1An in vivo study with an MRI tracer method reveals the biophysical properties of interstitial fluid in the rat brain显示文摘The nature of brain interstitial fluid (ISF) has long been a subject of controversy. Most of the previous studies on brain ISF were carried out in vitro. In the present study, a novel method was developed to characterize ISF in the living rat brain by magnetic resonance (MR) imaging using gadolinium-diethylenetriaminepentaacetic acid (Gd-DTPA) as a tracer. Sprague Dawley rats (n=8) were subjected to MR scanning before and after the introduction of Gd-DTPA into the caudate nucleus. A one-way drainage of brain ISF was demonstrated on the dynamic MR images. According to the traditional diffusion model, the diffusion and clearance rate constants of the tracer within brain extracellular space (ECS) were derived as (3.38±1.07)×10-4 mm 2 s-1 and (7.60±4.18)×10-5 s-1 . Both diffusion and bulk flow contributed to the drainage of ISF from the caudate nucleus, which demonstrated an ISF-cerebrospinal fluid confluence in the subarachnoid space at the lateral and ventral surface of the brain cortex at 3 h after the injection. 6Distribution of green algae micro-propagules and their function in the formation of the green tides in the coast of Qinhuangdao,the Bohai Sea, China显示文摘Since 2015, a novel green tide has been recurring in the coastal areas of Qinhuangdao at the western coast of the Bohai Sea in China, threatening the environment and ecosystem of the Beidaihe seaside holiday resort along the coast. Micro-propagules of the green algae including gametes, spores, micro-germlings and micro-vegetative fragments play an important role in the formation of green tides. They serve as a 'seed source' of green macroalgae, and their distributions could reflect and influence the 'algae source' of green tides. In this study,monthly surveys in the inshore and offshore areas of the Qinhuangdao coast were conducted from April to September 2016 and in January 2017 to investigate the tempo-spatial distribution patterns and the biomass variations of the green algae micro-propagules. The obtained results show that micro-propagules were mainly distributed in the inshore areas with a significantly decreasing abundance towards offshore areas. Their biomass was highest in July and August, and lowest in winter. The areas that were affected by the green tides showed a remarkably higher abundance of micro-propagules compared to other areas. These micro-propagules could serve as the 'seed' source of green tides. 8Origin of strong solid solution strengthening in the CrCoNi-W medium entropy alloy显示文摘Solid solution strengthening is one of the most conventional strategies for optimizing alloys strength,while the corresponding mechanisms can be more complicated than we traditionally thought specifically as heterogeneity of microstructure is involved.In this work,by comparing the change of chemical distribution,dislocation behaviors and mechanical properties after doping equivalent amount of tungsten(W)atoms in CrCoNi alloy and pure Ni,respectively,it is found that the alloying element W in CrCoNi alloy resulted in much stronger strengthening effect due to the significant increase of heterogeneity in chemical distribution after doping trace amount of W.The large atomic scale concentration fluctuation of all elements in CrCoNi-3W causes dislocation motion via strong nanoscale segment detrapping and severe dislocation pile up which is not the case in Ni-3W.The results revealed the high sensitivity of elements distribution in multi-principle element alloys to composition and the significant consequent influence in tuning the mechanical properties,giving insight for complex alloy design.Yujie Chen Yan Fang Xiaoqian Fu Yiping Lu Sijing Chen Hongbin Bei Qian Yu 2021Journal of Materials Science & Technology2021,,14:5
9Engineering an enthesis-like graft for rotator cuff repair:An approach to fabricate highly biomimetic scaffold capable of zone-specifically releasing stem cell differentiation inducers显示文摘Rotator cuff(RC)attaches to humerus across a triphasic yet continuous tissue zones(bone-fibrocartilage-tendon),termed“enthesis”.Regrettably,rapid and functional enthesis regeneration is challenging after RC tear.The existing grafts bioengineered for RC repair are insufficient,as they were engineered by a scaffold that did not mimic normal enthesis in morphology,composition,and tensile property,meanwhile cannot simultaneously stimulate the formation of bone-fibrocartilage-tendon tissues.Herein,an optimized decellularization approach based on a vacuum aspiration device(VAD)was developed to fabricate a book-shaped decellularized enthesis matrix(O-BDEM).Then,three recombinant growth factors(CBP-GFs)capable of binding collagen were synthesized by fusing a collagen-binding peptide(CBP)into the N-terminal of BMP-2,TGF-β3,or GDF-7,and zone-specifically tethered to the collagen of O-BDEM to fabricate a novel scaffold(CBP-GFs/O-BDEM)satisfying the above-mentioned requirements.After ensuring the low immunogenicity of CBP-GFs/O-BDEM by a novel single-cell mass cytometry in a mouse model,we interleaved urine-derived stem cell-sheets into this CBP-GFs/O-BDEM to bioengineer an enthesis-like graft.Its high-performance on regenerating enthesis was determined in a canine model.These findings indicate this CBP-GFs/O-BDEM may be an excellent scaffold for constructing enthesis-like graft to patch large/massive RC tears,and provide breakthroughs in fabricating graded interfacial tissue.Can Chen Qiang Shi Muzhi Li Yang Chen Tao Zhang Yan Xu Yunjie Liao Shulin Ding Zhanwen Wang Xing Li Chunfeng Zhao Lunquan Sun Jianzhong Hu Hongbin Lu 2022Bioactive Materials2022,7,10:4

