1Efficacy and safety of external application of Traditional Chinese Medicine for the treatment of acute gouty arthritis: a systematic review and Meta-analysis显示文摘OBJECTIVE:To evaluate the clinical efficacy and safety of external application of Traditional Chinese Medicine (EATCM) on acute gouty arthritis (AGA).METHODS:Six electronic data bases were retrieved from their inception to march 2017.15 randomized controlled clinical trials (RCTs) were included.The quality of the literatures was assessed according to Cochrane review criteria by using RevMan5.3 software;and related data was counted using Stata14.0 software.RESULTS:There was no significant difference in C-reactiveprotein (CRP) between experimental intervention group and control group [(WMD =1.093,95% C/(-2.514,4.701);P =0.553)].In terms of overall efficacy,the EATCM's treatment group was significantly superior to control group [(OR =3.692;95% CI (2.408,5.661);P <0.001)].The EATCM was better than western medicine treatments or other interventions in reducing the adverse reactions [(OR =0.135;95% CI (0.067,0.274);P < 0.001)].All these funnel plots showed unlikelihood of publishing bias.But due to excessive heterogeneity,the statistical results of serum uric acid and visual analogue scale and CRP between the two groups became uncertain.CONCLUSION:The results indicate that EATCM may have greater overall efficacy with fewer adverse drug reactions,although the evidence is weak owing to the low methodological quality and the small number of the included trials.Huang Xinxing Zhu Zonghua Wu Guang-hui Min Peng Fei Peng Zhu Tao Wang Xuezong Xiao Lianbo 2019Journal of Traditional Chinese Medicine2019,39,3:12
2Metabolic regulation of T cell development by Sinl-mTORC2 is mediated by pyruvate kinase M2显示文摘Glucose metabolism plays a key role in thymocyte development. The mammalian target of rapamycin complex 2 (mT0RC2) is a critical regulator of cell growth and metabolism, but its role in early thymocyte development and metabolism has not been fully studied. We show here that genetic ablation of Sinl, an essential component of mTORC2, in T lineage cells results in severely impaired thymocyte development at the CD4^- CD8^- double negative (DN) stages but not at the CD4^+ CD8^+ double positive (DP) or later stages. Notably, Sinl-deficient DN thymocytes show markedly reduced proliferation and glycolysis.Importantly, we discover that the M2 isoform of pyruvate kinase (PKM2) is a novel and crucial Sinl effector in promoting DN thymocyte development and metabolism. At the molecular level, we show that Sinl-mTORC2 controls PKM2 expression through an AKT-dependent PPAR-y nuclear translocation. 3MAP3K2 augments Th1 cell differentiation via IL-18 to promote T cell-mediated colitis显示文摘T cell-mediated immunity in the intestine is stringently controlled to ensure proper immunity against pathogenic microbes and to prevent autoimmunity,a known cause of inflammatory bowel disease.However,precisely how T cells regulate intestine immunity remains to be fully understood.In this study,we found that mitogen-activated protein kinase kinase kinase 2(MAP3K2)is required for the CD4^(+)T cell-mediated inflammation in the intestine.Using a T cell transfer colitis model,we found that MAP3K2-deficient naïve CD4^(+)T cells had a dramatically reduced ability to induce colitis compared to wild type T cells.In addition,significantly fewer IFN-γ-but more IL-17A-producing CD4^(+)T cells in the intestines of mice receiving MAP3K2-deficient T cells than in those from mice receiving wild type T cells was observed.Interestingly,under well-defined in vitro differentiation conditions,MAP3K2-deficient naïve T cells were not impaired in their ability to differentiate into Th1,Th17 and Treg.Furthermore,the MAP3K2-regulated colitis severity was mediated by Th1 but not Th17 cells in the intestine.At the molecular level,we showed that MAP3K2-mediated Th1 cell differentiation in the intestine was regulated by IL-18 and required specific JNK activation.Together,our study reveals a novel regulatory role of MAP3K2 in intestinal T cell immunity via the IL-18-MAP3K2-JNK axis and may provide a novel target for intervention in T cell-mediated colitis.Ningbo Wu Dongping Chen Hongxiang Sun Jianmei Tan Yao Zhang Tianyu Zhang Yuheng Han Hongzhi Liu Xinxing Ouyang Xiao-Dong Yang Xiaoyin Niu Jie Zhong Zhengting Wang Bing Su 2021Science China(Life Sciences)2021,64,3:3

