2Curcumin,a potent anti-tumor reagent,is a novel histone deacetylase inhibitor regulating B-NHL cell line Raji proliferation显示文摘Aim: To investigate curcumin (diferuloylmethane) induced apoptosis and its molecular mechanism of action in B-NHL cell line Raji cells. Methods: Raji cells were cultured in RPMI-1640 medium and treated with curcumin in different concentrations. 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium (MTT) assay was used to detect growth inhibition and Hoechst 33258 staining was used to detect apoptosis. Immunocytochemistry and Western blot were used to detect the expressions of histone deacetylase 1, 3, and 8 (HDAC1, HDAC3, and HDAC8) and acetylated histone H4 (Ac-histone H4) protein. Results: Curcumin inhibited the proliferation of B-NHL cell line Raji cells with a 36-h IC50 value of 24.1±2.0μmol/L. Hoechst 33258 staining showed that curcumin could induce Raji cell apoptosis.The expression levels of HDAC1, HDAC3, and HDAC8 proteins were downregulated following curcumin treatment in Raji cells, whereas Ac-histone H4 protein expression was upregulated after treatment with curcumin. Conclusion: Curcumin, as a new member of the histone deacetylase inhibitors, can inhibit the expression of class Ⅰ HDACs (HDAC1, HDAC3, and HDAC8), and can increase the expression of Ac-histone H4 in Raji cells. 3Expression of p16 gene and Rb protein in gastric carcinoma and their clinicopathological significance显示文摘AIM: To analyze the correlation between the protein expression of p16 and Rb genes in gastric carcinoma (GC),to investigate the role of p16 gene in invasion and lymph node metastasis of GC, and to examine the deletion and mutation in exon 2 of p16 gene in GC.METHODS: The protein expression of p16 and Rb genes was examined by streptavidin-peroxidase conjugated method (S-P) in normal gastric mucosa, dysplastic gastric mucosa and GC. The deletion and mutation of p16 gene were examined by polymerase chain reaction (PCR) and polymerase chain reaction single strand conformation polymorphism (PCR-SSCP) respectively in normal gastric mucosa and GC.RESULTS: The positive rates of P16 and Rb protein expression respectively were 96% (77/80) and 99%(79/80) in normal gastric mucosa, 92% (45/50) and 80%(40/50) in dysplastic gastric mucosa, 48% (58/122) and 60% (73/122) in GC. The positive rates of P16 and Rb protein expression in GC were significantly lower than that in normal gastric mucosa and dysplastic gastric mucosa (P<0.05). The positive rate of P16 protein expression in mucoid carcinoma (10%, 1/10) was significantly lower than that in poorly differentiated carcinoma (51%, 21/41),undifferentiated carcinoma (58%, 15/26) and signet ring cell carcinoma (62%, 10/16) (P<0.05). The positive rates of P16 protein in 30 cases of paired primary and lymph node metastatic GC were 47% (14/30) and 17% (5/30)respectively, being significantly lower in the later than in the former (P<0.05). There was no mutation in exon 2 of p16 gene in the 25 freshly resected primary GCs. But five cases in the 25 freshly resected primary GCs displayed deletion in exon 2 of p16 gene. The positive rate of both P16 and Rb proteins was 16% (14/90), and the negative rate of both P16 and Rb proteins was 8% (7/90) in 90GCs. The rate of positive P16 protein with negative Rb protein was 33% (30/90). The rate of negative P16 protein with positive Rb protein was 43% (39/90). There was reverse correlation between P16 and Rb expression in 90GCs (P<0.05).CONCLUSION; The loss protein expression of p16 and Rb genes is related to GC. The loss expression of P16protein is related to the histopathologic subtypes and lymph node metastasis of GC. Expression of P16 and Rb proteins in GC is reversely correlated. 6Regulating expressions of cyclin D1,pRb,and anti-cancer effects of deguelin on human Burkitt's lymphoma Daudi cells in vitro显示文摘Aim: To investigate anticancer effects and molecular mechanism of deguelin on human Burkitt' s lymphoma Daudi cells in vitro and compare the cytotoxicities of deguelin on Daudi ceils and human peripheral blood monocular ceils (PBMC). Methods: The effects of deguelin on the growth of Daudi cells were studied by 3-(4, 5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium (MTT) assay. Apoptos iswere dectected through Hoechst 33258 staining and Annexin V/PI double-labeled cytometry. The effect of deguelin on the cell cycle of Daudi cells were studied by a propidium iodide method. The expressions