| 1 | In vitro assessment of thyroid hormone receptor activity of four organophosphate esters显示文摘Previous animal experiments have implied that organophosphate esters(OPEs) have a disruption effect on the thyroid endocrine system. However, knowledge of the toxicological mechanism remains limited. In this study, the activities of four OPEs have been characterized against the thyroid hormone(TH) nuclear receptor(TR) using two in vitro models, with the aim of evaluating their toxicity mechanisms towards the TR. The results of a TH-dependent cell proliferation assay showed that tris(2-chloro-1-(chloromethyl)ethyl)phosphate(TDCPP) could induce cell growth, while the other three OPEs had no effect. The results of a luciferase reporter gene assay revealed that all four of the OPEs tested in the current study showed agonistic activity towards TRβ, with TDCPP being the most potent one. Moreover, molecular docking revealed that all the tested OPEs could fit into the ligand binding pocket of TRβ, with TDCPP binding more effectively than the other three OPEs. | 2 | Four-year dynamic observation and study on standardized management of elderly patients with type 2 diabetes in Beijing Yongding Road Community显示文摘Purpose:This study aimed to understand the joint standard of blood glucose,blood li-pids,and blood pressure in elderly patients>60 years of age with type 2 diabetes in the Beijing Yongding Road Community.Methods:One hundred ninety-two elderly patients>60 years of age who were diagnosed with type 2 diabetes in Yongding Road Community served as the investigation subjects,under-went standard management,and the dynamic changes in blood glucose,blood lipids,and blood pressure were monitored for 48 months.Results:At the end of the observation period,the standard rates of fasting blood glucose and postprandial blood glucose were 72.55%and 80.00%,respectively,which were increased compared with 55.73%and 56.08%at baseline(P<0.01).The standard rate of HbA1c was 59.81%,which was increased compared with 53.44%at baseline(P>0.05).The standard rates of TG and LDL-C were 76.71%and 60.38%,respectively,which were increased compared with 54.69%and 34.74%at base-line,and the standard rate of HDL-C was 13.64%,which was decreased compared with 40.10%at baseline(P<0.01).The standard rate of BP was 58.33%,which was increased compared with 38.54%at baseline(P<0.01).The optimal control rate of blood glucose for 48 months(the standard times of the total measurement times in 48 months≥75%)was higher;the fasting blood glucose was 52.17%,the 2-h postprandial blood glucose was 60.22%,and the HbA1c was 46.45%.The optimal control rate of blood lipids was lower;LDL-C was 17.49%and HDL-C was 13.59%.The optimal control rate of BP was 9.13%.At the end of the observation period,the levels of fasting blood glucose and postprandial blood glucose were decreased by 0.7 mmol/L and 1.48 mmol/L,respectively,compared with the base-line(P<0.01).The level of HbA1c was decreased by 0.18%compared with the baseline(P<0.05).The levels of LDL-C and HDL-C were decreased by 0.4 mmol/L and 0.23 mmol/L,respectively,compared with the baseline(P<0.01).The levels of SBP and DBP were decreased by 4 mmHg compared with the baseline(P<0.01).At the end of the observation period,the joint standard rate of the three indices of HbA1c,LDL-C,and BP was 24.72%,which was increased compared with 6.25%at baseline(P<0.01).Conclusion:Standardized management of elderly patients with diabetes in the Community can improve the joint standard rate of blood glucose,blood lipids,and blood pressure.The op-timal control rate and joint standard rate are the important indices for evaluating the quality of diabetes management. | Yue Li Dongyun Shen Wei Zhang Yimin Wang Hongmei Li Jie Geng Liying Yang Zhijun Xiao Yan Liu Zhigang Feng Linying Cao Yan Jin Lixia Shi | 2014 | Family Medicine and Community Health2014,2,2: | 4 |

