| 2 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. | 5 | Optimization of a serpentine flow field with variable channel heights and widths for PEM fuel cells显示文摘The present study proposes a modified serpentine flow field design in which the channel heights vary along each straight flow path to enhance reactant transport and liquid water removal. An optimization approach, combining a simplified conjugate-gradient method (inverse solver) and a three-dimensional, two-phase, non-isothermal fuel cell model (direct solver), has been developed to optimize the key geometric parameters. The optimal design has tapered channels for channels 1, 3 and 4 and increasing heights for channels 2 and 5 with the flow widths first increasing and then decreasing. The optimal channel heights and widths enhance the efficiency by 22.51% compared with the basic design having all heights and widths of 1 mm. The diverging channels have a greater impact on cell performance than fine adjustments of the channel widths for the present simulation conditions. The channel heights have more effect on the sub-rib convection, while the channel widths affect the uniformity of the fuel delivery more. The reduced channel heights of channels 2–4 significantly enhance the sub-rib convection to effectively transport oxygen to and liquid water out of the diffusion layer. | 6 | Influence of the NAO on Wintertime Surface Air Temperature over East Asia:Multidecadal Variability and Decadal Prediction显示文摘In this paper,we investigate the influence of the winter NAO on the multidecadal variability of winter East Asian surface air temperature(EASAT)and EASAT decadal prediction.The observational analysis shows that the winter EASAT and East Asian minimum SAT(EAmSAT)display strong in-phase fluctuations and a significant 60-80-year multidecadal variability,apart from a long-term warming trend.The winter EASAT experienced a decreasing trend in the last two decades,which is consistent with the occurrence of extremely cold events in East Asia winters in recent years.The winter NAO leads the detrended winter EASAT by 12-18 years with the greatest significant positive correlation at the lead time of 15 years.Further analysis shows that ENSO may affect winter EASAT interannual variability,but does not affect the robust lead relationship between the winter NAO and EASAT.We present the coupled oceanic-atmospheric bridge(COAB)mechanism of the NAO influences on winter EASAT multidecadal variability through its accumulated delayed effect of~15 years on the Atlantic Multidecadal Oscillation(AMO)and Africa-Asia multidecadal teleconnection(AAMT)pattern.An NAO-based linear model for predicting winter decadal EASAT is constructed on the principle of the COAB mechanism,with good hindcast performance.The winter EASAT for 2020-34 is predicted to keep on fluctuating downward until~2025,implying a high probability of occurrence of extremely cold events in coming winters in East Asia,followed by a sudden turn towards sharp warming.The predicted 2020/21 winter EASAT is almost the same as the 2019/20 winter. | Jianping LI Tiejun XIE Xinxin TANG Hao WANG Cheng SUN Juan FENG Fei ZHENG Ruiqiang DING | 2022 | Advances in Atmospheric Sciences2022,39,4: | 4 |
| 7 | Early assessment of the safety and immunogenicity of a third dose(booster)of COVID-19 immunization in Chinese adults显示文摘Inducing durable and effective immunity against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)via vaccination is essential to combat the current pandemic of coronavirus disease 2019(COVID-19).It has been noticed that the strength of anti-COVID-19 vaccination-induced immunity fades over time,which calls for an additional vaccination regime,as known as booster immunization,to restore immunity among previously vaccinated populations.Here we report a pilot open-label trial of a third dose of BBIBP-CorV,an inactivated SARS-CoV-2 vaccine(Vero cell),on 136 participants aged between 18 to 63 years.Safety and immunogenicity in terms of neutralizing antibody titers and cytokine/chemokine responses were analyzed as the main endpoint until day 28.While systemic reactogenicity was either absent or mild,SARS-CoV-2-specific neutralizing antibody titers rapidly arose in all participants within 4 weeks,surpassing the peak antibody titers elicited by the initial two-dose immunization regime.Broad increases of cellular immunity-associated cytokines and chemokines were also detected in the majority of participants after the third vaccination.Furthermore,in an exploratory study,a newly developed recombinant protein vaccine,NVSI-06-08(CHO Cells),was found to be safe and even more effective than BBIBP-CorV in eliciting humoral immune responses in BBIBP-CorV-primed individuals.Together,these results indicate that a third immunization schedule with either homologous or heterologous vaccine showed favorable safety profiles and restored potent SARS-CoV-2-specific immunity,providing support for further trials of booster vaccination in larger populations. | Yuntao Zhang Yunkai Yang Niu Qiao Xuewei Wang Ling Ding Xiujuan Zhu Yu Liang Zibo Han Feng Liu Xinxin Zhang Xiaoming Yang | 2022 | Frontiers of Medicine2022,16,1: | 3 |

