1A critical epitope in CD147 facilitates memory CD4^(+) T-cell hyper-activation in rheumatoid arthritis显示文摘The abnormal activation of CD4^(+)CD45RO+memory T(Tm)cells plays an important role in the pathogenesis of rheumatoid arthritis(RA).Previous studies have shown that CD147 participates in T-cell activation.However,it remains unclear whether CD147 is involved in abnormal Tm-cell activation in RA patients.In this study,we demonstrated that CD147 was predominantly upregulated in Tm cells derived from RA patients.The anti-CD147 mAb 5A12 specifically inhibited Tm-cell activation and proliferation and further restrained osteoclastogenesis.Using a structural–functional approach,we depicted the interface between 5A12 and CD147.This allowed us to identify two critical residues,Lys63 and Asp65,as potential targets for RA treatment,as the double mutation K63A/D65A inhibited Tm-cell activation,mimicking the neutralization by 5A12.This study provides not only a theoretical basis for a“CD147-Tm/Osteoclast-RA chain”for the potential prevention and treatment of RA or other T-cell-mediated autoimmune diseases but also a new target for related drug design and development.Na Guo Sheng Ye Kui Zhang Xiaoling Yu Hongyong Cui Xiangmin Yang Peng lin Minghua Lv Jinlin Miao Yang Zhang Qing Han Rongguang Zhang Zhinan Chen Ping Zhu 2019Cellular & Molecular Immunology2019,16,6:6
2Molecular characteristics and evolutionary analysis of a very virulent infectious bursal disease virus显示文摘Infectious bursal disease virus(IBDV) poses a significant threat to the poultry industry. Viral protein 2(VP2), the major structural protein of IBDV, has been subjected to frequent mutations that have imparted tremendous genetic diversity to the virus. To determine how amino acid mutations may affect the virulence of IBDV, we built a structural model of VP2 of a very virulent strain of IBDV identified in China, vv IBDV Gx, and performed a molecular dynamics simulation of the interaction between virulence sites. The study showed that the amino acid substitutions that distinguish vv IBDV from attenuated IBDV(H253Q and T284A) favor a hydrophobic and flexible conformation of ?-barrel loops in VP2, which could promote interactions between the virus and potential IBDV-specific receptors. Population sequence analysis revealed that the IBDV strains prevalent in East Asia show a significant signal of positive selection at virulence sites 253 and 284. In addition, a signal of co-evolution between sites 253 and 284 was identified. 3PIP5k1β controls bone homeostasis through modulating both osteoclast and osteoblast differentiation显示文摘PIP5k1βis crucial to the generation of phosphotidylinosotol(4,5)P2.PIP5k1βparticipates in numerous cellular activities,such as B cell and platelet activation,cell phagocytosis and endocytosis,cell apoptosis,and cytoskeletal organization.In the present work,we aimed to examine the function of PIP5k1βin osteoclastogenesis and osteogenesis to provide promising strategies for osteoporosis prevention and treatment.We discovered that PIP5k1β deletion in mice resulted in obvious bone loss and that PIP5k1β was highly expressed during both osteoclast and osteoblast differentiation.Deletion of the gene was found to enhance the proliferation and migration of bone marrow-derived macrophage-like cells to promote osteoclast differentiation.PIP5k1β-/-osteoclasts exhibited normal cytoskeleton architecture but stronger resorption activity.PIP5kip deficiency also promoted activation of mitogen-activated kinase and Akt signaling,enhanced TRAF6 and c-Fos expression,facilitated the expression and nuclear translocation of NFATC1,and upregulated Grb2 expression,thereby accelerating osteoclast differentiation and function.Finally,PIP5k1β enhanced osteoblast differentiation by upregulating master gene expression through triggering smad1/5/8 signaling.Therefore,PIP5k1βmodulates bone homeostasis and remodeling.Xiaoying Zhao Penglei Cui Guoli Hu Chuandong Wang Lei Jiang Jingyu Zhao Jiake Xu Xiaoling Zhang 2020Journal of Molecular Cell Biology2020,12,1:4
4Cholecystectomy is associated with higher risk of recurrence after microwave ablation of hepatocellular carcinoma:a propensity score matching analysis显示文摘Objective:To explore the association between cholecystectomy and the prognostic outcomes of patients with hepatocellular carcinoma(H CC)who underwent microwave ablation(MWA).Methods:Patients with HCC(«=921)who underwent MWA were included and divided into cholecystectomy(n=114)and non-cholecystectomy groups(n=807).After propensity score matching(PSM)at a 1:2 ratio,overall survival(OS)and disease-free survival(DFS)rates were analyzed to compare prognostic outcomes between the cholecystectomy(«=114)and non-cholecystectomy groups(n=228).Univariate and multivariate Cox analyses were performed to assess potential risk factors for OS and DFS.Major complications were also compared between the groups.Results:After