11Iterative learning control for nonlinear systems with uncertain state delay and arbitrary initial error显示文摘Most of the existing iterative learning control algorithms proposed for time-delay systems are based on the condition that the time-delay is precisely available, and the initial state is reset to the desired one or a fixed value at the start of each operation, which makes great limitation on the practical application of corresponding results. In this paper, a new iterative learning control algorithm is studied for a class of nonlinear system with uncertain state delay and arbitrary initial error. This algorithm needs to know only the boundary estimation of the state delay, and the initial state is updated, while the convergence of the system is guaranteed. Without state disturbance and output measurement noise, the system output will strictly track the desired trajectory after successive iteration. Furthermore, in the presence of state disturbance and measurement noise, the tracking error will be bounded uniformly. The convergence is strictly proved mathematically, and sufficient conditions are obtained. 12The contribution of attached Ulva prolifera on Pyropia aquaculture rafts to green tides in the Yellow Sea显示文摘Green tides caused by the unusual accumulation of high floating Ulva prolifera have occurred regularly in the Yellow Sea since 2007.The primary source of the Yellow Sea green tides is the attached algae on the Pyropia aquaculture rafts in the Subei Shoal.Ulva prolifera and Blidingia(Italic)sp.are the main species observed on Pyropia aquaculture rafts in the Subei Shoal.We found that U.prolifera has strong buoyancy and a rapid growth rate,which may explain why it is the dominant species of green tides that occur in the China's sea area of the Yellow Sea.The growth rate of floating U.prolifera was about 20%–31%d–1,which was much higher than Blidingia(Italic)sp.There were about 1.7×104 t of attached algae on the Pyropia aquaculture rafts in May 2012.We found that 39%of attached algae could float when the tide rose in the Subei Shoal,and U.prolifera accounted for 63%of the floating algae.Our analysis estimated that about 4000 t of attached U.prolifera floated into the surrounding waters of the Subei Shoal during the recycling period of aquaculture rafts.These results suggest that the initial floating biomass of large-scale green tides in the Yellow Sea is determined by the U.prolifera biomass attached to Pyropia aquaculture rafts,further impacting the scale of the green tide。Hongbin Han Shiliang Fan Wei Song Yan Li Jie Xiao Zongling Wang Xuelei Zhang Dewen Ding 2020Acta Oceanologica Sinica2020,39,2:3

14Surveillance of emerging infectious diseases for biosecurity显示文摘Emerging infectious diseases, such as COVID-19, continue to pose significant threats to human beings and their surroundings. In addition, biological warfare, bioterrorism, biological accidents, and harmful consequences arising from dual-use biotechnology also pose a challenge for global biosecurity. Improving the early surveillance capabilities is necessary for building a common biosecurity shield for the global community of health for all. Furthermore, surveillance could provide early warning and situational awareness of biosecurity risks. However, current surveillance systems face enormous challenges, including technical shortages, fragmented management, and limited international cooperation. Detecting emerging biological risks caused by unknown or novel pathogens is of particular concern. Surveillance systems must be enhanced to effectively mitigate biosecurity risks. Thus, a global strategy of meaningful cooperation based on efficient integration of surveillance at all levels, including interdisciplinary integration of techniques and interdepartmental integration for effective management, is urgently needed. In this paper, we review the biosecurity risks by analyzing potential factors at all levels globally. 