa-derived peptides show potent antimicrobial activities against methicillin-resistant Staphylococcus aureus(MRSA),which is the main pathogen with high morbidity and mortality that causes various infections in humans.These peptides exert bactericidal actions on MRSA by permeabilizing the cytoplasmic membranes and damaging membrane integrity.All of the four peptides exhibit excellent stability under harsh conditions,including extreme temperature and salts.Furthermore,they inhibit the formation of biofilm and eradicate mature biofilm of MRSA.The GHa-derived peptides decrease bacterial surface hydrophobicity,autoaggregation and polysaccharide intercellular adhesion synthesis in concentration-dependent manner.Real-time quantitative reverse transcription PCR analysis revealed that the peptides downregulatethe expression of adhesion genes icaADBC involved in biofilm formation.Except for GHaR7R,the other three peptides have low hemolytic toxicity against human erythrocytes.In the presence of human erythrocytes,GHaR7R,GHaR8R and GHaR9W interact with MRSA preferentially.GHaR6R,GHaR8R and GHaR9W show less toxicity toward normal cells HL-7702 and hFOB1.19.These results suggest that the GHa-derived peptides may be promising antimicrobial candidates against MRSA infections.Xiuchuan Tan Shenghong Xie Xiao Jin Ruiying Zhu Shuangshuang Wei Yanting Song Xi Xie Rong Wang Lushuang Li Manchuriga Wang Yingxia Zhang 2022Acta Biochimica et Biophysica Sinica2022,54,3:0

14Retrieval of Winter Wheat Canopy Carotenoid Content with Ground-and Airborne-Based Hyperspectral Data显示文摘Accurate assessment of canopy carotenoid content(CC_(x+c)C)in crops is central to monitor physiological conditions in plants and vegetation stress,and consequently supporting agronomic decisions.However,due to the overlap of absorption peaks of carotenoid(C_(x+c))and chlorophyll(C_(a+b)),accurate estimation of carotenoid using reflectance where carotenoid absorb is challenging.The objective of present study was to assess CC_(x+c)C in winter wheat(Triticum aestivum L.)with ground-and aircraft-based hyperspectral measurements in the visible and near-infrared spectrum.In-situ hyperspectral reflectance were measured and airborne hyperspectral data were acquired during major growth stages of winter wheat in five consecutive field experiments.At the canopy level,a remarkable linear relationship(R^(2)=0.95,p<0.001)existed between C_(x+c) and Ca+b,and correlation between CC_(x+c)C and wavelengths within 400 to 1000 nm range indicated that CC_(x+c)C could be estimated using reflectance ranging from visible to near-infrared wavebands.Results of Cx+c assessment based on chlorophyll and carotenoid indices showed that red edge chlorophyll index(CI red edge)performed with the highest accuracy(R^(2)=0.77,RMSE=22.27μg/cm^(2),MAE=4.97μg/cm^(2)).Applying partial least square regression(PLSR)in CC_(x+c)C retrieval emphasized the significance of reflectance within 700 to 750 nm range in CC_(x+c)C assessment.Based on CI red edge index,use of airborne hyperspectral imagery achieved satisfactory results in mapping the spatial distribution of CC_(x+c)C.This study demonstrates that it is feasible to accurately assess CC_(x+c)C in winter wheat with red edge chlorophyll index provided that C_(x+c) correlated well with C_(a+b) at the canopy scale.it is therefore a promising method for CC_(x+c)C retrieval at regional scale from aerial hyperspectral imagery.Ting Cui Xianfeng Zhou Yufeng Huang Yanting Guo Yunrui Lin Leyi Song Jingcheng Zhang 2023Phyton-International Journal of Experimental Botany2023,92,9:0