matching,no significant differences between groups were observed in baseline characteristics.The 1-,3-,and 5-year OS rates were 96.5%,82.1%,and 67.1%in the cholecystectomy group,and 97.4%,85.2%,and 74.4%in the non-cholecystectomy group(P=0.396);the 1-,3-,and 5-year DFS rates were 58.4%,34.5%,and 26.6%in the cholecystectomy group,and 73.6%,44.7%,and 32.2%in the non-cholecystectomy group(P=0.026),respectively.The intrahepatic distant recurrence rate in the cholecystectomy group was significantly higher than that in the non-cholecystectomy group(P=0.026),and the local tumor recurrence and extrahepatic recurrence rates did not significantly differ between the groups(P=0.609 and P=0.879).Multivariate analysis revealed that cholecystectomy(HR=1.364,95%Cl 1.023-1.819,P=0.035),number of tumors(2 vs.1:HR=2.744,95%Cl 1.925-3.912,P<0.001;3 vs.1:HR=3.411,95%Cl 2.021-5.759,P<0.001),and y-GT levels(HR=1.003,95%Cl 1.000-1.006,P<0.024)were independent risk factors for DFS.The best y-GT level cut-off value for predicting median DFS was 39.6 U/L(area under the curve=0.600,P<0.05).A positive correlation was observed between cholecystectomy and y-GT level(r=0.108,95%Cl-0.001-0.214,P=0.047).Subgroup analysis showed that the DFS rates were significantly higher in the non-cholecystectomy group than the cholecystectomy group when Y-GT>39.6 U/L(i3=0.044).The 5-,10-,15-,20-,and 25-year recurrence rates from the time of cholecystectomy were 2.63%,21.93%,42.11%,58.77%,and 65.79%,respectively.A significant positive correlation was observed between cholecystectomy and the time from cholecystectomy to recurrence(r=0.205,95%Cl 0.016-0.379,P=0.029).There were no significant differences in complications between groups(P=0.685).Conclusions:Patients with HCC who underwent cholecystectomy were more likely to develop intrahepatic distant recurrence after MWA,an outcome probably associated with increased y-GT levels.Moreover,the recurrence rates increased with time.Hongcai Yang Yi Yang Jianping Dou Rui Cui Zhigang Cheng Zhiyu Han Fangyi Liu Xiaoling Yu Xiang Zhou Jie Yu Ping Liang 2020Cancer Biology & Medicine2020,17,2:3
5Pairing superfluidity in spin-orbit coupled ultracold Fermi gases显示文摘We review some recent progresses on the study of ultracold Fermi gases with synthetic spin-orbit coupling.In particular,we focus on the pairing superfluidity in these systems at zero temperature.Recent studies have shown that different forms of spin-orbit coupling in various spatial dimensions can lead to a wealth of novel pairing superfluidity.A common theme of these variations is the emergence of new pairing mechanisms which are direct results of spin-orbit-coupling-modified single-particle dispersion spectra.As different configurations can give rise to single-particle dispersion spectra with drastic differences in symmetry,spin dependence and low-energy density of states,spin-orbit coupling is potentially a powerful tool of quantum control,which,when combined with other available control schemes in ultracold atomic gases,will enable us to engineer novel states of matter.YI Wei ZHANG Wei CUI XiaoLing 2015Science China(Physics,Mechanics & Astronomy)2015,58,1:3
6A single mutation in the PBC loop of VP2 is involved in the in vitro replication of infectious bursal disease virus显示文摘To test whether amino acid mutations in the PBC and PHI loops of VP2 are involved in the replication and virulence of infectious bursal disease virus(IBDV), a pair of viruses, namely the moderately virulent IBDV(rG x-F9VP2) and the attenuated strain(rGt), were used. Residue mutations A222P(P_(BC)) and S330R(PHI), selected by sequence comparison, were introduced individually into r Gx-F9VP2 by using a reverse genetics system. In addition, the reverse mutation of either P222 A or R330 S was introduced into r Gt. The four modified viruses were then rescued and evaluated in vitro(CEF cells) and in vivo(SPF chickens). Results showed that A222 P elevated the replication efficiency of rG x-F9VP2 while P222 A reduced that of rG t in CEF cells. A mutation at residue 330 did not alter IBDV replication. In addition, animal experiments showed that a single mutation at either residue 222 or 330 did not significantly influence the virulence of IBDV. 7Nanoengineered hydrogels as 3D biomimetic extracellular matrix with injectable and sustained delivery capability for cartilage regeneration显示文摘The regeneration of articular cartilage remains a great challenge due to the difficulty in effectively enhancing spontaneous healing.Recently,the combination of implanted stem cells,suitable biomaterials and bioactive molecules has attracted attention for tissue regeneration.In this study,a novel injectable nanocomposite was rationally designed as a sustained release platform for enhanced cartilage regeneration through integration of a chitosan-based hydrogel,articular cartilage stem cells(ACSCs)and mesoporous SiO_(2)nanoparticles loaded with