5HPV16E7-HSP70 Hybrid DNA Vaccine Induces E7-Specific Cytotoxic T Cells and Antitumor Immunity显示文摘Using human papillomavirus type 16 (HPV16) E7 as an antigen and Heat Shock Protein 70 as adjuvant, we constructed a DNA vaccine by linking HSP70 gene to E7~ C91G gene. Mice, after being immunized with E7~ C91G -HSP70, E7~ C91G /HSP70, E7~ C91G , and wild E7 DNA vaccines respectively, produced E7 specific CD8^+ T-cell precursor frequencies of 280.33±2.52, 144.34±4.04, 164.34±5.13 and 82.33±3.51 respectively within every 1×10~5 mouse splenocytes. This proves that E7~ C91G -HSP70 fusion vaccine can significantly enhance the E7 specific cellular immunity within the mice body(p<0.01). After being immunized with E7~ C91G -HSP70 fusion vaccine, tumor-bearing mice of the group being treated have significantly longer latency and survival periods, comparing with other three categories of E7 vaccines. Experiment shows that this vaccine has a significant effect on enhancing E7 positive tumor-treatment within mice body. After being immunized with E7~ C91G -HSP70 vaccine, there were no pathological changes found in livers, kidneys and spleens of the mice, which proves that the vaccine is quite safe. 6Distributed cache replacement method for geospatial data using spatiotemporal localitybased sequence显示文摘Specific features of tile access patterns can be applied in a cache replacement strategy to a limited distributed high-speed cache for the cloud-based networked geographic information services(NGISs),aiming to adapt to changes in the access distribution of hotspots.By taking advantage of the spatiotemporal locality,the sequential features in tile access patterns,and the cache reading performance in the burst mode,this article proposes a tile sequence replacement method,which involves structuring a Least Recently Used(LRU)stack into three portions for the different functions in cache replacement and deriving an expression for the temporal locality and popularity of the relevant tile to facilitate the replacement process.Based on the spatial characteristics of both the tiles and the cache burst mode with regard to reading data,the proposed method generates multiple tile sequences to reflect spatiotemporal locality in tile access patterns.Then,we measure the caching value by a technique based on a weighted-based method.This technique draws on the recent access popularity and low caching costs of tile sequences,with the aim of balancing the temporal and spatial localities in tile access.It ranks tile sequences in a replacement queue to adapt to the changes in accessed hotspots while reducing the replacement frequency.Experimental results show that the proposed method effectively improves the hit rate and utilization rate for a limited distributed cache while achieving satisfactory response performance and high throughput for users in an NGIS.Therefore,it can be adapted to handle numerous data access requests in NGISs in a cloud-based environment.Rui LI Jiapei FAN Xinxing WANG Zhen ZHOU Huayi WU 2015Geo-Spatial Information Science2015,18,4:1

14Analysis on Radar Characteristics of a Short-time Heavy Rainfall Event in Wuchuan County显示文摘Using minute rainfall data of automatic ground station and a variety of products from new generation Doppler weather radar in Wuchuan, the characteristics of a short-time heavy precipitation process on April 23, 2022 were analyzed. The results showed that the appearance of differential reflectivity(ZDR) column and big-value zone of high-elevation ZDR had better indication on short-term heavy rainfall process in Shichao station. Ice phase process played a very important role in particle growth. The storm tracking information product can predict the path of the storm 15 min in advance. The storm stayed and moved less or even turned back to more than two to three scanning volumes in one place, indicating the occurrence of short-term heavy rainfall. One-hour accumulated precipitation(OHP) had a good effect on the estimation of continuous precipitation in a large area where the hourly rainfall exceeded 50 mm for more than two stations. 16HLA-B*6701 as a Subtype Performing Post-Selected Marking Gene for Cervical Cancer显示文摘A method of sequence-based typing(SBT) has been adopted to assort types of exons 2 and 3,which have the most polymorphism,of HLA-B locus of the Tujia nation group in Hubei province.The correlation among the HLA-B alleles,human papillomavirus(HPV) infection and cervical cancer risk has also been investigated.Under the condition of resident location and age,race unified,100 specimens of cancer patients were sampled as a case group,of which 86 were HPV positive and were screened for HLA-B alleles;while 187 specimens were taken from healthy people,of which 92 were HPV negative as a control group.The result shows that by comparing the above mentioned 86 HPV positive cervical cancer group and 92 HPV negative normal group,it was concluded that HLA-B*6701 was only found in the cervical cancer group(p<0.034),which shows that HLA-B*6701 can be used as an important candidate biological marking gene for generation of cervical cancer in Wufeng county of Hubei province.QIU Xiaoping TAO Ning TAN Yun WU Xinxing 2010Wuhan University Journal of Natural Sciences2010,15,6:0

18On the thermo-mechanical properties of unsaturated soils显示文摘The establishment of energy balance equation is necessary to study the thermo-mechanical properties of unsaturated soils.To solve this equation,the determination of two fundamental parameters as volumetric specific parameter and thermal conductivity coefficient is essential.In this paper,the effective thermal conductivity coefficient of dry soil grain is analyzed for soils with different compositions,and the thermo-mechanical properties of porous media with water and gas are studied by considering the soil water retention curve(SWRC).Different methods,i.e.volumetric average method,self-consistent method,Hashin-Strikman method,are employed to calculate thermal conductivity coefficients,and a new method is proposed to determine the thermo-mechanical parameters.Comparison of the results obtained by different methods shows that the proposed method is in a good agreement with the experimental results and is suitable for describing the main properties of the thermo-mechanical behaviors of soils.The relationship between the SWRC and the seepage curve is further studied by the natural proportional rule.The characteristics of the SWRC,its differential coefficient and the seepage curve,are investigated by considering the physico-mechanical mechanism;the limit scopes of the indices of the SWRC and the seepage curve are also given.Yingfa Lu Xinxing Wu Yujun Cui 2010Journal of Rock Mechanics and Geotechnical Engineering2010,2,2:0