| 5 | Combination of a biopharmaceutic classification system and physiologically based pharmacokinetic models to predict absorption properties of baicalein in vitro and in vivo显示文摘Objective: To determine the in vitro and in vivo absorption properties of active ingredients of the Chinese medicine, baicalein, to enrich mechanistic understanding of oral drug absorption.Methods: The Biopharmaceutic Classification System(BCS) category was determined using equilibrium solubility, intrinsic dissolution rate, and intestinal permeability to evaluate intestinal absorption mechanisms of baicalein in rats in vitro. Physiologically based pharmacokinetic(PBPK) model commercial software GastroPlus~(TM) was used to predict oral absorption of baicalein in vivo.Results: Based on equilibrium solubility, intrinsic dissolution rate, and permeability values of main absorptive segments in the duodenum, jejunum, and ileum, baicalein was classified as a drug with low solubility and high permeability. Intestinal perfusion with venous sampling(IPVS) revealed that baicalein was extensively metabolized in the body, which corresponded to the low bioavailability predicted by the PBPK model. Further, the PBPK model predicted the key indicators of BCS, leading to reclassification as BCS-II. Predicted values of peak plasma concentration of the drug(C_(max)) and area under the curve(AUC)fell within two times of the error of the measured results, highlighting the superior prediction of absorption of baicalein in rats, beagles, and humans. The PBPK model supported in vitro and in vivo evidence and provided excellent prediction for this BCS class II drug.Conclusion: BCS and PBPK are complementary methods that enable comprehensive research of BCS parameters, intestinal absorption rate, metabolism, prediction of human absorption fraction and bioavailability, simulation of PK, and drug absorption in various intestinal segments across species. | 16 | Assimilation of Radar and Cloud-to-Ground Lightning Data Using WRF-3DVar Combined with the Physical Initialization Method——A Case Study of a Mesoscale Convective System显示文摘Radar data, which have incomparably high temporal and spatial resolution, and lightning data, which are great indicators of severe convection, have been used to improve the initial field and increase the accuracies of nowcasting and short-term forecasting. Physical initialization combined with the three-dimensional variational data assimilation method(PI3 DVarrh) is used in this study to assimilate two kinds of observation data simultaneously, in which radar data are dominant and lightning data are introduced as constraint conditions. In this way, the advantages of dual observations are adopted. To verify the effect of assimilating radar and lightning data using the PI3 DVarrh method, a severe convective activity that occurred on 5 June 2009 is utilized, and five assimilation experiments are designed based on the Weather Research and Forecasting(WRF) model. The assimilation of radar and lightning data results in moister conditions below cloud top, where severe convection occurs;thus, wet forecasts are generated in this study.The results show that the control experiment has poor prediction accuracy. Radar data assimilation using the PI3 DVarrh method improves the location prediction of reflectivity and precipitation, especially in the last 3-h prediction, although the reflectivity and precipitation are notably overestimated. The introduction of lightning data effectively thins the radar data, reduces the overestimates in radar data assimilation, and results in better spatial pattern and intensity predictions. | 17 | A Computational Study of Asymmetric Hydrogenation of 2-Phenyl Acrylic Acids Catalyzed by a Rh(l) Catalyst with Ferrocenyl Chiral Bisphosphorus Ligand: The Role of Ion-Pair Interaction显示文摘Asymmetric hydrogenation reaction is one of the most efficient synthetic methods to form useful chiral compounds for synthetic chemistry,medicinal chemistry and material chemistry.Generally,the enantioselectivity of many hydrogenation reactions is controlled by steric hindrance between the chiral ligand and substrate.Recently,Zhang group developed a highly asymmetric hydrogenation of 2-aryl and 2-alkyl acrylic acids catalyzed by a Rh(l)catalyst with a chiral Wudaphos ligand.The excellent enantioselectivity of this asymmetric reaction was proposed to be controlled by ion-pair interaction between the substrate and chiral ligand.In this study,a systematic density functional theory study has been carried out to investigate the reaction mechanism and origin of the enantioselectivity.Our computational results suggest that this reaction follows the classic mechanism involving oxidative addition of H_(2),migratory insertion and reductive elimination.Different from the C=C coordination to the metal in the common oxidative addition step,our study found that the chelation of the carboxyl group of the substrate to the cationic Rh(l)metal is more favorable in this oxidative addition step.The high enantioselectivity is proposed to be dictated by a better catalyst/substrate geometric complementarity in the major pathway to have less distortion of the catalyst for a strong ion-pair interaction. | Xiangru Fan Lini Zheng Yuhong Yang Xiu-Qin Dong Xumu Zhang Lung-Wa Chung | 2021 | Chinese Journal of Chemistry2021,39,6: | 0 |
