125 × 50 Gbps wavelength division multiplexing silicon photonics receiver chip based on a silicon nanowire-arrayed waveguide grating显示文摘A high-performance monolithic integrated wavelength division multiplexing silicon(Si) photonics receiver chip is fabricated on a silicon-on-insulator platform. The receiver chip has a 25-channel Si nanowire-arrayed waveguide grating, and each channel is integrated with a high-speed waveguide Ge-on-Si photodetector. The central wavelength, optical insertion loss, and cross talk of the array waveguide grating are 1550.6 nm, 5–8 dB, and-12 –-15 dB, respectively. The photodetectors show low dark current density of 16.9 mA∕cm^2 at-1 V and a high responsivity of 0.82 A/W at 1550 nm. High bandwidths of 23 and 29 GHz are achieved at 0 and-1 V, respectively. 4Dl-3-n butylphthalide attenuates brain injury caused by cortical infarction accompanied by cranial venous drainage disturbance显示文摘Background Cerebral venous disorder may have a harmful effect on ischaemic stroke;however,the underlying mechanism remains to be elucidated.Although Dl-3-n butylphthalide is a multitarget agent for antiischaemic stroke,its neuroprotective role in brain ischaemia accompanied by brain venous disturbance remains unclear.In this study,we induced cerebral venous disturbance by the occlusion of bilateral external jugular veins(EJVs)to explore the potential mechanism of the adverse effects of cerebrovenous disorders in cerebral infarction and explore the protective effect of Dl-3-n butylphthalide on cerebral infarction accompanied through cerebral venous disturbance.Methods Cerebral venous disturbance was induced in Sprague-Dawley rats through the permanent occlusion of bilateral EJVs,and cerebral ischaemic stroke was induced through the permanent occlusion of the right cortical branches of the middle cerebral artery.2,3,5-triphenyltetrazolium chloride staining,MRI,Evans blue extravasation and behavioural test were performed to evaluate infarction volume,cerebral blood flow(CBF),blood-brain barrier(BBB)integrity and neurological function.Immunofluorescence staining and western blot analysis were performed to detect loss of neuron,endothelial cells,pericytes and tight junctions.Results Bilateral EJVs occlusion did not cause cerebral infarction;however,it increased the infarction volume compared with the simple middle cerebral artery occlusion(MCAO)group,accompanied by severe neuron loss,worse neurological function,lower CBF,increased EJVs pressure,exacerbated Evans blue extravasation and brain oedema,as well as attenuated angiogenesis.Dl-3-n butylphthalide displayed a neuroprotective effect in rats with MCAO accompanied by EJVs occlusion by reducing neuron loss,accelerating CBF restoration,promoting angiogenesis and relieving BBB damage.Conclusion Bilateral EJVs occlusion did not significantly affect normal rats but aggravated brain damage in the case of ischaemic stroke.Dl-3-n butylphthalide treatment plays a neuroprotective role in rats with MCAO accompanied by EJVs occlusion,mainly due to the promotion of CBF restoration and BBB protection.Kangping Song Xiuli Zeng Xiaomei Xie Rongxuan Zhu Jianye Liang Guobing Chen Li’an Huang 2022Stroke & Vascular Neurology2022,7,3:1

6Erythroblast island macrophages: recent discovery and future perspectives显示文摘Erythroblastic island(EBI),composed of a central macrophage surrounded by developing erythroid cells,is a structure found in hematopoietic tissues such as fetal liver and bone marrow.It is the first described hematopoietic niche that predominantly supports erythropoiesis.Although it is well accepted that EBIs and EBI macrophage play important roles during erythropoiesis,the mechanisms by which they support erythropoiesis remain largely unclear due to our inability to identify and isolate EBI macrophages.Earlier efforts to identify surface markers for EBI macrophages have focused on the adhesion molecules which are involved in macrophage’s interaction with erythroblasts.These include EMP,Vcam1,CD169,CD163,and aV integrin.Findings from these earlier studies suggested that combination of Vcam1,CD169,and mouse macrophage surface marker F4/80 can be used to define mouse EBI macrophage.We found that not all F4/80+Vcam1+CD169+macrophages are EBI macrophages.Instead,we discovered that EBI macrophages are characterized by the expression of Epor in both mouse and man.RNA-seq analyses of the newly identified EBI macrophages revealed that EBI macrophages have involved specialized function in supporting erythropoiesis.Our findings provide foundation for future studies.Here we will review current knowledge of EBI macrophages and discuss future perspectives.Wei Li Yaomei Wang Lixiang Chen Xiuli An 2019Blood Science2019,1,1:1
