| 1 | Molecular basis of CONSTANS oligomerization in FLOWERING LOCUS T activation显示文摘The transcription factor CONSTANS(CO)integrates day-length information to induce the expression of florigen FLOWERING LOCUS T(FT)in Arabidopsis.We recently reported that the C-terminal CCT domain of CO forms a complex with NUCLEAR FACTOR-YB/YC to recognize multiple cis-elements in the FT promoter,and the N-terminal tandem B-box domains form a homomultimeric assembly.However,the mechanism and biological function of CO multimerization remained unclear.Here,we report that CO takes on a head-to-tail oligomeric configuration via its B-boxes to mediate FT activation in long days.The crystal structure of B-boxesCOreveals a closely connected tandem B-box fold forming a continuous head-to-tail assembly through unique CDHH zinc fingers.Mutating the key residues involved in CO oligomerization resulted in a non-functional CO,as evidenced by the inability to rescue co mutants.By contrast,a transgene encoding a human p53-derived tetrameric peptide in place of the B-boxesCOrescued co mutant,emphasizing the essential role of BboxesCO-mediated oligomerization.Furthermore,we found that the four TGTG-bearing cis-elements in FT proximal promoter are required for FT activation in long days.Our results suggest that CO forms a multimer to bind to the four TGTG motifs in the FT promoter to mediate FT activation. | Xiaolin Zeng Xinchen Lv Rui Liu Hang He Shiqi Liang Lixian Chen Fan Zhang Liu Chen Yuehui He Jiamu Du | 2022 | Journal of Integrative Plant Biology2022,64,3: | 2 |
| 2 | Fine-grained analysis of cellular smartphone usage characteristics based on massive network traffic显示文摘Increased adoption of smartphones leads to the explosive growth of mobile network traffic. Understanding the traffic characteristics of mobile network is important for Internet service providers(ISPs) to optimize network resources. In this paper, we conduct a detailed measurement study on the hyper text transfer protocol(HTTP) traffic characteristics of cellular network among different operating systems(OSs) as well as different device-types. Firstly, we propose a probability-based method to identify the installed OS of each smartphone. Then we analyze the traffic characteristics of these smartphones in terms of OS and device-type based on a dataset across 31 days(a billing cycle). Finally, we identify the installed apps of each smartphone and compare the usage of apps on the dimensions of OS and device-type. | 5 | Probabilistic Top-k Query:Model and Application on Web Traffic Analysis显示文摘Top-k ranking of websites according to traffic volume is important for Internet Service Providers(ISPs) to understand network status and optimize network resources. However, the ranking result always has a big deviation with actual rank for the existence of unknown web traffic, which cannot be identified accurately under current techniques. In this paper, we introduce a novel method to approximate the actual rank. This method associates unknown web traffic with websites according to statistical probabilities. Then, we construct a probabilistic top-k query model to rank websites. We conduct several experiments by using real HTTP traffic traces collected from a commercial ISP covering an entire city in northern China. Experimental results show that the proposed techniques can reduce the deviation existing between the ground truth and the ranking results vastly. In addition, we find that the websites providing video service have higher ratio of unknown IP as well as higher ratio of unknown traffic than the websites providing text web page service. Specifically, we find that the top-3 video websites have more than 90% of unknown web traffic. | 7 | c-Fos expression in rat brainstem following intake of sucrose or saccharin显示文摘To examine whether the activation of brainstem neurons during intake of a sweet tastant is due to orosensory signals or post-ingestive factors,we compared the distribution of c-Fos-like immunoreactivity(c-FLI)in the nucleus of the solitary tract(NST)and parabrachial nucleus(PBN)of brainstem following ingestion of 0.25 M sucrose or 0.005 M saccharin solutions.Immunopositive neurons were localized mainly in the middle zone of the PBN and four rostral-caudal subregions of the NST.Intake of sucrose increased the number of FLI neurons in