anhydroicaritin(AHI).The biocompatible engineered nanocomposite acting as a novel 3D biomimetic extracellular matrix exhibited a remarkable sustained release effect due to the synergistic regulation of the organic hydrogel framework and mesopore channels of inorganic mSiO_(2)nanoparticles(mSiO_(2)NPs).Histological assessment and biomechanical tests showed that the nanocomposites exhibited superior performance in inducing ACSCs proliferation and differentiation in vitro and promoting extracellular matrix(ECM)production and cartilage regeneration in vivo.Such a novel multifunctional biocompatible platform was demonstrated to significantly enhance cartilage regeneration based on the sustained release of AHI,an efficient bioactive natural small molecule for ACSCs chondrogenesis,within the hybrid matrix of hydrogel and mSiO_(2)NPs.Hence,the injectable nanocomposite holds great promise for use as a 3D biomimetic extracellular matrix for tissue regeneration in clinical diagnostics.Penglei Cui Panpan Pan Ling Qin Xinluan Wang Xiaodong Chen Yonghui Deng Xiaoling Zhang 2023Bioactive Materials2023,,1:2
8Improving DFA attacks on AES with unknown and random faults显示文摘Differential fault analysis(DFA) aiming at the advanced encryption standard(AES) hardware implementations has become a widely research topic. Unlike theoretical model, in real attack scenarios, popular and practical fault injection methods like supply voltage variation will introduce faults with random locations,unknown values and multibyte. For analyzing this kind of faults, the previous fault model needed six pairs of correct and faulty ciphertexts to recover the secret round-key. In this paper, on the premise of accuracy, a more efficient DFA attack with unknown and random faults is proposed. We introduce the concept of theoretical candidate number in the fault analysis. Based on this concept, the correct round-key can be identified in advance, so the proposed attack method can always use the least pairs of correct and faulty ciphertexts to accomplish the DFA attacks. To further support our opinion, random fault attacks based on voltage violation were taken on an FPGA board. Experiment results showed that about 97.3% of the attacks can be completed within 3 pairs of correct and faulty ciphertexts. Moreover, on average only 2.17 pairs of correct and faulty ciphertexts were needed to find out the correct round-key, showing significant advantage of efficiency compared with previous fault models. 9Macroporous-mesoporous C-,S-,N-doped titania microspheres via the polyHIPE microspheres templates显示文摘Doping and increasing specific surface area by forming highly porous structures are two effective ways to enhance the photocatalytic performances of TiO_(2) particles.He re for the first time,we report a new facile method to prepare the macroporous-mesoporous C-,S-,N-doped TiO_(2)(C/S/N-TiO_(2))microspheres via polyHIPE microsphe res as templates.The chemical and crystalline structure s of these hierarchical porous TiO_(2) microspheres are analyzed with FTIR,XPS,EDS,and XRD.The macroporous-mesoporous structures are confirmed with SEM observation and BET analysis.UV-vis DRS spectra analysis shows that the band gaps of C doped TiO_(2),C/N doped TiO_(2),C/S doped TiO_(2) and C/S/N doped TiO_(2) are estimated to be 3.07,3.01,2.94 and 2.81 eV,respectively,which are significantly narrower than that of TiO_(2) nanoparticles(3.23 eV).Photoluminescence spectra demonstrate that the recombination of electrons and holes in these macroporous-mesoporous TiO_(2) microspheres is also suppressed.The hierarchical porous C/S/N-TiO_(2) microspheres show high visible-light catalytic efficiency and excellent cycling stability to degrade RhB dye.Xiaoling Cui Huarong Liu Xingyuan Zhang Hewen Liu 2021Chinese Chemical Letters2021,32,3:1

11Microalgal Cultivation in Secondary Effluents:Enhancement of Algal Biomass,Nutrient Removal,and Lipid Productivity显示文摘The growth performance,nutrient removal,lipid accumulation and morphological changes of Cyanobacterium aponinum OUC1 and Scenedesmus obliquus which were cultured in secondary effluents from two wastewater treatment plants:Tuandao Wastewater Treatment Plant(ETD)and Licun River Wastewater Treatment Plant(ELR)were investigated.The results showed that both C.aponinum OUC1 and S.obliquus have superior growth performances in both undiluted effluents,while the better of them was that in ETD effluent,with cell densities of C.aponinum OUC1 and S.obliquus increased by 159%and 66%over that of BG11(control),respectively.Regarding nutrient removal,S.obliquus could completely remove inorganic phosphorus,and decrease ammonia nitrogen in ETD effluent by 81%.In addition,both C.aponinum OUC1 and S.obliquus cultivated in ETD exhibited extraordinary potential for biofuel production,increasing lipid productivities by 133%and 89%of that cultivated in ELR,respectively.As to ultrastructural changes,the differences in the lipoidal globules and glycogen granules of S.obliquus and C.aponinum OUC1 among the ETD and ELR treatments were mostly related to phosphorus limitations.The findings from this research reveal the probability using the secondary effluents as cultivation media to enhance algal biomass,nutrient removal and lipid productivity.ZHANG Bo MENG Fanping CUI Hongwu DOU Xiang DU Shuhao PENG Xiaoling 2020Journal of Ocean University of China2020,19,6:1