| 9 | Hyperthermia based individual in situ recombinant vaccine enhances lymph nodes drainage for de novo antitumor immunity显示文摘The continuing challenges that limit effectiveness of tumor therapeutic vaccines were high heterogeneity of tumor immunogenicity, low bioactivity of antigens, as well as insufficient lymph nodes(LNs) drainage of antigens and adjuvants. Transportation of in situ neoantigens and adjuvants to LNs may be an effective approach to solve the abovementioned problems. Therefore, an FA-TSL/AuNCs/SV nanoplatform was constructed by integrating simvastatin(SV) adjuvant loaded Au nanocages(AuNCs)as cores(AuNCs/SV) and folic acid modified thermal-sensitive liposomes(FA-TSL) as shells to enhance de novo antitumor immunity. After accumulation in tumor guided by FA, AuNCs mediated photothermal therapy(PTT) induced the release of tumor-derived protein antigens(TDPAs) and the shedding of FATSL. Exposed AuNCs/SV soon captured TDPAs to form in situ recombinant vaccine(AuNCs/SV/TDPAs). Subsequently, AuNCs/SV/TDPAs could efficiently transport to draining LNs owing to the hyperthermia induced vasodilation effect and small particle size, achieving co-delivery of antigens and adjuvant for initiation of specific T cell response. In melanoma bearing mice, FA-TSL/AuNCs/SV and laser irradiation effectively ablated primary tumor, against metastatic tumors and induced immunological memory. In order to validate the mathematical model developed for the cooling process of real CDQ shaft, the one-seventh scale experimental setup was established. With the air as the working fluid and the coke as the packed bed, the coke and gas temperatures and the pressure drop in the cooling shaft were measured. A mathematical model based on the non-Darcian and non-thermal equilibrium model for the cooling process of the CDQ experimental shaft was presented. The computational results were compared to the experimental data. | 12 | Comprehensive Characteristics of MicroRNA Expression Profile Conferring to Rhizoctonia solani in Rice显示文摘MicroRNAs(miRNAs)are about 22 nucleotides regulatory non-coding RNAs that play versatile roles in reprogramming plant responses to biotic and abiotic stresses.However,it remains unknown whether miRNAs confer the resistance to necrotrophic fungus Rhizoctonia solani in rice.To investigate whether miRNAs regulate the resistance to R.solani,we constructed 12 small RNA libraries from susceptible and resistant rice cultivars treated with water/pathogen at 5 h post inoculation(hpi),10 hpi and 20 hpi,respectively.By taking the advantage of next-generation sequencing,we totally collected 400–450 known miRNAs and 450–620 novel miRNAs from the libraries.Expression analysis of miRNAs demonstrated different patterns for known and novel miRNAs upon R.solani challenge.Thirty-four miRNA families were identified to be expressed specifically in rice,and most of them were involved in plant disease resistance.A particular Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis result revealed that a great majority of target genes of regulated miRNAs belonged to the pathway of plant-pathogen interaction.Moreover,miR444b.2,miR531a,mir1861i,novel_miR1956 and novel_miR135 conferred response to R.solani infection confirmed by Northern blot.Our global understanding of miRNA profiling revealed that the regulation of miRNAs may be implicated in the control of rice immunity to R.solani.Analysis of the expression of miRNAs will offer the community with a direction to generate appropriate strategies for controlling rice sheath blight disease. | CAO Wenlei CAO Xinxin ZHAO Jianhua ZHANG Zhaoyang FENG Zhiming OUYANG Shouqiang ZUO Shimin | 2020 | Rice science2020,27,2: | 2 |

| 14 | Assessing the genetic diversity of the critically endangered Chinese sturgeon Acipenser sinensis using mitochondrial markers and genome-wide single-nucleotide polymorphisms from RAD-seq显示文摘As a living fossil, the endangered Chinese sturgeon(Acipenser sinensis) has been considered a national treasure in China. Here,the famous Gezhouba Dam and Three Gorges Dam on the Yangtze River were built in 1988 and 2006, for economic purposes.The natural population of Chinese sturgeon has declined since then, as these dams block its migratory route to the original spawning grounds in the middle reaches of the Yangtze River. In 2013 and 2014, there was an absence of spawning where it typically happened near the Gezhouba Dam. Nevertheless, from April to June in 2015, over 1,000 larvae with different body lengths(10–35 cm) were detected along the Shanghai Yangtze Estuary; but only little is currently known about the population genetic structure of the Chinese sturgeon. Herein, we inferred population genetic parameters from 462 available Chinese sturgeon specimens based on a 421-bp fragment of the mitochondrial DNA(mtDNA) D-loop region and 1,481,620 SNPs(singlenucleotide polymorphisms) generated by restriction site-associated DNA sequencing(RAD-seq). For the D-loop dataset, 15 haplotypes were determined. Randomly picked 23 individuals, representing the 15 D-loop haplotype groups, were subsequently used for further RAD-seq validation. The average nucleotide diversity calculated from the mtDNA and RAD datasets was 0.0086 and 0.000478, respectively. The overall effective female population size was calculated to be 1,255 to 2,607, and the long-term effective population size was estimated to range from 11,950 to 119,500. We observed that the genetic variability and the effective female population size of the current population in the Yangtze River are severely low, which are similar to the data reported over 10 years ago.