almost every subnucleus of the PBN(F_((2,13))=7.610,P=0.023),in addition to the caudal NST at the level of the area postrema(F_((2,13))=10.777,P=0.003)and the NST intermediate zone(F_((2,13))=7.193,P=0.014).No significant increase in the number of c-Fos positive neurons was detected in response to saccharin ingestion,although there was a trend towards a modest increase in a few select NST and PBN nuclei.These results suggest that the PBN and NST may be involved in sweet taste perception and modulation of sweet tastant intake,but the significantly enhanced intensity of Fos expression induced by sucrose indicates that PBN/NST neuronal activity is driven by the integrated effects of sweet taste sensation and post-ingestive signals. | Ke Chen Jianqun Yan Jinrong Li Bo Lv Xiaolin Zhao | 2011 | Frontiers of Medicine2011,5,3: | 0 |
| 8 | How variable progenitor clones construct a largely invariant neocortex显示文摘The neocortex contains a vast collection of diverse neurons organized into distinct layers. While nearly all neocortical neurons are generated by radial glial progenitors(RGPs), it remains largely unclear how a complex yet organized neocortex is constructed reliably and robustly. Here, we show that the division behavior and neuronal output of RGPs are highly constrained with patterned variabilities to support the reliable and robust construction of the mouse neocortex. The neurogenic process of RGPs can be well-approximated by a consistent Poisson-like process unfolding over time, producing deep to superficial layer neurons progressively. The exact neuronal outputs regarding layer occupation are variable;yet, this variability is constrained systematically to support all layer formation, largely reflecting the variable intermediate progenitor generation and RGP neurogenic entry and exit timing differences. | 10 | National incidence of joint dislocation in China:a retrospective survey of 512,187 individuals显示文摘Background:Joint dislocations significantly impact public health.However,a comprehensive study on the incidence,distribution,and risk factors for joint dislocations in China is lacking.We conducted the China National Joint Dislocation Study,which is a part of the China National Fracture Study conducted to obtain the national incidence and risk factors for traumatic fractures,and to investigate the incidence and risk factors for joint dislocations.Methods:For this national retrospective epidemiological study,512,187 participants were recruited using stratified random sampling and probability-proportional-to-size method from January 19 to May 16,2015.Participants who sustained joint dislocations of the trunk,arms,or legs(skull,sternum,and ribs being excluded)in 2014 were personally interviewed to obtain data on age,educational background,ethnic origin,occupation,geographic region,and urbanization degree.The joint-dislocation incidence was calculated based on age,sex,body site,and demographic factors.The risk factors for different groups were examined using multiple logistic regression.Results:One hundred and nineteen participants sustained 121 joint dislocations in 2014.The population-weighted incidence rate of joint dislocations of the trunk,arms,or legs was 0.22(95%confidence interval[CI]:0.16,0.27)per 1000 population in 2014(men,0.27[0.20,0.34];women,0.16[0.10,0.23]).For all ages,previous dislocation history(male:OR 42.33,95%confidence interval[CI]:12.03-148.90;female:OR 54.43,95%CI:17.37-170.50)and alcohol consumption(male:OR 3.50,95%CI:1.49-8.22;female:OR 2.65,95%CI:1.08-6.50)were risk factors for joint dislocation.Sleeping less than 7 h/day was a risk factor for men.Compared with children,women aged≥15 years(female 15-64 years:OR 0.16,95%CI:0.04-0.61;female≥65 years:OR 0.06,95%CI:0.01-0.41)were less likely to sustain joint dislocations.Women with more than three children were at higher dislocation risk than women without children(OR 6.92,95%CI:1.18-40.78).Conclusions:The up-to-date data on joint dislocation incidence,distribution,and risk factors can be used as a reference for national healthcare,prevention,and management in China.Specific strategies for decreasing alcohol consumption and encouraging adequate sleeping hours should be developed to prevent or reduce dislocation incidents.Trial Registration:Chinese Clinical Trial Registry,ChiCTR-EPR-15005878. | Hongzhi Lv Wei Chen Zhiyong Hou Siming Jia Yanbin Zhu Bo Liu Xiao Chen Guang Yang Lei Liu Tao Zhang Haili Wang Bing Yin Song Liu Jialiang Guo Xiaolin Zhang Yichong Li Yingze Zhang | 2022 | Chinese Medical Journal2022,,14: | 0 |