15Effects of ethanol extracts of scorpion on hippocampal apoptosis and caspase-3 expression in lithium chloride-pilocarpine-induced status epilepticus rats显示文摘BACKGROUND: Previous studies have demonstrated that scorpion venom in the scorpion can inhibit epilepsy and apoptosis.However,it remains unclear whether ethanol extracts of scorpion (EES) exhibit similar effects.OBJECTIVE: To investigate the effects of EES on hippocampal apoptosis and caspase-3 expression,and to compare the effects on sodium valproate (positive control drug) in a rat model of status epilepticus induced by lithium chloride-pilocarpine.DESIGN,TIME AND SETTING: This randomized,controlled study was conducted at the Drug Research and Development Center,Kanghong Pharmaceuticals Group,and the Department of Pathology,Sichuan Academy of Medical Sciences & Sichuan Provincial People’s Hospital,China from May 2007 to April 2008.MATERIALS: EES were prepared by Huashen Pharmaceutical,China.Sodium valproate (Hunan Xiangzhong Pharmaceutical,China) and lithium chloride-pilocarpine (Sigma,USA) were also used in the present study.METHODS: From a total of 156 rats,six served as normal controls.The remaining rats were intraperitoneally injected with lithium chloride-pilocarpine to establish status epilepticus models,and then assigned to five groups (n = 30,respectively).Animals in each group were administered drugs at 15 minutes after epileptic seizure by gavage.i.e.in the normal control and model groups,rats were treated with 1 mL/0.1 kg saline.The sodium valproate group was administered 120 mg/kg/d sodium valproate.The low-,moderate-,and high-dose EES groups received treatments of 290,580,and 1 160 mg/kg/d EES.The dispensed concentration was 1 mL/0.1 kg.Rat seizure behavior was observed.If status epilepticus did not terminated after 1 hour,the rats were intraperitoneally administered atropine (1 mg/kg) and diazepam (10 mg/kg) to terminate seizure.These rats were continuously observed for 6 hours to ensure seizure termination.Then rats were treated with the above-mentioned drugs at 8: 00 am each day until sacrifice,which took place 4 hours after drug administration.MAIN OUTCOME MEASURES: Terminal dUTP nick end labeling (TUNEL)-positive cells and caspase-3 expression were,respectively,determined by TUNEL and immunohistochemistry at 6,24,48,and 72 hours,as well as 7 days,after status epilepticus.Behavioral changes were also measured.RESULTS: A few caspase-3-positive cells were observed.TUNEL-and caspase-3-positive cells were mainly visible in the hippocampal CA1 and CA3 regions 6 hours following status epilepticus in the model and drug intervention groups.The number of TUNEL-positive cells reached a peak at 48 hours following status epilepticus in the sodium valproate group,as well as the moderate-and high-dose EES groups,and number of TUNEL-positive cells reached a peak at 72 hours in the model and low-dose EES groups.The number of caspase-3-positive cells reached a peak at 48 hours in each group.Following treatment of sodium valproate and EES,the number of TUNEL-and caspase-3-positive cells significantly decreased compared with the model group at various time points (P < 0.05).The number of TUNEL-and caspase-3-positive cells was greatest in the low-dose EES group,followed by the moderate-and high-dose EES groups.The number of TUNEL-and caspase-3-positive cells was similar between the sodium valproate and high-dose EES groups.Epileptic seizure was significantly improved in the sodium valproate group,as well as the moderate-and high-dose EES groups,compared with the model group (P < 0.05 or P < 0.01).Treatment with sodium valproate and high-dose EES resulted in the best outcome,although the results were similar (P > 0.05).CONCLUSION: A dose of 1 160 mg/kg/d EES significantly inhibited status epilepticus.This outcome corresponded to a decreased number of apoptotic cells and caspase-3-positive cells,which was similar to sodium valproate.These results suggest that it is not necessary to extract a component from the scorpion for the treatment of epilepsy.The high dose of EES significantly inhibited epilepsy,which correlated with decreased hippocampal caspase-3 expression.Liang Yu Hongbin Sun Yi Liang Yan Xie Baoming He Fei Xu 2010Neural Regeneration Research2010,5,2:2
16Discovery and identification of EIF2AK2 as a direct key target of berberine for anti-inflammatory effects显示文摘Using chemoproteomic techniques,we first identified EIF2AK2,eEF1A1,PRDX3 and VPS4B as direct targets of berberine(BBR)for its synergistically anti-inflammatory effects.Of them,BBR has the strongest affinity with EIF2AK2 via two ionic bonds,and regulates several key inflammatory pathways through EIF2AK2,indicating the dominant role of EIF2AK2.Also,BBR could subtly inhibit the dimerization of EIF2AK2,rather than its enzyme activity,to selectively modulate its downstream pathways including JNK,NF-κB,AKT and NLRP3,with an advantage of good safety profile.In EIF2AK2 gene knockdown mice,the inhibitory IL-1β,IL-6,IL-18 and TNF-a secretion of BBR was obviously attenuated,confirming an EIF2AK2-dependent anti-inflammatory efficacy.The results highlight the BBR's network mechanism on anti-inflammatory effects in which EIF2AK2 is a key target,and inhibition of EIF2AK2 dimerization has a potential to be a therapeutic strategy against inflammationrelated disorders.Wei Wei Qingxuan Zeng Yan Wang Xixi Guo Tianyun Fan Yinghong Li Hongbin Deng Liping Zhao Xintong Zhang Yonghua Liu Yulong Shi Jingyang Zhu Xican Ma Yanxiang Wang Jiandong Jiang Danqing Song 2023Acta Pharmaceutica Sinica B2023,13,5:2