13Biological indicators of sub-optimal health status显示文摘OBJECTIVE:To investigate biological indicators of sub-optimal health status and provide means of objective assessment of sub-optimal health status.METHODS:We set the unified standards for diagnosing a SHS.We tested various laboratory indicators in 407 cases that we selected randomly from2807 subjects and collected 15 mL of fasting venous blood from each case.We measured serum immunoglobulin A(IgA)and immunoglobulin G(IgG)concentrations,serum beta endorphins(β-EP),cortisol(C),testosterone(T),plasma adrenocorticotropic hormone(ACTH)and serum T lymphocyte subsets CD3+and CD4+.RESULTS:Mean serum testosterone concentrations and their ratio to cortisol(C)concentrations weresignificantly higher in the healthy group than in those with sub-optimal health status(P<0.01).Mean serum CD3+concentrations were significantly higher in those with sub-optimal health status than in the healthy group(P<0.05).CONCLUSION:Decreased serum testosterone/cortisol ratio may be an objective indication of sub-optimal health status.Changes in neuroendocrine and immunological indicators may explain some of the symptoms,including malaise and poor work performance,attributable to persistent or relapsing fatigue in subjects with sub-optimal health status.Limin Wang Xin Zhao Jiaxu Chen Xiaoling Guo Xin Liang Danhui Yi Huating Cui Yueyun Liu 2013Journal of Traditional Chinese Medicine2013,33,5:1

17Efficient evaluation model including interconnect resistance effect for large scale RRAM crossbar array matrix computing显示文摘Crossbar architecture has been considered as an efficient means to execute a matrix-vector multiplication computation. An efficient evaluation model for this computation including the interconnect resistance effect on the high density resistive random access memmory(RRAM) crossbar array is proposed in this paper. The proposed model considers the interconnect resistance impacts on the columns and rows separately. The simulation results indicate that the computing speed of the proposed model can be boosted by over three orders of magnitude with the computation deviation of 7.7% in comparison with the precise comprehensive model in the 64 kb crossbar array fabricated at the 14 nm technology node. 19Mechanism of high-concentration electrolyte inhibiting the destructive effect of Mn(Ⅱ)on the performance of lithium-ion batteries显示文摘By optimizing electrolyte formulation to inhibit the deposition of transition metal ions(TMIs) on the surface of the graphite anode is an effective way to improve the electrochemical performance of lithium-ion batteries.At present,it is generally believed the formation of an effective interfacial film on the surface of the anode electrode is the leading factor in reducing the dissolution of TMIs and prevent TMIs from being embedded in the electrode.It ignores the influence of the solvation structures in the electrolyte system with different composition,and is not conducive to the design of the electrolyte formulation from the perspective of changing the concentration and the preferred solvent to inhibit the degradation of battery performance caused by TMIs deposition.In this work,by analyzing the special solvation structures of the high-concentra tion electrolyte,we study the main reason why high-concentration electrolyte inhibits the destructive effect of Mn(Ⅱ) on the electrochemical performance of LIBs.By combining the potentialresolved in-situ electrochemical impedance spectroscopy technology(PRIs-EIS) and density functional theory(DFT) calculation,we find that Mn(Ⅱ) mainly exists in the form of contact ions pairs(CIPs) and aggregates(AGGs) in high-concentration electrolyte.These solvation structures can reduce the destructive effect of Mn(Ⅱ) on battery performance from two aspects:on the one hand,it can rise the lowest unoccupied orbital(LUMO) value of the solvation structures of Mn(Ⅱ),thereby reducing the chance of its reduction;on the other hand,the decrease of Mn2+ions reduction can reduce the deposition of metallic manganese in the solid electrolyte interphase(SEI),thereby avoiding the continuous growth of the SEI.This study can be provided inspiration for the design of electrolytes to inhibit the destructive effect of TMls on LIBs.Xiaoling Cui Jinlong Sun Dongni Zhao Jingjing Zhang Jie Wang Hong Dong Peng Wang Junwei Zhang Shumin Wu Linhu Song Ningshuang Zhang Chunlei Li Shiyou Li 2023Journal of Energy Chemistry2023,,3:1