7One-step rapid synthesis,crystal structure and 3.3 microseconds long excited-state lifetime of Pd1Ag28 nanocluster显示文摘Doping foreign atom(s)in metal nanoclusters is an effective strategy to engineer the properties and functionalities of metal nanoclusters.However,until now,to dope Pd atom into Ag nanoclusters remains a huge challenge.Here we develop a one-step rapid method to synthesize the Pd-doped Ag nanocluster with high yield.The prepared Pd1Ag28 nanocluster was characterized by mass spectroscopy,X-ray photoelectron spectroscopy,X-ray crystallography,fluorescence spectroscopy,ultraviolet-visible absorption spectroscopy and transient absorption spectroscopy.The nanocluster exhibits a perfect face-centered cubic(FCC)kernel structure with a tetrahedron-like shell.Of note,Pd1Ag28 nanocluster had an unexpectedly long excited-state lifetime of 3.3 microseconds,which is the longest excited-state lifetime for Ag-based nanoclusters S0 far.Meanwhile,the excellent near-infrared luminescence indicated the nanocluster has the potential in fluorescent bio-imaging.Besides,it was revealed that Pd1Ag28 nanocluster could be transformed into Au1Ag28 nanocluster via ion exchange reaction of AuPPhzCl with Pd1Ag28 nanocluster.This work provides an efficient synthetic protocol of alloy nanoclusters and wil contribute to study the effect of foreign atom on the properties of metal nanoclusters.Xinzhang Lin Hengjiang Cong Keju Sun Xuemei Fu Wanchao Kang Xiuli Wang Shengye Jin Ren'an Wu Chao Liu Jiahui Huang 2020Nano Research2020,13,2:1
8Comparison of two methods for evaluating lower urinary tract symptoms in cervical cancer patients following radical hysterectomy显示文摘Objective:To compare the degree of agreement and consistency of urodynamic studies(UDS)with low urinary tract symptoms(LUTS)questionnaires for evaluating LUTS in cervical cancer patients following radical hysterectomy(RH)and pelvic lymphadenectomy.Methods:From January 2012 to March 2015,204 cervical cancer patients who underwent RH in 13 hospitals were evaluated using the Incontinence Questionnaire-Female Lower Urinary Tract Symptoms(ICIQ-FLUTS)and the Overactive Bladder Symptom Score(OABSS).Urodynamic tests were also performed on these patients during the same period.Results:Study participants’age ranged from 23 to 75 years,with a mean(standard deviation)of 48.0±9.3 years.Using questionnaires,the prevalence of patients with LUTS symptoms,including storage symptoms,voiding symptoms,stress urinary incontinence(SUI)and overactive bladder(OAB)was 86.3%,77.0%,62.7%,52.9%and 14.7%,respectively.For UDS,the corresponding prevalence was 89.7%,70.1%,66.7%,46.6%and 13.2%,respectively.The diagnostic concordance of questionnaires and UDS for storage symptoms,voiding symptoms,SUI and OAB was 79.9%,66.7%,66.7%,57.4%and 79.9%,respectively.For voiding symptoms,the correlation coefficient was 0.272,which was higher than that of storage symptoms,SUI and OAB.Fang An Sha Wang Zhiqi Wang Lingying Wu Qiubo Lv Aiming Lv Hongwu Wen Jinsong Han Yumei Wu Yunong Gao Qing Liu Sumei Wang Hongxia Li Luwen Wang Yanlong Wang Ruifang Wu Huan Li Xiuli Sun 2021Gynecology and Obstetrics Clinical Medicine2021,1,2:0