of cyclin D1 and pRb were checked by Western blot. Results: The proliferation of Daudi cells were decreased in deguelin-treated group with a 24-h IC50 value of 51.55 nmol/L. Deguelin induced Daudi cells apoptosis was in a time- and dose-dependent manner. G0/G1 phase increased and S phase decreased in Daudi cells treated with deguelin. With deguelin 0, 5, 10, 20, and 40 nmol/L treatment for 24 h, G0/G1 phase increased from 37.34% to 56.56%, whereas S phase decreased from 37.72% to 21.36%. PBMC was less sensitive to the cytotoxic effect of deguelin than Daudi cells. The expression of cyclin D1 and pRb protein were decreased sharply in Daudi cells treated with deguelin. Conclusion: Deguelin is able to inhibit the proliferation of Daudi cells by regulating the cell cycle that arrested cells at Go/G, phase and inducing the cell apoptosis. Moreover, deguelin selectively induced apoptosis of Daudi cells with low toxicity in PBMC. 7Composition optimized trimetallic PtNiRu dendritic nanostructures as I versatile and active electrocatalysts for alcohol oxidation显示文摘Platinum-based nano crystals are the most effective electrocatalysts for accelerati ng the chemical tran sformatio ns on the anode in direct alcohol fuel cells. To facilitate practical applications and overcome the drawbacks of diverse alcohols, it is significant to develop electrocatalysts with high activities and a wide fuel flexibility. Here, we demonstrate a practicable solution method for fabricating composition tun able trimetallic PtNiRu den dritic nano structures (DNSs) which can serve as versatile and active catalysts for electrooxidatio n of a variety of liquid alcohols. A series of trimetallic DNSs with tun able Pt/Ni/Ru atomic ratios were successfully syn thesized by simply adjusti ng the feeding of precursors. Detailed electrochemical test indicates that, among other compositions, the Pt66Ni27Ru7 DNSs present much superior electroactivity in catalyzing electrooxidation of liquid alcohols in acidic mediums. Specifically, the mass activity and specific activity on the Pt66Ni27Ru7 DNSs, for electrooxidation of methanol, ethanol, and ethylene glycol, are 4.57 and 4.34 times, 3.55 and 3.42 times, and 2.37 and 2.28 times that of the commercial Pt black, respectively. X-ray photoelectron spectroscopy and CO stripping studies reveal the adsorption of CO on these PtNiRu DNSs is much weaker than on pure Pt. Meanwhile, the surface Ru sites can provide neighbouring -OH groups to facilitate the oxidati on and removal of the adsorbed in termediates (-CO) on the surface Pt sites, effectively improvi ng the CO tolera nee of the catalysts. 8WSe2 2D p-type semiconductor-based electronic devices for information technology:Design,preparation,and applications显示文摘The pioneering exfoliation of monolayer tungsten diselenide has greatly inspired researchers toward semiconducting applications.WSe2 belongs to a family of transition-metal dichalcogenides.Similar to graphene,WSe2 and analogous dichalcogenides have layered structures with weak van der Waals interactions between two adjacent layers.First,the readers are presented with the fundamentals of WSe2,such as types,morphologies,and properties.Here,we report the characterization principles and practices such as microscopy,spectroscopy,and diffraction.Second,the methods for obtaining high-quality WSe2,such as exfoliation,hydrothermal and chemical vapor deposition,are briefly listed.With advantages of light weight,flexibility,and high quantum efficiency,2D materials may have a niche in optoelectronics as building blocks in p-n junctions.Therefore,we introduce a state-of-the-art demonstration of heterostructure devices employing the p-type WSe2 semiconductor.The device architectures include field-effect transistors,photodetectors,gas sensors,and photovoltaic solar cells.Due to its unique electronic,optical,and energy band properties,WSe2 has been increasingly investigated due to the conductivity of the p-type charge carrier upon palladium contact.Eventually,the dynamic research on WSe2 and van der Waals heterostructures is summarized to arouse the passion of the 2D research community.Qilin Cheng Jinbo Pang Dehui Sun Jingang Wang Shu Zhang Fan Liu Yuke Chen Ruiqi Yang Na Liang Xiheng Lu Yanchen Ji Jian Wang Congcong Zhang Yuanhua Sang Hong Liu Weijia Zhou 2020InfoMat2020,2,4:5