| 12 | Interaction of AcMADS68 with transcription factors regulates anthocyanin biosynthesis in red-fleshed kiwifruit显示文摘In red-fleshed kiwifruit,anthocyanin pigmentation is a crucial commercial trait.The MYB-bHLH-WD40(MBW)complex and other transcription factors regulate its accumulation.Herein,a new SEP gene,AcMADS68,was identified as a regulatory candidate for anthocyanin biosynthesis in the kiwifruit by transcriptome data and bioinformatic analyses.AcMADS68 alone could not induce the accumulation of anthocyanin both in Actinidia arguta fruit and tobacco leaves.However,in combination with AcMYBF110,AcMYB123,and AcbHLH1,AcMADS68 co-overexpression increased anthocyanin biosynthesis,whereas its silencing reduced anthocyanin accumulation.The results of the dual-luciferase reporter,firefly luciferase complementation,yeast two-hybrid and co-immunoprecipitation assays showed that AcMADS68 could interact with both AcMYBF110 and AcMYB123 but not with AcbHLH1,thereby co-regulating anthocyanin biosynthesis by promoting the activation of the target genes,including AcANS,AcF3GT1,and AcGST1.Moreover,AcMADS68 also could activate the promoter of AcbHLH1 surported by dual-luciferase reporter and yeast one-hybrid assays,thereby further amplifying the regulation signals from the MBW complex,thus resulting in enhanced anthocyanin accumulation in the kiwifruit.These findings may facilitate better elucidation of various regulatory mechanisms underlying anthocyanin accumulation and contribute to the quality enhancement of red-fleshed kiwifruit. | Yanfei Liu Guowen Lv Yaqi Yang Kangxun Ma Xiaolin Ren Mingjun Li Zhande Liu | 2023 | Horticulture Research2023,10,2: | 0 |

| 15 | Immunogenicity and safety of a booster COVID-19 vaccination in patients with chronic liver disease:A multicenter study显示文摘Aim:Patients with chronic liver disease(CLD),especially cirrhosis,are at a high risk of severe illness or death from coronavirus disease-2019(COVID-19)and may have a suboptimal immune response to the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccine.This study aimed to evaluate the safety and immunogenicity of the COVID-19 booster vaccination in patients with CLD.Methods:The study protocol was prospectively registered at ClinicalTrials.gov(No.NCT05204602)after approval by the Ethics Committee.Adult participants with CLD were enrolled in this multicenter prospective study.They completed two doses of the inactivated COVID-19 vaccine and received booster doses at least 6 months later.Adverse reactions were recorded within 14 days after the booster dose.Serum samples of the enrolled patients were collected before and after booster vaccination and tested for SARS-CoV-2 receptor-binding domain(RBD)immunoglobulin G and neutralizing anti-bodies.The chi-squared or Fisher's exact test was used to compare categorical data,and the Mann-Whitney U test was used to compare continuous variables.Two-sided p<0.05 were considered statistically significant.Results:In total,63 patients were enrolled from four hospitals in China,including 29 patients with cirrhosis.The median age of all patients was 55 years,and 61.9%(39/63)were male.The vaccines were well tolerated;most adverse reactions were mild and transient,and injection site pain(6.4%;4/63)and fatigue(3.2%,2/63)were the most frequent local and systemic adverse events.Following the booster vaccination,our results showed that in the whole cohort,the levels and positive rates of anti-RBD IgG and neutralizing antibodies were significantly higher than baseline levels(all p<0.05).Conclusions:The inactivated COVID-19 booster vaccine was safe and significantly increased antibody levels and positivity rates following standard vaccination regimens in patients with CLD,especially those with cirrhosis. | Jitao Wang Jingwen Ai Huiling Xiang Yanliang Zhang Zhiyun Hou Qiran Zhang Jiaojian Lv Shubo Chen Chuan Liu Qianqian Li Jing Liang Faren Xie Shujun Jiang Nina Zhang Aiguo Zhang Xiaolin Lan Xuying Zhang Jinlong Li Dengxiang Liu Wenchuan Wang Wei Rao Zhang Qun Qiuju Tian Xiaolong Qi Wenhong Zhang | 2023 | Portal Hypertension & Cirrhosis2023,2,3: | 0 |