17Scorpion ethanol extract and valproic acid effects on hippocampal glial fibrillary acidic protein expression in a rat model of chronic-kindling epilepsy induced by lithium chloride-pilocarpine显示文摘The present study analyzed the effects of ethanol extracts of scorpion on epilepsy prevention and hippocampal expression of glial fibrillary acidic protein in a lithium chloride-pilocarpine epileptic rat model. Results were subsequently compared with valproic acid. Results showed gradually- increased hippocampal glial fibrillary acidic protein expression following model establishment; glial fibrillary acidic protein mRNA expression was significantly increased at 3 days, reached a peak at 7 days, and then gradually decreased thereafter. Ethanol extracts of scorpion doses of 580 and 1 160 mg/kg, as well as 120 mg/kg valproic acid, led to a decreased number of glial fibrillary acidic protein-positive cells and glial fibrillary acidic protein mRNA expression, as well as decreased seizure grades and frequency of spontaneously recurrent seizures. The effects of 1 160 mg/kg ethanol extracts of scorpion were equal to those of 120 mg/kg valproic acid. 18Optimal time for subarachnoid transplantation of neural progenitor cells in the treatment of contusive spinal cord injury显示文摘This study aimed to identify the optimal neural progenitor cell transplantation time for spinal cord injury in rats via the subarachnoid space. Cultured neural progenitor cells from 14-day embryonic rats, constitutively expressing enhanced green fluorescence protein, or media alone, were injected into the subarachnoid space of adult rats at 1 hour (acute stage), 7 days (subacute stage) and 28 days (chronic stage) after contusive spinal cord injury. Results showed that grafted neural progenitor cells migrated and aggregated around the blood vessels of the injured region, and infiltrated the spinal cord parenchyma along the tissue spaces in the acute stage transplantation group. However, this was not observed in subacute and chronic stage transplantation groups. O4- and glial fibrillary acidic protein-positive cells, representing oligodendrocytes and astrocytes respectively, were detected in the core of the grafted cluster attached to the cauda equina pia surface in the chronic stage transplantation group 8 weeks after transplantation. Both acute and subacute stage transplantation groups were negative for O4 and glial fibrillary acidic protein cells. Basso, Beattie and Bresnahan scale score comparisons indicated that rat hind limb locomotor activity showed better recovery after acute stage transplantation than after subacute and chronic transplantation. Our experimental findings suggest that the subarachnoid route could be useful for transplantation of neural progenitor cells at the acute stage of spinal cord injury. Although grafted cells survived only for a short time and did not differentiate into astrocytes or neurons, they were able to reach the parenchyma of the injured spinal cord and improve neurological function in rats. 19P2X4 receptor and brain-derived neurotrophic factor in neuropathic pain显示文摘Objective:To investigate whether the activation of p38MAPK is involved in the neuropathic pain induced by P2X4 receptor,and the effects of activated P2X4 receptor and p38MAPK on expression of brain-derived neurotrophic factor (BDNF) in the chronic neuropathic pain.Methods:Lumbar intrathecal catheters were chronically implanted in male Sprague-Dawley rats.The right sciatic nerve was loosely ligated proximal to the sciatica's trifurcation at approximately 1.0 mm intervals with 4-0 silk sutures.The microglia inhibitor minocycline,P2X4 antagonist (TNP-ATP) and p38MAPK inhibitor (SB203580) were intrathecally administered every 12 h,3 d post-chronic constriction injury (CCI).Mechanical nociceptive thresholds were assessed with the paw withdrawal threshold (PWT) to von Frey filaments.The expression of P2X4 and BDNF were assessed by both immunohistochemical analysis and RT-PCR.Results:Intrathecal injection of minocycline or TNP-ATP or SB203580 significantly attenuated CCI-induced mechanical allodynia.The time courses of P2X4 receptor and BDNF expression were increased at all points after CCI and reached a peak level on postoperative d 7.Intrathecal injection of minocycline or TNP-ATP or SB203580 markedly suppressed the increase of CCI-induced P2X4 receptor and BDNF expression in the spinal cord.Conclusion:The activation of P2X4 receptor BDNF pathways contributes to neuropathic pain in CCI rats,and the activation of p38MAPK is involved in the neuropathic pain induced by P2X4 receptor.Yan Xiaodi Zhou Shuangqiong Chen Qianbo Chen Chengwen Yuan Hongbin 2010Journal of Medical Colleges of PLA(China)2010,25,5:2