9Structural comparisons of meptazinol with opioid analgesics显示文摘Aim: To investigate the mechanism of action of a potent analgesic, (±)-meptazinol. Methods: The structures of meptazinol enantiomers were compared with opioid pharmacophore and tramadol. Results: Neither enantiomer of meptazinol fitted any patterns among the opioid pharmacophore and tramadol, although they did share some structural and pharmacological similarities. However, the structure superpositions implied that both enantiomers of meptazinol might share some similar analgesic mechanisms with typical opiate analgesics. 10Hierarchical yolk-shell structured Li-rich cathode boosting cycling and voltage stabled LIBs显示文摘Despite the high energy density of lithium-rich(Li-rich)cathodes,their implementation is hampered by the unsatisfied rate capacity and poor cycling performance accompanied with substantial voltage decay.To address these issues,the hierarchical yolk-shell structured Li_(1.2)Mn_(0.54)Ni_(0.13)Co_(0.13)O_(2)cathodes(YK-LMNCO)was proposed and synthesized through a facile glycerol assisted solvothermal approach and the following lithiation process.Benefitting from the shortened lithium diffusion lengths and the enhanced tolerance to the large volume variation upon lithium ions intercalation/de-intercalation,the unique structure reciprocates an initial coulombic efficiency of 85.8%,an outstanding capacity retention rate of 89.1%after cycling at 2.0 C for 200 cycles with a minor voltage drop,and a capacity retention rate of 93.8%after cycling at 10.0 C for 500 cycles,85.2%for 1,000 cycles.When assembled with graphite as anode,the YK-LMNCO//graphite full cell shows a remarkable capacity retention rate of 87.2%after cycling at 5.0 C for 50 cycles.Our facile strategy for constructing the yolk-shell structured Li-rich cathodes with high capacity and voltage stability sheds light on synthesizing other lithium storage materials.Yanchen Liu Yafen Chen Jing Wang Wei Wang Zhiyu Ding Leyuan Li Yang Zhang Yida Deng JunweiWu Yanan Chen 2022Nano Research2022,15,4:3
11A preference-based multi-objective building performance optimization method for early design stage显示文摘A large number of cases show that the multi-objective optimization method can significantly improve building performance.The method for multi-objective building performance optimization(BPO)design has achieved rapid development in recent years.However,the BPO method still needs to be improved.Specifically,weak interaction between the optimization process and the decision-making process results in low optimization efficiency,which limits the widespread application of the optimization method in early design stage.In this paper,a new interactive BPO mode is explored to strengthen the interaction between the optimization process and decisionmaking process,and a preference-based multi-objective BPO method is proposed to account for designers'decision preferences during the optimization process,making the objective more controllable,improving the optimization efficiency and ensuring the diversity of solutions.Firstly,this paper illustrates the proposed method in detail,defines the concept of performance preference,expounds the flow of the preference-based multi-objective optimization algorithm,and proposes three indicators to evaluate the algorithm,which includes convergence speed,preference satisfaction rate,and diversity measurement.Secondly,through testing and comparison,it is found that the proposed preference-based algorithm has advantages over the non-preference optimization algorithm(represented by the NSGA-II algorithm).The proposed method leads to faster convergence and higher preference satisfaction,so it is more suitable for the BPO process in the early design stage.Specially,the proposed method can achieve 100%preference satisfaction rate with only 2400 simulations,while the non-preference method can only achieve 20%preference satisfaction rate after 5800 simulations.In this paper,a preference-based multi-objective BPO method is proposed to make the optimization process closely interact with the decision-making process and make the design preferences be accounted during the BPO process,thereby improving the optimization efficiency.In addition,this study first proposes two indicators to measure the quality of optimization results:preference satisfaction rate and diversity measurement.This study aims to guide the development of BPO methods towards providing high satisfaction rate and high quality optimization results.Borong Lin Hongzhong Chen Yanchen Liu Qiushi He Ziwei Li 2021Building Simulation2021,14,3:3