| 18 | Robust Benchmark Structural Variant Calls of An Asian Using State-of-the-art Long-read Sequencing Technologies显示文摘The importance of structural variants(SVs)for human phenotypes and diseases is now recognized.Although a variety of SV detection platforms and strategies that vary in sensitivity and specificity have been developed,few benchmarking procedures are available to confidently assess their performances in biological and clinical research.To facilitate the validation and application of these SV detection approaches,we established an Asian reference material by characterizing the genome of an Epstein-Barr virus(EBV)-immortalized B lymphocyte line along with identified benchmark regions and high-confidence SV calls.We established a high-confidence SV callset with 8938 SVs by integrating four alignment-based SV callers,including 109×Pacific Bio sciences(PacBio)continuous long reads(CLRs),22×PacBio circular consensus sequencing(CCS)reads,104×Oxford Nanopore Technologies(ONT)long reads,and 114×Bionano optical mapping platform,and one de novo assembly-based SV caller using CCS reads.A total of 544 randomly selected SVs were validated by PCR amplification and Sanger sequencing,demonstrating the robustness of our SV calls.Combining trio-binning-based haplotype assemblies,we established an SV benchmark for identifying false negatives and false positives by constructing the continuous high-confidence regions(CHCRs),which covered 1.46 gigabase pairs(Gb)and 6882 SVs supported by at least one diploid haplotype assembly.Establishing high-confidence SV calls for a benchmark sample that has been characterized by multiple technologies provides a valuable resource for investigating SVs in human biology,disease,and clinical research. | Xiao Du Lili Li Fan Liang Sanyang Liu Wenxin Zhang Shuai Sun Yuhui Sun Fei Fan Linying Wang Xinming Liang Weijin Qiu Guangyi Fan Ou Wang Weifei Yang Jiezhong Zhang Yuhui Xiao Yang Wang Depeng Wang Shoufang Qu Fang Chen Jie Huang | 2022 | Genomics, Proteomics & Bioinformatics2022,20,1: | 0 |
| 19 | Network pharmacology and subsequent experimental validation reveal the synergistic myocardial protection mechanism of Salvia miltiorrhiza Bge.and Carthamus tinctorius L.显示文摘Objective:To reveal the molecular mechanism underlying the compatibility of Salvia miltiorrhiza Bge(S.miltiorrhiza,Dan Shen)and C.tinctorius L.(C.tinctorius,Hong Hua)as an herb pair through network pharmacology and subsequent experimental validation.Methods:Network pharmacology was applied to construct an active ingredient-efficacy target-disease protein network to reveal the unique regulation pattern of s.miltiorrhiza and C.tinctorius as herb pair.Molecular docking was used to verify the binding of the components of these herbs and their potential targets.An H9c2 glucose hypoxia model was used to evaluate the efficacy of the components and their synergistic effects,which were evaluated using the combination index.Western blot was performed to detect the protein expression of these targets.Results:Network pharmacology analysis revealed 5 pathways and 8 core targets of s.miltiorrhiza and C.tinctorius in myocardial protection.Five of the core targets were enriched in the hypoxia-inducible factor-1(HIF-1)signaling pathway.S.miltiorrhiza-C.tinctorius achieved vascular tone mainly by regulating the target genes of the HIF-1 pathway.As an upstream gene of the HIF-1 pathway,STAT3 can be activated by the active ingredients cryptotanshinone(Ctan),salvianolic acid B(Sal.B),and myricetin(Myric).Cell experiments revealed that Myric,Sal.B,and Ctan also exhibited synergistic myocardial protective activity.Molecular docking verified the strong binding of Myric,Sal.B,and Ctan to STAT3.Western blot further showed that the active ingredients synergistically upregulated the protein expressionof STAT3.Conclusion:The pharmacodynamic transmission analysis revealed that the active ingredients of S.miltiorrhiza and C.tinctorius can synergistically resist ischemia through various targets and pathways.This study provides a methodological reference for interpreting traditional Chinese medicine compatibility. | Linying Zhong Ling Dong Jing Sun Jie Yang Zhiying Yu Ping He Bo Zhu Yuxin Zhu Siyuan Li Wenjuan Xu | 2024 | Journal of Traditional Chinese Medical Sciences2024,11,1: | 0 |