11Loss of 4.1N in epithelial ovarian cancer results in EMT and matrix-detached cell death resistance显示文摘Epithelial ovarian cancer(EOC)is one of the leading causes of death from gynecologic cancers and peritoneal dissemination is the major cause of death in patients with EOC.Although the loss of 4.1N is associated with increased risk of malignancy,its association with EOC remains unclear.To explore the underlying mechanism of the loss of 4.1N in constitutive activation of epithelial-mesenchymal transition(EMT)and matrixdetached cell death resistance,we investigated samples from 268 formalin-fixed EOC tissues and performed various in vitro and in vivo assays.We report that the loss of 4.1N correlated with progress in clinical stage,as well as poor survival in EOC patients.The loss of 4.1N induces EMT in adherent EOC cells and its expression inhibits anoikis resistance and EMT by directly binding and accelerating the degradation of 14-3-3 in suspension EOC cells.Furthermore,the loss of 4.1N could increase the rate of entosis,which aggravates cell death resistance in suspension EOC cells.Moreover,xenograft tumors in nude mice also show that the loss of 4.1N can aggravate peritoneal dissemination of EOC cells.Single-agent and combination therapy with a ROCK inhibitor and a 14-3-3 antagonist can reduce tumor spread to varying degrees.Our results not only define the vital role of 4.1N loss in inducing EMT,anoikis resistance,and entosis-induced cell death resistance in EOC,but also suggest that individual or combined application of 4.1N,14-3-3 antagonists,and entosis inhibitors may be a promising therapeutic approach for the treatment of EOC.Dandan Wang Letian Zhang Ajin Hu Yuxiang Wang Yan Liu Jing Yang Ningning Du Xiuli An Congying Wu Congrong Liu 2021Protein & Cell2021,12,2:0
12Inhibition of aryl hydrocarbon receptor signaling promotes the terminal differentiation of human erythroblasts显示文摘The aryl hydrocarbon receptor(AHR)plays an important role during mammalian embryo development.Inhibition of AHR signaling promotes the development of hematopoietic stem/progenitor cells.AHR also regulates the functional maturation of blood cells,such as T cells and megakaryocytes.However,little is known about the role of AHR modulation during the development of erythroid cells.In this study,we used the AHR antagonist StemRegenin 1(SR1)and the AHR agonist 2,3,7,8-tetrachlorodibenzo-p-dioxin during different stages of human erythropoiesis to elucidate the function of AHR.We found that antagonizing AHR signaling improved the production of human embryonic stem cell derived erythrocytes and enhanced erythroid terminal differentiation.RNA sequencing showed that SR1 treatment of proerythroblasts upregulated the expression of erythrocyte differentiation-related genes and downregulated actin organization-associated genes.We found that SR1 accelerated F-actin remodeling in terminally differentiated erythrocytes,favoring their maturation of the cytoskeleton and enucleation.We demonstrated that the effects of AHR inhibition on erythroid maturation were associated with F-actin remodeling.Our findings help uncover the mechanism for AHRmediated human erythroid cell differentiation.We also provide a new approach toward the large-scale production of functionally mature human pluripotent stem cell-derived erythrocytes for use in translational applications.Yijin Chen Yong Dong Xulin Lu Wanjing Li Yimeng Zhang Bin Mao Xu Pan Xiaohong Li Ya Zhou Quanming An Fangxin Xie Shihui Wang Yuan Xue Xinping Cai Mowen Lai Qiongxiu Zhou Yan Yan Ruohan Fu Hong Wang Tatsutoshi Nakahata Xiuli An Lihong Shi Yonggang Zhang Feng Ma 2022Journal of Molecular Cell Biology2022,14,2:0
13Novel methods for studying normal and disordered erythropoiesis显示文摘Erythropoiesis is a process during which multipotential hematopoietic stem cells proliferate, differentiate and eventually form mature erythrocytes. Interestingly, unlike most cell types, an important feature of erythropoiesis is that following each mitosis the daughter cells are morphologically and functionally different from the parent cell from which they are derived, demonstrating the need to study erythropoiesis in a stage-specific manner. This has been impossible until recently due to lack of methods for isolating erythroid cells at each distinct developmental stage. This review summarizes recent advances in the development of methods for isolating both murine and human erythroid cells and their applications.