13Ultrasensitive and stable all graphene field-effect transistor-based Hg^(2+)sensor constructed by using different covalently bonded RGO films assembled by different conjugate linking molecules显示文摘As'molecular bridge,'coupling agents can not only realize the covalent connection of composites,but also affect their properties,thus affecting the properties of devices based on them.Herein,leveraging differences in charge conduction properties of the(3-aminopropyl)trimethoxysilane and 5,10,15,20-tetrakis(4-aminophenyl)-21H,23H-porphine caused by conjugacy structural differences,two kinds of layerby-layer assembled smart carbon materials with different electrical properties are obtained at the same reduction temperature.The two graphene ultrathin films are then“planted”on Si/SiO2 substrates,respectively,as semiconductor layer and source/drain electrodes to fabricate an ultra-stable all-graphene field effect transistor(AG-FET).Enabled by the covalent functionalized configuration and the functionally diverse of coupling agents,the AG-FET obtained by this simple method won the high electrical characteristics,the hole,electron mobility,and the shelflife could reach 3.79 cm2/(V·s),3.78 cm2/(V·s),and 18months,respectively.In addition,good material stability and excellent device structure endow the device exceptional stability,electrical stability,and solvent resistance,improving its application prospect in solution phase sensing/detection.Such characteristics could be used to sense,transduce,and respond to external stimuli,especially in solution phase to monitor the important analytes,such as Hg^(2+)in a flowing sewage environment.We believe that such easy-to-manufacture AG-FETs with ultrahigh performance and ultrahigh stability could also show great application prospects in other significant fields.Mingyuan Sun Congcong Zhang Duo Chen Jian Wang Yanchen Ji Na Liang Haoyang Gao Shanshan Cheng Hong Liu 2021SmartMat2021,2,2:2
14Theoretical investigation of lithium ions’ nucleation performance on metal-doped Cu surfaces显示文摘Lithium metal batteries(LMBs)of an ultrahigh theoretical energy density have attracted lots of attentions for a wide range of practical applications.However,there are still numerous challenges in LMBs system,such as poor cycling performance,complicated interfacial reactions,low Coulombic efficiency,and uncontrollable lithium dendrites.Understanding Li^+ions’nucleation mechanism is essential to tackle the uncontrolled growth of lithium dendrites.However,the nucleation behavior of Li+ions is interfered by the structural complexities of existing substrates during the reduplicative plating/stripping process and the rational mechanism of uniform nucleation of Li^+ions has not been clearly understood from the theoretical point of view.In our work,first-principles theoretical calculations are carried out to investigate the Li^+ions nucleation performance on metal-doped Cu surfaces(MDCSs)and the key descriptors that determines the properties of various MDCSs are systematically summarized.It is found that the introduction of heterogeneous doping Ag and Zn atoms will induce a gradient adsorption energy on MDCSs,and such gradient deposition sites can reduce the diffusion barriers and accelerate the diffusion rates of Li+ions dynamically.By maneuvering the Li+ions nucleation on MDCSs,a dendrite-free lithium metal anode can be achieved without the use of porous matrixes and complex synthesis process,which can be attributed to suppress the uncontrollable lithium dendrites for realizing the high-efficiency LMBs.Yanchen Fan Tianshuai Wang Dominik Legut Qianfan Zhang 2019Journal of Energy Chemistry2019,28,12:2
15Noninvasive ultrasound deep brain stimulation of nucleus accumbens induces behavioral avoidance显示文摘Ultrasound stimulation is an emerging noninvasive option in treating neuropsychiatric disorders. The present study investigates the behavioral alterations resulting from ultrasound stimulation on the nucleus accumbens(NAc) in freely moving mice. Our results show that an acute ultrasound stimulation on the NAc, rather than the visual cortex or auditory cortex, led to a pronounced avoidance behavior, while repeated NAc ultrasound stimulation resulted in an obvious conditioned place aversion with changes in synaptic protein(Glu A1/2 subunit) expression. Notably, NAc ultrasound stimulation suppressed the morphine-induced conditioned place preference.
